Saccharomyces cerevisiae KNU5377 is a constitutively thermotolerant, fermentative strain at high temperatures over 4$0^{\circ}C$. The exposure to 0.5 M NaCl caused S. cerevisiae KNU5377 to be lost its constitutive thermotolerance. Furthermore, the NaCl adaptation beyond 0.3 M during the overnight culture forced the strain-specific fermentation ability of S. cerevisiae KNU5377 to be disappeared. However, these phenomena did not occur in the reference, Saccharomyces cerevisiae ATCC24858. As a result, this adaptation led both strains to show the closely similar thermotolerance level and alcohol fermentation ability, implying the NaCl adaptation eliminated its strain-specific characteristics of S. cerevisiae KNU5377 Therefore it indicated that the superior intrinsic characteristics of S. cerevisiae KNU5377 must be related to the NaCl adaptation. On the other hand, the heat adaptation elevated alcohol productivity for both strains, but surprisingly did it for KNU5377 at the rate of two times higher than the reference's one; this suggests that KNU5377 possesses more efficient system enough to cause the difference. Consequently, these characteristics of S. cerevisiae KNU5377 must be interesting targets for further study to understand on how KNU5377 could acquire the constitutive thermotolerance and the outstanding fermentative capacity at high temperatures.
Endophytic fungal strains were isolated from the roots of six species plants in the Dokdo islands. Native plant samples, such as Artemisia japonica, Chenopodium album and Solanum nigrum were isolated from Dongdo, and those such as Cyrtomium falcatum, Dianthus longicalyx and Tetragonia tetragonoides were isolated from Seodo. In total, thirty two fungal strains were isolated from these native plants. To identify the fungal strains, polymerase chain reaction (PCR) amplification of internal transcribed spacer (ITS: containing ITS1, 5.8s and ITS2 region) regions was done with universal primers ITS1 and ITS4. Endophytic fungi of four species were isolated from A. japonica, eight species from C. album, three species from S. nigrum, three species from C. falcatum, three species from D. longicalyx and eleven species from T. tetragonoides. Culture filtrates (CF) of isolated endophytic fungi were used to treatwaito-c rice seedlings to test plant growth-promoting activity. As a result of bioassay, Ca-5-2-2 strain isolated from C. album expressed highest plant growth-promotion activity. Of all the endophytic fungi isolated, Penicillium sp., Fusarium sp. and Aspergillus sp. were the most abundantly distributed fungal strains in the six plants used in this study.
The nature of science has been recognized in a great deal in the field of science education. However, only few innovative programs are offered for science-gifted students to improve their recognition of the nature of science. The current study describes and analyzes science-gifted students' understandings of the nature of science (NOS). In addition, the study looks into contradictory views among the aspects of NOS, which are fundamental data in constructing target programs on NOS for science gifted students. Data used in this study were collected from 73 middle school science-gifted students using an open-ended questionnaire, VNOS. The results of this study showed that the participants' understanding of NOS was significantly distributed on naive or transition view except for 'tentative NOS', and the results revealed inconsistent view among the aspects of NOS. This study proposes two suggestions to enhance the recognition of science-gifted on NOS of science to informed state and to have consistent perspectives with other areas. First, the role of experiment has to be changed-it should be the process in constructing scientific knowledge rather than an instrument to check scientific knowledge to transform perspective on experimental data and scientific knowledge. Second, various opportunities must be provided to science-gifted students, so they can experience the culture and community of scientists and science to gain a wider insight of science.
The purpose of this study was to find out the removal capacity of harmful ammonia by different filter media in the submerged biological filters in a given space of chamber. Four materials, pile cloth, corrugated skylight roofing plate, embossed plastic plate, and gravel, were used as the experimental filter media. Each filter medium was placed in two aquariums, each aquarium measuring $90cm\times60cm\times60cm\;(depth)$. Under the normal operating condition, the average of mean ammonia removal rates during the first and second functioning periods by each filter material which occupied tile space in the filter chamber (aquarium) was as follows: 1. Pile cloth: $8.381\;g{\cdot}m^{-3}.\;day^{-1}$ 2. Corrugated skylight roofing plate: $7.834\;g{\cdot}m^{-3}.\;day^{-1}$ 3. Embossed plastic plate: $7.797\;g{\cdot}m^{-3}.\;day^{-1}$ 4. Gravel: $7.051\;g{\cdot}m^{-3}.\;day^{-1}$ Thus, there were no significant differences between the media, but at the time of practical application of these materials, some other factors such as investment cost, easiness for the removal of excess detritus accumulated in tile interstices of filter media, etc. should be fallen into consideration. When large units are required, in particular, removal of excess detritus from tile gravel bed is extremely difficult, and in case of pile cloth filters the installation work is much complicated and a problem in supporting the structure when drained also exists. In these respects, corrugated skylight roofing plate and embossed plastic plate seem to be more optimal, but again in practice the local situation for the availability and the price of the materials should be rechecked and the fitness of tile materials in the particular filter chambers under use or under consideration for construction must be taken into account.
We explored the possibilities of using the freshwater rotifer Brachionus angularis as a live food for small fishes cultured in fresh- or brackish waters. Brachionus angularis were collected from a reservoir for isolation and laboratory culture. Length and width of the lorica were $102.3{\mu}m$ and $76.6{\mu}m$, respectively, and those of amictic eggs were $64.4{\mu}m\;and\;47.9{\mu}m$, respectively. When their growth rates were examined at six different temperatures, i.e., 15, 20, 25, 30, 35, and $40^{\circ}C$, the highest daily growth rate of 0.801 was observed at $35^{\circ}C$, and growth was lower with decreasing temperature. Adaptation to salinity change was evaluated with two different modes of salinity increase: step-wise elevation lasting for short durations of 5 to 30 min or a long duration of 24 h. With the short duration modes, no individuals survived salinity higher than 10 psu, and the number of live individuals did not increase throughout the experiment. However, in the 24-h elevation, the number of individuals increased when salinity was elevated by 1 to 2 psu per day for the first 2 or 3 days, while no increase in number occurred at salinity increments higher than 3 psu per day. In addition, to assess the effect of different diets, four single-component diets (Chlorella vulgaris, Nannochloris sp., baker's yeast, or dry yeast) and three combination diets (C. vulgaris + Nannochloris sp. + baker's yeast + dry yeast; C. vulgaris 70% + baker's yeast 30%; C. vulgaris 30% + baker's yeast 70%) were used. The specific growth rates of B. angularis fed combination diets were higher than those of rotifers fed any single-component diet, with the highest rate of 0.648 in B. angularis fed a mixture of C. vulgaris, Nannochloris sp., baker's yeast, and dry yeast, and the lowest rate of 0.200 in those fed dry yeast only. Our results indicate that the freshwater rotifer B. angularis can be used for seedling production of both freshwater and brackish-water fishes that require small (less than about $120{\mu}m$) live food during their early stages.
A recirculating water system without filter bed was used for a high density experimental rearing of common carp, Cyprinus carpio, during the summer of 1981. Of the water system, 3 tanks were used for stocking each tank with 10,000, 20,000 and 30,000 common carp fingerlings which averaged 0.6 g. The water in the system was maintained in dark green colour by heavy growth of phytoplankton during the most period of the experiment. In this experiment, the best daily growth rate, $3.7\%$, was obtained from the 10,000 fish group followed by $2.8\%\;and\;3.0\%$ from the 20,000 and 30,000 fish groups respectively. Thus there was no significant difference between the latter two groups. Especially, notably high concentration of total ammonia, $5\sim7$ ppm ammonia-N, in the culture water appeared during the most experimental period, but this did not seem to have affected feeding and growth of the fish when the water was kept at heavy bloom. It was also obvious that the outbreak of columnaris disease was effectively depressed in this green water recirculating system compared to that in clarified recirculating system. One tank ($7m^2$) of this system was stocked with 446 common carp of intermediate size averaging 352.2 g and after 40 day rearing they grew to 486.3 g in average with a daily growth rate of $0.8\%$. This result was a little inferior to that reared in a tank of the clarified water system as control which showed $1.0\%$ daily growth rate. Therefore, before an initiation of the commercial production of fish in this method, further studies concerning the amount of ammonium with its effect on the fish under rearing in this system and the columnaris disease problem should be carried out.
Objectives: This job analysis of a staff in charge of quit-smoking policy at public health centers aims at providing fundamental information to establish strategies supporting various quit-smoking. Methods: The job analysis of a staff in charge of quit-smoking policy at public health centers was carried out through DACUM(Development of Curriculum) method from April through May 2006. Three experts had developed job description with staffs in charge of quit-smoking policy at public health centers through two workshops. The survey was practiced for staff in charge of quit-smoking policy at the other public health centers. The characteristics of the staffs such as age, years for working at public health center, years for charging with quit-smoking work, the proportion of responsibility for quit-smoking work, were surveyed. Results: The research has reached the conclusions below. 1. The job description have been developed considering input-process-outcome axis and plan-do-evaluation axis for quit-smoking policy at public health centers. The final job description is composed of 3 missions, 7 accountabilities, 20 sub-work items. 2. The quit-smoking activity mostly focused at direct education and counselling. But planing and evaluation activity for quit-smoking have been under-achieved. 3. The staffs for quit-smoking policy were feel it is easy to educate and counsel to comer to public health centers for quit-smoking. But having the high proportion of responsibility for quit-smoking policy have usually difficult to do that. So they want to education about counselling for smoker. 4. The staffs who worked over the 2 years for quit-smoking policy the public health center have responded that investigate the smoking rate of the jurisdiction community and the problem of the culture about smoking and smoking policy is important. Conclusions: The study helps reinforcing the initiatives of central government for quit-smoking policy at public health centers. Especially staffs want education in technology area for counselling smoker. And they want nationwide supporting for investigating smoking rate and related factors at the local level.

Purpose : The purpose of this study was to examine the expression of various angiogenic factors during osteoblastic differentiation of periostealderived cells and the effects of osteogenic inductive medium of periosteal-derived cells on the proliferation of endothelial progenitor cells. Materials and methods : Periosteal-derived cells were obtained from mandibular periosteums and introduced into the cell culture. After passage 3, the cells were divided into two groups and cultured for 21 days. In one group, the cells were cultured in the DMEM supplemented with osteogenic inductive agent, including 50g/ml L-ascorbic acid 2-phosphate, 10 nM dexamethasone and 10 mM -glycerophosphate. In the other group, they were cultured in DMEM supplemented without osteogenic inductive agent. VEGF isoforms, VEGFR-1, VEGFR-2, and neuropilin-1 mRNA expression was observed. Human umbilical cord blood-derived endothelial progenitor cell proliferation was also observed. Results : The expression of VEGF isoforms was higher in osteogenic inductive medium than in non-osteogenic inductive medium. The expression of VEGFR-2 was also higher in osteogenic inductive medium than in non-osteogenic inductive medium. However, the expression of VEGFR-1 and neuropilin-1 was similar in both osteogenic inductive medium and non-osteogenic inductive medium. In addition, conditioned medium from differentiated periosteal-derived cells stimulated human umbilical cord blood-derived endothelial progenitor cell numbers compared to conditioned medium from non-differentiated periosteal-derived cells. Conclusion : These results suggest that in vitro osteoblastic differentiation of periosteal-derived cells has angiogenic capacity to support endothelial progenitor cell numbers.
The fibroblasts are the principal cells in the periodontal ligament of peridontium. As the periodontal ligament fibroblasts (PDLF) show similar phenotype with osteoblasts, the PDLF are thought to play an important role in alveolar bone remodeling. Cell-to-cell contacted signaling is crucial for osteoclast formation. Recently it has been reported that PDLJ enhance the bone resorbing activity of osteoclasts differentiated from hematopoietic preosteoclasts. The aims of this study were to $clarify\;^{1)}$ the mechanism of PDLF-induced osteoclastogenesis $and\;^{2)}$ whether we can use preosteoclast cell line instead of primary hematopoietic preosteoclast cells for studying the mechanism of PDLF-induced osteoclastogenesis. Osteoclastic differentiation of mouse macrophage cell line RAW264.7 was compared with that of mouse bone marrow-derived M-CSF dependent cell (MDBM), a well-known hematopoietic preosteoclast model, by examining, 1) osteoclast-specific gene expression such as calcitonin receptor, M-CSF receptor (c-fms), cathepsin K, receptoractivator nuclear factor kappa B (RANK) ,2) generation of TRAP(+) multinucleated cells (MNCs), and 3) generation of resorption pit on the $OAAS^{TM}$ plate. RAW264.7 cultured in the medium containing of soluble osteoclast differentiation Factor (sODF) showed similar phenotype with MDBM-derived osteoclasts, those are mRNA expression pattern of osteoclast-specific genes, TRAP(+) MNCs generation, and bone resorbing abivity. Formation of resorption pits by osteoclastic MNCs differentiated from sODF-treated RAW264.7, was completely blocked by the addition of osteoprotegerin (OPG), a soluble decoy receptor for ODF, to the sODF-containing culture me야um. The effects of PDLF on differentiation of RAW264.7 into the TRAP(+) multinucleated osteoclast-like cells were examined using coculture system. PDLF were fxed with paraformaldehyde, followed by coculture with RAW264.7, which induced formation of TRAP(+) MNCs in the absence of additional treatment of sODF. When compared with untreated and fixed PDLF (fPDLF), IL-1 ${\beta}$-treated, or lipopolysaccha-ride-treated and then fixed PDLF showed two-folld increase in the supporting activity of osteoclastogenesis from RAW264.7 coculture system. There were no TRAP(+) MNCs formation in coculture system of RAW264.7 with PDLF of no fixation. These findigs suggested that we can replace the primary hematopoietic preosteoclasts for RAW264. 7 cell line for studying the mechanism of PDLF-induced osteoclastogenesis, and we hypothesize that PDLF control osteoclastogenesis through ODF expression which might be enhanced by inflammatory signals.

This article presents the effects of estimation activities related to speed and density on students' concept-understanding, answer-prediction, and answer-calculation in problem solving procedure with quantitative and qualitative methods. Participants were one hundred and ninety two seventh graders from one coeducational school in Seoul. Half of them participated in the estimation activities and the other half did in the measurement activities. Discussions of three students during estimation activities on density and their post-interviews were tape-recorded. Pre- and post-assessment scores were analyzed for the whole classes, and students' discussions and interviews served this research as evidences for the case analysis. Results of scores indicated that students in the estimation activities were significantly better than those in the measurement activities for predicting answers, but not for understanding concepts. Analysis of the cases revealed that estimation activity helped them to understand the relations of mass, volume and density, empirically, which enhanced their prediction ability. Furthermore, the ability could help a student with low calculation ability to comprehend the calculation problems. Thus, it is concluded that estimation activities could influence students' empirical learning on quantitative concepts, which enhanced their prediction ability.
