Targeting of COX-2 Expression by Recombinant Adenovirus shRNA Attenuates the Malignant Biological Behavior of Breast Cancer Cells

  • Background: Cyclooxygenase-2 (COX-2), considered to have tumor-promoting potential, is highly expressed in a variety of tumors, including breast cancer. Since the functions and action mechanisms of COX-2 in breast cancer have not been fully elucidated, in the present study, the effects of target inhibiting COX-2 with recombinant adenovirus Ad-COX-2-shRNA on malignant biological behavior were investigated in representative cell lines. Materials and Methods: Breast cancer MDA-MB-231 and MCF-7 cells were transfected with Ad-COX-2-shRNA and COX-2 expression was tested by RT-PCR and Western blotting. Changes in proliferation, apoptosis and invasion of breast cancer cells were detected with various assays including MTT, colony forming, flowcytometry and Transwell invasion tests. The expression of related proteins involved in the cell cycle, apoptosis, invasion and signaling pathways was assessed by Western blotting. Results: COX-2 expression was significantly reduced in both breast cancer cell lines infected with Ad-COX-2-shRNA, with obvious inhibition of proliferation, colony forming rate, G2/M phase passage and invasion, as well as induction of apoptosis, in MDA-MB-231 and MCF-7 cells, respectively. At the same time, proteins related to the cell cycle, anti-apoptosis and invasion were significantly downregulated. In addition, c-myc expression and phosphorylation activation of Wnt/${\beta}$-catenin and p38MAPK pathways were reduced by the Ad-COX-2-shRNA. Conclusions: COX-2 expression is associated with proliferation, apoptosis and invasion of breast cancer cells, and its mechanisms of action involve regulating expression of c-myc through the p38MAPK and Wnt/${\beta}$-catenin pathways.

Microbiological cleaning and disinfection efficacy of a three-stage ultrasonic processing protocol for CAD-CAM implant abutments

  • PURPOSE. Computer-aided design and manufacturing (CAD-CAM) of implant abutments has been shown to result in surface contamination from site-specific milling and fabrication processes. If not removed, these contaminants can have a potentially adverse effect and may trigger inflammatory responses of the peri-implant tissues. The aim of the present study was to evaluate the bacterial disinfection and cleaning efficacy of ultrasonic reprocessing in approved disinfectants to reduce the microbial load of CAD-CAM abutments. MATERIALS AND METHODS. Four different types of custom implant abutments (total N = 32) with eight specimens in each test group (type I to IV) were CAD-CAM manufactured. In two separate contamination experiments, specimens were contaminated with heparinized sheep blood alone and with heparinized sheep blood and the test bacterium Enterococcus faecium. Abutments in the test group were processed according to a three-stage ultrasonic protocol and assessed qualitatively and quantitatively by determination of residual protein. Ultrasonicated specimens contaminated with sheep blood and E. faecium were additionally eluted and the dilutions were incubated on agar plates for seven days. The determined bacterial counts were expressed as colony-forming units (CFU). RESULTS. Ultrasonic reprocessing resulted in a substantial decrease in residual bacterial protein to less than 80 ㎍ and a reduction in microbiota of more than 7 log levels of CFU for all abutment types, exceeding the effect required for disinfection. CONCLUSION. A three-stage ultrasonic cleaning and disinfection protocol results in effective bacterial decontamination. The procedure is reproducible and complies with the standardized reprocessing and disinfection specifications for one- or two-piece CAD-CAM implant abutments.

  • A numerical simulation is carried out on the directivity synthesis of ultrasonic transducers by point source array. Gauss elimination method is practiced by means of a directive method to realize the desired directivity. Desired directivity is chosen to be that of a directivity of line source, a beam width and a direction arbitrary specified. On the numerical result, Gauss elimination method is showed high speed ca- lculative simulation and stability of system more than iterative method(LMS, DFP). Numerical simulations are carried out by PC(CPU:80486 DX2, RAM 16Mbyte).

  • Ceramic tapes were fabricated by ultrasonically spraying slurries of monodispersed spherical and alumina powders. Effects of slurry compositions on tape forming were investigated. A relatively fast rate of solvent evaporation caused pores and cracks to be formed. A good chemical affinity between solvent and binder gave rise to binder separation resulting in inhomogeneous distribution of binder. Defect-free silica tapes with uniform distribution of particle packing and the binder were obtained from the solvent having a low chemical affinity and a slow evaporation rate and containing appropriate amounts of the binder and the plasticizer. Tape thickness could be controlled by adjusting solids loadings and slurry feed rates. It was possible to fabricate a tape in 15 $\mu\textrm{m}$ thickness from 7 vol% alumina slurry.

A plane strain punch stretching test for evaluating stamping formability (평면변형장출실험을 이용한 스탬핑 성형성 평가)

  • Plane strain punch stretching test (PSST) was developed to evaluate stamping formability of sheet materials. In this test, the rectangular specimen of sheet material is uniformly stretched up to fracture by raising a specially designed punch to certainly assure plane strain stretching deformation along the longitudinal direction of the specimen. The stamping formability was evaluated by limit punch height(LPH) in plane strain punch stretching test compared to limit dome height(LDH) in hemispherical punch stretching test. LPH-value in PSST well ranks the stamping formability of various material and correlates with press performance. Moreover by using ultrasonic thickness gauge the plane strain intercept-limit plane strain(FLCo)-in forming limit curve can be accurately determined from thickness measurement around the fracture area. The FLCo derived from thickness measurement well correlates with the results from circle grid analysis for the deformed circle grid marked on the surface of the specimen.

  • Lab-on-a-disc is a circular disc shape of cartridge that can be used for blood-based liquid biopsy to diagnose an early stage of cancer. Currently, liquid biopsies are regarded as a time-consuming process, and require sophisticated skills to precisely separate cell-free DNA (cfDNA) and circulating tumor cells (CTCs) floating in the bloodstream for accurate diagnosis. However, by applying the lab-on-a-disc to liquid biopsy, the entire process can be operated automatically. To do so, the lab-on-a-disc should be designed to prevent blood leakage during the centrifugation, transport, and dilution of blood inside the lab-on-a-disc in the process of liquid biopsy. In this study, the main components of lab-on-a-disc for liquid biopsy are fabricated by injection molding for mass production, and ultrasonic welding is employed to ensure the bonding strength between the components. To guarantee accurate ultrasonic welding, the flatness of the components is optimized numerically by using the response surface methodology with four main injection molding processing parameters, including the mold & resin temperatures, the injection speed, and the packing pressure. The 27 times finite element analyses using Moldflow® reveal that the injection time and the packing pressure are the critical factors affecting the flatness of the components with an optimal set of values for all four processing parameters. To further improve the flatness of the lab-on-a-disc components for stable mass production, a quarter-disc shape of lab-on-a-disc with a radius of 75 mm is used instead of a full circular shape of the disc, and this significantly decreases the standard deviation of flatness to 30% due to the reduced overall length of the injection molded components by one-half. Moreover, it is also beneficial to use a quarter disc shape to manage the deviation of flatness under 3 sigma limits.

The effect of chlorhexidine on reduction of viable organisms in aerosol produced by ultrasonic scaler (클로르헥시딘이 초음파 치석제거기에 의해 발생된 에어로졸 내 세균 수에 미치는 영향)

  • Purpose: The aerosol generated by ultrasonic scaler can contain bacteria or virus which can penetrate into body through respiratory systems of dentists, dental hygienist or patients. The aim of this study is to evaluate the effect of chlorhexidine digluconate as preoperative mouthrinse or lavage for ultrasonic scaler on the reduction of viable organisms in aerosol produced during periodontal treatment using ultrasonic scaler. Methods: 30 patients with moderate chronic periodontitis were included and divided into 3 groups: Control (no preoperative mouthrinse and tap water as lavage), CHG (preoperative mouthrinse with 0.1% chlorhexidine digluconate and tap water as lavage), CHL (no preoperative mouthrinse and 0.1% chlorhexidine digluconate as lavage). Each patient received scaling or subgingival curettage for 30 min. In CHG group, mouthrinse with chlorhexidine digluconate was performed for 1 min. before treatment. Before, during and after scaling or subgingival curettage, air sampling was performed for 7 min. each (1000 L/7 min.) with trypticase-soy agar plate. Agar plates were incubated in $37^{\circ}C$ aerobically. The numbers of colony-forming units (CFU) were counted and compared. Results: The numbers of CFUs of the samples obtained during treatment were $97{\pm}14.0$ in control, $73.1{\pm}14.9$ in CHG group and $44.5{\pm}9.0$ in CHL group. The difference among the 3 groups was determined to be statistically significant (one-way ANOVA with Bonferroni's correction, p-value: 0.0003). In contrast, the numbers of CFU of samples obtained before and after treatment were not significantly different among the groups. Conclusions: Chlorhexidine digluconate used as preoperative mouthrinse or lavage for ultrasonic scaler can reduce the microorganisms in aerosol produced during periodontal treatment using ultrasonic scaler. Less number of microorganisms were detected when chlorhexidine was used as lavage for ultrasonic scaler.

Preparation and Electrical Conductivity of Scandia Stabilized Zirconia by using Ultrasonic Spray Pyrolysis (초음파 분무 열분해법을 이용한 스칸디아 안정화 지르니코니아의 제조와 전기 전도도)

  • Scandia stabilized zirconia (ScSZ) is adapted for electrolyte material of solid oxide fuel cell (SOFC) because of its high ionic conductivity and chemical stability. ScMnSZ1 powder having a composition of $((ZrO_2)_{0.89}(Sc_2O_3)_{0.1}(MnO_2)_{0.01})$ is synthesized by ultrasonic spray pyrolysis (USP) method. Porous ScMnSZ1 powder is obtained by using a pore forming agent. Microstructure and morphology, particle size distribution of porous powder synthesized with 3wt% pore forming agent are investigated. Sintered ScMnSZ1 sample with ground fine powder are also investigated their microstructure and electrical conductivity. The electrical conductivity of sintered ScMnSZ1 samples with ground fine powder was 0.082 S/cm, 0.127 S/cm and 0.249 S/cm at $750^{\circ}C$, $800^{\circ}C$ and $900^{\circ}C$, respectively.

Development of Ceramic Roll Materials for Food Grinding Processing and Evaluation of Mechanical Behavior (식품분쇄용 세라믹 롤 재료 개발과 기계적 특성평가)

  • In order to prevent the possibility of mixing of metal powder during food grinding processing with the metal roll mill this study was conducted to develope the materials of ceramics roll as a substitute of gray cast iron mill. Since the ceramics is brittle material and can be broken easily by a crack, it was needed to develope engineering ceramics roll materials with high elastic modulus and fracture toughness. Adding 0∼50 wt% Al$_2$O$_3$as densification additives to porcelain body material and forming the ceramics an different condition, mechanical properties were evaluated. The material structure’s densification process was analyzed by SEM and XRD. The evaluation of the mechanical properties of ceramics roll materials were compared and analyzed by non-destructive test using Young’s modulus and destructive test using 3-point bending strength and fracture toughness. The results showed several correlative results. Porcelain body material with 40 wt% Al$_2$O$_3$content heated at 1,200$\^{C}$ for 5h was high bulk density of 2.77, Young’s modulus of 118.4Gpa, 3-point bending strength of 137 MPa and fracture toughness of 2.88 MPa$.$m$\^$$\sfrac{1}{2}$/ . After analyzing the relationship between non-destructive test and destructive test, the coefficient of determination was more than 0.9. Therefore, the evaluation of non-destructive test by ultrasonic was turned out to be feasible in evaluating the mechanical properties of ceramics.

  • In chemistry, the study of sonochemistry is concerned with understanding the effect of sonic waves and wave properties on chemical systems. In the area of chemical kinetics, it has been observed that ultrasound can greatly enhance chemical reactivity in a number of systems by as much as a million-fold. Nano-technology is a super microscopic technology in which structures of 100 nanometers or smaller can be investigated. This technology has been used to develop $TiO_2$ materials and $TiO_2$ devices of that size. Thus far, electrochemistry methods and photochemistry methods have generally been used to create $TiO_2$ nano-size particles. However, these methods are complicated and create pollutants as a by-product. In the present study, nano-scale silver particles (5 nm) were prepared in a sonochemistry method. Sonochemistry deals with mechanical energy that is provided by the collapse of cavitation bubbles that form in solutions during exposure to ultrasound. $TiO_2$ powders 25 nm in size doped with Ag were formed using an ultrasonic sound technique. The experimental results showed the high possibility of removing pollution through the action of a photocatalyst. This powder synthesis technique can be considered as an environmentally friendly powder-forming processing owing to its energy saving characteristics.
