Anti-Oxidant Property and Inhibition of Melanin Synthesis of Eight Plant Extracts (수종의 식물수출물의 항산화 및 Melanin 합성 억제효과)

  • Plants extracts are good resources to find functional compounds for human health. The following eight plants were collected and total phenolic contents were determined. Acer psedo-siebolianum showed the highest phenolic contents, 16.4 mg/g, whereas Cercidiphyllum japonica showed the lowest contents, 1.9 mg/g. The DPPH free radical scavenging capacities of the plant extracts showed high activity in following order : Acer ginnala ($21.3\;{\mu}g/mL$) > Cornus walteri ($23.9\;{\mu}g/mL$) > Distylum racemosum ($29.2\;{\mu}g/mL$) > Castanopsis cuspidata var. Thunbergii ($31.7\;{\mu}g/mL$) > Acer psedo-siebolianum ($34.6\;{\mu}g/mL$) > Thuijopsis dolabrata cv. Aurea ($53.1\;{\mu}g/mL$) > Cercidiphyllum Japonica ($115.2\;{\mu}g/mL$). Also the mushroom tyrosinase inhibitory activities of total extracts were determined at different concentration. D. racemosum extract showed highest (49.1% at 1,000 mg) in inhibitory activity than other seven extracts. The ethanol fraction $IC_{50}$ value: $118.1\;{\mu}g/mL$) from D. racemosum showed more inhibitory activity than ethyl acetate fraction ($IC_{50}$ value: $203\;{\mu}g/mL$). The ethanol fraction on showed no significant cytotoxicity in B16/F1 cells line up to $60\;{\mu}g/mL$. Over $80\;{\mu}g/mL$ of ethanol fraction showed cytotoxicity in B16/F1 cells. The melanin contents of cells were significantly attenuated by ethanol fraction in a dose-dependent manner. The $IC_{50}$ value of ethanol fraction was $75.4\;{\mu}g/mL$.

Effect of Extraction Methods on Antioxidant Activities of Mori ramulus (추출방법에 따른 상지 추출물의 항산화 활성)

  • The objective of this study was to examine the antioxidant activities of extracts from Mori ramulus using different extraction methods (HE, hot water extraction; EE, 50% ethanol extraction; UE, ultrasonic extraction; PE, pressured extraction). The extraction yield of PE (3.07%) was higher than that of UE (1.43%), EE (1.18%), and HE (1.07%). The total phenolic and flavonoid contents of EE were 334.66 mg/g, and 35.64 mg/g, respectively. The oxygen radical absorbance capacity (ORAC) of EE ($3,483.37{\mu}M/g$ FW) was higher than those of HE ($2,687.52{\mu}M/g$ FW), UE ($2,300.45{\mu}M/g$ FW), and PE ($2,117.62{\mu}M/g$ FW). The DPPH and ABTS radical scavenging activities of EE at $1,000{\mu}g/mL$ were 65.84%, and 97.52%, respectively. The superoxide radical scavenging activity of EE was 67.77~98.74% ($100{\sim}500{\mu}g/mL$) higher than those of other extracts. The ferric reducing antioxidant power and reducing power of EE were $189.00{\sim}974.80{\mu}M$, and 0.12~0.82, respectively. The tyrosinase inhibitory activity of EE (23.25~67.20%) improved with an increase treatment concentration. The antioxidant and tyrosinase inhibitory activities of EE were significantly higher than those of other extracts. In conclusion, we provided experimental evidence that extracts from Mori ramulus have potential as functional materials.

Comparison of Melanogenesis-Inhibiting Activity by Extracts of Prunus persica Flower and Calyx (도화 및 도화악 추출물의 미백활성 비교)

  • The antioxidant activity and whitening effect of the distilled water (DW) and ethanol extracts of the Prunus persica flower and calyx were studied. In the oxygen radical absorption capacity (ORAC) assay for antioxidant activity measurement, it was confirmed that the flower extract was stronger than the calyx extract, and that the ethanol extract was relatively stronger than the DW extract. To define the whitening effect, an experiment was conducted involving tyrosinase inhibitory assay and measurement of the melanin content of B16F10. As a result of the use of tyrosinase, the DW extract of calyx showed 53% inhibition as the highest activity. The melanin content inhibitory rates were defined as 57% for the ethanol flower extract and 63% of the ethanol calyx extract, based on a $10{\mu}g/mL$ concentration. Based on these results, mixture with the whitening effect in the extract of P. persica and another compounds should be researched for development as a cosmetic ingredient.

Protective effect of Mori Fructus in HaCaT human keratinocytes exposed to UVB (UVB에 노출된 HaCaT 인간 각질세포에서 상심자의 보호효과)

  • Objective : Mori Fructus has been used to relieve thirst, and treat dizziness, tinnitus, and insomnia caused by poor constitution. This study was performed to investigate the protective effect of the ethanol extract of Mori Fructus (MF) in ultraviolet B (UVB)-induced apoptotic cell death in human keratinocyte cells. Methods : MF was prepared by extracting 100 g of Mori Fructus in 1 L of 100% ethanol for 48 h. 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay was used to measure cell viability. Apoptosis was determined by terminal deoxynucleotidyl transferase dUTP nick end labeling (TUNEL) assay, and the expression of apoptosis-related proteins was observed by Western blot. Tyrosinase activity was measured with a colorimetric commercial kit. Results : MF promoted cell vitality and inhibited apoptosis of UVB-induced HaCaT cells. MF pretreatment reduced TUNEL-positive cells and increased the expression of caspase-3 and -9. MF also displayed antioxidant effect with high radical scavenging ability. At 2 ㎎/㎖ concentration, the 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid (ABTS) and 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical inhibition rates were 55.3 ± 4.6% and 48.5 ± 1.3%, respectively. Furthermore, MF showed a concentration-dependent inhibitory effect on tyrosinase. Conclusion : These results suggest that MF functions as a protective regulator in UVB-induced HaCaT cells by regulating apoptosis and partially exerting antioxidant effects. In addition, the tyrosinase inhibitory effect of MF shows the potential for MF to be used for skin pigmentation.

Whitening Effect of Androsace umbellata Extract (봄맞이 추출물의 미백 효능 연구)

  • The purpose of this study is to identify the whitening effect of Androsace umbellata extract. To discover the anti-pigmentation effective, we investigated Androsace umbellata extract on tyrosinase and melanogenesis inhibition. As results, it reduced tyrosinase activity and melanin contents in B16F1 melanoma cells in a dose-dependent manner with decreased to about 32% at a concentration of $25{\mu}g/mL$. To reveal how it works in inner-cellular level, we performed Western blot method. We found out that it also inhibited the protein expression in tyrosinase, tyrosinase related protein 1 (TRP-1), and microphthalmia associated transcription factor (MITF) in melanocytes. Therefore, we successfully identified the whitening effect of Androsace umbellata extract, and this finding suggested that Androsace umbellata extract is a considerable potent cosmetics ingredient for whitening. Based on this, we anticipated further researches about Androsace umbellata extract for gene levels and additional mechanisms to develop not only for functional cosmetics but for medicines or healthcare food.

Effect of Gamma-Irradiation on the Whitening Activity of β-Glucan (감마선 조사가 베타글루칸의 미백활성에 미치는 영향)

  • This study evaluated the change in whitening activity of ${\beta}-glucan$ by gamma-irradiation. Tyrosinase inhibition was significantly increased in the samples with 30, 50, 100 kGy irradiated ${\beta}-glucan$. Melanin synthesis of irradiated ${\beta}-glucan$ was measured from B16BL6 melanoma cell line treated with ${\alpha}-melanin$ stimulating hormone. Melanin synthesis was increased in the ${\alpha}-melanin$ stimulating hormone added group. However, it was decreased in the groups of 30, 50 and 100 kGy gamma-irradiated ${\beta}-glucan$ treated with ${\alpha}-melanin$ stimulating hormone. These results indicate that gamma irradiated ${\beta}-glucan$ may elevate the whitening activity. Therefore, gamma-irradiated ${\beta}-glucan$ could be used for nutraceutical foods in cosmetic industry.

Antioxidative Activity and Chemical Characteristics of Cordycepin-enriched Cordyceps militaris JLM0636 Powder (Cordycepin 고함유 Cordyceps militaris JLM0636 용매별 추출물의 이화학적 특성 및 항산화 효과)

  • The antioxidative activity and bioactivity of water, ethanol, and methanol extracts from Paecilomyces japonica (PJ), Cordyceps militaris (CM), and cordycepin-enriched C. militaris JLM0636 ($CM{\alpha}$) were tested in in vitro experimental models. The PJ water extract showed the highest extraction yield (42.53%). The highest content of phenolic compounds and flavonoids were found in the water extract of PJ, 2.72% and 1.73%, respectively. The major minerals were K, Mg, and Ca. The water extracts of PJ also showed the highest DPPH free radical scavenging activity and reducing power. Linoleic acid peroxidation and antioxidative activities were strong in $CM{\alpha}$. The methanol extracts of PJ showed the highest inhibition activity against tyrosinase. Fibriolytic activity was higher in $CM{\alpha}$ than in CM. These results may provide basic data to understand the biological activities of bioactive materials derived from $CM{\alpha}$ for the development of functional foods, cosmetics, and antithrombotics.

Comparison of Biological Activity between Nelumbo nucifera G. Extracts and Cosmetics Adding Nelumbo nucifera G. (백련(Nelumbo nucifera G.) 추출물 및 화장품에 첨가 시 생리활성 비교)

  • The solvent extracts of Nelumbo nucifera G. were investigated for antioxidant activities, whitening and anti-wrinkle effects to apply as a functional ingredient in cosmetic products. For their industrial application, the cosmetic products were also prepared with advanced formulation techniques such as W/O/W multiple emulsion. Total phenolic and flavonoids contents increased in Nelumbo nucifera G.-Leaf (NN-L). The electron donating ability of Nelumbo nucifera G.-Flower (NN-F) or Nelumbo nucifera G.-Leaf (NN-L) extracts were above 85% at a concentration of 500 ppm. The superoxide dismutase (SOD)-like activity of Nelumbo nucifera G. (NN-L) extracts was about 60% at a concentration of 1,000 ppm. The xanthine oxidase inhibitory effect of NN-L extract was higher than that of NN-F and NN-S extracts. The tyrosinase inhibitory effect, which is related to skin-whitening, was 36% in NN-F at 1,000 ppm. For anti-wrinkle effect, the elastase inhibition activity of NN-L was about 30% at 1,000 ppm. The results of stability test showed that W/O/W multiple emulsion (ME) containing Nelumbo nucifera G. extracts. The electron donating ability of the ME containing NN-F and NN-L were about 60% at a concentration of 100 ppm. The superoxide dismutase (SOD)-like activity of the ME containing NN-L was 30% at 1,000 ppm. The tyrosinase inhibitory effect, which is related to skin-whitening, was 34% in the ME containing NN-F at 1,000 ppm. In anti-wrinkle effect, the elastase inhibition activity of the ME containing NN-L was about 55% at 1,000 ppm.

Biological Activities of Extracts from Cornus kousa Fruit (산딸나무(Cornus kousa) 열매 추출물의 생리활성)

  • The extracted phenolic compounds from Cornus kousa fruit for biological activities as functional resources were examined. The phenolic compounds which were extracted with water and 40% ethanol from Cornus kousa fruit were $7.04{\pm}0.27$ and $4.47{\pm}0.18mg/g$, respectively. The 1,1-diphenyl-2-picrylhydrazyl free radical scavenging activity of water and ethanol extracts were 84% and 86% at $50{\mu}g/mL$phenolics, respectively. The 2,2'-Azinobis-(3-ethylbenzothiazoline-6-sulfonic acid radical decolorization activity of water and ethanol extracts were 84 and 95% at $100{\mu}g/mL$ phenolics, respectively. Antioxidant protection factor in water and ethanol extracts at $50{\mu}g/mL$ phenolics were 1.93 and 1.82 PF, respectively. Thiobarbituric acid reactive substance were 69% in water extracts and 89% in ethanol extracts at $150{\mu}g/mL$ phenolics. The inhibition activity on xanthine oxidase in water and ethanol extracts was 34 and 60%, respectively. The inhibition activity on ${\alpha}$-glucosidase was 29% in water extracts and 87% in ethanol extracts. The tyrosinase inhibitory activity was 19% in ethanol extracts. The collagenase inhibition activity of anti-wrinkle effect showed an excellent wrinkle improvement effect as 53% in water extracts and 77% in ethanol extracts at $200{\mu}g/mL$ phenolics. The hyaluronidase inhibition activity as antiinflammation effect of water extracts was confirmed to 34% of inhibition at $200{\mu}g/mL$ phenolic. The results can be expected extracts from Cornus kousa fruit to use as functional resource for antioxidant, antigout, inhibitor of carbohydrate degradation, antiwrinkle activity and antiinflammation activity.

A Study of the Antioxidant Activities and Whitening Activities of Areca semen Extracts as Cosmetic Ingredient (화장품 소재로서 빈랑자의 항산화 및 미백활성에 관한 효과)

  • Herbal plant extracts are good resources to find functional compounds for cosmetic ingredient. In this study, the extract of Areca semen (A. semen) was studied for melanogenesis inhibition and antioxidant activity. The results showed that ethyl acetate fraction of A. semen contained phenolic contents, $301.35{\pm}0.88{\mu}g/mg$, and exhibited potent antioxidant activity with $IC_{50}$ value of $1.02{\pm}0.07{\mu}g/mg$. Further, FRAP value exhibited potent antioxidant activity with $9.07{\pm}0.36mM$. Disk diffusion assay was performed for antibacterial activity. Ethyl acetate fraction of A. semen showed antibacterial activity against Staphylococcus aureus (S. aureus) and Staphylococcus epidermidis (S. epidermidis) at $80{\mu}g/mL$, whereas it showed no significant antibacterial activity against Escherichia coli (E. coli). The results of cell viability indicated that ethyl acetate fraction did not show cytotoxicity to B16/F1 cells at $80{\mu}g/mL$ and showed significant cytotoxicity at $100{\mu}g/mL$ of concentration and showed inhibition of melanin synthesis inhibitory, $29.78{\pm}0.31%$ at $80{\mu}g/mL$. Furthermore, mRNA expressions of tyrosinase and MITF were decreased after treatment with ethyl acetate fraction in a dose-dependent manner. As a result, the ethyl acetate fraction of A. semen could be considered as potential as whitening agents.
