Introduction of VP6 Gene into Potato Plant by Agrobacterium-mediated Transformation and Analysis of VP6 Expression in Transgenic Potatoes (Rotavirus VP6 유전자의 감자식물체내로의 도입과 형질전환체의 발현분석)

  • A VP6 fragments was subcloned with BamHI in the binary pMBP-1 vector under Califlower Mosaic Virus (CaMV) 355 promoter and neomycin phosphotransferase II (npt II) gene. The recombinant binary vector was mobilized into Agrobacterium-tumefaciens LBA4404 by the freeze-thaw method and potato (Solanum tubensum L. cv Desiree) was transformed by modified leaf-disc cocultivation. Shoots were induced on MS medium with 0.01 mg/L NAA, 0.1 mg/L GA$_3$, 2.0 mg/L Zeatin, 100.0 mg/L kanamycin, 500.0 mg/L carbenicillin. In order to identify the copy number of VP6 into potato plant, total genomic DNA was isolated from transgenic potato and analysed by Southern blotting. Genomic DNA and total mRNA analysis demonstrated the incorporation of the foreign gene into the potato genome, as well as their transcription.

Image Evaluation and Exposure Dose with the Application of Tube Voltage and Adaptive Statistical Iterative Reconstruction of Low Dose Computed Tomography (저 선량 전산화단층촬영의 관전압과 적응식 통계적 반복 재구성법 적용에 따른 영상평가 및 피폭선량)

  • The study has attempted to evaluate and compare the image evaluation and exposure dose by respectively applying filter back projection (FBP), the existing test method, and adaptive statistical iterative reconstruction (ASIR) with different values of tube voltage during the low dose computed tomography (LDCT). With the image reconstruction method as basis, chest phantom was utilized with the FBP and ASIR set at 10%, 20% respectively, and the change of tube voltage (100 kVp, 120 kVp). For image evaluation, back ground noise, signal-noise ratio (SNR) and contrast-noise ratio (CNR) were measured, and, for dose assessment, CTDIvol and DLP were measured respectively. In terms of image evaluation, there was significant difference in ascending aorta (AA) SNR and inpraspinatus muscle (IM) SNR with the different amount of tube voltage (p < 0.05). In terms of CTDIvol, the measured values with the same tube voltage of 120 kVp were 2.6 mGy with no-ASIR and 2.17 mGy with 20%-ASIR respectively, decreased by 0.43 mGy, and the values with 100 kVp were 1.61 mGy with no-ASIR and 1.34 mGy with 20%-ASIR, decreased by 0.27 mGy. In terms of DLP, the measured values with 120 kVp were $103.21mGy{\cdot}cm$ with no-ASIR and $85.94mGy{\cdot}cm$ with 20%-ASIR, decreased by $17.27mGy{\cdot}cm$ (about 16.7%), and the values with 100 kVp were $63.84mGy{\cdot}cm$ with no-ASIR and $53.25mGy{\cdot}cm$ with 20%-ASIR, a decrease by $10.62mGy{\cdot}cm$ (about 16.7%). At lower tube voltage, the rate of dose significantly decreased, but the negative effects on image evaluation was shown due to the increase of noise.

cDNA Cloning and Expression of Human Rotavirus Outer Capsid Protein VP7 in Insect Cells

  • Rotavirus is a major cause of severe gastroenteritis in young children and animals throughout the world. The VP7 of rotavirus is thought to induce the synthesis of neutralizing antibodies and to be responsible for determining viral serotypes. The cDNA coding for the VP7 capsid protein of human rotavirus, obtained from Korean patients (HRV-Y14), was cloned and its nucleotide sequence was determined. Comparative analysis of the nucleotide sequences between VP7 of Y14 and that of other foreign isolates showed $92.7~95.2\%$ homology to G1 serotypes (RV-4, KU, K8, WA), $74.2\%$ homolgy to G2 serotype HU-5, $76.4\%$ homology to G3 serotype SA-11, and $77.6\%$ homology to G4 serotype A01321. These data suggest that HRV-Y14 can be classified as a G1 serotype. cDNA coding for VP7 of HRV-YI4 was subcloned into the baculovirus vector and the VP7 glycoprotein was expressed in insect cells. The expressed proteins in Sf9 cell extract and tissue culture fluid were separated on SDS-PAGE, and Western blot analysis with monoclonal antibody raised against the synthetic peptide containing 21 amino acids within the VP7 conserved region was performed. The molecular weight of recombinant VP7 was estimated to be 36 kDa which is about the same size as the native VP7. Addition of tunicamycin in the culture media caused a reduction of the molecular weight of the recombinant VP7 indicating that the expressed protein was glycosylated.

  • Electronic personal dosimeter (EPD) provide real time monitoring and a direct indication of the accumulated dose or dose rate in terms of personal dose. Most EPD do not perform well in low energy photon radiation fields present in medical radiation environments. It has poor responsibility and large error rate for low energy photon radiation of medical radiation environments. This study evaluated to optimal additional filtration for EPD using silicon PIN photodiode detector form 40 to 120 kVp range in medical radiation environments. From 40 to 80 kVp energy range, Al 0.2 mm and Sn 1.0 mm overlapped filtration showed good responsibility to dose rate and from 80 kVp to 120 kVp energy range, Al 0.2 mm and Sn 1.6 mm overlapped filtration showed good responsibility to dose rate.

The Evaluation of the Radiation Dose and Image Quality Through the Change of the Tube Voltage in Cerebral CT Angiography (전산화단층촬영장치를 이용한 뇌 혈관조영 검사에서 관전압 변화에 따른 방사선량과 영상의 질 평가)

  • To image diagnosis in neurovascular diseases using Multi-Detector Computed Tomography(MDCT), injected the same contrast material when inspecting Brain Computed Tomography Angiography(BCTA) to examine radiation dose and Image quality on changing Cerebral Artery CT number by tube voltage. Executed an examination with same condition[Beam Collimation $128{\times}0.6mm$, Pitch 0.6, Rotation Time 0.5s, Slice Thickness 5.0mm, Increment 5.0mm, Delay Time 3.0sec, Care Dose 4D(Demension ; D)] except for tube voltage on 50 call patients for BCTA and divided them into two groups (25 people for a group, group A: 80, group B: 120kVp). From all the acquired images, set a ROI(Region of Interest) on four spots such as left cerebral artery, right cerebral artery, posterior cerebral artery and cerebral parenchyma to compare quantitative evaluation, qualitative evaluation and effective dose after measuring CT number value from Picture Archiving Communications System(PACS). Evaluating images with CT number acquired from BCTA examination, images with 80 kVp was 18% higher in Signal to Noise Ratio and 19% in Contrast to Noise Ratio than those with 120 kVp. It was seen that expose dose was decreased by over 50% with tube voltage 80 kVp than with 120 kVp. Group A (25 patients) was examination with tube voltage 80kVp while group B with 120 kVp to examine radiation dose and Image quality. It is considered effective to inspect with lower tube voltage than with conventional high kVp, which can reduce radiation dose without any affect on diagnosis.

An Assessment of the Radiation Dose from Radiography with the Change in Air Gap (공극(기극(氣隙)) 변화에 따른 방사선촬영 선량평가)

  • This study aims to propose a method for reducing radiation dose in high-voltage radiography using air gap technique while maintaining the same image quality as when using grids. For an experiment, air gaps were set at 10 cm, 15 cm, 20 cm, 25 cm, and 30 cm with a focus-receptor distance of 180 cm; with each air gap distance, tube current was set at 15 mAs, and tube voltage was set at 80 kVp, 85 kVp, 90 kVp, 95 kVp and 100 kVp. Then, radiographs were taken. In a situation of employing a conventional method of using grids, radiographs were taken at 15 mAs and 107 kVp with a focus-receptor distance of 180 cm. According to the results of the experiment, the surface radiation dose from radiography using grids was 0.130 R; the surface radiation dose at a 20cm air gap was 0.124 R; PSNR between these two images was 10.65 [dB]. In conclusion, the air gap distance, which could maintain the image quality similar to that of a case where scattered radiation was removed and grids were used with a small surface radiation dose, was 20 cm. The result of this study is thought to be used as an indicator to remove surface radiation dose in radiography using air gap.

Dose and Image Assessment according to Radiologic Factors Variation at Digital Humerus X-ray Examination (디지털 환경에서 Humerus 검사 시 촬영인자 변화에 따른 선량 및 화질 평가)

  • Purpose : We aim at presenting the optimum radiologic factor through the evaluation of dose variation and of image quality through the use of a grid in Humerus examination and the change of dose because of the change of radiologic factor. Materials and Methods : We divided it in 3 cases: when using a grid or not and when using IP(Image Plate) in a digital system. Also, as fixing kVp to 70kVp it changed mAs, and fixing mAs to 10 it changed kVp, we put up resolution chart and Burger rose phantom on the acrylic phantom of 7cm (the same level of Humerus) to evaluate the dose and image. We used Image J program to evaluate the quantitative resolution of the obtained image, and made the qualitative evaluation and statistical analysis of the image saved in PACS for 20 radiologic technologist with more than 10 years of experience in order of evaluate its contrast. We used SPSS10(SPSS Inc. Chicago, Illinois) for statistical analysis. Results : We observed the analytic result of resolution by the change of kVp that it was $4.539dGycm^2$ in 60kVp and $757.472dGycm^2$ in 75kVp, which increased about 64.6% of dose, while for the resolution it had the pixel value 30.7% better with 851 in 60kVp than 651 in 75kVp. Also, we analyzed the result of resolution by the change of mAs that it was $3.106dGycm^2$ in 5mAs, and $12.470dGycm^2$ in 20mAs, which increased about 400% of dose, while for the resolution DR had 678 in 5mAs, and 724 in 20mAs that increased about 6.8% of resolution. We made the qualitative evaluation of contrast by the change of kVp that DR showed the higher quality than CR, but the contrast by the change of kVp had no special different at the moment of visual evaluation, nor statistically significant difference(P>0.05). We observed the qualitative evaluation of contraste by the change of mAs that the contrast increased as DR increased mAs, and had statistically significant difference(P<0.05). On the other hand, CR had no significant difference for more than 10mAs nor statistically significant difference(P>0.05). Conclusion : In case of some patients with radiographic exposure by the repeated examination such as emergent patient or Follow up patient, they are considered to try to limit the use of a grid, to set kVp under 65kVp in fixed mode, to select less than 10mAs and to reduce the possibility of patient being bombed.

Cloning and nucleotide sequence analysis of VP7 genes of bovine rotaviruses isolated in Korea (국내에서 분리된 소 로타바이러스의 VP7 유전자 크로닝 및 염기서열 분석)

  • Bovine rotaviruses(A, 288, 55086 strains) isolated from fecal samples in Korea were propagated onto MA104 cells and were confirmed tentatively as G6, G8, and G10, respectively, by RFLP analysis. Full-length VP7 gene of these isolates was amplified by reverse transcriptase polymerase chain reaction(RT-PCR) using VP7 specific primers and cloned into TA vector. Nucleotide and deduced amino acid sequences of VP7 genes of the isolates were determined and compared with those of bovine rotavirus reference strains(NCDV; G6, UK; G6, Cody I-801; G8 and B223; G10). A, 288 and 55086 isolates showed high degree of nucleotide sequence homology with NCDV and UK(93% and 94%), Cody I-801(86%) and B223(97%), respectively, However, they showed 71~74% of nucleotide sequence homlogy with bovine rotavirus reference strains which belong to different serotypes. From the results of deduced amino acid sequence homology analysis, three isolates showed 94~96% of homology with the same serotype reference strains but 80~84% of homology with the different serotype reference strains. Three bovine rotavirus isolates, A, 288 and 55086 strains, were confirmed as G6, G8, and G10, respectively, by nucleotide and deduced amino acid sequence analysis.

Clinical Outcomes after Percutaneous Vertebroplasty for Pathologic Compression Fractures in Osteolytic Metastatic Spinal Disease

  • Objective : Percutaneous vertebroplasty (VP) can provide immediate stabilization in pathologic fractures of spinal tumors. However, long term follow-up data in cases of pathologic fractures are lacking. The authors report follow-up results of VP in 185 pathologic fractures of 102 spinal tumor patients. Methods : Percutaneous VP was performed at 185 vertebral bodies of 102 patients from 2001 to 2007. Retrospective analysis was done with medical records and radiological data. The change of visual analogue score (VAS), vertebral body (VB) height and kyphotic angle were measured preoperatively and on postoperative one day and at 3, 6, and 12 months. Results : The patients were composed of metastatic spine tumors (81%) and multiple myeloma (19%). Involved spinal segments were between T6 and L5. Mean follow-up period was 12.2 months. VAS for back pain was 8.24 preoperatively, 3.59 (postoperative one day), 4.08 (three months) and 5.22 (one year). VB compression ratio changed from 21.33% preoperatively to 13.82% (postoperative one day), 14.36% (three month), and 16.04% (one year). Kyphotic angle changed from $15.35^{\circ}$ preoperatively to $12.03^{\circ}$ (postoperative one day), $13.64^{\circ}$ (three month), and $15.61^{\circ}$ (one year). Conclusion : Immediate pain relief was definite after VP in pathologic compression fracture of osteolytic spinal disease. Although VAS was slightly increased on one year follow-up, VP effect was maintained without significant change. These results indicate that VP could be a safe and effective procedure as a palliative treatment of the spinal tumor patients.

Quality and Shelf-life of Chilled Smoked Pork Belly Depending on Packaging Methods (포장방법에 따른 냉장 훈제삼겹살의 품질과 저장성)

  • The changes in quality and shelf life of smoked pork belly according to packaging methods were investigated during storage at $5{\pm}1^{\circ}C$ up to 12 days. Three packaging treatments including air-containing packaging (AC), vacuum packaging (VP), and packaging incorporated with oxygen scavenger (OS) were applied in this experiment. In all treatments, the initial total aerobic plate count (TPC) was 2.4 log cfu/g which was increased with storage period. The rapidest increase of TPC was observed in the AC samples, followed by OS and VP. The VP samples showed inhibiting effects on the growth of Pseudomonas spp. and coliforms over the storage. The TBA values were increased in the order of AC-OS-VP. The VBN values of the VP and OS samples tended to be increased slower than the AC samples during the storage. According to the sensory evaluations, the point of losing marketability in the raw AC, OS, and VP samples was determined to be the day 8, 10, and 12, respectively. However, the evaluations for the cooked samples showed the AC and OS samples preserved the marketing value until the day 10 and 12, while the VP samples until the day 12 except in the parameter of texture. In conclusion, it was found that the most appropriate packaging method for preserving the quality and extending the shelf life of chilled smoked pork belly is vacuum packaging.
