Characteristics of $A_1\;and\;A_2$ Adenosine Receptors upon the Acetylcholine Release in the Rat Hippocampus

  • As it has been reported that the depolarization induced acetylcholine (ACh) release is modulated by activation of presynaptic $A_1$ adenosine heteroreceptor and various lines of evidence suggest the $A_2$ adenosine receptor is present in the hippocampus. The present study was undertaken to delineate the role of adenosine receptors on the hippocampal ACh release. Slices from the rat hippocampus were equilibrated with $[^3H]choline$ and then the release amount of the labelled product, $[^3H]ACh$, which was evoked by electrical stimulation (rectangular pulses, 3 Hz, 2 ms, 24 mA, $5\;V/cm^{-1}$, 2 min), was measured, and the influence of various adenosine receptor-related agents on the evoked tritium outflow was investigated. And also, the drug-receptor binding assay was performed in order to confirm the presence of $A_1$ and $A_2$ adenosine receptors in the rat hippocampus. N-ethylcarboxamidoadenosine (NECA), a potent adenosine receptor agonist with nearly equal affinity at $A_1$ and $A_2$ adenosine receptors, in concentrations ranging from $1{\sim}30\;{\mu}M$, decreased the electrically-evoked $[^3H]ACh$ release in a concentration-dependent manner without affecting the basal rate of release. And the effect of NECA was significantly inhibited by 8-cyclopentyl-1,3-dipropylxanthine (DPCPX, 2 ${\mu}M$), a selective $A_1$ adenosine receptor antagonist, but was not influenced by 3,7-dimethyl-1-propargylxanthine (DMPX, 5 ${\mu}M$), a specific $A_2$ adenosine receptor antagonist. $N^6-cyclopentyladenosine$ (CPA), a selective $A_1$ adenosine receptor agonist, in doses ranging from 0.1 to 10 ${\mu}M$, reduced evoked $[^3H]ACh$ release in a dose-dependent manner without the change of the basal release. And the effect of CPA was significantly inhibited by 2 ${\mu}M$ DPCPX treatment. 2-P-(2-carboxyethyl)-phenethylamino-5'-N-ethylcarboxamidoadenosine hydrochloride (CGS-21680C), a potent $A_2$ adenosine receptor agonist, in concentrations ranging from 0.1 to 10 ${\mu}M$, did not alter the evoked ACh release. In the drug-receptor binding assay, the binding of $[^3H]2-chloro-N^6-cyclopentyladenosine$ ($[^3H]$CCPA) to the $A_1$ adenosine receptor of rat hippocampal membranes was inhibited by CPA ($K_i$ = 1.22 nM), NECA ($K_i=10.17 nM$) and DPCPX ($K_i=161.86 nM$), but not by CGS-21680C ($K_i=2,380 nM$) and DMPX ($K_i=22,367 nM$). However, the specific binding of $[^3H]CGS-21680C$ to the $A_2$ adenosine receptor was not observed. These results suggest that the $A_1$ adenosine heteroreceptor play an important role in evoked ACh release, but the presence of $A_2$ adenosine receptor is not confirmed in this study.

Comparisons of Developmental Potential and Gene Expression Level in Porcine Nuclear Transfer, Parthenogenetic and Fertilized Embryos

  • This study was conducted to detect the apoptosis incidence in blastocysts and to compare the abundance of Bax, Bcl2L1, VEGF and FGFR2 in in vitro fertilized (IVF), parthenogenetic (PAT) and nuclear transfer (NT) embryos. Oocytes matured for 40 hr were enucleated and reconstructed with confluenced fetal fibroblasts (FFs) derived from a ${\sim}45$ day fetus. Reconstructed eggs were then fused with 2 DC pulses (2.0 kV/cm, $30{\mu}sec$) and cultured with $7.5{\mu}g/ml$ cytochalasin B for 3 hr. Parthenotes (PAT) were produced with the same electric strength and culture for NT eggs. The embryos were cultured in NCSU-23 medium at $39^{\circ}C,\;5%\;CO_2,\;5%\l;O_2$ in air. In 3 runs, set of 10 embryos at the 4-cell to blastocyst stages were used to extract total RNA for analyzing the gene expression patterns of pro-apoptotic (Bax), anti-apoptotic (Bcl2L1), vasculogenesis (VEGF), implantation (FGFR2III) using real-time quantitative PCR. Cleavage and blastocyst rates were significantly higher (P<0.05) in IVF and PAT ($79.3{\pm}8.5\;and\;25.5{\pm}6.1,\;and\;85.0{\pm}6.4\;and\;38.6{\pm}5.5$, respectively)than NT counterparts ($65.1{\pm}5.2\;and\;15.6{\pm}3.0$, respectively). Significantly higher (P<0.05) total cells were observed in IVF controls and PAT ($34.7{\pm}5.8\;and\;38.1{\pm}4.1$) than NT embryos ($24.8{\pm}3.2$). Apoptosis index was significantly lower (P<0.05) in IVF than NT embryos. The Relative abundances (RA) of Bax and VEGF were significantly higher (P<0.05) at blastocyst stage in NT than IVF control. The RA of Bcl2L1 and FGFR2III were significantly higher (P<0.05) at blastocyst stage in IVF than NT. The present study observed the abnormal gene expressions in NT embryos at various developmental stages, suggesting certain clues to find out the cause of the low efficiency of NT to term.

기기 중성자방사화 분석을 이용한 대전 지하철 객차 내 PM10과 미량성분의 특성 (PM10 and Associated Trace Elements in the Subway Cabin of Daejeon by Instrumental Neutron Activation Analysis)

  • 본 연구는 기기중성자방사화분석법을 이용하여 다중이용시설 중 지하철 객차 내 실내공기 중 미세먼지(PM10)의 미량원소 분포특성과 실내공기질을 평가하고, 향후 실내공기질의 효율적인 제어를 위한 객차 내 PM10의 오염원을 평가하고자 하였다. 대전광역시에서 운행 중인 지하철 객차 내에서 채취한 PM10의 평균농도는 $59.3{\pm}14.5{\mu}g/m^3$이었고, 지하철 객차 내 PM10 중 24종의 원소성분을 정량분석한 결과, 농도는 $10^{-3}{\sim}10^5ng/m^3$ 범위에 걸쳐 넓게 분포하였으며, Fe ($12.5{\mu}g/m^3$)의 농도가 특히 높았다. 이는 지하철 브레이크 시스템과 철로 및 차륜 마찰과 같은 지하철 운행에 의하여 Fe가 상당히 부유되기 때문인 것으로 판단된다. 분석된 미량원소 농도를 바탕으로 오염원을 평가한 결과, brake-nonferrous metal particle, resuspended rail dust, fuel combustion, vehicle exhaust, black carbon, Cr-related가 주요 오염원인 것으로 나타났다.

Family history of chronic renal failure is associated with malnutrition in Korean hemodialysis patients

  • The present study was to investigate the nutritional status and factors related to malnutrition in end-stage renal disease (ESRD) patients requiring hemodialysis (HD) in South Korea. Subjects were ESRD outpatients from general hospitals or HD centers in Seoul referred to the dialysis clinic for maintenance HD care. A total of 110 patients (46 men and 64 women; mean ages $58.6{\pm}1.0y$) were eligible for this study. The family history of chronic renal failure (CRF) was considered positive if a patient reported having either a first-degree or second-degree relative with CRF. Malnutrition was defined as a triceps skinfold thickness or mid-ann muscle circumference below the fifth percentile for age and sex and forty-seven of the 110 patients were malnourished. Almost all (94%) patients had anemia (hemoglobin: <13 g/dL for men and <12 g/dL for women). Energy intake was below the recommended intake levels of energy [30-35 kcal/kg ideal body weight (IBW)] and protein (1.2 g/kg IBW) in 60% of patients. The duration of HD was longer in malnourished HD patients (P=0.0095). Malnutrition was more prevalent in women (P=0.0014), those who never smoked (P=0.0007), nondiabetic patients (P=0.0113), and patients with bone diseases (P=0.0427), adequate HD (spKt/$V{\geq}1.2$) (P=0.0178), and those with a family history of CRF (P=0.0255). Multiple logistic regression was used to examine the relationship between malnutrition and potential risk factors. After adjusting for age, sex, and other putative risk factors for malnutrition, the OR for malnutrition was greater in HD patients with a family history of CRF (OR, 3.290; 95% CI, $1.003{sim}10.793$). Active nutrition monitoring is needed to improve the nutritional status of HD patients. A family history of CRF may be an independent risk factor for malnutrition in Korean HD patients. A follow-up study is needed to investigate whether there is a causal relationship between a family history of CRF and malnutrition in Korean ESRD patients.

소나무 잎과 가지의 정유와 그 성분들의 항산화 및 항노화 작용 (Anti-oxidant and Anti-aging Activities of Essential Oils of Pinus densiflora Needles and Twigs)

  • To investigate effects of pine essential oil isolated by steam distillation from Pinus densiflora needles (PN) and twigs (PT) on anti-oxidant and anti-aging activities, the experiments were carried out to determine anti-oxidant and antiaging activities on DPPH radical scavenging activity, NBT/xanthine oxidase-superoxide scavenging activity, silica-induced intracellular $H_2O_2$ and hydroxyl radical generation in RAW264.7 cells, hyaluronidase and elastase activities in vitro. Essential oil of PN and PT were contained 0.225 and 0.176 (w/v) %, respectively. PN was contained with 30 kinds of essential oil and its major constituent is $\alpha$-pinene (21.5%). Further PT was contained with 40 kinds of essential oil and its major constituent is $\beta$-pinene (22.4%) in GC/MS assay. Other essential oils of PN were $\beta$-pinene, camphene, myrcene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene and PT were $\alpha$-pinene, camphene, phellandrene, limonene, terpinolene, $\alpha$-terpineol, bornyl acetate, $\alpha$-copaene, caryophellene and humulene. The essential oil of PT have more active than that of PN in anti-oxidant activity which has significant DPPH radical and superoxide scavenging activity, and significant inhibitory activities on silica-induced intracellular $H_2O_2$ and hydroxyl radical generation, as well as, significantly inhibited elastase and hyaluronidase activities. Further, phellandrene, myrcene and $\alpha$-pinene have DPPH radical and superoxide scavenging activities, $\beta$-pinene, terpinolene, myrcene and phellandrene inhibited silica-induced intracellular $H_2O_2$ and hydroxyl radical generations. And also phellandrene and $\beta$-pinene inhibited hyaluronidase and elastase activities. In conclusion, the essential oils isolated from PN and PT have anti-oxidant and anti-aging activities.

화악산(경북 ${\cdot}$ 청도)의 관속식물 분포현황 (Distribution of Vascular Plants in Mt. Hwaak (Cheongdo-gun, Gyeongsangbuk-do))

  • 본 연구는 화악산(931m)의 관속식물 분포를 파악하기 위하여 수행되었다. 2006년 5월부터 2007년 10월까지 6회에 걸쳐 채집된 관속식물은 총 92과 248속 281종 1아종 47변종 9품종의 총 338종류로 정리되었으며, 이는 한반도의 관속식물 4,881종류의 6.9%에 해당되었다. 조사 지역에서 분포하고 있는 338종류의 식물자원 유용도를 분석한 결과 총 278종류의 자원식물로 분류되었으며, 식용자원(E)은 154종류(55.3%), 약용자원(M)은 163종류(58.6%), 관상자원(O)은 58종류(20.8%), 초지자원(P)은 91종류(32.7%), 공업원료자원(I)은 22종류(7.9%), 목재자원(T)은 9종류(3.2%)로 나타났다. 특기할만한 식물로는 한국특산식물이 9종류, 산림청 지정희귀 및 멸종위기식물이 1종류, 식물구계학적 특정식물이 21종류, 귀화식물이 19종류로서 귀화율은 5.6%, 도시화지수는 6.8%로 나타났다.

  • We developed an 4.5" $192{\times}64$ active matrix organic light-emitting diode display on a glass using organic thin-film transistor (OTFT) switching-arrays with two transistors and a capacitor in each sub-pixel. The OTFTs has bottom contact structure with a unique gate insulator and pentacene for the active layer. The width and length of the switching OTFT is $800{\mu}m$ and $10{\mu}m$ respectively and the driving OTFT has $1200{\mu}m$ channel width with the same channel length. On/off ratio, mobility, on-current of switching OTFT and on-current of driving OTFT were $10^6,0.3{\sim}0.5\;cm^2/V{\cdot}sec$, order of 10 ${\mu}A$ and over 100 ${\mu}A$, respectively. AMOLEDs composed of the OTFT switching arrays and OLEDs made using vacuum deposition method were fabricated and driven to make moving images, successfully.

Apert syndrome : 증례보고 (APERT SYNDROME : CASE REPORT)

  • Apert syndrome은 관상봉합의 조기유합에 의한 첨두증(acrocephaly)과 합지증(syndactyly)이 함께 나타나는 선천성 유전 질환으로서, 1906년 Apert에 의해 보고된 증후군이다. 약 $65,000{\sim}160,000$ 당 1명꼴로 발생되며, FGFR2(fibroblast growth factor receptor 2)의 돌연변이로 생겨난다고 알려져 있고, 상염색체 우성 유전을 한다. 임상적으로 첨단두증(acrobrachycephaly)을 보이고, 특징적으로 손과 발의 합지증을 보이며, 높은 이마와 평평한 뒤통수, 후퇴된 중안면부, 안구돌출, 시력이상, 양안격리증, 밑으로 쳐진 안검열, 상악골 형성부전, 상대적인 하악 전돌의 양상 등을 보인다. 감소된 비강인두와 좁아진 후비공으로 인한 구호흡과 전방부 개교를 보이며, 이완시에 입술의 모양은 사다리꼴 형태를 보인다. 중이염이 흔하고, 청각장애를 유발하기도 하며, 정신지체가 높은 비율로 나타난다. 특징적인 구내소견으로 연구개열 또는 구개수열과 가성 경구개열, V자 모양의 악궁과 치열의 총생 등이 관찰되며, 전치부 개방교합과 구치부 교차교합을 동반한 Class III 부정교합을 보인다. 본 증례는 강릉대학교 치과병원 소아치과에 내원한 6세 3개월의 남아로 충치 치료를 받고 싶다는 것을 주소로 내원하였다. 이 환자에서 보이는 Apert syndrome의 특징적인 치과적 소견에 대해 보고하는 바이다.

방사선(放射線) 치료(治療)의 신속정확(迅速正確)을 위한 저온용융(低溫熔融) 차폐물(遮蔽物)의 제작(製作)과 응용(應用) (Rapidly and Accurately Processing of Low Melting Block for Shielding of Radiotherapy)

  • 고(高)에너지 방사선(放射線) 치료(治療)에 있어서 정상조직(正常組織)의 완전차폐(完全遮蔽)를 위하여 $5{\sim}8cm$ 납두께의 부정형(不定形) 차폐(遮蔽)벽돌을 제작(製作)해야하는 난점(難點)이 있었다. 저자(著者)들은 납 30.0%, 주석 11.5% 비스므스 48.5%, 카드미늄 10.0%를 사중(四重) 공정결합(共晶結合)시켜 밀도(密度)가 $9.8g/cm^3$ 용융온도(熔融溫度)가 $68^{\circ}C$인 저용융(低熔融) 차폐물질(遮蔽物質)을 개발(開發)하여 이를 Lead Y라고 명명(名命)하였다. 제작(製作)된 Lead Y Block을 $68^{\circ}C$에서 용융(熔融)시켜 보호(保護)해야할 중요(重要)한 장기(臟器)의 형태(形態)대로 제작(製作)된 styrofoam 음형(陰形)에 부어서 차폐효과(遮蔽效果)가 큰 차폐(遮蔽)벽돌을 쉽고 안전(安全)하게 제작(製作)할 수 있었고 납보다 더 단단하고 재현성(再現性)이 크며 저렴(低廉)한 가격(價格)으로 구입(購入)이 가능(可能)하므로 방사선(放射線) 치료효과(治療效果)에 큰 도움을 줄 수 있었다.

버찌(Fruit of Prunus serrulata L. var. spontanea Max. wils.) 분말을 첨가한 요구르트의 저장 중 품질 특성 (Quality Characteristics of Yogurt Prepared with Flowering Cherry (Prunus serrulata L. var. spontanea Max. wils.) Fruit Powder during Storage)

  • 버찌분말을 첨가한 요구르트(0.0, 0.5, 1.0, 2.0%)를 제조하여 $4^{\circ}C$에 저장하면서 14일간 품질특성을 조사하였다. 요구르트의 pH는 버찌분말 첨가에 따라 유의적으로 감소하였으며, 저장기간의 경과에 따라서는 대조구 및 버찌분말 첨가구모두에서 낮아지는 경향을 나타내었다. 적정산도는 대조구에 비해 버찌분말 첨가 요구르트에서 높은 값을 나타내었으며, 대조구 및 버찌분말 첨가구 모두 저장기간의 경과에 따라서 산도가 증가하는 것으로 나타나 pH 변화와 비슷한 양상을 나타내었다. 버찌분말 첨가에 따라 요구르트의 당도는 높아졌으며, 점도는 감소하는 경향을 나타내었다. 요구르트의 색도는 버찌분말 첨가량이 증가할수록 명도와 황색도가 감소하고 적색도는 증가하는 유의적인 차이를 나타내었다. 항산화활성은 버찌분말의 첨가에 의해 유의적으로 증가하였다. 버찌분말 요구르트의 유산균수는 대체적으로 대조구에 비해 버찌분말 첨가 요구르트에서 높게 측정되었고 저장기간 동안 모든 시료구에서 증가하는 경향을 나타내었다. 관능평가 결과 색에 대한 선호도를 제외하고 냄새, 점성, 맛, 전반적인 기호도에서 1.0% 버찌분말 첨가구에서 높은 선호도를 나타내었다. 따라서 요구르트에 대한 버찌분말의 첨가는 저장동안의 품질 및 관능 특성, 기능성을 고려했을 경우 1.0% 첨가가 최적일 것으로 판단되었다.
