Effect of Ulmi radicis Cortex Extract on Systemic and Local Anaphaylaxis in Rats (유근피(楡根皮)가 전신적(全身的) 및 국소적(局所的) 아나필락시스에 미치는 효과(效果))

  • Ulmi radicis cortex is a herb medicine which has been used for the treatment of such allergic disease as urticaria, allergic rhinitis and athma. To assess the contribution of an aqueous extract of Ulmi radicis cortex(URC) in systemic anaphylaxis, we used compound 48/80 as a fatal anaphylaxis inducer in rats. URC inhibited anaphylactic shock 100% with a dose of 1.0 mg/g body weight (BW) 1 hr before injection of compound 48/80. URC significantly inhibited serum histamine levels induced by compound 48/80. URC (1.0 mg/g BW) also inhibited to 79.1% passive cutaneous anaphylaxis activated by anti-dinitrophenyl (DNP) IgE. URC dose-dependently inhibited the histamine release from the rat peritoneal mast cells (RPMC) by compound 48/80. Moreover, URC had a significant inhibitory effect on anti-DNP IgE-induced histamine release or tumor necrosis $factor-{\alpha}$ production from RPMC. The level of cAMP in RPMC, when URC was added, significantly increased compared with that of normal control. These results indicate that URC may possess strong antianaphylactic effect.

Effects of Forsythiae Fructus, Ulmi Cortex and JwaGwiEum in Allergic Rhinitis Induced by Ovalbumin in Mice (난알부민으로 유발된 생쥐의 알레르기 비염에 연교, 유근피, 좌귀음이 미치는 영향)

  • Objectives In the past, studies have been shown that Forsythiae Fructus (FF), Ulmi Cortex (UC) and JwaGwiEum (JGE) are effective in treating allergic reactions. However, no report shows the difference in effects of FF, UC and JGE in allergic rhinitis (AR). Therefore, the purpose of this study was to differentiate the effects of FF, UC and JGE in AR induced by ovalbumin (OVA) in mice. Methods From this experiment, the effects of FF, UC and JGE were several - changes in body weight, hematologic changes such as WBC, RBC, Hb and PLT counts, immunological changes such as levels of histamine, IgE, IL-4 and expressions of iNOS & COX-2 mRNA. Moreover, histological change of nasal mucosa was also investigated. Results FF administration group and JGE administration group significantly inhibited level of IgE compared to the control group. Also, FF administration group, UC administration group and JGE administration group inhibited IL-4 and expressions of iNOS & COX-2 mRNA. In histological assessment, FF administration group, UC administration group and JGE administration group showed reduced amount of subepithelial edema, desquamated mucosa and hyperplasia of basement membrane from OVA.

Review on the medicinal origin of Ulmi Semen Pasta (무이(蕪荑)의 약용 연원에 관한 고찰)

  • Objective : Mu-yi (蕪荑, Ulmi Semen Pasta) is the fermented product of the seeds of Ulmus macrocarpa Hance (Ulmaceae) or other species of Ulmus genus mixed with adjuvant materials. The medicinal origin and preparing method of Ulmi Semen Pasta are different from those in the past literature. Therefore, medicinal origin of Ulmi Semen Pasta and its preparation method were investigated by literature study. Methods : The classic and contemporary literatures from Korea, China, and Japan regarding to Ulmi Semem Pasta were searched to find the time-based changes of Ulmi Semen Pasta preparation and its origin. Results : The fruit or seed of U. macrocarpa has been medicinally used as Mu-yi (蕪荑) or Mu-yi-in (蕪荑仁) respectively in Korea, China, and Japan until early in 20th century. However, from the middle of the 20th century, the fermentation was performed in medicinal preparation of the fruit or seed of U. macrocarpa mixed with various adjuvant materials such as clay, Chrysanthmi Flos, Ulmi Cortex, etc. The roles or evidences of those adjuvant materials have not been clearly explained, which could not guarantee the consistent quality of Ulmi Semen Pasta. Conclusion : Current preparation method of Ulmi Semen Pasta have challenges in terms of quality maintenance, standardization, and marketing. Therefore, it is suggested that apparent evidence of current preparation method should be established or intact botanical part (seed or fruit) could be used as medicinal part.

Studies on the Apoptosis-Inducing Effect of Ulmi Pumilae Cortex on Human Leukemia HL-60 Cells

  • The antiproliferative effect of the water extract of the branch and root bark of Ulmi Pumilae Cortex(WEUPC) was investigated on the p53-negative human leukemia cell line (HL-60). A dose- and time-dependent inhibition of cell growth was observed; this effect appears to be due to induction of apoptosis. Involvement of oxidative stress is indicated by a dose-dependent increase in intracellular reactive oxygen species levels. In addition. anti-apoptic effect was observed in the cells simultaneously treated with WEUPC and the anti-oxidant N-acetylcysteine. WEUPC did not affect the anti-apoptotic Bcl-2 and the pro-apoptotic Bax, whereas p21/sup WAF1/CIPl/ was enhanced in a dose- and time-dependent fashion; this effect was partially inhibited by N-acetylcysteine. The increase in p21/sup WAF1/CIPl/ was accompanied by a parallel accumulation of cells in the G1 phase of the cycle. These results suggest that the p53-independent induction of p21/sup WAF1/CIP/ and the induction of apoptosis may mediate the anti proliferative effect of WEUPC at least in this study; on the basis of this observation, WEUPC could be proposed as an useful adjunct to the treatment of p53-deficient tumors, which are often refractory to standard chemotherapy.

Effects of Ulmi Pumilae Cortex on AGS Gastric Cancer Cells

  • Objective: Ulmi Pumilae Cortex (UPC) is a deciduous tree with uneven pinnate leaves and is classified as a subfamily of Ulmuceae and contains many pharmacologically active constituents. The aim of this study was to investigate the effects of UPC on the growth and survival of AGS cells, the most common human gastric adenocarcinoma cell lines. Methods: The AGS cells were treated with varying concentrations of UPC. Analyses of the sub G1, caspase-3 activity, and mitochondrial depolarization were conducted to determine whether AGS cell death occured by apoptosis. Furthermore, to identify the role of the transient receptor potential melastatin (TRPM) 7 channels in AGS cell growth and survival, we used human embryonic kidney (HEK) 293 cells overexpressed with TRPM7 channels. Results: The addition of UPC to a culture medium inhibited AGS cell growth and survival. Experimental results showed that the sub G1, caspase-3 activity, and mitochondrial depolarization were increased. Furthermore, TRPM7 channel overexpression in HEK 293 cells exacerbated UPC-induced cell death. Conclusion: These findings indicate that UPC inhibits the growth and survival of gastric cancer cells due to a blockade of the TRPM7 channel activity. Therefore, UPC is a potential drug for treatment of gastric cancer, and TRPM7 channels may play an important role in survival in cases of gastric cancer.

Effects of Cortex Ulmi pumilae on Human HaCaT keratinocyte and its antioxidant and anti-cancer effect (유백피(楡白皮) 추출물이 인간의 피부 세포 재생 및 악성 흑색종 세포에 미치는 영향)

  • Objective : Cortex of Ulmi pumilae(CUP) has been used to treat several diseases including boil, swelling, and scabies etc. Recently, CUP was known to have wrinkle care and whitening actions. But, It's exact mechanisms are unclear. Methods : The present study was designed to investigate effects of CUP on Human HaCaT keratinocyte and malignant melanoma cells such as SK-MEL-2 and B16F10 in terms of cell viabilities, proliferations, DPPH free radical scavenging activities, oxygen free radical productions and inhibitory action on elastase activities. Results : CUP accelerated proliferation of HaCaT keratinocytes in the lower concentration. CUP also prevented cell death of HaCaT induced by Hydrogen peroxide, which products oxygen free radicals. On the contrary, CUP did not affect proliferations of SK-MEL-2 or B16F10. Futhermore, CUP showed inhibitory action against SK-MEL-2 proliferation at the concentration of $500{\mu}g/m{\ell}$ In addition, CUP was shown to have DPPH free radical scavenging activities and also have inhibitory effects on elastase activities too. On the fluorescent examinations, the present author knows that CUP elevated production levels of oxygen free radicals in malignant melanoma cell, SK-MEL-2. Conclusions : These results suggest that CUP has possibilities of usage for functional cosmetics which have wrinkle care and whitening activities and related mechanisms are involved in inhibition of elastase action and acceleration of oxidative stress in melanoma cell.

Antioxidant, anti-inflammatory, antibacterial and ovoprotective effects of mixture of Ulmi cortex and Smilacis rhizoma extracts (유백피, 토복령 추출물 혼합물의 항산화, 항염, 항균 및 난소세포 보호효과)

  • Objectives : US extract is a mixture of each extract of Ulmi cortex and Smilacis rhizoma. In this study, we investigated the antioxidant, anti-inflammatory, antibacterial, and ovoprotective effects of US extract in in vitro model to identify potential candidates for improving female reproductive function. Methods : The antioxidant activity of US extract was measured using 1,1-diphenyl- 2-picrylhydrazyl free radical and superoxide anion radical scavenging assays. The anti-inflammatory effect of US extract on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells were determined with a nitric oxide (NO) assay, enzyme linked immunosorbent assays, and western blots analysis. The antibacterial activity of US extract against vaginitis infection microorganisms were determined with disc diffusion and minimum inhibitory concentration assays. The ovoprotective effect of US extract on 4-vinylcyclohexene diepoxide (VCD)-induced ovotoxicity in CHO-K1 cells were evaluated with a cell viability assay. Result : US extract showed good antioxidant capacity and inhibited LPS-induced NO production as well as iNOS and COX-2 expression and secretion of pro-inflammatory cytokine IL-6 without affecting the cell viability. It showed significant clear zones for Staphylococcus aureus and Candida albicans but did not indicate the clear zones for Escherichia coli and Enterococcus faecium. VCD-induced ovotoxicity in CHO-K1 cells was significantly reduced by US extract pre-treatment. Conclusions : These results demonstrate that US extract has antioxidant activity, anti-inflammatory effects on the LPS-stimulated macrophages, antibacterial activity against vaginitis infection microorganisms, and protective effects on the ovarian cells against VCD-induced ovotoxicity. These findings suggest that the US extract can be used as new prescriptions, supplements, functional foods, and cosmetics for improving female reproductive function.

Ulmi Cortex Prevents Cisplatin-Induced Apoptosis in Mice (시스플라틴에 의한 세포고사에서 유근피(楡根皮)의 효과)

  • Objectives : The purpose of this study is to investigate the effect of Ulmi Cortex(UC) on the cisplatin-induced cell death. Materials and Methods : I examined several kinds of cell populations such as $CD4^+$ T cells, $CD8^+$ T cells, macrophages and dendritic cells in spleen. Result : When cisplatin was injected to mice, UC recovered total number of cells in spleen and also the number of T cells, macrophages and dendritic cells. UC also effected the activation of $CD4^+$ and $CD8^+$ T cells such as $CD25^+$, $CD69^+$ cells. To further investigate the effect of UC on the cisplatin-induced cell death, I examined the death of splenocyte and total T cells. UC inhibited cisplatin-induced cell death. Conclusion : Taken together, my results suggest that UC may be a beneficial oriental medicine for side effects during anti-tumor therapy.

Anti-proliferative Effect of Ulmi Pumilae Cortex Extracts on MCF-7 cells (유근피(楡根皮) 추출물의 유방암 세포주 MCF-7 생장 억제 효과)

  • Purpose: This study was conducted to investigate the anti-proliferative effects of Ulmi Pumilae Cortex Extracts(UPCE) on MCF-7(human, breast, adenocaecinoma) and NIH3T3 (human, murine, fibroblast). Methods: MCF-7 cells and NIH3T3 cells were cultured and seeded in cell culture plates, respectively. UPC was extracted with hot water and then further fractionated it into five types: hexane, chloroform, ethyl acetate, butanol, and water soluable fractions. These five different fractions from UPCE were tested for their anti-proliferative effects on MCF-7 cells and NIH3T3 cells by MMT assay. Results: Among the five solvent-fractions of UPCE, n-hexane fraction and ethyl acetate fraction showed a strong anti-proliferative effects on MCF-7 cells but they displayed significant cytotoxicity on NIH3T3 cells, too. On the other hand, chloroform fraction showed a marked anti-proliferative effects on MCF-7 cells and low cytotoxicity on NIH3T3 cells. Conclusion: Chloloform fraction from UPCE showed selective anti-cancer activities on human breast cancer cell MCF-7 relatively to the other fractions.
