8-isoprostane (8-$isoPGF_{2{\alpha}}$) is a reliable marker and considered a gold standard for lipid peroxidation. There are very few reports of 8-isoprostane levels in cancer patients, and in patients undergoing chemotherapy. Oxidative stress is however expected and has been observed in patients with cancer. This study measured 8-isoprostane levels in urine by ELISA of 25 patients undergoing chemotherapy for advanced non-small cell lung cancer, at cycles 1, 2, and 3 of treatment. It considers the creatinine clearance of the patients, and correction of 8-isoprostane levels by creatinine clearance, and overnight urine volume methods. The average 8-isoprostane levels in urine increased more than 6 to 12 fold on chemotherapy treatment, from $532{\pm}587$ pg/mL at cycle $1,6181{\pm}4334$ at cycle 2, and $5511{\pm}2055$ at cycle 3. Similar results were obtained if 8-isoprostane levels were corrected for overnight urine volume, giving averages of $285{\pm}244{\mu}g$ at cycle $1,4122{\pm}3349$ at cycle 2, and $3266{\pm}1200$ at cycle 3. No significant difference was seen in average total overnight urine volume or number of urinations between chemotherapy cycles except for a large variation in urine volume between cycle 2 and 3. Creatinine levels were significantly different only between cycles 1 and 2 (p=0.016). In conclusion, cisplatin therapy has been shown to induce high levels of lipid peroxidation in lung cancer patients and can be assessed from the 8-isoprostane marker in overnight urine, with or without urine volume correction.

Objective: The aim of the study was to conduct metabolic profiling of dairy cattle serum and urine using proton nuclear magnetic resonance (1H-NMR) spectroscopy and to compare the results obtained with those of other dairy cattle herds worldwide so as to provide a basic dataset to facilitate research on metabolites in serum and urine. Methods: Six dairy cattle were used in this study; all animals were fed the same diet, which was composed of total mixed ration; the fed amounts were based on voluntary intake. Blood from the jugular neck vein of each steer was collected at the same time using a separate serum tube. Urine samples were collected by hand sweeping the perineum. The metabolites were determined by 1H-NMR spectroscopy, and the obtained data were statistically analyzed by performing principal component analysis, partial least squares-discriminant analysis, variable importance in projection scores, and metabolic pathway data using Metaboanalyst 4.0. Results: The total number of metabolites in the serum and urine was measured to be 115 and 193, respectively, of which 47 and 81, respectively were quantified. Lactate (classified as an organic acid) and urea (classified as an aliphatic acylic compound) exhibited the highest concentrations in serum and urine, respectively. Some metabolites that have been associated with diseases such as ketosis, bovine respiratory disease, and metritis, and metabolites associated with heat stress were also found in the serum and urine samples. Conclusion: The metabolites measured in the serum and urine could potentially be used to detect diseases and heat stress in dairy cattle. The results could also be useful for metabolomic research on the serum and urine of ruminants in Korea.
Constituents of animal biofluids such as milk, blood and urine contain information specifically related to metabolic and health status of the ruminant animals. Some changes in composition of biofluids can be attributed to disease response of the animals. Mastitis is a major problem for the global dairy industry and causes substantial economic losses from decreasing milk production and reducing milk quality. The purpose of this study was to investigate potential of NIRS combined with multivariate analysis for cow's mastitis diagnosis based on NIR spectra of milk, blood and urine. A total of 112 bulk milk, urine and blood samples from 4 Holstein cows were analyzed. The milk samples were collected from morning milking. The urine samples were collected before morning milking and stored at -35$^{\circ}C$ until spectral analysis. The blood samples were collected before morning milking using a catheter inserted into the carotid vein. Heparin was added to blood samples to prevent coagulation. All milk samples were analyzed for somatic cell count (SCC). The SCC content in milk was used as indicator of mastitis and as quantitative parameter for respective urine and blood samples collected at same time. NIR spectra of blood and milk samples were obtained by InfraAlyzer 500 spectrophotometer, using a transflectance mode. NIR spectra of urine samples were obtained by NIR System 6500 spectrophotometer, using 1 mm sample thickness. All samples were divided into calibration set and test set. Class variable was assigned for each sample as follow: healthy (class 1) and mastitic (class 2), based on milk SCC content. SIMCA was implemented to create models of the respective classes based on NIR spectra of milk, blood or urine. For the calibration set of samples, SIMCA models (model for samples from healthy cows and model for samples from mastitic cows), correctly classified from 97.33 to 98.67% of milk samples, from 97.33 to 98.61% of urine samples and from 96.00 to 94.67% of blood samples. From samples in the test set, the percent of correctly classified samples varied from 70.27 to 89.19, depending mainly on spectral data pretreatment. The best results for all data sets were obtained when first derivative spectral data pretreatment was used. The incorrect classified samples were 5 from milk samples,5 and 4 from urine and blood samples, respectively. The analysis of changes in the loading of first PC factor for group of samples from healthy cows and group of samples from mastitic cows showed, that separation between classes was indirect and based on influence of mastitis on the milk, blood and urine components. Results from the present investigation showed that the changes that occur when a cow gets mastitis influence her milk, urine and blood spectra in a specific way. SIMCA allowed extraction of available spectral information from the milk, urine and blood spectra connected with mastitis. The obtained results could be used for development of a new method for mastitis detection.
이 실험의 목적은 탄수화물 부하식이요법시의 삼출건비방 투여가 달리기 선수의 뇨중 대사성분변화에 미치는 영향에 대해 관찰 연구한 것이다. 탄수화물 부하식이시 삼출건비탕(蔘朮健脾湯) 투여는 전해질대사 관련 변인에 있어서 $Na^+$, $Cl^-$, $K^+$, $Ca^{+ +}$에 유의성 있는 영향을 미쳤으며, 이는 세포막 투과성의 변화를 의미하는 것으로 $Na^+$-$K^+$ pump의 활성도가 증가하여 운동수행능력을 향상시킬 것으로 기대되나 추후 전해질 대사와 관련하여 renin, aldosterone 등의 호르몬 분비 기전에 관해서도 지속적인 연구가 진행되어야 할 것으로 사려된다.
In hospitals, nurses are subjectively determining the urine status to check the kidneys and circulatory system of patients whose statuses are related to patients with kidney disease, critically ill patients, and nursing homes before and after surgery. To improve this problem, this paper proposes a urine spectrum analysis system which clusters urine test results based on a hybrid machine learning model consists of unsupervised learning and supervised learning. The proposed system clusters the spectral data using unsupervised learning in the first part, and classifies them using supervised learning in the second part. The results of the proposed urine spectrum analysis system using a mixed model are evaluated with the results of pure supervised learning. This paper is expected to provide better services than existing medical services to patients by solving the shortage of nurses, shortening of examination time, and subjective evaluation in hospitals.
본 연구에서 방광 내벽과 외벽사이의 거리를 측정하여 뇨량을 계산하는 시스템에 적용하기 위한 방광용 초음파 변환기를 설계하고 개발하였다. 뇨량 측정 시스템은 방광에서 오줌을 배출하기 전의 양을 측정함으로써 뇨실금 환자의 치료 과정을 도와주게 된다. 이 실험의 결과로, 뇨량을 더 정확하게 측정할 수 있고 또한 뇨실금 방지 시스템을 개발할 수 있다. 본 연구를 통하여 개발되는 시스템은 뇨실금과 야뇨증 환자를 더 오래 더 건강한 삶을 살 수 있도록 해줄 수 있을 것이다.
The alterations in serum and urine potassium were studied in twenty two patients who underwent open heart surgery using extracorporeal circulation from June 1990 to August 1990 at the Department of Thoracic and Cardiovascular Surgery, School of Medicine, Kei-myung University. There were fifteen cases congenital heart disease and seven acquired heart disease. The serum and urine potassium levels were measured pre-, intra- and postoperatively until seventh postoperative day using ionic selective electrode measuring method. After general anesthesia, the serum potassium level decreased significantly but slowly increased during.cardiopulmonary bypass and returned preoperative level after operation. The urine potassium level decreased slowly from general anesthesia to cardiopulmonary bypass weaning but returned preoperative level following operation. During cardiopulmonary bypass, serum and urine potassium levels in diuretic group were lower than that of non diuretic group. There was no remarkable difference in the serum potassium level between single RA cannulation group and bicaval cannulation group preoperatively, but the serum potassium level in single RA cannulation group was much higher than that of bicaval cannulation group. There was no significant difference in the urine potassium level between single RA can-nulation group and bicaval cannulation group postoperatively.
A gas chromatography with flame ionization detection was developed to measure tranylcypromine in rat urine. The method involves extraction of the drug and the internal standard, phenylpropylamine from the urine using ethyl acetate and back extraction into 0.5N $H_{2}SO_{4}$. Following final extraction using dichloromethane, both the drug and the internal standard were converted to trifluoroacetyl derivatives and analyzed using a column of 3% SE-30 on 80/100 mesh Chromosorb W(HP). A calibration curve was constructed in the range of $5~50{\mu}g$tranylcypromine sulfate in 0.5ml urine and found to be linear. The detection limit was $2{\mu}g$. The tranylcypromine could be analyzed with the percent recovery of $100.81{\pm}8.13$ (SD) ina concentration range of $8-40{\mu}g$ in 0.5ml urine. When 0.4mmol/kg dose of the drug was administered through, an oral route, excretion percent of tranylcypromine in rat urine over 36hr was found to be $11.90{\pm}6.04$ (SD) for tranyleypromine sulfate and $2.23{\pm}0.63$ (SD) for benzyl trans-2-phenylcyclopropanecarbamate.
A method was described for the graphite furnace atomic absorption spectrophotometric determination of trace cadmium, copper, chromium and lead in urine samples. The elements were directly determined without any other treatments. The ash temperature was intensively optimized to improve the large background by the removal of organic materials and alkali and alkali earth metals in urine samples. Two kinds of standard solutions were used to plot calibration curves. From the recovery data, it could be confirmed that the analytical results with the synthetic urine matrix similar to real urine were more accurate than with a deionized water matrix.
