  • Ultrastructural changes in Z-line, M-line and myofilaments of bovine psoas muscle produced by leukocyte lysosomal enzymes in vitro at different pH values (pH 7.0 and 4.0), temperatures (37 and $4^{\circ}C$) and time intervals (12, 24 hours at $37^{\circ}C$ and 36, 168 hours at $4^{\circ}C$) were studied by transmission electron microscope. Muscle incubated with leukocyte lysosomal enzymes at pH 7.0 produced distinguishable degradation of Z-line, M-Line and H-zone at both temperatures but at acidic pH (pH 4.0), Z-line were very stable and myofilaments were severely disintegrated.

  • Surface ultrastructural changes in endomysial connective tissue, sarcolemma and transverse ridges of bovine psoas muscle produced by leukocyte lysosomal enzymes in vitro at different pH (pH 7.0 and 4.0), temperature (37 and $4^{\circ}C$) and time interval (12, 24 hours at $37^{\circ}C$ and 36, 168 hours at $4^{\circ}C$ were studied by scanning electron microscope. Muscle incubated with leukocyte lysosomal enzymes at pH 7.0 produced severe degradation of endomysial and sarcolemmal connective tissue and transverse ridges but at pH 4.0 endomysial and sarcolemmal structures remain moderately stable and tranverse ridges are very stable even after 24 hours incubation at $37^{\circ}C$ and 7 days incubation at $4^{\circ}C$.

Some Observations on the fine Structure of Suncus Murinus Liver (Suncus Murinus 간장의 미세구조적 관찰)

  • This study was designed to observe the ultrastructural characteristics of hepatocytes, endothelial cells, Kupffer cells and Ito cells in the liver of Suncus Murinus. The specimens were processed for electron microscopy, following the immersing in the Karnovsky fixative for 8 hrs and 1% osmium tetroxide for 2 hrs. Hepatocytes contained large number of lipid droplets and large mitochondria. Spaces between the hepatocytes were narrow and in some area were irregular in contours with long and slender microvilli of the hepatocytes. Endothelial cells may contain lysosomes as Kupffer cells do. Bile canaliculi were relatively wide and easily seen in the lobule. Bile ductules located in the space of Disse were formed by the hepatocytes and small cells with or without lipid droplets. The results suggest that the Suncus Murinus liver shows some ultrastructural characteristics of aquatic animal livers, endothelial and Kupffer cells may be the same one only in the different functional states, and the bile ductule may be partly formed by the Ito cell.

  • This study was carried out to investigate the histomorphological changes and the electrophoretic patterns of egg components, obtained from 100 of 2-year-old female catfishes (Silurus asotus). Female Korean catfishes collected in the vicinity of Chollabukdo have synchronous ovaries which discontinously ovulate eggs during the breeding season (from late May until early July). The fishes were killed by severing the spinal cord just posterior to the head after immobilization with tricaine methanesulfonate (MS 222). Especially, the light microscopic and ultrastructural changes of ooplasm and follicular membranes of oocytes, were examined by means of light and transmission electron microscopy. The size of the nucleoli and number of the yolk granules increased as the oocyte groved. Yolk granules were deposited in the oocyte as fluid Due to tile presence of large early and late maturing oocytes, their ovaries were enlarged, transparent, granular and greenish in color. As the percentages of fish in late maturing oocyte (LMO) and ripe oocyte (RO) stage increased from March to April, mean gonadosomatic index (GSI) values (19.95%) increased. Zona radiata changed from squamous into cuboid in shape in the early maturing oocyte (EMO) stage. Processes, microvilli, from the zona radiata and from the oocyte grow, and make contact with each other in the pore canals of the zona radiata during vitellogenesis, but are withdrawn as the zona radials becomes more compact and devoid of pore canals during oocyte maturation. Seasonal changes in the microscopic appearance of the ovaries were well correlated with those in both gonadosomatic index and macroscopic appearance. The main cytological changes such as increase in size of cell, nucleus, nucleolus, and increase in number of nucleoli and mitochondria demonstrated with electron microscopy in the previtellogenic oocytes of Korean catfish, provided evidences for important synthetic processes in an early preparatory phase of oocyte development. The electrophoretic pattern of major band in mature stage was much thicker (24 k, 66 k, 90-110k dalton) than that in previtellogenic phase.

Dose-dependent Ultrastructural Changes of the Odontoblasts in the Neonatal Rat after Ingestion of the Fluoride to the Pregnant Rat (음용수 불소농도에 따른 흰쥐태아 상아모세포의 미세구조적 변화)

  • The pregnant rats were given a drinking water administration of the sodium fluoride and normal saline for control animals. The sodium fluoride produced cellular changes of odontoblast with consistent response. Compare to control group, the odontoblasts that were administrated by sodium fluoride, showed significantly ultrastructural differences including large number of free ribosomes and swelled mitochondria in dose-dependent manner (300 ppm). From fine structural and morphological investigations of the changes in odontoblast, there were three distinctive structural changes: (1) destruction of the endoplasmic reticulum, (2) swelling of the mitochondria, and (3) severe cellular derangement of the endoplasmic reticulum and mitochondria. From this consecutive structural change, we observed that sodium fluoride temporarily affects the cell organelles in odontoblasts (100, 200 ppm), suggesting it is important that optimal concentration of the sodium fluoride in developing fetus of the rat.

Ultrastructural changes in cristae of lymphoblasts in acute lymphoblastic leukemia parallel alterations in biogenesis markers

  • We explored the link between mitochondrial biogenesis and mitochondrial morphology using transmission electron microscopy (TEM) in lymphoblasts of pediatric acute lymphoblastic leukemia (ALL) patients and compared these characteristics between tumors and control samples. Gene expression of mitochondrial biogenesis markers was analysed in 23 ALL patients and 18 controls and TEM for morphology analysis was done in 15 ALL patients and 9 healthy controls. The area occupied by mitochondria per cell and the cristae cross-sectional area was observed to be significantly higher in patients than in controls (p-value=0.0468 and p-value<0.0001, respectively). The mtDNA copy numbers, TFAM, POLG, and c-myc gene expression were significantly higher in ALL patients than controls (all p-values<0.01). Gene Expression of PGC-1α was higher in tumor samples. The analysis of the correlation between PGC-1α expression and morphology parameters i.e., both M/C ratio and cristae cross-sectional area revealed a positive trend (r=0.3, p=0.1). The increased area occupied by mitochondria and increased cristae area support the occurrence of cristae remodelling in ALL. These changes might reflect alterations in cristae dynamics to support the metabolic state of the cells by forming a more condensed network. Ultrastructural imaging can be useful for affirming changes occurring at a subcellular organellar level.

Evaluation by Contrast-Enhanced MR Imaging of the Lateral Border Zone in Reperfused Myocardial Infarction in a Cat Model

  • Objective: To identify and evaluate the lateral border zone by comparing the size and distribution of the abnormal signal area demonstrated by MR imaging with the infarct area revealed by pathological examination in a reperfused myocardial infarction cat model. Materials and Methods: In eight cats, the left anterior descending coronary artery was occluded for 90 minutes, and this was followed by 90 minutes of reperfusion. ECG-triggered breath-hold turbo spin-echo T2-weighted MR images were initially obtained along the short axis of the heart before the administration of contrast media. After the injection of Gadomer-17 and Gadophrin-2, contrast-enhanced T1-weighted MR images were obtained for three hours. The size of the abnormal signal area seen on each image was compared with that of the infarct area after TTC staining. To assess ultrastructural changes in the myocardium at the infarct area, lateral border zone and normal myocardium, electron microscopic examination was performed. Results: The high signal area seen on T2-weighted images and the enhanced area seen on Gadomer-17-enhanced T1WI were larger than the enhanced area on Gadophrin-2-enhanced T1WI and the infarct area revealed by TTC staining; the difference was expressed as a percentage of the size of the total left ventricle mass (T2= 39.2 %; Gadomer-17 =37.25 % vs Gadophrin-2 = 29.6 %; TTC staining = 28.2 %; p < 0.05). The ultrastructural changes seen at the lateral border zone were compatible with reversible myocardial damage. Conclusion: In a reperfused myocardial infarction cat model, the presence and size of the lateral border zone can be determined by means of Gadomer-17- and Gadophrin-2-enhanced MR imaging.

Histidine-tryptophan-ketoglutarate Versus Blood Cardioplegic Solutions: A Prospective, Myocardial Ultrastructural Study (선천성 심장기형의 수술 후 Histidine-tryptophan-ketoglutarate 심정지액과 혈성 심정지액의 전자현미경적 심근 구조의 비교 관찰)

  • Background: We performed a prospective clinical study to evaluate the ultrastructural integrity of the myocardium after using Histidine-Tryptophan-Ketoglutarate (HTK) solution in comparison with blood cardioplegic solution during congenital heart surgery. Material and Method: Twenty two patients with acyanotic heart disease, who were scheduled for elective open heart surgery, were randomized into two groups. The HTK Group (n=11) received HTK cardioplegic solution; the blood group (n=11) received conventional blood cardioplegic solution during surgery. The preoperative diagnoses included ventricular septal defect (n=9) and atrial septal defect (n=2) in each group. A small biopsy specimen was taken from the right ventricle's myocardium, and this was processed for ultrastructural examination at the end of 30 minutes of reperfusion. Semiquantitative electron microscopy was carried out 'blindly' in 4 areas per specimen and in 5 test fields per area by 'random systematic sampling' and 'point and intersection counting'. The morphology of the mitochondrial membrane and cristae were then scored. The interstitial edema of the myocardium was also graded. Result: The semiquantitative score of the mitochondrial morphology was $19.65{\pm}4.75$ in the blood group and $25.25{\pm}5.85$ in the HTK group (p=0.03). 6 patients (54.5%) in the blood group and 3 patients (27.3%) in the HTK group were grade 3 or more for the interstitial edema of the myocardium. Conclusion: The ultrastructural integrity was preserved even better with HTK solution than with conventional blood cardioplegic solution.

Ultrastructural Study on the Cleistothecium Development in Aspergillus nidulans

  • Cleistothecial development in Aspergillus nidulans(teleomorph, Emericella nidulans) was examined with the transmission electron microscopy. Cleistothecial initial was a small coiled lump of cells, ca. 6 ${\mu}m$ in diameter, which was consisted of a slightly swollen core with a short "tail" hypha. Initials were wrapped with a loose layer of hyphae. Core cells of cleistothecial initials were broad and multinucleated at first, then formed dikaryotic ascogenous cells, followed by post-meiotic tetra-nucleate or octa-nucleate protoasci and finally mature ascospores. Croziers were formed early during cleistothecium development. The peridial layer of mature cleistothecia was derived from the wrapping hyphae which originally invested the young cleistothecium. Completion of peridial layers development was associated with the depositing of a non-enzyme reactive material around peridial cells. $H\ddot{u}lle$ cell formation during the cleistothecial development appeared to be somewhat coordinated with the developmental stages of cleistothecium.

Fixation of Cellular Ultrastructure by the Microwave Irradiation (마이크로파 조사에 의한 세포 미세구조의 고정 효과)

  • The microwave fixator has recently been introduced in morphological research. The present study was carried out to investigate the ultrastructural effects of microwave fixation of rat brain. kidney, liver and skeletal muscle tissues. The results are as follows: In the case of microwave fixed cerebrum. the cytoplasmic processes of neurons and the various membranous organelles such as nuclear envelope, mitochondria, rough endoplasmic reticulum and Golgi apparatus were well preserved, The myelin sheath wrapping neuronal axon was prominent. Microwave fixed hepatocytes showed the microvilli on the free surface of bile canaliculus, the evident nucleolar components, and typical organelles. In nephron, ultrastructures of glomerulus and Bowman's capsule were preserved, and also tubular wall were structurally observed. Among the skeletal muscle cells, plentiful collagen fibers were appeared, myofibrils and mitochondria were typically observed. In conclusion, the microwave fixation procedures result in an good preservation of the tissues and would be time- and reagent-saving.
