  • Purpose: The aim of this study is to investigate the changes in hardness and microstructure of a dental multipurpose alloy after simulated complete firing with controlled cooling rate and holding time by characterizing the changes in hardness and microstructure after simulated firing with various cooling rates and holding times. Methods: Before hardness testing, the specimens were solution treated and then were rapidly quenched into ice brine. The specimens were completely fired in furnace. Hardness measurements were made using a Vickers microhardness tester. The specimens were examined at 15 kV using a field emission scanning electron microscope. Results: The maximum hardness value was obtained at stage 0 after simulated firing with various cooling rates (quick cooling, stage 0, stage 1, stage 2, stage 3). By the repetitive firing, the hardness of the tested alloy decreased gradually. By holding the specimen at $500^{\circ}C$ for 10-20min after simulated firing, the hardness increased apparently. However, to hold the alloy for long periods of time in the relatively high temperature after simulated firing resulted in the formation of thick oxidation layer. The oxide film formed on the surface of the alloy after simulated complete firing with controlled cooling rate, which was mainly composed of O and Zn. Conclusion: It is reasonable to hold the alloy at $500^{\circ}C$ for 10-20min after complete firing in other to improve the final hardness of the alloy.

  • In order to investigate the organoleptic properties of rapid processed sardine sauce which was prepared using sardine autolysates and sardine meal koji, the taste endowing compounds were analysed. The chemical compositions of the rapid processed sardine sauce fermented for 15 days at $40^{\circ}C$ were $1.98\%$ in total nitrogen, $1.08\%$ in amino type nitrogen, $5,296.8mg\%$ in free amino acid, and the salinity was $13.0\%$ (W/V). the predominant free amino acid in the sauce were glutamic acid, lysine, aspartic acid, valine and leucine. Those amino acids held $53.9\%$ of total free amino acids. The content of IMP was $20.8mg\%$. The rapid processed sardine sauce was comparable to traditional anchovy sauce which was fermented at ambient temperature for one year in the distribution and concentrations of nitrogenous compounds and organoleptic properties.

  • Bacillus stearothermophilus No. 236 isolated from the soil is a strong xylan degrader producing all the xylanolytic enzymes. However, the strain was discovered to be highly intractable to its transformation. In the present study, we have developed a reliable method for transformation of B. stearothermophilus No. 236 by a systematic examination of several factors which might have an influence on the efficiency of electrotransformation. Notably, we found that the most critical factor influencing the transformation efficiency (TE) was the incubation temperature after pulsing, with its optimum incubation of $37^{\circ}C.\; At\; 50^{\circ}C$, the optimum growth temperature of the B. stearothermophilus strain, the transformants could not be obtained at a recognizable level. The combination of field strength of 7.5 kV/cm along with pulse duration of 10 msec (resistance of $400{\Omega}\; and\; capacitance\; of\; 25{\mu}F$) was shown to be the best electrical parameters at the incubation temperature of $37^{\circ}$. A higher TE was obtained when the cells were harvested at an early-exponential phase. Twenty percent of PEG-8000 in a suspension buffer and an addition of 0.1% glycine in the growth medium resulted in about 4-fold and 3-fold increases in TE, respectively. We also found that the plasmid DNA which had been cycled through the host B. stearothermophilus cells enhanced TE by one order of magnitude higher. Under the presently described conditions, $2.5{\times}10^{5} transformants per ${\mu}g$ DNA was attained.

  • A study detennined whether certain biochemical and physiological variables were altered during marginal Zn deficiency. Ten weaned crossbred Hereford Angus heifer calves, weighing $163{\pm}2kg$, were utilized. Five calves were fed a Zn - deficient (- Zn) brome-alfalfa hay diet containing 17 mg Zn/kg diet DM, and five calves were fed a Zn-adequate (+Zn) diet with 23 mg Zn/kg diet DM from $ZnSO_4$ added to the - Zn diet (total diet, 40 mg Zn/kg diet DM), for 32 d. At 21 d the - Zn calves had a reduction (p < .05) in feed efficiency. By 25 d, plasma Zn and alkaline phosphatase concentrations were reduced (p < .05) in the - Zn calves. Blood urea nitrogen, glucose, insulin, IGF-I, Cu plasma concentration and Zn and Cu concentrations of red blood cell (RBC) and liver were not altered (p > .05) by the - Zn diet through 25 d. In response to a single i. m. injection of dexamethasone (20 mg) on d 25, calves fed the two dietary Zn amounts showed no changes (p > .05) in plasma or RBC Zn and Cu concentrations, serum IGF-I, insulin, and glucose when measured at 6, 12, 24, 48, 72, and 96 h after injection. In response to an intradermal injection of phytohemagglutinin on d 30, cell mediated immune (CMI) response was reduced (p < .05) in the - Zn calves. These observations indicate that during a marginal Zn deficiency in calves, there was a decrease in feed efficiency, plasma Zn, serum alkaline phosphatase, and CMI response.

  • Optimum conditions for production of casein phosphopeptides (CPP) from sodium casenate by immobilized cell culture of Streptococcus faecalis var. liquefaciens were investigated. Immobilized cells were made by mixing 60% sodium alginate solution with an equal volume of culture broth at the end of exponential phase and subsequently dropping the mixture into $CaCl_{2}$ solution. Optimum conditions for CPP production by the immobilized cells were the same as those ($50^{\circ}C$, pH 7.0, and 10% substrate concentration) by the crude enzyme solution from the supernatant of culture broth. Optimum loading volume of the immobilized cells into a batch reactor was 30% (w/v). Using a continuous reactor loaded by the immobilized cells under the identified optimal conditions, we were able to produce CPP continuously up to 30 days with a maximum CPP conversion efficiency of 20%.

  • Production from copepodite IV to adult of two euchaetid species Euchaeta plana and Paraeuchaeta russelli was measured at the southeastern sea of Korea from April to November, 2014. The mean density was $2.0ind\;m^{-3}$ for E. plana and $4.1ind\;m^{-3}$ for P. russelli, with the high contribution of copepodite V to total density. The densities of total individuals, adult females and eggs were highest in November for both species. The mean egg production rate (EPR) was $1.7eggs\;female^{-1}\;d^{-1}$ for E. plana and $3.1eggs\;female^{-1}\;d^{-1}$ for P. russelli. Both of them showed the highest EPR in September but zero EPR in summer. The mean weight-specific EPR was $0.038d^{-1}$ for E. plana and $0.079d^{-1}$ for P. russelli. The mean total production rates of E. plana and P. russelli were $5.3{\mu}g\;C\;m^{-3}\;d^{-1}$ and $17.8{\mu}g\;C\;m^{-3}\;d^{-1}$, respectively, with the largest production in November. The mean Production/Biomass ratio was $0.06d^{-1}$ for E. plana and $0.07d^{-1}$ for P. russelli, with its peak in September for both. The total production of E. plana and P. russelli was positively correlated with the density of a copepod Oncaea venusta, rather than chl-a concentration, indicating that the two copepods might be carnivores. This study evaluates the contribution of euchaetids to the copepod community in the southeastern sea of Korea.

  • Edible offal is easily contaminated by Escherichia coli (E. coli) and fluoroquinolone (FQ)-resistant E. coli is considered a serious public health problem, thus, this study investigated the genetic characteristics of FQ-resistant E. coli from edible offal. A total of 22 FQ-resistant E. coli isolates were tested. A double mutation in each gyrA and parC led the highest MIC. Four (18.2%) isolates carried plasmid-mediated quinolone resistance genes. The fimH, eaeA, escV, astA, and iucC genes were confirmed. Seventeen isolates (77.3%) were positive for plasmid replicons. The isolates showed high genetic heterogeneity based on pulsed-field gel electrophoresis patterns.

  • In this study, we isolated 32 strains of kerosene degrading bacteria from oil contaminated soil by enrichment culture. Isolates were screened for kerosene degradation efficiencies and K14 were selected which had the highest removal efficiency for 1,000 mg/L of kerosene. K14 were identified as Pseudomonas aeruginosa by morphological, biochemical test and 16S rDNA analysis. The optimal culture condition were determined as initial inoculated cell concentration, 1.0 g/L; substrate concentration, 1,000 mg/L; temperature $30^{\circ}C$; pH 7. When we enforced batch test in this condition, K14 degraded 72% of kerosene with 1,000 mg/L during 72 hr. And, at low concentration (200 mg/L), K14 degraded 95.8% of kerosene during 48 hr. As a result, kerosene biodegradation by Pseudomonas aeruginosa K14 could be useful for clean up of groundwater and soil contaminated with crude oil.
