The Effectiveness of Ulmus Davidiana Planch Herbal Acupuncture to Inhibit $NF-{\kappa}B$ Activation on Type II Collagen-induced Arthritis in Mice (유근피(楡根皮) 약침의 $NF-{\kappa}B$ 활성 억제능이 생쥐의 Type II Collagen 유발 관절염에 미치는 영향)

  • Objectives : The purpose of this study is to investigate the effectiveness of Ulmus davidiana Planch herbal acupuncture(UA) to inhibit nuclear $factor(NF)-{\kappa}B$ activation on type II collagen-induced arthritis (CIA) in mice. Methods : Using an in vitro test, the synoviocytes picked out from the experimental CIA mice were subcultured. The synoviocyte cells were treated with phorbol-12-myristate-13-acetate(PMA) for 1 hour prior to the addition of indicated concentrations($0.4\;-\;1.0mg/m{\ell}$) of UA, and the cells were further incubated for 24 hours. The in vivo test, $NF-{\kappa}B$ p65, inducible nitric oxide synthase(iNOS), cyclooxygenase-2(COX-2), vascular cell adhesion molecule(VCAM)-1 production and apoptosis was observed by immunohistochemical staining. Results : The PMA-induced $I{\kappa}B$ kinase(IKK), iNOS and COX-2 mRNA expression were dose-dependently decreased in UA treated synoviocytes. Using the in vivo test, the number of eosinophils in mice treated with UA noticeably decreased in the the CIA group(P<0.05 using student t test). In mice treated with UA, there was less cartilage erosion. less bone destruction, mild synovial hyperplasia, mild fibrosis, and mild angiogenesis with less MIP-2 production. By immunohistochemical staining, suppression of $NF-{\kappa}B$ p65, iNOS production, inhibition of COX-2 production, inhibition of VCAM-1 production and inducing apoptosis were observed. Conclusions : These results suggest that UA might be applicable to the therapy of RA to suppress $NF-{\kappa}B$ activation.

  • The effect of the extract of Ulmus davidiana root on the activity of enzymes related to the removal of reactive oxygen species was investigated in the B6C3F1 mouse kidney. B6C3F1 mice were divided into five groups and fed for 20 weeks. Reduced xanthine of oxidase activity was observed in groups 4 (group fed with U. davidiana extract after N,N-diethylnitrosamine (DEN) treatment and 5 (group fed with U. davidiana extract from the beginning of DEN treatment) compared to group 2 (group treated with DEN). The level of Mn-superoxidase dismutase tended to increase in the groups after DEN treatment. In group 5, the catalase activity increased and the other groups exhibited an unchanged or slightly decreased level of enzyme. Similar effects were found far glutathione peroxidase. A lower degree of TBARS (thiobarbituric acid reactive substance) formation was estimated in groups 4 and 5, compared to that in DEN treated group 2.

Change in the Seed Characteristics and Germination Properties of Ulmus davidiana var. Japonica According to Seed Collection Time (채취 시기에 따른 느릅나무의 종자 형질 및 발아 특성 변화)

  • This study was conducted to examine the changes of seed characteristics and germination properties and to determine the most suitable collection time in Ulmus davidiana var. japonica. Seeds were collected six times with the interval of seven days after April 21, 2005 at Jeong-seon. There were differences in seed characteristics and germination properties among seed collection times. The sizes of seed and fruit increased with the collection time, but seed moisture contents decreased with the collection time after increasing until May 5. The 1,000 seeds weight decreased with the collection time after increasing until May 12. Collection time showed high positive correlations (r=0.790, p<0.01) with seed size and negative correlations (r=-0.919, p<0.01) with moisture contents. Percent germination (PG) presented the highest value in seeds collected at May 5, and decreased with the collection time. Mean germination time(MGT), germination speed(GS), and germination performance index(GPI) also showed the highest values in seeds collected at May 5. PG, MGT, and GS had correlations with other seed characteristics except the 1,000 seed weight. The relative growth rate of height and root collar diameter of seedlings presented the highest values in seedling germinated from seed collected at May 5, and seedlings germinated from seeds collected at May 5 and 12 showed the highest values in chlorophyll contents. Consequently it will be possible to product a number of seedlings when the seeds of U. davidiana var. japonica are collected just before drying morphologically.

Effect of Euonymus alatus and Ulmus clavidiana var japonica on the immune system (화살나무 및 느릅나무 추출물이 면역계세포의 활성에 미치는 영향)

  • We have previously shown that crude water extract of Euonymus alatus (EA) had strong prophylactic effect against chemically induced-and tumor cell implanted-cancer, and that the mechanisms responsible for its antitumor effects were due to nonspecific enhancement of the NK cell activities and the cell mediated immunity. However, it was unknown that any components of crude extract did work so, since it consisted of several components. In this paper, we fractionated the crude watar EA-extract into several fraction such as hexane-, ethylether-, ethyl acetate-, n-butanol- and water soluble-fraction, and screened the immune regulating activities of each fraction by the evaluation of lymphokine production and activated lymphocyte proliferation. As a result of the component fraction of EA-extract, it was found that n-butanol fraction was a potent immunostimulator, and the remained water soluble fraction also contained some stimulator, But, other fraction did not showed any remarkable effect. It is therefore suggested that EA-glycosides in n-butanol fraction may be new one of the potent biological response modifiers. The present study was also undertaken in an efforts to investigate the effects of elm-bark(EB, Ulmus clavidiana var japonica), which has been used for curing ulcer and inflammation as a folk medicine without any kind of experimental evidence to support this, on the cellular- and humoral-immune responses, lymphocyte function and NK cell activities in mice. Regardless of time and duration of EB-treatment, Arthus reaction and antibody response to SRBC were not modified by EB, but delayed hypersensitivity to SRBC was significantly enhanced only when EB was treated prior to SRBC-sensitization. EB slightly inhibited the proliferation responses of splenocytes to PHA-stimulation, but it significantly augmented the responses of these cells to S aureus Cowan 1 and Con A-activation, and these effects were manifested only when EB was added at culture initiation. EB did not influence Ig secretion of spleen cells but it significantly augmented the Con A-induced 1L 2 and MIF production of splenocytes. NK cell activities of splenocytes were markedly riled when effector cells were pretreated with EB and this augmentation was dine to the increase of binding affinity of effector cells to target cells and the target cell lytic activities of effector cells. These results led to the conclusion that EB triggers increase of cellular immune responses, such as delayed hypersensitivitiy, lymphokine production and NK cell activities. Also these results suggested that EB contains potent immune stimulants, which may provide the rational basis for their therapeutic use as one of the new biological response modifiers.

Effect of Ulmus davidiana Planch herbal acupuncture Solution on proinflammatory cytokine IL-$1{\beta}$ and $TF-{\alpha}$ a production in collagen-induced arthritis of rats

  • Objective: We have evaluated UDHA into the joint for its effectiveness on immune responses to CII in the rat CIA. In an attempt to gain further insight into the mode of action of UDHA, we also investigated the effects of UDHA on the incidence and development of arthritis in rat CIA with 2 different regimens: (1) started prior to a primary immunization, (2) started on the day of a primary immunization. Methods : Male rats were immunized with an emulsion of $200\;{\mu}g/100g$ of CII and complete Freund's adjuvant (CFA). The rats were then given intraperitoneal(i.p) stimulation of Ulmus davidiana Planch herbal acupuncture(UDHA) or saline during the experiment. Lymph node cells were obtained from rats 14 days after immunization and cultured in vitro with CII. When compared with rats treated with saline as control, UDHA at doses of more than $20\;{\mu}g/100\;g$ rat once a day for 7 days inhibited the ability of inguinal lymph node cells to produce T cell cytokines interleukin-$1{\beta}$, tumor necrosis $factor-{\alpha}$ $(TNF-{\alpha})$. When rats were injected intraperitoneally with SRBC, hemaglutination titers in UD-treated and control rats did not differ significantly when low doses of UD was given to rats. However, i.p injection of UD at doses of more than $10\;{\mu}g/100\;g/day$ for 7 days slightly suppressed antibody production. Results : The present results show that treatment with UDHA can inhibit the onset and development of arthritis and the immune responses to collagen. Conclusion: Therapeutic i.p injection with UD affect the clinical course of the disease and the immune responses to CII.

  • In this study, polysaccharide-poly(vinyl alcohol) (PVA) hydrogels were prepared by using ${\gamma}$-ray and evaluated for potential application as an anti-inflammation patch. Ulmus davidiana var. japonica (UD), one of polysaccharides has been particularly used as an oriental remedy for the treatment of inflammation and ulcers. PVA as a biocompatible polymer and glycerin as a moisturizer were blended with the UD, and its hydrogels were prepared by radiation crosslinking. Characterizations for UD hydrogels were performed by using cytotoxicity assay, antioxidant activity test, and physicochemical test such as gel fraction ratio, and swelling behavior. The results showed that these UD hydrogels had excellent physical properties, anti-inflammation activity, and non-cytotoxicity on the cells. Therefore, these polysaccharide based-UD hydrogels can be effectively used as an inflammation patch.

Effects of Nitrogen Fertilization on Growth of Populus sibirica and Ulmus pumila Seedlings and Soil Properties in a Semi-Arid Area, Mongolia (몽골 반건조지에서 질소 시비가 백양나무와 비술나무 묘목의 생장 및 토양 특성에 미치는 영향)

  • This study was conducted to investigate the effects of different levels and types of nitrogen fertilizer on seedlings and soil chemical properties in a semi-arid area, Mongolia. 2-year-old Populus sibirica and 4-year-old Ulmus pumila seedlings were planted in May 2014. Six treatments with three levels of nitrogen (low-level: urea $5g\;tree^{-1}$; medium-level: urea $15g\;tree^{-1}$, ammonium sulfate $33g\;tree^{-1}$, urea $15g\;tree^{-1}$ with potassium phosphate $10g\;tree^{-1}$; high-level: urea $30g\;tree^{-1}$) were applied and for the medium-level of nitrogen, different types of fertilizer were treated. Survival rate, root collar diameter (RCD) growth rate, leaf nitrogen concentration of seedlings, and soil chemical properties were determined in August 2014. The seedling survival rate of both species decreased as the level of nitrogen increased. This result can be explained by water stress caused by nitrogen fertilization in arid regions. The RCD growth rate of P. sibirica was significantly decreased by the treatment of high-level of nitrogen due to excessive nitrogen fertilization, and was increased by the treatment of ammonium sulfate due to sulfur which might promote nitrogen uptake. The leaf nitrogen concentration of P. sibirica did not change by the treatment of low-level of nitrogen, and was increased by the treatment of medium-level of nitrogen. There were no significant differences in the RCD growth rate and the leaf nitrogen concentration of U. pumila among the six treatments. None of soil chemical properties was affected by nitrogen fertilization. Overall, the low-level of nitrogen showed no effect on seedlings and soil chemical properties, except on survival rate of U. pumila and the high-level of nitrogen was considered excessive fertilization. Continuous monitoring of medium-level nitrogen fertilization including the ammonium sulfate, which increased early growth of seedlings, would be needed to elucidate the effect of fertilization on seedling growth and soil properties in a semi-arid region.

Validation of Analytical Methods for Unriped Rubus crataegifolius, Gardenia jasminoides and Ulmus macrocarpa Marker Compounds for Standardization of Natural Complex LS-RUG-com Preparation as Functional Ingredient (천연복합소재 LS-RUG-com의 기능성원료 표준화를 위한 산딸기 미숙과, 치자 및 유백피 지표성분의 분석법 검증)

  • LS-RUG-com preparation in a complex extract from mixture of three natural plants. Rubus crataegifolius/unriped, Gardenia jasminoides and Ulmus marcrocarpa that have been widely used in traditional functional health food. This study was conducted to establish the HPLC analysis methods that can be used to establish quantitative analysis of R. crataegifolius, G. jasminoides and U. macrocarpa for standardization of LS-RUG-com preparations. HPLC analysis methods for simultaneous determination of ellagic acid and geniposide and single determination of catechin-7-O-β-D-apiofuranoside were established for the quality control of natural plants complex (LS-RUG-com). Validation of HPLC analysis were performed by checking specificity, accuracy, precision, limit of detection and quantitation, and linearity following ICH (International Council for Harmonisation) guideline. As the result of quantitative analysis, the contents of ellagic acid, geniposide and catechin-7-O-β-D-apiofuranoside in each plant extracts were 11.2 mg/g (ellagic acid) and 72 mg/g (geniposide) and 10.2 mg/g (catechin-7-O-β-D-apiofuranoside). The contents of ellagic acid, geniposide and catechin-7-O-β-D-apiofuranoside in LS-RUG-com were 4.62~6.82 mg/g (ellagic acid), 19.2~28.8 mg/g (geniposide) and 1.36~2.04 mg/g (catechin-7-O-β-D-apiofuranoside) respectively.
