Antimicrobial Activities and Adherence Inhibition on Streptococcus mutans by Ethyl Acetate Extract from Caesalpinia sappan L. (소목(Caesalpinia sappan L.)의 에틸아세테이트 분획물이 Streptococcus mutans에 대한 항균활성 및 부착 억제)

  • Somok, the heart wood of Caesalpinia sappan is used in traditional Chinese medicine. Adherence of S. mutans to the tooth surface can result in the formation of a dental plaque. This study was performed to investigate the antibacterial activity and bacterial adhesion of ethyl acetate extract from C. sappan against S. mutans ATCC 25175. The bacteria were cultured in brain heart infusion(BHI) broth, and then incubated under 5% $CO_2$ at $37^{\circ}C$ for 18~24 hours. The antimicrobial activity of the ethyl acetate extract of C. sappan was then examined using the paper disc methods and MIC. In addition, bacterial adherence to hydroxyapatite was also examined. The ethyl acetate extract was shown to produce inhibitory effects and had MIC values of 125 mg/ml against S. mutans ATCC 25175. The ethyl acetate extract inhibited adhesion of S. mutans to saliva coated-hydroxyapatite beads(S-HA). At 24 hr, the ethyl acetate extract significantly reduced the adherence of S. mutans to S-HA beads relative to the control. The isolated active substance was identified as brazilin($C_{16}H_{14}O_5$) by $^1H-NMR$ and $^{13}C-NMR$. Thus, the application of C. sappan can be considered a useful and practical method for the prevention of dental caries.

  • Water line은 UV 처리된 호스의 내부에 압력보상 기능이 첨부된 경질의 미로 튜브를 끼워 사용하도록 제작되어 있다. 점적 호스의 외경은 15.8mm이며 대부분 0.5~2.5bar 사이의 압력에 1.5~3.0 L/hr의 유량을 공급한다. 따라서 전적 호스의 내부에 위치한 경질 미로 튜브의 설계는 유량의 압력보상에 밀접한 관계가 있다. 본 연구에서는 경질 미로 튜브를 통과하는 작은 돌기의 수가 압력과 유량에 어떤 영향을 미치는지에 대한 기초적인 실험을 수행하였으며 경질 미로의 압력과 유량 특성에 따른 이론적 해석과의 비교분석을 통하여 새로운 미로의 형상 설계에 활용할 기초적 연구를 수행하고자 하였다. 점적 호스의 성능은 길이(보통 300~400m)에 대한 유량의 균등성을 유지하는 것이며 균등성은 압력보상에 의해 이루어진다. 원통형 미로는 점적 호스의 내부에 삽입되어 원통 부분에 형성된 미로의 길이에 따른 유량의 균등성을 유지하도록 설계되어 있다. 점적 호스의 원통 주변에는 8개의 미로로 구성되어 있으며 미로의 길이를 금속 평면으로 금형 제작하여 투명판에서 미로의 형상을 보이게 하였다. 또한 입구부분에 물을 공급하고 미로의 각 위치별 배출구를 설치하여 압력에 따른 유출량을 전자저울에 측정하여 압력별 유량의 균등성에 대한 데이터를 측정하였다. 유입구의 압력을 0.5, 1.5, 2.5, 3.0bar로 각각 공급하였으며 유출량은 미로의 8개 구간에 따라 측정하였다. 경질 미로의 유출량은 미로의 길이가 짧을수록, 압력이 커질수록 유출량이 높게 나타났다. 또한 미로의 길이가 짧을 경우에는 유출량의 차이가 비교적 크게 나타났으나 미로의 길이가 길수록 압력에 대한 유출량에 대한 차이가 크지 않음을 알 수 있었다. 따라서 경질 미로에서 미로의 길이를 길게 할 경우에는 유출량을 비교적 균일하게 할 수 있음을 보여주고 있다. 반면에 미로의 길이가 길어질수록 압력변화에 따른 유출량이 지수적으로 감소함을 나타내고 있다. 미로의 길이가 7번과 8번에서는 상당히 균일한 성능을 보여주므로 경질 미로의 배출구를 전체적으로 모두 활용함으로써 압력보상을 비교적 균일하게 할 수 있음을 보여주고 있다. 압력보상은 탄성을 가진 실리콘과 미로로 구성될 경우에 가장 큰 효과를 나타내지만 점적 호스와 같이 경질 미로만으로 압력보상을 해야 할 경우에는 경질 미로의 길이가 매우 중요하다.

  • The mutant strain M6 derived from Acetobacter sp. A9, which produces high levels of the bacterial cellulose derived by random mutagenesis with N-methyl-N'-nitro-N-nitrosoguanidine or UV treatment, was selected by a Hestrin and Schramm medium (HSB) plate assay. The characterization of the cellulose production was studied in flask culture to improve the productivity of bacterial cellulose by $Acetobacter$ sp. A9 and mutant strain M6. The yield of cellulose production was superior to mutant M6 than $Acetobacter$ sp. A9. Cellulose was produced 0.12 g $L^{-1}$ by $Acetobacter$ sp. A9 at HS medium and the mutant M6 produced the cellulose 6.95 g $L^{-1}$at HS medium. Strain M6 produced less amount of gluconic acid than A9, thus showing that cellulose production is negatively relted with the gluconic acid production.

  • Six analytial methods for determing the available nitrogen in soils were tested to predict the optimum fertilizer rate for the flue-cured tobacco and to test the fertility level of soils for tobacco. All methods, nitrifiable $NO_3-N$ value for 2 and 4 weeks incubation, UV absorption value at 260nm and N-value in acid digestion of 0.01 M-$NaHCO_3$ extracts, N-value extracted in boiling with $CaCl_2$ solution, and autoclave-extractable $NH_4-N$ value in 0.01 M-$CaCl_2$, were closely correlated with total nitrogen uptake as well as yield. Therefore available nitrogen indices determined from above 6 analysis method could be used for the predicting of tobacco yield without fertilizer, criteria for fertility class, and recommendable range of optimum fertilization.

  • As the first step of domestic developmint of the nucleic acid-related compounds, purine base required auxotrophs from Brevibacterium ammoniagenes ATCC 6872 were derived by the ultraviolet irradiation or the treatment of N-methyl-N'-nitro-N-nitroso guanidine (NTG), diethyl sulfate (DES), and ethylme-thyl sulfate (EMS). The optimum conditions of mutation by means of several mutagens were induced respectively. The yield of mutants was 0.083% by the ultraviolet irradiation, 0.67% by the NTG treatment, 1.1% by the DES treatment, and 0.45% by the EMS treatment. Six strains among 239 auxotrophs were screened out to accumulate 5'-lMP in the culture broth. Cry-stalline 5'-lMP was isolated from the culture broth of Brevifbacterium ammoniagenes adnine-guanine less mutant D-21530 by the use of anion exchange resin, Amberlite IRA-402, and it was identified physically and chemically as 5'-inosinic acid.

  • 전통 날염방식(Silk Screen)과 디지털날염(Digital Textile Printing)을 비교해보면, 디지털날염은 디자인에서부터 날염까지의 전공정을 완전히 디지털화함으로서 다양하게 디자인할 수 있고 미묘한 색감의 흐름을 표현할 수 있어서 자연스러운 이미지의 표현이 가능하다. 그러나 현재 디지털날염을 피혁이나 직물에 적용하기에는 프린팅속도가 느려서 생산성이 낮고 이미지 내구성(세탁, 승화, 일광, 및 마찰견뢰도)이 낮은 등 많은 단점을 가지고 있다. 또한 신발산업은 현재 중국이나 동남아시아 등 개발도상국의 등장으로 수출의 국제적 경쟁시대에 접어들면서 대량생산이나 획일적인 디자인으로부터 벗어나 고부가가치 상품을 생산해야하는 과제를 안게 되었다. 한편 가죽에 응용가능한 잉크는 크게 수성, 유성, 용제형과 자외선 경화형의 잉크가 있으며 자외선 경화형 잉크는 자외선을 이용하여 기존의 잉크의 단점을 보완한 것이다. 자외선 경화형 잉크는 자외선 램프를 열원으로 하여 이온 또는 라디칼 중합, 광중합을 하며, 에너지 소비가 적어 경제적이며 시스템 가동시 비활성 기체가 필요하지 않다. 또한 자외선 경화형 기술은 상온에서 이용할 수 있으며 잉크의 경우 건조하는 데 걸리는 시간이 필요하지 않다. 또한 시스템을 가동 시키거나 중지 시키는 것이 쉬우며 고속으로 연속적인 일을 처리할 수 있다. 프린터의 크기가 작아 공간 효율이 유리하며 용제나 $CO_2$가 발생하지 않아 친환경적인 기술이라 할 수 있다. 따라서 본 연구에서는 자외선 경화형 시스템을 잉크에 적용하여 피혁 표면에 프린팅한 다음 그 견뢰도를 측정함으로써 그 효과를 알아보았다. 세탁에 의한 Matlab 측정값으로부터 용제형 잉크의 경우는 세탁 전, 후 line width가 큰 차이를 보이나 자외선 경화형 잉크의 경우는 그 차이가 적음으로부터 자외선 경화형 잉크가 용제형에 비해 색상 탈리가 적었음을 말한다. 또한 raggedness값 역시 마찬가지 결과를 보여 안료 색소와 피혁의 접착성이 향상된 것을 알 수 있다. SEM을 이용한 피혁의 표면과 단면 사진을 보면 용제형보다 자외선 경화형 잉크를 사용한 피혁이 좀 더 매끈한 표면을 보이는데 이것은 접착기능을 하는 아크릴레이트에 의해 균일한 날염이 가능하기 때문이라 고 생각된다. 이상의 결과로부터 자외선 경화형 잉크가 세탁, 마찰, 일광 견뢰도가 우수함을 알 수 있었으며 기존의 용제형 잉크를 대체할 수 있는 좋은 재료라고 생각된다.

  • Photolysis experiments were conducted to investigate the stability of a new insecticide, [O,O-Diethyl O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl) thiophosphoric acid ester: KH-502] under the various conditions. In the photolysis experiment, KH-502 was, after being added into the acetone or acetonitrile solution, irradiated under the sunlight or UV lamp $(300{\sim}350nm)$, where acetone or acetonitrile solution was varied with water and $O_2$contents and was treated with humic acid, rosebengal or tryptophan. Results for stability and degradation pattern of KH-502 from the above experiment can be summarized as follows: 1. The significant difference in KH-502 decomposition due to photolysis was shown for between KH-502s irradiated at $300{\sim}350$ nm and non-irradiated. KH-502 was photolyzed in the acetone by the sensitizing effect, but was stable in the acetonitrile. 2. The degradation pattern of KH-502 in the photolysis was different as compared to that in the thermal decomposition, and the decomposed products were O,O-Diethyl O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl)phosphoric acid ester (KH-502 oxo form), O,S-Diethyl O-(1-phenyl-3-trifluoromethyl-5-pyrazoyl)phosphorothiolate(S-ethyl KH-502), 1-Phenyl-3-trifluoromethyl-5-hydroxy pyrazole (PTMHP) and several unknown compounds. 3. Treatments of acetone or acetonitrile solution with humic acid, rosebengal or tryptophan revealed no-sensitizing effect on the photolysis of KH-502. Dissolved oxygen in the acetone played as a cosensitizer with acetone competitively to enhance the photolysis of KH-502. 4. Treatments of acetone with humic acid or paddy soil water collected from fields decreased the photolysis of KH-502.

Study of the Floc Size Distribution for the Efficiency Assessment of Flocculation Process in Water Works (플럭크기를 이용한 응집공정 진단에 관한 연구)

  • An assessment of flocculation was performed by measuring the distribution of the floc size during sedimentation in water works. The size and number of flocs have a greater effect on an evaluation of the efficiency of flocculation rather than the turbidity. The data was collected in situ using particle image velocimetry and image analysis. The measurements were carried out at a water depth of 1m. The removal efficiency of the total organic compounds, UV absorbance and turbidity depended on the size and floc size distribution in flocculation as the G value. The G value of 50 sec-1, 30 sec-1 and 10 sec-1 showed the highest degree of efficiency in the case of an inlet water turbidity of 5 NTU, and the highest degree of efficiency was observed at a G value of 65 sec-1, 40 sec-1 and 10 sec-1 when the inlet water turbidity was 263 NTU. The number of flocs with a distribution of above $1,200{\mu}m$ was 14. The dynamics between two important growth mechanisms were investigated as the energy input changed. This is a certain method that makes use of the size and number of flocs as an efficiency assessment.

Cell Fusion of Cellulolytic Fungi, Aspergillus sp. HB1 (섬유소 분해균 Aspergillus sp. HB 1의 세포융합에 관한 연구)

  • The author isolated high cellulolytic fungi from natural sources and determined optimal condition of protoplast formation and fusion as fundamental step for improvement of the isolated it's cellulolytic ability. Three different cellulolytic fungi, such as Aspergillus sp., Penicillium sp. and Trichoderma sp., were isolated from soil. Their cellulolytic activities were compared with that of Aspergillus niger which was useful industrially and had cellulase activity. It was Aspergillus sp. that showed the highest activity of all these four fungi. And then it was followed by Penicillium sp., Trichoderma sp., and Aspergillus niger in order. An auxotrophic mutant of Aspergillus sp. was obtained by UV mutagenesis method. Having try to produce protoplast from mycelia, the author found that ${\beta}-glucuronidase$, at pH 6.0, was effective cell-wall lytic enzyme. And the optimal concentration of this enzyme was 5,000 unit/ml. Regeneration rates of wild type, met. auxotroph and arg. auxotroph, in presence of osmotic stabilizer, were 7. 0%, 7. 5% and 5.2%, respectively. PEG with M.W. 6,000 was effective stimulator for protoplast fusion in the concentration of 30% (W IV). In such a condition, we obtained 1.2% cell fusion rate.

Synthesis of TiO2/active carbon composites via hydrothermal process and their photocatalytic performance (수열합성법에 의한 TiO2/active carbon 복합체의 제조 및 광촉매특성)

  • Granular bamboo-derived active carbons (AC) were impregnated (or coated) with $TiO_2$ nano crystalline powders. The photocatalytic activity of the $TiO_2$-impregnated active carbons ($TiO_2$/AC) were determined on the basis of the degradation rate of methylene-blue aqueous solution under UV irradiation. The active compounds of $TiO_2$ were impregnated onto the AC under moderate hydrothermal conditions (${\leq}200^{\circ}C$, pH 11). The mean size of $TiO_2$ particles calculated from BET surface area were found to be as 50 nm. The $TiO_2$ precipitates were coated on the cavities or pores on the surfaces of highly activated carbons. Since the hydrothermal process led to a lowering of the on-set temperature of the anatase-to-rutile transition of $TiO_2$ as low as $200^{\circ}C$, $TiO_2$ crystallites of a pure anatase or a mixed form with rutile were successfully coated on the AC depending on the synthesis temperatures.
