STUDY ON THE EXPRESSION OF mRNA OF TUMOR NECROSIS FACTOR-α AND INTERLEUKIN-6 IN THE CELL LINES OF SQUAMOUS CELL CARCINOMA (구강 편평상피세포암종 세포주에서 Tumor Necrosis Factor-α와 Interleukin-6의 mRNA 발현에 관한 연구)

  • The purpose of this study was to examine the mRNA levels of TNF-${\alpha}$ and IL-6 in the cell lines of normal oral keratocyte and oral squamous cell carcinoma. Total RNA was extracted from these cell lines, observed under UV light, developed by radiographic films of PCR products via reverse transcriptase polymerase chain reaction(RT-PCR) amplication, and measured with densitometer. Each mRNA level of these cell lines divided by ${\beta}$-actin mRNA level was compared to that of normal control group. The results were as follows: 1. Higher mRNA expression of TNF-${\alpha}$ than IL-6 in the normal oral epithelial cell line. 2. In general, expression of mRNA of IL-6 appeared 3-4 times more in tumor cell lines than in control group. 3. mRNA expression of TNF-${\alpha}$ showed variable expression in tumor cell lines, unlike normal cell line. 4. There are no special connections between differentiation of oral cancer cell lines and mRNA expression of TNF-${\alpha}$ and IL-6. From the above results, expression of mRNA of IL-6 in the cell lines of squamous cell carcinoma used in this study has higher than the normal oral epithelial cell line, but there are no relationship between the differentiation of oral cancer cell lines and the expression of mRNA of TNF-${\alpha}$ and IL-6.

  • Photochemical patterning of self-assembled mono layers (SAMs) has been performed by diode pumped solid state (DPSS) 3rd harmonic Nd:$YVO_4$ laser with wavelength of 355 nm. SAMs patternings of parallel lines have subsequently been used either to generate compositional chemical patterns or fabricate microstructures by a wet etching. This paper describes a selective etching process with patterned SAMs of alkanetiolate molecules on the surface of gold. SAMs formed by the adsorption of alkanethiols onto gold substrate employs as very thin photoresists. In this paper, the influence of the interaction between the functional group of SAMs and the etching solution is studied with optimal laser irradiation conditions. The results show that hydrophobic functional groups of SAMs are more effective for selective chemical etching than the hydrophilic ones.

Selection of Three (E)UV Channels for Solar Satellite Missions by Deep Learning

  • We address a question of what are three main channels that can best translate other channels in ultraviolet (UV) and extreme UV (EUV) observations. For this, we compare the image translations among the nine channels of the Atmospheric Imaging Assembly on the Solar Dynamics Observatory using a deep learning model based on conditional generative adversarial networks. In this study, we develop 170 deep learning models: 72 models for single-channel input, 56 models for double-channel input, and 42 models for triple-channel input. All models have a single-channel output. Then we evaluate the model results by pixel-to-pixel correlation coefficients (CCs) within the solar disk. Major results from this study are as follows. First, the model with 131 Å shows the best performance (average CC = 0.84) among single-channel models. Second, the model with 131 and 1600 Å shows the best translation (average CC = 0.95) among double-channel models. Third, among the triple-channel models with the highest average CC (0.97), the model with 131, 1600, and 304 Å is suggested in that the minimum CC (0.96) is the highest. Interestingly they are representative coronal, photospheric, and chromospheric lines, respectively. Our results may be used as a secondary perspective in addition to primary scientific purposes in selecting a few channels of an UV/EUV imaging instrument for future solar satellite missions.

Ultraviolet Radiation-Induced Apoptosis is Inversely Correlated with the Expression Level of Poly(ADP-ribose) Polymerase

  • The present study was conducted to elucidate whether the expression level of poly(ADP-ribose) polymerase (PARP) is related to the ultraviolet radiation (UV)-induced apoptosis. After treatment of the mammalian cell lines HeLa S3 and Chinese hamster ovary (CHO) with 50 J/m2 UV, induction of apoptosis was determined by several means during 24 h post-incubation. Incidence of apoptosis was much lower in CHO than HeLa S3 cells based on the percentage of apoptotic cells in terms of morphological changes in nucleus or direct counting of viable cells and qualitative or quantitative DNA fragmentation. Interestingly, when the expression level of PARP was measured by western blotting, the amounts of PARP that was retained at each time point inversely correlated with the incidences of apoptosis in these cells. Concomitant with generation of the 85 kDa fragment, 116 kDa PARP disappeared in HeLa S3 within 6 h after UV treatment, whereas a fair amounts of 116 kDa band was still retained in CHO cells at 36 h post-incubation. This inverse relationship was also observed in the adaptive response system, in which cells weve treated with a high dose of UV after pretreatment with a low dose. As expected, typical adaptive responses appeared in CHO cells but not in HeLa cells, showing greater cell viability and lesser DNA fragmentation. During the adaptive response in CHO cells, PARP was expressed at much higher level compared to the single, high dose-treated cells. Interestingly, even though PARP was induced at 6 h post-incubation In both cell types, its expression was more prominent in CHO cells. Thus, our data indicate that the retained level of intact PARP against UV damage inversely correlates with incidence of apoptosis in mammalian cells, and also suggest that a machinery to protect the PARP degradation against UV damage exists in CHO but not in HeLa S3 cells.

X-Ray, UV and Optical Observations of Classical Cepheids: New Insights into Cepheid Evolution, and the Heating and Dynamics of Their Atmospheres

  • To broaden the understanding of classical Cepheid structure, evolution and atmospheres, we have extended our continuing secret lives of Cepheids program by obtaining XMM/Chandra X-ray observations, and Hubble space telescope (HST) / cosmic origins spectrograph (COS) FUV-UV spectra of the bright, nearby Cepheids Polaris, ${\delta}$ Cep and ${\beta}$ Dor. Previous studies made with the international ultraviolet explorer (IUE) showed a limited number of UV emission lines in Cepheids. The well-known problem presented by scattered light contamination in IUE spectra for bright stars, along with the excellent sensitivity & resolution combination offered by HST/COS, motivated this study, and the spectra obtained were much more rich and complex than we had ever anticipated. Numerous emission lines, indicating $10^4$ K up to ${\sim}3{\times}10^5$ K plasmas, have been observed, showing Cepheids to have complex, dynamic outer atmospheres that also vary with the photospheric pulsation period. The FUV line emissions peak in the phase range ${\varphi}{\approx}0.8-1.0$ and vary by factors as large as $10{\times}$. A more complete picture of Cepheid outer atmospheres is accomplished when the HST/COS results are combined with X-ray observations that we have obtained of the same stars with XMM-Newton & Chandra. The Cepheids detected to date have X-ray luminosities of log $L_X{\approx}28.5-29.1$ ergs/sec, and plasma temperatures in the $2-8{\times}106$ K range. Given the phase-timing of the enhanced emissions, the most plausible explanation is the formation of a pulsation-induced shocks that excite (and heat) the atmospheric plasmas surrounding the photosphere. A pulsation-driven ${\alpha}^2$ equivalent dynamo mechanism is also a viable and interesting alternative. However, the tight phase-space of enhanced emission (peaking near 0.8-1.0 ${\varphi}$) favor the shock heating mechanism hypothesis.

The effect of Daehwanggogasangbakpi(大黃膏加桑白皮) on the skin damage induced by ultraviolet irradiation (가미대황고(加味大黃膏)의 멜라닌세포 활성억제가 자외선 조사로 인한 피부 손상 완화에 미치는 영향)

  • Objective : As a result of increasing amount of ultraviolet ray, skin problems including sunburn, rapid skin aging, melanoma, and even skin cancer continue to rise. In the present study, the effect of oriental herbal extract, Daehwanggo(大黃膏,DH) and Daehwanggogasnagbakpi(大黃膏加桑白皮,DS), as external application, on the skin damage, was investigated. Methods : 30 mice were equally distributed into 3 groups : control, UVB-control and UVB-irradiated and DS-treated group. Also mouse melanoma cell lines were cultured. Tyrosinase inhibition was measured to analyze the UN-protection effect. Melanogenesis in the UV-irradiated melanoma cell lines was compared in DS-treated cell line and control cell line. Sample skin from the ear tissue of the 3 groups were analyzed to observe the inflammatory response, T cell differentiation, apoptosis of keratinocytes. Results : The tyrosinase was more significantly inhibited in the DS group compared to DH group. Antioxidative effects was more prominent in DS group when superoxide dismutase was measured. Both the DS- and DH-treated cell lines showed significantly reduced melanogenesis. The reduction of external skin damage including erythematous papule, eczema, keratinocyte, pyopoiesis was observed in the DS- and DH-treated sample cells. In terms of the effect on the skin damage, sunburn cell, activated skin mast cells, secretion of IL-12, manifestation of HSP70, hyperplasia of epithelial cells, MMP-9 and destruction of the collagen were all significantly improved in the DS-treated sample cells. Melanin cells and the apoptosis in the melanoma cell line were decreased. Conclusion : DH and DS were traditionally applied externally for the scald in the oriental medicine. The present study elucidated the possibility of herbal extracts to be used as ultraviolet protectives. Further investigations are needed to assure the clinical application.

Enhanced Salt Stress Tolerance in Transgenic Potato Plants Expressing IbMYB1, a Sweet Potato Transcription Factor

  • IbMYB1, a transcription factor (TF) for R2R3-type MYB TFs, is a key regulator of anthocyanin biosynthesis during storage of sweet potatoes. Anthocyanins provide important antioxidants of nutritional value to humans, and also protect plants from oxidative stress. This study aimed to increase transgenic potatoes' (Solanum tuberosum cv. LongShu No.3) tolerance to environmental stress and enhance their nutritional value. Transgenic potato plants expressing IbMYB1 genes under the control of an oxidative stress-inducible peroxidase (SWPA2) promoter (referred to as SM plants) were successfully generated through Agrobacterium-mediated transformation. Two representative transgenic SM5 and SM12 lines were evaluated for enhanced tolerance to salinity, UV-B rays, and drought conditions. Following treatment of 100 mM NaCl, seedlings of SM5 and SM12 lines showed less root damage and more shoot growth than control lines expressing only an empty vector. Transgenic potato plants in pots treated with 400 mM NaCl showed high amounts of secondary metabolites, including phenols, anthocyanins, and flavonoids, compared with control plants. After treatment of 400 mM NaCl, transgenic potato plants also showed high DDPH radical scavenging activity and high PS II photochemical efficiency compared with the control line. Furthermore, following treatment of NaCl, UV-B, and drought stress, the expression levels of IbMYB1 and several structural genes in the flavonoid biosynthesis such as CHS, DFR, and ANS in transgenic plants were found to be correlated with plant phenotype. The results suggest that enhanced IbMYB1 expression affects secondary metabolism, which leads to improved tolerance ability in transgenic potatoes.

Raman scattering Wings of Hydrogen in Active Galactic Nuclei

  • Active galactic nuclei (AGNs) are powered by a supermassive black hole with an accretion disk and exhibit prominent broad and narrow emission lines. The unification model AGNs requires the presence of a geometrically and optically thick torus component that hides the broad line region from observers lying in the equatorial direction. The strong far UV radiation characterizing AGN spectra is expected to be scattered inelastically in the torus region to reappear around hydrogen Balmer lines or Paschen lines in the form of broad wings. Adopting a Monte Carlo technique we produce broad wings around $H{\alpha}$, $H{\beta}$ and $Pa{\alpha}$ that are formed through Raman scattering. The widths of the wings are mainly affected by the neutral column density of the torus, and the overall strengths are primarily determined by the covering factor and the column density of the neutral region. It is concluded that deep spectroscopy of AGNs of broad wings around hydrogen emission lines may shed much light on the AGN unification model.

  • Recently polymeric insulators and arresters are being used for outdoor high voltage applications. Polymeric insulators for transmission line have significant advantages over porcelain and glass insulators, especially for ultra-high voltage(UHV) transmission lines. Their advantages are light weight, vandalism resistance and hydrophobicity. Polymeric insulator kind are a relatively new technology, but their expected life is still unknown. In this paper, the material property for polymeric insulating material such as silicone rubber and EPBM is investigated by cross-link time and the relation between tracking resistance and W resistance is analyzed by IEC 60587 and W aging experiment.

Characteristics of a Corona between a Wiring Clamp (Dead End Clamp) and a Porcelain Insulator Used in a 154kV Power Receptacle

  • The occurrence of a corona is that electrical discharge due to the heterogeneity that occurs when an electrical field is concentrated in an electrode due to a cusp formed on said electrode. Wire treatment at the end of a 154kV dead end clamp for end users accelerates the occurrence of corona, which in turn leads to power loss and noise. In this study, the characteristics of the corona which occurs between porcelain insulators and support clamps of overhead lines used in l54kV power receiving facilities for end users were investigated. The corona, which cannot be identified by one common method, was measured utilizing a UV image camera. A risk assessment for fire damage and its status was suggested. The stress distribution of the electrical field by length of bare wire was suggested by means of the finite element method (FEMLAB). As a result, it was found to affect a porcelain insulators. These results can be utilized for the enhancement of clamp installation and safety in power facilities.
