Inhibitory Effects of Plant Extracts on Tyrosinase Activity and Melanin Synthesis

  • In order to develop a new skin whitening agent, approximately 100 plant extracts were evaluated for their inhibitory activities against melanin biosynthesis in cultured mouse melanocyte melan-a cells. As a result, seven extracts exhibited over 50% inhibition of melanin synthesis compared to control at a concentration of $20\;{\mu}g/ml$. In particular, Aster ageratoides Turcz. var. ageratoides (branch, root, aerial, flower; $IC_{50}$ = 17.3, 6.1, 13.6, $12.9\;{\mu}g/ml$, respectively) and Physalis alkekengi var. francheti (leaf, unripen fructus, aerial; $IC_{50}$ = 6.5, 28.3, $23.9\;{\mu}g/ml$) markedly inhibited melanin synthesis. In addition, tyrosinase activity was monitored by the measurement of dopachrome formation from the oxidation of L-3,4-dihydroxyphenylalanine. Extracts of A. ageratoides Turcz. var. ageratoides (flower) and P. alkekengi var. francheti (leaf) showed the most potent tyrosinase inhibitory activity. These plants might be the potential candidate sources in the development of novel skin-whitening products.

Diarylpropionitrile inhibits melanogenesis via protein kinase A/cAMP-response element-binding protein/microphthalmiaassociated transcription factor signaling pathway in α-MSH-stimulated B16F10 melanoma cells

  • Diarylpropionitrile (DPN), a selective agonist for estrogen receptor β (ERβ), has been reported to regulate various hormonal responses through activation of ERβ in tissues including the mammary gland and brain. However, the effect of DPN on melanogenesis independent of ERβ has not been studied. The aim of this study is to examine the possibility of anti-melanogenic effect of DPN and its underlying mechanism. Melanin contents and cellular tyrosinase activity assay indicated that DPN inhibited melanin biosynthesis in alpha-melanocyte stimulating hormone-stimulated B16F10 melanoma cell line. However, DPN had no direct influence on in vitro tyrosinase catalytic activity. On the other hand, 17β-estradiol had no effect on inhibition of melanogenesis, suggesting that the DPN-mediated suppression of melanin production was not related with estrogen signaling pathway. Immunoblotting analysis showed that DPN down-regulated the expression of microphthalmia-associated transcription factor (MITF), a central transcription factor of melanogenesis and its down-stream genes including tyrosinase, tyrosinase-related protein (TRP)-1, and TRP-2. Also, DPN attenuated the phosphorylation of protein kinase A (PKA) and cAMP-response element-binding protein (CREB). Additionally, DPN suppressed the melanin synthesis in UVB-irradiated HaCaT conditioned media culture system suggesting that DPN has potential as an anti-melanogenic activity in physiological conditions. Collectively, our data show that DPN inhibits melanogenesis via downregulation of PKA/CREB/MITF signaling pathway.

Functional Properties of Hydrolysates Prepared from Agar Treated with Organic Acids (한천 유기산 가수분해물의 생리활성)

  • This study was concerning various physiological activities of agar hydrolysates. All agar hydrolysates showed strong antimicrobial activity against Bacillus subtilis and Bacillus cereus. Also, the agar hydrolysates prepared at the temperature of $110^{\circ}C$ or $120^{\circ}C$ showed antimicrobial activity against St. aureus and E. coli. Among the agar hydrolysates, several hydrolysates treated with citrate or malate at $110^{\circ}C$ or $120^{\circ}C$ conditions showed tyrosinase activity inhibition, and their inhibition rates of tyrosinase activity were about 80%. Some tested samples treated with 0.5% organic acid at $100^{\circ}C$ or $110^{\circ}C$ inhibited the growth of cancer cell. Two agar hydrolysates prepared with 0.5% citrate and lactate at $110^{\circ}C$ for $180^{\circ}C$ min had relatively high cancer cell growth inhibition among the tested samples. The agar hydrolysates treated with citrate and lactate at $110^{\circ}C$ for 180 min obtained the main peaks of six and seven from Sephadex G-15 column chromatography. Among the main peaks, the cancer cell growth inhibition of C-3 and L-3 fractions were higher than that of other fractions.

The Study on Depigmentation Effects of Salt, Acetum, Sesami Semen Nigrum (염(鹽), 초(醋), 향유(香油)의 미백효과에 관한 연구)

  • Objectives: This study was performed to investigate the depigmentation effects of Salt, Acetum, Sesami Semen Nigrum. Methods: Inhibition of tyrosinase activity, melanin production & cell viability in cultured B16 melanoma cells, UV screen and cytoprotective effects on PC12 cells injured by hydrogen peroxide were measured. Results: Salt, Acetum, Sesami Semen Nigrum did not show any inhibitory effects of tyrosinase activity & melanin production in melanoma cells, UV screen and cytoprotective effects on PC12 cells injured by hydrogen peroxide. Conclusions: This study shows that Salt, Acetum, Sesami Semen Nigrum which were generally used for the addition in external application do not have the depigmentation effects. Following this, the depigmentation effects of the other addition which were generally used for external application shoud be examined.

  • Clematis mandshurica (Ranunculaceae) has traditionally been used as a remedy for antidiuretic, antifungal, rheumatic conditions and alleviate pain. We carried out to evaluate the anti-oxidative effect, anti-inflammatory effect and anti-melanogenic effect of ethanol extract and solvent fractions of Clematis mandshurica. The ethanol extract and the dichloromethane fraction of Clematis mandshurica showed an anti-oxidative effect in DPPH assay, the inhibitory activity of nitric oxide (NO) production in lipopolysaccharide (LPS) activated RAW 264.7 cell, and melanin synthesis and tyrosinase activity of B16F10 melanoma cells. They reduced NO production and melanin content in a dose-dependent manner at concentrations of $2.5{\sim}10{\mu}g/ml$. They also suppressed iNOS and tyrosinase protein and m-RNA expressions dose dependently, assayed by western blot analysis and RT-PCR experiment.

The Study on Depigmentation of Kamikwibi-Tang (가미귀비탕의 미백효과에 관한 연구)

  • Objective: This study was performed to investigate the depigmentation effects of the extracts of Kamikwibi-Tang. Methods Inhibition of tyrosinase activity, melanin production & cell viability in cultured B16 melanoma cells, UV screen and cytoprotective effects on PC12 cells injured by hydrogen peroxide were measured. Results: The extracts of Kamikwibi-Tang did not have any inhibitory effects on tyrosinase activity and did not show any inhibitory effects on melanin production in melanoma cells and also did not have any inhibitory effects on UV screen. But the extracts showed high cytoprotective effects on PC12 cells injured by hydrogen peroxide. Conclusion : These results suggest that Kamikwibi-Tang indrectly inhibits melanin biosynthesis which is involved in hyperpigmentation and could be used as a whitening agent for the skin.

Inhibitory Effects of Hydrolysable Tannins on Tyrosinase Activities in B16 Mouse Melanoma Cells

  • To investigate skin whitening natural substances, the effects on melanogenesis by measuring the tyrosinase activity and the melanin contents of three hydrolysable tannins, $1,2,6-tri-O-galloyl-{\beta}-D-glucose$ (1), 2,3-(S)-HHDP-D-glucose (2) and pedunculagin (3) in B16 melanoma cells were examined. $1,2,6-Tri-O-galloyl-{\beta}-D-glucose$ (1), 2,3-(S)-HHDP-D-glucose (2) and pedunculagin (3) inhibited tyrosinase activity in B16 melanoma cells in a dose-dependent manner.

Antioxidative Activities of Water Extracts from Different Parts of Taraxacum officinale (민들레의 부위별 열수 추출물의 항산화 활성)

  • This study was conducted to examine antioxidative and physiological activities of water extracts from different parts (flower, leaf, root, and the whole plant) of Taraxacum officinale. The water extracts from different parts were measured to obtain total flavonoids content, total polyphenol content, electron donating ability, SOD-like activity, nitrite-scavenging ability and tyrosinase inhibition effects. Total flavonoids and total polyphenol contents in flower extract were 32.91 mg/g and 49.31 mg/g, respectively, which were much higher than those of any other parts. The electron donating abilities of flower, leaf, the whole plant, and root extracts were 87.07%, 87.66%, 81.06% and 66.20%, respectively at a concentration of 1 mg/mL. The activities increased in a dose-dependent manner. The SOD-like activity of water extracts from different parts showed 9.07~10.97% at a concentration of 1 mg/mL. The nitrite-scavenging abilities of flower and leaf extracts measured at pH 1.2 were 36.34% and 38.16%, respectively at a concentration of 1 mg/mL. Tyrosinase inhibition activity of the leaf extract at a concentration of 1 mg/mL was the highest (34.19%) and that of the whole plant and root extracts was shown to be more than 20%. These results suggest that water extracts from different parts of Taraxacum officinale could be used as an antioxidative functional food source.

Physicochemical Properties and Biological Activities of Collagens with Different Molecular Weights from Alaska Pollack (Theragra chalcogramma) Skin (명태 껍질 유래 콜라겐의 분자량에 따른 이화학적 특성 및 생리활성)

  • This study was conducted to investigate the physicochemical properties and biological activities of collagens with different molecular weights from Alaska pollack (Theragra chalcogramma) skin as well as their efficacies as functional materials. The molecular weights of collagens were between 1~10 kDa (below 1 kDa (AP1), 1~3 kDa (AP2), 3~10 kDa (AP3), and above 10 kDa (AP4). The protein content of AP4 (40.19 g/100 g) was the highest. Collagen contents of AP1, AP2, AP3, and AP4 were 36.43, 32.23, 19.23, and 14.89%, respectively. The free amino acid and essential amino acid contents of AP1 were higher than those of AP2, AP3, and AP4. Fourier transform infrared spectroscopy spectra of collagens with different molecular weights showed wavenumbers representing the regions of amide I, amide II, amide III, and amide A, respectively. The electron-donating ability (29.51%) and SOD-like activity (38.45%) of AP1 were higher than those of AP2, AP3, and AP4. Tyrosinase inhibition activity of AP1 improved with higher treatment concentration. The rate of inhibition of MMP-1 production in HS68 cells exposed to UVB was suppressed by treatment with AP1 (29.78%) and AP2 (26.49%) at 1 mg/mL. Furthermore, there was a strong correlation between DPPH, superoxide dismutase, tyrosinase activity, and MMP-1 inhibition rate of collagens with different molecular weights.

Screening of Biological Activity for Phenolic Fraction from Onion (양파 페놀성분획물의 생리활성기능탐색)

  • To develop a material for functional food, the fraction Ⅰ,II and Ⅲ of onion extract were separated by Sephadex LH-20 column. The revealed purification levels of each fraction were 5.2%-6.5%, 12.2%-14.5% and 82.6%-86.7%, respectively. A functional activity test was performed with fraction Ⅲ. The ACE inhibition activity was about 50% at 5mg and about 80% at 10mg. The XOase inhibition activity of Korean Changnyong Daego was 84%, which was the highest. At the experiment of tyrosinase inhibition was detected 21-36% at 10mg. The cholesterol dose in mice blood was as low as low as 79$\pm$5(mg/100 mL) after the intake of fraction Ⅲ. Cholestrol dose in liver was decreased to below 50%. The results show that Sephadex LH-20 column method was very useful as a fraction method for the development of functional food material nsing onions.
