Degradation of synthetic dye in water by solution plasma process

  • In this study, the solution plasma process was utilized with the aim of degrading synthetic dyes in water at atmospheric pressure. The experiments were conducted in a batch-type reactor consisting of a symmetric wire-wire electrode configuration with rhodamine B (RhB) as the target synthetic dye. The effects of the plasma treatment time and initial dye concentration on the RhB degradation were investigated by monitoring the change in absorbance of RhB solutions. The RhB solutions turned lighter in color and finally colorless with prolonged plasma treatment time, indicating the destruction of dye molecules. The RhB solutions were found to have degraded, following the first-order kinetic process. However, for high initial RhB concentrations, another kinetic process or factor seems to play a dominant role at the initial degradation stage. The fitted first-order rate constant decreased as the initial concentration increased. This result suggests that the degradation behavior and kinetic process of the RhB solution strongly depends on its initial concentration. The RhB degradation is considered to be due to a combination of factors, including the formation of chemically oxidative species, as well as the emission of intense UV radiation and high-energy electrons from the plasma. We believe that the solution plasma process may prove to be an effective and environment-friendly method for the degradation or remediation of synthetic dye in wastewater.

The Signaling of UV-induced Apoptosis in Melanocytes

  • Ultraviolet B (UVB) radiation may activate or deteriorate cultured human epidermal melanocytes, depending on the doses and culture conditions. In this study, we examined whether apoptosis of melanocytes can be induced by physiologic doses of UVB irradiation. PI staining for DNA condensation and flow cytometric analyses demonstrated the apoptotic cell death of melanocytes after UVB irradiation. The level of p53 and Bax revealed a dose-dependent increase with increasing dose of UVB, but the level of Bcl-2 remained unchanged. Confocal microscopic examination showed that Bax moved trom a diffuse to a punctate distribution after UVB irradiation. However, there were no changes in the pattern of Bcl-2. We next examined the downstream targets of apoptosis. Our results showed that a precursor form of caspase-3 disappeared with increasing doses of UVB. We also observed cleavage of poly(ADP-ribose) polymerase (PARP) after UVB irradiation. In addition, UVB irradiation resulted in a remarkable activation of c-Jun N-terminal kinase (JNK). These results indicate that UVB may induce apoptosis via JNK activation in human melanocytes.

X-ray / gamma ray radiation shielding properties of α-Bi2O3 synthesized by low temperature solution combustion method

  • In the present communication, pure and stable α-Bismuth Oxide (Bi2O3) nanoparticles (NPs) were synthesized by low temperature solution combustion method using urea as a fuel and calcined at 500℃. The synthesized sample was characterized by using powder X-ray Diffraction (PXRD), Scanning Electron Microscopy (SEM), Energy dispersive X-ray analysis (EDAX), Transmission Electron Microscopy (TEM), Fourier Transform Infrared Spectroscopy (FTIR) and UV-Visible absorption spectroscopy. The PXRD pattern confirms the formation of mono-clinic, stable and low temperature phase α-Bi2O3. The direct optical energy band gap was estimated by using Wood and Tauc's relation which was found to be 2.81 eV. The characterized sample was studied for X-ray/gamma ray shielding properties in the energy range 0.081-1.332 MeV using NaI (Tl) detector and multi channel analyzer (MCA). The measured shielding parameters agrees well with the theory, whereas, slight deviation up to 20% is observed below 356 keV. This deviation is mainly due to the influence of atomic size of the target medium. Furthermore an accurate theory is necessary to explain the interaction of X-ray/gamma ray with the NPs.The present work opens new window to use this facile, economical, efficient, low temperature method to synthesize nanomaterials for X-ray/gamma ray shielding purpose.

Optical, thermal and gamma ray attenuation characteristics of tungsten oxide modified: B2O3-SrCO3-TeO2-ZnO glass series

  • The glass series modified by tungsten oxide was created using the compounds (75-x) B2O3- 10SrCO3- 8TeO2- 7ZnO - xWO3, where x = 0, 1, 5, 10, 22, 27, 34, and 40% mole percentage. A UV-visible spectrophotometer and thermogravimetric-differential thermal analysis (TG-DTA) methods were employed to characterize the specimen's optical and phase transition attributes, respectively. The mass-attenuation coefficient (AC) of all created glasses from BSTZW0 to BSTZ7 was estimated using Geant4 code from 0.05 to 3 MeV and compared to the XCOM software results, with a relative difference of less than 2% between the two results. The increase of WO3 percentage lead to an increase in the Linear-AC at each studied energy, and this is mainly due to the fact that the higher the percentage of WO3 in the glass increases its density which causes an increase in the Linear-AC, so an energy of 0.06 MeV, as an example, the values of the Linear-AC was 4.009, 4.509, 5.442, 6812, 8.564, 9.856, 10.999 and 11.628 cm-1 form BSTZW0 too BSTZW7, respectively. The Half-VL (value layer), Mean-FP (free path), Tenth-VL, and Radiation attenuation performance (RAP) were also calculated for the current BSTZW-glass samples and revealed that BSTZW7 had the best gamma ray attenuation performance at all discussed energies when compared to other studied glass samples.

Synthesis of New Uraci1-5-Sulphonamide-p-Phenyl Derivatives and Their Effect on Biomphalaria alexandrina Snail's Nucleoproteins

  • In continuation of the previous work (Fathalla, 1992) on the synthesis of some heterocycles containing uracil moiety, we report herein the incorporation of uracil moiety into cyan-opyridine thione, thiosemecarbazone, semicarbazone, cyanopyridine, ami nocyano pyridine, isoxazoline, pyrazoline, pyrimidine, triazolo pyrimidine, pyran, selena and thiazole derivatives which might modify their biological activities. The biological studies revealed that the chemical compound III f showed high molluscicdal activity than other compounds. The profile of the nucleoprotein extracted from chemically (compound IIIc, e, f and g) treated or UV-irradiated B.alexandrina snails did not show appreciable differences when compared to non-treated (native) snails by using SDS-PAGE, where no obvious qualitative or quantitative differences were observed. Immunization of experimental animals with the nucleoprotein extracted from native, chemically (compound III f & g) treated or physically treated B.alexandrina snails induced significant protection against challenge with normal S.mansoni cercariae, as compared to the non-immunized challenged control. As well as , a decrease in the number of granuloma formation and the size range of granuloma was also observed in immunized animals. It is concluded that, compounds III f and g have a potent molluscicidal activity. They also induced chemical modification comparable to that induced by physical treatment in the snail's nucleoprotein, which could possibly be used in immunization against S. mansoni infection.

자외선B 조사에 의한 모발 외부와 내부의 광산화에 관한 분광학적 비교 (Spectroscopic Comparison of Photo-oxidation of Outside and Inside of Hair by UVB Irradiation)

  • 모발은 여러 가지 아미노산들을 포함하는 단백질로 이루어져 있다. 자외선(UV)은 태양광선중에서 모발손상에 가장 큰 영향을 미치며 모발 노화에 주된 역할을 한다. 본 연구의 목적은 전자현미경(SEM), 공초점현미경(CLSM) 및 적외선 현미경분광법(IR micro spectroscopy)을 이용하여 정상모발에 UVB를 조사한 후 특징적인 형태학적 및 화학적 구조변화를 알아보는 것이다. 에너지 분산형 X선 분광기가 부착된 전자현미경은 자외선 조사모발의 표면이 정상모발과 비교했을 때 거칠고 높은 산소원소의 함량을 보였다. 형광 및 3차원 위상 이미지를 CLSM으로 분석한 결과 정상모발의 초록색 형광방출이 UVB 조사모발에 비해 매우 높았다. 또한 fluorescamine 형광 염색법을 통해 UVB 조사모발은 정상모발에 비해 펩타이드 결합의 파괴로 생성된 자유 아미노기가 많음을 확인할 수 있었다. UVB 조사모발의 강한 푸른색 형광은 아미노기의 함량이 높다는 것을 의미하며, 이는 CLSM에서도 관찰되었다. 따라서 fluorescamine은 UVB 조사모발에서 펩타이드 결합의 파괴를 관찰하는데 유용한 도구가 될 수 있다. 정상모발과 UVB 조사모발의 단면을 IR micro-spectroscopy를 통해 이미지 맵핑(mapping)한 결과, UVB 조사모발은 정상 모발에 비해 모발의 표면은 물론 내부에 걸쳐 디설파이드 결합(disulfide bond)의 산화가 일어나고 있음을 확인할 수 있었다. 이러한 분광학적 방법은 단독 또는 다른 분석법과 함께 모발화장품의 개발에 응용될 수 있을 것이다.

멜라노마세포(B16F10)에서 노간주나무의 tyrosinase, MITF, TRP-1 및 TRP-2 발현 저해능 (Down-regulation of Tyrosinase, MITF, TRP-1, and TRP-2 Expressions by Juniperus rigida Sieb. in Murine B16F10 Melanoma)

  • 사람의 피부는 자외선, 오염된 공기, 화학제품 등 환경에 끊임없이 노출된다. 특히 자외선은 피부에 다양한 형태로 영향을 주어 주름, 잔주름, 피부거침, 피부건조와 같은 현상을 발생시켜 피부노화를 유발시킨다. 미백효과를 확인하기 위하여 tyrosinase 저해활성을 측정한 결과 열수 추출물에서 49.4%, 에탄올 추출물에서 80.0%의 효과를 나타내어 열수 추출물에 비해 에탄올 추출물의 효과가 높게 나타났으며, 노간주나무 에탄올 추출물이 B16F10 흑세포종에 대하여 멜라닌 색소 생성 억제 및 tyrosinase, MITF, TRP-1, TRP-2의 억제 효과를 가지고 있는 것을 확인하였다. 이와 같은 결과로 미루어 보아 열수 추출물보다 에탄올 추출물이 더 높은 효과를 나타냄을 확인할 수 있었으며, 노간주나무 추출물의 미백효과를 이용하여 기능성 화장품 소재로서의 가능성을 확인할 수 있었다.

잘피 추출물의 UVB로 손상을 유도한 각질형성세포에 대한 항염 효능 (Anti-inflammation effect of extract from Zostera marina using UVB-induced damage on keratinocytes)

  • Objectives : In order to confirm whether extracts of different parts of Zostera marina (ZM), a marine flowering plant, can be used as cosmetic ingredients, this study evaluated their cytotoxicity and cytoprotective effects against ultraviolet B (UVB). Inflammatory responses induced by UV stimuli are also associated with the aging of the skin.Methods : We investigated the effects of ZM extracts on cells through the water soluble tetrazolium salt-1(WST-1) assay for cell viability. In order to investigate the anti-inflammatory effects, we evaluated the suppression of Cyclooxygenase-2 (COX-2) expression by ZM extracts in HaCaT cells with UVB-induced damages, and also evaluated the production of Prostaglandin E2 (PGE2) in RAW 264.7 cells with LPS-induced damages.Results : High cell viabilities above 90% were observed in all types of ZM extracts, except for whole ZM extract at 0.5 mg/ml; in keratinocytes with UVB-induced damages, the cell viabilities were above 80% when treated with all types of ZM extracts. We confirmed their anti-inflammatory effects by investigating the suppression of inflammatory mediators. In keratinocytes with UVB-induced damages, COX-2 expression decreased in the experimental group treated with ZM extract. Similarly, in RAW 264.7 cells where inflammation was induced with LPS, the biosynthesis of PGE2 was inhibited.Conclusion : These results suggest that ethanol extracts from Zostera marina may have value as the potential anti-inflammatory medicinal plant. Also based on the abovementioned results, ZM extract protects skin cells from UV-induced damages, and thus can be used in topically applied products for skin protection.

Anti-wrinkling effects of "L-Skin Care" and molecular mechanisms on hairless mouse skin caused by chronic ultraviolet B irradiation.

  • Background: Naturally occurring antioxidants were used to regulate the skin damage caused by ultraviolet (UV) radiation because several antioxidants have demonstrated that they can inhibit wrinkle formation through prevention of matrix metalloproteinases (MMPs) and/or increase of collagen synthesis. We examined the effect of oral administration of the antioxidant mixture ("L-Skin Care") on UVB-induced wrinkle formation. In addition, we investigated the possible molecular mechanisms of photoprotection against UVB through inhibition of collagen-degrading MMP activity or through enhancing of pro collagen synthesis in mouse dorsal skin. Methods: Female SKH-l hairless mice were orally administrated "L-Skin Care" (test group) or vehicle (control group) for 10 weeks with UVB irradiation by three times a week. The intensity of irradiation was gradually increased from 30 to $180mJ/cm^2$. Microtopographic and histological assessments of the dorsal skins were carried out at the end of 10 weeks to evaluate wrinkle formation. Western blot analysis and EMSA were also carried out to investigate the changes in the balance of collagen synthesis and collagen degradation. Results: Our "L-Skin Care" significantly reduced UVB-induced wrinkle formation, accompanied by significant reduction of epidermal thickness, and UVB-induced hyperplasia, acanthosis and hyperkeratosis. Oral administration of "L-Skin Care" significantly prevented UVB-induced expressions of MMPs, mitogen-activated protein (MAP) kinases and activation of activator protein (AP)-1 transcriptional factor in addition to enhanced type I procollagen and transforming growth factor-$\beta$ (TGF-$\beta$) expression. Conclusion: Oral administration of "L-Skin Care" significantly inhibited wrinkle formation caused by chronic UVB irradiation through significant inhibition of UVB-induced MMP activity accompanied with enhancement of collagen synthesis.
