Artificial Reestablishment of the Kelp and Red Algal Symbiosis

  • A type of symbiosis was previously described from nature in which the gametophytes of Laminariales were endophytic in filamentous red algae. Here we reconstruct this symbiosis for the first time in laboratory culture using zoospores of the kelp, Undaria pinnatifida, and the red alga, Aglaothamnion oosumiense. Zoospores of U. pinnatifida readily attached to A. oosumiense. In 48 h these spores germinated and the initial germ tube penetrated into the host cell wall leaving only an empty zoospore wall outside the host. Within ten days, four to five-celled endophytic gametophytes were present. Zoospores of Laminaria religiosa which were also inoculated into cultures of A. oosumiense rarely attached to the red alga and never became endophytic. Within ten days the free-living gametophytes of L. religiosa on cover slips became fertile and produced young sporophytes. These observations demonstrate the ability of U. pinnatifida to become endophytic, and show differences in host specificity among kelp species.

Host Specificity of Endophytic Kelp Gametophytes (내부공생 켈프 배우체의 숙주 선택)

  • Farmed kelp gametophytes were previously observed to be living endophytically in filamentous red algae. The interactions of two farmed kelp species and six red algae were examined in laboratory culture. Undaria pinnatifida (Harvey) Suringar and Laminaria religiosa Miyabe demonstrated the differing abilities of zoospores to become endophytic in four host red algae and neither kelp became endophytic in two non-filamentous red algae. There was a strong seasonal component regarding infectiousness that is associated with the changes in fron erosion in U. pinnatifida from April to June. At the same time, L. religiosa showed no significant changes in frond erosion, and there were no apparent changes in infection levels in the two species they were able infect. This study indicated clear differences between two keip species with regard to their symbiotic relationship to red algae in terms of host specificity and preference of kelp gametophytes.

Characteristics of Lead Biosorption by Biosorbents of Marine Brown Algae (해양 갈조류를 생물흡착제로 이용한 납흡착 특성 연구)

  • Lead sorption performances by biomass of nonliving, dried marine brown algae Undaria phnaunda, Hlzikia hsyormls. and Sugassum fulvellum used as biosorbent materials were investigated. As the amount of biosorbent materials added was increased, the lead removal by biosorbent materials Increased but the lead biosorption capacities decreased. However, with increasing Initial lead concentration the lead biosorption capacities by the biosorbent materials Increased but lead removal efficiencies decreased. In the range of Initial lead concentration(Co) 10-500 mg/L the lead biosorption capacities and removal efficiencies by the biosorbent materials Increased with increasing pH. Among the biosorbent materials used in this study, the lead biosorption capacities decreased in the following sequence: U. plilnaunda > H. fusiformis > S. fulvellum. The lead biosorption by biosorbent materials were expressed by the Langmuir Isotherm better than the Freundlich Isotherm. The biosorption rate could be expressed by the first order reaction rate equation for initial lead concentration like that rad : 0.288Co for U. phanda, rad = 0.255Co for H. fusiformis, and rad : 0.161Co for S. fulvellum. Community Dynamics of Benthic Marine Algae in the Intertidal and Subtidal Rocky Shore of Samyang, Jejudo Island

  • Community structure and vertical distribution of benthic marine algae were investigated in the intertidal and subtidal rocky shore of Samyang, Jejudo Island. The total number of 62 algal species composed of 6 Cyanophyta, 6 Chlorophyta, 19 Phaeophyta, and 31 Rhodophyta were observed. The dominant species were melobesioidean algae, Sargassum thunbergii, Hizikia fusiformis, and Ulva pertusa in the intertidal zone and melobesioidean algae, Ulva pertusa, Corallina spp., and Undaria pinnatifida in the subtidal zone. The vertical algal distribution was represented by the melobesioidean algae in the whole rocky shore, Caulacanthus ustulatus in the upper intertidal zone, Sargassum thunbergii and Hizikia fusiformis in the middle and lower zone, Ulva pertusa in the lower zone and Ulva pertusa, Corallina spp., and Undaria prinnatifida in the subtidal zone. The composition of dominant species and pattern of the vertical distribution of algae in the present study were significantly different from the previous reports. Especially, distribution of crustose coralline algae was significantly extended.

Anti-arteriosclerotic and Anti-hyperlipidemic Effects of Sea Mustard (Undaria pinnatifida) in Sprague-Dawley rats

  • This study investigated the inhibitory effects of sea mustard on high-fat diet-induced obesity and hyperlipidemia in Sprague-Dawley rats. Sea mustard (Undaria pinnatifida) powder, sea mustard ethanol extract, and sea mustard ethanol-extracted residue were tested. The ethanol extracted residue had the most beneficial anti-hyperlipidemic activity. Alginate in the sea mustard was considered to be the key component. The ethanol-extracted residue of sea mustard also had antioxidant activity, which may be effective in preventing hyperlipidemia by increasing the enzymatic activity of superoxide dismutase, which can remove active oxygen from the bloodstream.

Lead Biosorption by Biosorbent Materials of Marine Brown Algae U. pinnatifida, H. fusiformis, and S. fulvellum

  • Biosorbents of nonliving, dried marine brown algae Undaria pinnatifida, Hizikia fusiformis, and Sargassum fulvellum were investigated for their lead biosorption performances. As the amount of biosorbent added was increased, the lead removal by biosorbent materials increased but the lead biosorption capacities decreased. However the lead biosorption capacity by the biosorbent materials increased with increasing initial lead concentration and pH in the range of $C_o\;10\~500\;mg/L$. Among the biosorbent materials used in this study, the lead biosorption capacity in the solutions with no pH adjustment decreased in the following sequence: U. pinnatifida > H. fusiformis > S. fulvellum. Equilibrium parameters based on Langmuir and Freundlich isotherm were determined. It was found that the lead biosorption by biosorbent materials were expressed by the Langmuir isotherm better than the Freundlich isotherm.

Isolation and Characterization of Marine Bacterial Strain Degrading Fucoidan from Korean Undaria pinnatifida Sporophylls

  • In spite of an increasing interest in fucoidans as biologically active compounds, no convenient commercial sources with fucoidanase activity are yet available. A marine bacterial strain that showed confluent growth on a minimal medium containing fucoidan, prepared from Korean Undaria pinnatifida sporophylls, as the sole carbon source was isolated and identified based on a 16S rDNA sequence analysis as a strain of Sphingomonas paucimobilis, and named Sphingomonas paucimobilis PF-1. The strain depolymerized fucoidan into more than 7 distinct low-molecular-mass fucose-containing oligosaccharides, ranging from 305 to 3,749 Da. The enzyme activity was shown to be associated with the whole cell, suggesting the possibility of a surface display of the enzyme. However, a whole-cell enzyme preparation neither released the monomer L-fucose from the fucoidan nor hydrolyzed the chromogenic substrate p-nitrophenyl-${\alpha}$-L-fucoside, indicating that the enzyme may be an endo-acting fucoidanase rather than an ${\alpha}$-L-fucosidase. Therefore, this would appear to be the first report on fucoidanolytic activity by a Sphingomonas species and also the first report on the enzymatic degradation of the Korean Undaria pinnatifida sporophyll fucoidan. Moreover, this enzyme activity may be very useful for structural analyses of fucose-containing polysaccharides and the production of bioactive fucooligosaccharides.

미역(Undaria pinnatifida) 6품종간의 형질상관분석

  • 명천산, 카마이시산, 나루산, 풍남산, 기장산, 울산산 미역(Undaria pinnatifida)의 성숙 포자체는 각각 2001년 5월에 명천 배양장에서 채묘하였다. 인공 채묘와 채묘의 주요방법은 손(1993)에 따랐다. 재배미역의 번식생태학과 생산량이 우수한 형질을 규명하기 위하여 2001년 12월부터 2002 4월까지 명천산, 카마이시산, 나루산, 풍남산, 기장산, 울산산 미역(Undaria pinnatifida)에 대해서 고흥군 명천 어장과 계도 어장에서 양식실험을 실시하였다. 형태적 형질에 있어서 명천, 계도 어장에서 전장은 3월까지 급격한 성장을 보였으나 4월 이후 성장이 점점 완만해 지면서 감소하였다. 품종간에도 전장의 성장도가 달리 나타났으며 그중 가장 큰 것은 가마이시산이었고 가장 작은 것은 울산산이었다. 경장은 3월까지 지속적인 증가를 보였으며 6품종중 1품종은 증감하였다. 품종별로 경장의 변화는 달리 나타났다. 각 어장별 품종별로 줄기길이는 대부분이 월별로 증가하다 4월부터 감소하였다. 그러나 명천산은 4월까지 증가 하였다. 성실엽장은 6품종 모두 4월까지 지속적인 증가를 보였으며 특히 풍남산과 가마이시산이 두 어장에서 크게 나왔으며, 울산산이 두 어장에서 가장 작게 나타났다. 명천 어장에서 6품종의 전장과 경장의 상관계수는 카마이시산이 가장 크게 나타났고, 울산산이 가장 작게 나타났다. 계도 어장에서도 카마이시산이 가장 크게 나타났으며 울산산이 가장 작에 나타났다. 전장과 줄기길이의 상관계수는 명천어장에서는 풍남산이 가장 크게 나타났고, 울산산이 가장 작게 나타났으며, 계도 어장에서는 카마이시산이 가장 크게 나타났고 울산산이 가장 작게 나타났다. 명천 어장에서 전장과 성실엽장의 상관관계를 보면 명천산의 상관 계수가 가장 크게 나타났으며 나루토산이 가장 작게 나타났다. 계도 어장에서는 카마이시산이 가장 크게 나루토산이 가장 작게 나타났다. 명천, 계도 어장에서 전장과 각 형질간 ANOVA 분석은 1%~5% 수준에서 품종별로 모두 유의성 있게 나타났다.
