첩포시험을 이용한 치과용 재료의 알러지 평가 (Evaluation of Hypersensitivity Reaction to Dental Materials with a Patch Test)

  • Both immune reaction and hypersensitivity reaction are occurred by the same mechanism, the antigen and antibody reaction. The favorable result of this reaction towards a host is called clinically an immune reaction and the opposite results is called an hypersensitivity reaction. Type IV hypersensitivity reaction is a delayed type which is related to the cellular immune reaction and a contact hypersensitivity is included in this type. Various dental materials such as metal (mercury, nickel, chrome, cobalt), resin and eugenol are etiologic substances. Patch test kit is composed of test substance with a controlled concentration which respond only to a susceptible patient and an aluminum chamber, and etiologic substances for hypersensitivity can be easily and comfortably found just by applying the kit to the patient's skin. In this case report, the patch test was performed to a patients with oral lichen planus and the allergen, restorative material was found. After removal of the matching restoration from the patient's mouth, the symptom was improved.

소자강기탕(蘇子降氣湯)과 가미소자강기탕(加味蘇子降氣湯)이 I형(型) 및 IV형(型) 알레르기 반응(反應)과 폐손상(肺損傷)에 미치는 영향(影響) (The effects of Sojagangkitang and Gamisojagangkitang on the Type I and IV hypersensitivities and on the experimental lung damage)

  • This study attempted to investigate the effects of Sojagangkitang and Gamisojagangkitang on the variation of lung thiobarbituric acid(TBA) value, tracheal glycoprotein, serum sodium ion$(Na^+)$ contents, serum potassium ion$(K^+)$ contents ; immediatly type allergy reaction, delayed type allergy reaction in rats and mice. The results were as follows: 1. Sojagangkitang and Gami-sojagangkitang revealed significant effect on immediatly type hypersensitivity responds to histamine. 2. Sojagangkitang and Gami-sojagangkitang revealed significant effect on delayed type hypersensitivity responds to picryl chloride. 3. Sojagangkitang and Gami-sojagangkitang revealed significant effect on delayed type hypersensitivity responds to SRBC, effect of Gami-sojagangkitang was outstanding. 4. Lung thiobarbituric acid(TBA) value was decreased with statistical significance. 5. Sojagangkitang and Gami-sojagangkitang revealed decreasing effect on Tracheal glycoprotein contents, effect of Gami-sojagangkitang was outstanding. 6. Sojagangkitang and Gami-sojagangkitang revealed decreasing effect on phenol red excretion of respiratory tract. 7. Viscosity of mucine solution was decreased in proportion to increasing dosage of the Sample. 8. Serum $Na^+$ contents was not recognized significance. 9. Sojagangkitang and Gami-sojagangkitang revealed decreasing effect on Serum $K^+$ contents, effect of Gami-sojagangkitang were outstanding. According to the above results, it seems that Sojagangkitang and Gami-sojagangkitang can be applied for asthma, chronic obstructive pulmonary diseases, allergic respiratory diseases.

소음인(少陰人) 곽향정기산(藿香正氣散)의 항(抗) Allergy 작용(作用) (Anti-allergy Action of Soeumin Kwakhyangjeonggisan)

  • The purpose of this research was to investigate the effects of Kwakhyangjeonggisan (KJS) on the anti-allergic action. In the present study, we examined the effect of KJS on type I and type IV allergic reaction. KJS inhibited the systemic anaphylaxis induced by compound 48/80 and platelet activating factor (PAF), and inhibited the passive cutaneous anaphylaxis (PCA) induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin (HSA) in vivo. In addition, KJS dose-dependently inhibited the release of histamine from peritoneal mast cells in rat. Also, KJS inhibited the delayed type hypersensitivity (DTH) induced by SRBC and the contact dermatitis induced by dinitrofluorobenzene (DNFB). KJS inhibited the proliferation of splenocytes, the subpopulation of B220+ cells and CD4+CD8-(Th) cells in splenocytes and the production of γ-interferon in serum and splenocytes. These findings suggest that KJS prevented the type I allergy by the inhibition of histamine release from mast cells and the type IV allergy by the inhibition of γ-interferon production and B lymphocytes subpopulation. These results indicate that KJS may be useful for the prevention and treatment of type I and type IV allergy related disease.

식용식물 열수 추출물에 의한 지연형 알레르기 반응의 억제효과 (Inhibitory Effect on Delayed-type Hypersensitivity by the Hot Water Extracts from Medicinal Herbs)

  • 5가지 식용식물인 신이(Flos magnoliae), 지실(Poncirus trifoliata), 시호(Bupleurum falcatum), 황금(Scutellaria baicalensis), 및 오미자(Schisandra chinensis)의 열수 추출물(10 mg/20 g B.W.)이 제IV형 알레르기 반응에 미치는 억제효과를 관찰하기 위하여 DNFB로 감작된 6주령의 웅성 BALB/C mouse에 1, 2차 시료를 투여한 후, 유도된 접촉성 피부염에 의한 부종도를 측정하였으며 대조 약물로는 cyclosporin(0.005 mg/20 g B.W)을 사용하였다. DNFB의 피부염증반응에 대한 귀 두께 측정은 1차 감작에서 황금, 오미자, 지실 군이 대조 군에 비하여 유의한 감소를 나타냈고(p<0.05), 약물 대조군인 cyclosporin과 같은 억제효과를 보였다. 시호, 신이 군은 대조 군에 비해 감소하는 경향을 보였으나 유의한 차이는 없었다. 억제 효과는 황금>오미자>지실>시호>신이의 순으로서 황금 군의 활성이 가장 높았다. 2차 감작에서는 오미자, 지실, 황금, 신이 군이 대조 군에 비하여 유의한 감소를 나타냈고 오미자와 지실은 약물 대조군인 cyclosporin과는 유의적인 차이는 없었지만 억제 효과가 더 컸다. 시호군은 대조 군과 유의적인 차이가 없었으며 오미자>지실>황금>신이>시호의 순으로서 오미자 군이 가장 높은 억제효과를 나타냈다.

보폐원탕(補肺元湯)이 알러지반응에 미치는 영향 (Effect of Bopaewon-tang on Allergic Reaction)

  • The purpose of this research was to investigate the effects of Bopaewon-tang (BT) on allergic reaction. In the present study, we examined the effect of BT on type 1 and type tV allergic reaction. BT (500 mg/kg) did not affect the systemic anaphylaxis induced by compound 48180 and the passive cutaneous anaphylaxis induced by anti-dinitrophenyl (DNP)-IgE and DNP-human serum albumin in vivo. Also, BT did not affect the release of histamine from peritoneal mast cells in rats. In addition, BT did not affect the permeability of evans blue into peritoneal cavity, but inhibited the writhing syndrome induced by acetic acid. BT inhibited the delayed type hypersensitivity induced by SRBC and the contact dermatitis induced by dinitrofluorobenzene. These results indicate that BT may be useful for the prevention and treatment of type IV allergy related disease.

주귀음(酒歸飮) 및 주귀음가미방(酒歸飮加味方)의 항(抗)알레르기에 관(關)한 실험적(實驗的) 효과(?果) (Study on the effects of Juguieum and Juguieum-gamibang on the Anti-allergic effect)

  • Experimental studies were done to research the clinical effects of Juguieum and Juguieum-gamibang(Juguieum with Lonicerae Flos and Forsythiae Fructus added) on the Antiallergic effect. The results obtained as follows; 1. In the effects of Jugieum and Jugieum-gamibang on vascular permeability responses to intradermal histamine, Juguieum group revealed none significant effect, but Jugieum-gamibang group revealed significant effect. 2. In the effects of Juguieum and Juguieum-gamibang on vascular permeability responses to intradermal serotonin, Juguieum and Juguieum-gamibang groups revealed significant effect. 3. In the 48hrs homologous passive cutaneous amaphylaxis provoked by the IgE-like antibody against egg albumin, Juguieum and Juguieum-gamibang groups revealed significant effect. 4. In the delayed type hypersensitivity responses to Picryl Chloride, Juguieum and Juguieum-gamibang groups revealed significant effect. 5. In the delayed type hypersensitivity responses to SRBC, Juguieum group revealed significant effect, but Jugieum-gamibang group revealed none significant effect. According to above-stated results, Juguieum and Juguieum-gamibang are concluded to be effective as anti-allergic regimen and recommended to be used for the treatment of type I and type IV allergic disease.

탁리소독음(托裏消毒飮)의 항(抗)알레르기 효과(效果)에 관(關)한 실험적(實驗的) 연구(硏究) (An Experimental Study on the Anti-allergic effect of the Taklisodok-um)

  • This Experimental study was done to investigate the effect of the Taklisodok-um on the Anti-allergic response. The results were obtained as follows: 1. On vascular permeability response to the intradermal injected Histamine, the Taklisodok-um treated group revealed more significant decrease than control group. 2. On vascular permeability response to the intradermal injected Serotonin, the Taklisodok-um treated group revealed more significant decrease than control group. 3. In the 48hrs homologous passive cutaneous anaphylaxis provoked by the IgE-like antibody against white egg albumin, there was no significant difference between the Taklisodok-um treated group and control group. 4. The Taklisodok-um treated group revealed more significant inhibitory effect than control group in the delayed type hypersensitivity response to Picryl chloride. 5. The Taklisodok-um treated group revealed more significant inhibitory effect than control group in the delayed type hypersensitivity response to SRBC. According to the above-stated results, it is considered that the Taklisodok-um could be applied widely to the type I and IV allergic diseases.

산사(山楂)가 DNCB로 유발된 생쥐의 알레르기성 접촉피부염에 미치는 영향 (Effects of Crataegus Pinnatifida (CP) on Allergic Contact Dermatitis (ACD) Induced by DNCB in Mice)

  • Allergic contact dermatitis (ACD) is a type IV delayed hypersensitivity reaction that results from exposures and subsequent sensitization to an environmental chemical. Crataegus Pinnatifida (CP) is commonly used to improve spleen function, remove retention of food, and promote blood circulation. This study is designed to investigate the effects of CP on ACD induced by 2,4-dinitrochlorobenzene (DNCB) in mice. In this experiment, the effects of CP on changes in body weights, ear and dorsum skin thicknesses, ear weights, clinical aspects on the dorsum skin, histopathological changes, spleen weights, cytokines were investigated. In addition, the effects on the proliferation rates of splenocytes were also investigated in vivo and vitro study. In results, CP spread (CPS) group and CP spread and administered (CPS+Adm) group showed decrease in spleen weights. In CPS+Adm group, dorsum skin thicknesses were decreased significantly compared to control group. CP treatment diminished erythema, desquamation and keratosis which were induced by repeated painting of DNCB. In histopathological observation, spongiosis and edema were diminished in CPS and CPS+Adm group. CP led to decrease in the proliferation rates of splenocytes in vivo and vitro. In conclusion, these data suggest that CP can decrease symptoms of ACD, so CP is useful to treat patient with ACD.

  • Experiments were performed on mice to investigate the effects of a polycyclic aromatic hydrocarbon, 3-methylcholanthrene (MCA) on Arthus reaction, delayed-type hypersensitivity (DTH) and antibody production to sheep red blood cells (SRBC). Mice were sensitized iv with 0.1ml of 1% SRBC suspension were treated with a single ip injection of olive oil alone or with different doses of MCA in oil (0.5~50mg/Kg) at various time before (-) or after (+) sensitization (day 0) and were challenged at 4 days after SRBC. Arthus reaction was measured at 3 hours after challenge and other responses at 24 hours. Treatment with MCA inhibited Arthus reaction and DTH to SRBC, measured by footpad swelling reaction, and this immunosuppressing effect was dependent on the dose and time of MCA treatment in relation to SRBC sensitization. Humoral immune responses as measured by serum hemagglutinin-and hemolysin-titers to SRBC were significantly depressed when MCA was injected before or at the same time of sensitization. However, the response was slightly depressed when injected after SRBC. These results indicate that MCA suppress the function of the cells involved in immune responses.

Effects of Panax ginseng on Morphine-induced Immune Suppression

  • To investigate the possibility of Panax ginseng as a therapeutic agent for the immune suppression, ginseng total saponin (GTS) extracted from korean red ginseng was tested on immune functions from morphine-induced immune suppressed mice. To study how immune functions are affected by morphine and also to test whether GTS can be an useful therapeutic agent for morphine toxicity, several parameters were employed, body weight, immune organ weight, B cell functions, and T cell function. Morphine impaired the development of body weight and immune organ weight such as spleen and thymus. Morphine also depressed a B-cell function, antibody production. T-cell functions studied by type IV hypersensitivity test were most markedly affected by morphine treatment. GTS restored most of morphine-induced immune suppression. GTS restored the morphine-induced decrease in spleen weight to body weight ratio in a dose dependent manner, but not the body weight decrease. Also all of the morphine-induced impairments of B cell functions and cellmediated immunity were fully recovered by GTS. These results suggest that ginseng product could be very helpful for the treatment of immune suppression occurring in morphine abusers.
