Tuber yield and content of general component and diosgenin which is a main bioactive property were investigated in order to determine the growth characteristics of round typed yam(Dioscorea opposita L.) and the potential of artificial culture at Suwon, Korea. Tubers of round yam were initiated to form at 60 days after planting and then enlargement of tubers lasted by 160 days after planting. Compared to short typed yam(108g), tuber weight of round yam was higher(127g) on the basis of dry weight at 200 days after planting. In comparison of general component between round yam and short yam, protein of round yam$(3.62\%)$ was higher than short yam$(2.10\%)$. Water content in round yam$(64.5\%)$ was lower in short yam$(79.4\%)$, indicating a higher dry weight ratio of round yam. Hardness of round yam was 2787.6 while short yam showed about two times higher hardness(4946.9). Lightness was higher in round yam(77.4). In tuber extracts analysis, diosgenin content was respectively $3.32\%$ in round yam and $2.61\%$ in short yam.

Metagenomics is a powerful tool to isolate novel biocatalyst and biomolecules directly from the environmental DNA libraries. Since the metagenomics approach bypasses cultivation of microorganisms, un-cultured microorganisms that are majority of exists can be the richest reservoir for natural products discovery. To discover novel herbicidal substances from soil metagenome, we established three easy, simple and effective high throughput screening methods such as cucumber cotyledon leaf disc assay, microalgae assay and seed germination assay. Employing the methods, we isolated two active single clones (9-G1 and 9-G12) expressing herbicidal activity which whitened leaf discs, inhibited growth of microalgae and inhibited root growth of germinated Arabidopsis seeds. Spraying butanol fraction of the isolated active clones' culture broth led to growth retardation or desiccation of Digitalia sanguinalis (L) Scop. in vivo. These results represent that the screening methods established in this study are useful to screen herbicidal substances from metagenome libraries. Further identifying molecular structure of the herbicidal active substances and analyzing gene clusters encoding synthesis systems for the active substances are in progress.
We have employed comparative RFL,P(Restriction Fragment Ixngth Pol~iniorphism) analysis and molecular phylogenetic techniques to investigate the diversity of uncultured microorganisms associated with the anaerobic PCP degradation in PCP-adapted enrichment cultures inoculated by samples from anaerobic cewage sludgc(Jangrim, Pusan) and leachate of landfill site(Kimhae). 16s rDNA cloncs were obtairted by PCR amplification of mixed population DNAs extracted directly from the nonactive and active stage ol each PCP-adapted culture. After three rounds of comparative RFLP analyses. two RFLP types. designated as Ala and Hld, were found prevalent and common in both active stage samples. Thc analysis of phylogenctic diversity bawd on the 5'-terminal 180 nt of sequences from whole clones of the Ala and Bld RFLP types showed close similarity among themselves. In case of Bld clones, 7XQ of them shared identical sequences. Thcse resuliq suggest that the clones of both RFLP types wcre originated from highly affiliated microorganisms which are e~iriched as a result of metabolic activity to PCP. The full-length 16s rRNA sequence of each representative clone from both RFLP types was determined. and an Ala clone w i n found to he related to Clo.strrdiurn ulfutzac~(Genk~ank No. Z69203) and a Bld clone to Thermobacteroides proteolyticus(Genbank No. X09335), with sequence similarities of 89%' and 97%. respectively.

The objective of this work was to evaluate a solid-state fermentation process for the practical production of arachidonic acid(AA) by Mortierella alpina ATCC 32222. In the present investigation, batch culture kinetics for both submerged- and solid-state fermentations was carried out at $25^{\circ}C$ to identify the relationship between growth and arachidonic acid (AA) production. Glucose and yeast extract were used in submerged fermentations by using flasks, while rice bran was used as a sole raw material in the other type of fermentations by using a series of Petri dishes. It was evident that a mixed-growth associated pattern existed between the two variables, irrespective of modes of fermentations. The effect of carbon to nitrogen (CfN) ratio on AA production in solid-state fermentation was studied in the range of 6.5 - 20. As a result, an optimum condition was found to be 6.5. Supplementary carbon source was not necessary to meet the optimum C/N ratio. Unlike the Previous results obtained by other researchers, a supplement of sodium glutamate up to $4\%$ (w/w) to the rice bran medium did not have a positive effect on the AA productivity. However, an increase in AA productivity was obtained with the rice bran medium supplemented with sesame oil.

National Records Designation System is designed to protect valuable civilian records from loss or damage. It also intends that government administrates important civilian records to raise public concerns civilian records and to foster archival culture in Korea. This study investigates the current states of service fo the designated record through the web page of National Archive of Korea. Major findings are as follows. First, the information of designated records is dispersed in two web pages by the National Archive of Korea, an introductive web page of every collection in the National Archive of Korea and a web page of designated record service. Second, the web page of designated record service provides information of designated records only at collection level, so it is not easy for users to understand the contents of the records. In order to improve the service for the designated record service of the National Archive of Korea, this study proposed the unification of dispersed web pages to provide information of the designated records consistently. It also suggested a facet based directory service and word cloud service to give access to the contents of each designated record collection. The facet based directory and word cloud service will help users to understand the designated records in more detail.

There is evidence to suggest that cranberry juice supplements improve the health of the urinary tract by inhibiting the binding of fimbriated uropathogenic E. coli to the bladder mucosa. In patients with neurogenic bladders, urinary tract infections (UTI) are particularly common and often poorly managed by antibiotic treatment. A double-blind, randomized, placebo-controlled trial was undertaken on 29 geriatric and spinal cord injured patients with dysfunctional bladders. They received three times daily at mealtimes a 4 oz bottle of cranberry juice (Ocean Spray Cranberries, USA) or a specially prepared synthetic placebo drink. Two episodes of UTI arose in week one of cranberry intake and none thereafter, compared to four episodes of UTI in 4 placebo patients in weeks four, six and 10. Mean bacterial adhesion counts on bladder cells of the patients rose during the first month of treatment in 71 % of the placebo patients compared to only 31 % of cranberry patients (p < 0.001). The difference persisted to some extent for the second and third months. Bacterial adhesion levels correlated with culture findings (higher adhesion and higher viable counts in urine) (p < 0.001), positive leukocyte nitrite tests (136$\pm$131 bacteria per cell versus 52$\pm$86 in negative tests) (p < 0.001), and higher white blood cell counts (> 10) per high power field (126$\pm$125 versus 48$\pm$85 bacteria per cell) (p<0.001). E. coli was the most frequently isolated organism (40% samples) followed by K. pneumoniae (17%) and a number of other uropathogens. Group B Streptococci, and coagulase negative Staphylococcus were recovered from urine in 4 samples but were not associated with any red blood cell presence. The daily intake of cranberry juice, in amounts which are not detrimental to long term compliance, appeared to have a role in reducing the risk of bladder colonization and infection in a highly susceptible patient population.
To improve the efficiency of production of cloned embryos and animals by nuclear transplantation in the rabbit, the effect of cell cycle of donor nuclei and type of recipient cytoplasm on the in vitro developmental potential and production efficiency of offspring was determined. The embryos of 16-cell stage were collected from the mated does at 48h post-hCG injection and they were synchronized to G$_1$ phase of 32-cell stage. The oocytes collected at 14h post-hCG injection were freed from cumulus cells and then enucleated. One group of the enucleated cytoplasms was activated by electrical stimulation prior to injection of donor nucleus, and the other group was not pre-activated. The separated G$_1$phase blastomeres of 32-cell stage embryos were injected into the perivitelline space of recipient cytoplasms. After culture for 20h post-hCG injection, the nuclear transplant oocytes were electrofused and activated by electrical stimulation and the fused nuclear transplant embryos were co-cultured for 120h and the nuclear transplant embryos developed to blastocyst stage were stained with Hoechst 33342 dye and their blastomeres were counted. Some of the nuclear transplant embryos developed in vitro to 2- to 4-cell stage were transferred into the oviducts of synchronized recipient does. The electrofusion rate was similar between the types of donor nuclei and recipient cytoplasms used. However, the nuclear transplant embryos using G$_1$ phase donor nuclei were developed to blastocyst at higher rate(60.3%) than those using S phase ones(24.7%). Also, when non-preactivated oocytes were used as recipient cytplasms, the develop-mental rates of nuclear transplant embryos to blastocysts were significantly(P< 0.05) higher(57.1%) than those using preactivated ones(20.8%). The cell counts of nuclear transplant embryos developed to blastosyst stage were increased signficantly(P<0.05) more in the non-preactivated recipient cytoplasm(163.7 cells), as compared whit the preactivated recipient cytoplasm(85.4 cells), A total of 49 nuclear transplant embryos were tranferrid into 5 recipient does, of which two offsprings were produced from a foster mother 31 days after embryo transfer. these results showed that the blastomeres of G1 phase and non-preactivated oocytes might be utillzed efficiently as donor nuclei and recipient cytoplasms in the nuclear transplant procedure, thought the offspring production remained still low.
The aim of this study was to measure and compare nutrient intake, anthropometric measurements and serum indices by percent body fat as one of the index of obesity degree in female college students. Additionally we attempted to investigate percentile distribution of fat free mass index (FFMI) and fat mass index (FMI) for developing reference values for these two parameters. The subjects were 91 female college students who were classified to 4 groups according to the percentile of percent body fat (Group1: 25 th < percentile of percent body fat, Group 2 : 25 th ${\le}$ percentile of percent body fat < 50 th, Group 3 : 50 th ${\le}$ percentile of percent body fat < 75 th, Group 4 : percentile of percent body fat ${\ge}$ 75 th). The mean percent body fat and body mass index were 28.2%, $20.5 kg/m^2$ respectively. The mean energy intake was 1707 kcal(81% of KDRIs) and vitamin C, folate, Ca and Zn intake were 73.9%, 54.7%, 79.6%, 97.5% of KDRIs respectively. Most nutrient intake (energy, carbohydrate, cholesterol, fatty acid, Ca, Fe) of G4 was lower than that of G1, G2 and G3. Serum HDL-cholesterol concentration was significantly lower in G4 than G1, G2, G3 and it tended to increase as percent body fat decreased. LDL/HDL, A1 of G4 were significantly the highest among the 4 groups and increased as percent body fat increased. The mean fat free mass index and fat mass index were $14.5 kg/m^2$, $6.0 kg/m^2$ respectively. The criteria of sarcopenic obesity which has been defined as under 25 th percentile of FFMI and below 75 th percentile of FMI were shown $12.8 kg/m^2$, $8.2 kg/m^2$ respectively in this study. In conclusion, we should continue to more systematically research on the studies of new obesity measurement which includes FFMI and FMI as one of the variables. And the public education for weight control that emphasizes both the understanding of body composition and the importance of nutrition balance is also required.
The crisis in profit and funding for animation is in evidence around the world. The change in media environment has forced broadcasting advertising revenue down and animation viewers spread out. Now, animation players in the strongest position are either the US major studios, or producers benefiting from domestic support schemes. Government support is available in many countries because many governments believe that support for domestic animation carries both economic and cultural benefits. This paper is designed to suggest new policy schemes for Korean animation industry. The new paradigm of animation policy needs a new perspective on content industry as a whole, not centered on the animation itself. The researchers on public policy for culture, audiovisual and content industry argued that the government should, (i) play the role of facilitator for virtuous cycle of industry value chain, (ii) provide fiscal support through automatic and selective schemes, (iii) provide tax benefit to strengthen the competitiveness of industry, and (iv) enforce the broadcasters to contribute to domestic programming and financing. Comparative analysis on French and Canadian audiovisual policy supports such arguments, and animation industry of two countries are enjoying the high audience ratings and sustainable production volume. From the analysis, this paper suggests the new government schemes for Korean animation industry, which are, (i) securing the public funding for fiscal support, (ii) introduction of automatic production support, (iii) modification of broadcasting quota, (iv) broadcaster's performance envelope and production quota, and (v) tax benefit as indirect support.
Hallyu in Malaysia is considered to be in the growth stage as in countries such as Philippine, Myanmar, Hungary, and Uzbekistan. In the growth stage, Hallyu becomes increasingly popular and familiar to the general public and the sales of Hallyu-related products starts to increase. The purpose of this research is to compare how Hallyu is perceived among Malaysian youths and adults. Analysis is carried out using FGD. The two groups are asked about what they think of Korea and the success of Hallyu, how much interest they have in foreign cultures in general, and how they come into contact with them. Based on these results, problems currently facing Hallyu and possible solutions are examined. The analysis shows that the interest in Hallyu among Malaysians varies according to age and gender. Both male and female participants in their 20's have shown a much greater interest in Hallyu than the participants over 30's. But Malaysians are known to be relatively open-minded about accepting foreign cultures, and this is reflected in the current study. They view the popularity of Korean drama, movies, and music as a trend, rather than being intolerant of them. There is no concern for negativity toward Hallyu in Malaysia, but it is likely that Hallyu may be replaced by another foreign culture unless its influence is constantly maintained in the area. Therefore, it is essential to develop measures and models to not only retain, but strengthen Hallyu's influence.
