Separation of Protein and Fatty Acids from Tuna Viscera Using Supercritical Carbon Dioxide

  • Supercritical carbon dioxide extraction was investigated as a method for removing lipids and bad flavor from tuna viscera. To find the optimum conditions, different experimental variables, such as pressure, temperature, flow rate of solvent and sample size, were evaluated for the effective removal of lipids and the undesirable smell. Ethanol was used as the entrainer, with a $3\%$ by vol $CO_2$ flow rate. By increasing the pressure at constant temperature, the efficiency of the lipid removal was improved and the protein was concentrated without denaturalization. The main fatty acids extracted from the tuna viscera were palmitic acid (16:0), heptadecanoic acid (17:1), oleic acid (18:1) and docosahexaenoic acid (22:6). The major amino acids in the tuna viscera treated by supercritical carbon dioxide were glutamic acid, leucine and lysine, and the free amino acids were L-proline, taurine and L-$\alpha$-aminoadipic acid.

Biological Activities of Low Molecular Weight Substances Fractionated from the Seasoned and Smoked Products of Skipjack Tuna (Euthynus pelamis) (가다랑어 훈연조미제품 유래 저분자물질의 생리활성)

  • We studied the anticancer, antioxidative, ACE inhibitory, antithrombic, and cerebral ischemia inhibitory activities, of low molecular weight substances fractionated from the seasoned and smoked products of skipjack tuna (Euthynus pelamis). Low molecular weight substances fractionated from the liquid extract of seasoned and smoked skipjack tuna powder showed 7 peaks on gel permeation chromatography. Anticancer activity was the highest at Peak 3 $(54.23%)$ followed by Peak 4 $(41.58\%)$. The potency order of antioxidative activity was the highest at Peak 3 $(94.14\%)$ followed by Peak 2 $(82.07\%)$ and Peak 5 $(81.24\%)$. ACE inhibitory activity was the highest at Peak 3 $(67.27\%)$ followed by Peak 5 $(45.62\%)$. But there was weak anti thrombotic effect (PT and APTT) in the low molecular weight substances of the seasoned and smoked products of skipjack tuna. Cerebral ischemia inhibitory activity was the highest at Peak 3 $(80.0\%)$ followed by Peak 5 $(40.0\%)$.

Fishing efficiency of Korean regular and deep longline gears and vertical distribution of tunas in the Indian Ocean (인도양에서의 한국재래식 및 심층연승의 어획효과와 다랑어류의 연직분포)

  • Yellowfin and bigeye tunas have been targeting and the most important species for the Korean tuna longline fishery in the Indian Ocean. This study is aimed to analyse the fishing efficiency of the regular and the deep longlines and the vortical distribution of tunas, and the weight composition by fishing depth based on the data from Korean tuna longline fishery from 1973 to 1980 and from 1984 to 1986 in the Indian Ocean. It was found that the deep longline gear on bigeye tuna was significantly different from the regular longline gear on yellowfin tuna in the whole Indian Ocean. Yellowfin tuna and billfishes were chiefly distributed at the shallow layer and bigeye at the deep layer. The weight composition of yellowfin and bigeye tunas by depth showed that the deeper the depth, the larger the bigeye distributed.

Bioadhesive Characteristics of Biofilm Manufactured from Gelatin Derived from Acetic Acid-treated Skin of the Yellowfin Tuna Thunnus albacares (아세트산처리 황다랑어(Thunnus albacares) 껍질 유래 젤라틴으로 제조한 바이오필름의 생체 접착 특성)

  • This study aimed to demonstrate the bioadhesive characteristics of gelatin biofilm to rat skin. The biofilm was manufactured from gelatin extracted from the acetic acid treated-skin of the yellowfin tuna Thunnus albacares. The bioadhesive strength of tuna gelatin biofilm was compared to that of porcine gelatin biofilm. The tuna gelatin biofilm exhibited a higher bioadhesive strength than the porcine gelatin biofilm. Gelatin biofilm was subjected to glutaraldehyde treatment at different concentrations, temperatures and pH in order to improve its bioadhesive strength. Glutaraldehyde treatment improved the bioadhesive strength of gelatin biofilm up to three-fold. The bioadhesive strength of glutaraldehyde treated-biofilm was significantly decreased by application of sodium borohydride.

Analysis of Indonesian Tuna Fish Export to Twelve Main Destination Countries: A Panel Gravity Model

  • Purpose: This study purposes to analyze the determinants of the volume of Indonesian tuna exports. Research design, data and methodology: The framework was developed from the gravity model for trade, which was expanded with additional variables of competitiveness, exchange rate, and industrial share of the destination country. The data sources used in this study are UN Comtrade and the World Bank. The data used is yearly data from 12 countries in 2001-2019. The scope of the study is limited to exports to the twelve main export destinations. Panel data regression analysis is used to determine the factors that affect the volume of Indonesian tuna exports. Results: The results show that according to the theory, Indonesia's GDP has a positive effect and economic distance has a negative effect on the volume of the exports. Meanwhile, the GDPs of the destination countries are not proven to have a positive effect. However, the higher the industrial share in the country, the higher the export volume tends to be. Conclusions: The conclusion obtained from this study is that Indonesia's GDP, economic distance, real exchange rate, industrial GDP share of the destination country, and the RCA index affect the volume of Indonesian tuna exports.

Studies on the Heavy Metal Contents in Canned Fishes (어류통조림중의 중금속 함량에 관한 연구)

  • This study was carried out to determine the contamination level of heavy mentals in canned fishes on the market. Contents of tin, lead, iron and zinc were analyzed by Atomic Absorption Spectrophtometer. Contents of total mercury were determined by mercury analyzer. The results were as follows. 1. The highest value of tin was $1.7706{\pm}0.3216$pprn in canned yellow pin & skip jack(mix) tuna, the lowest was 1.2109 ${\pm}$ 0.0769ppm in canned mackerel pike. 2. The highest value of lead was 0.1657 ${\pm}$ 0.0530 pprn in canned yellow pin tuna, the lowest was 0.0335k 0.0100 pprn in canned mackerel pike. 3. The highest value of iron and zinc was 22.5943 ${\pm}$ 0.7945 ppm and 13.4556 ${\pm}$ 0.6774 pprn in canned sardine fish, the lowest was 4.3514 ${\pm}$ 0.3607 pprn and 3.8773 ${\pm}$ 0.1914 pprn in canned albacore tuna. 4. The total mercury contents was detected 0.0531~0.3386 pprn in three kinds of canned tuna and could not be detected with the others.

Study on Acute Subcutaneous Toxicity of Hydroxyapatite Sinter Produced from Tuna Bone in Sprague-Dawley Rats (참치뼈로부터 제조한 Hydroxyapatite 소결체의 랫드에 대한 급성피하독성시험)

  • This study was performed to evaluate the actue sbucutaneous toxicity of hydroxyapatite sinter produced from tuna bone in Sprague-Dawley(SD) rats. Hydroxyapatite sinter was administrated at dose levels of 5000, 2500, 1250, 625, 312.5 and 0 mg/kg. After single subcutaneous adiminstration to both sexes to both sexes SD rats, we observed rats for 14 days. Hydroxyapatite sinter did not induce any toxic signs inmortalities, clinical findings, body weights and gross findings of the rats. In view of result, it was impossible to estimate LD/ sub 50/ values in SD rats. In conclusion, these results suggest that hydroxyapatite sinter produced from tuna bone has no effect on acute subcutaneous toxicity in SD rats.

Evaluation of Mucous Membrane lrritation by Hydroxyapatite Sinter Produced from Tuna Bone in Syrian Hamsters (참치뼈로부터 Hydroxyapatite 소결체의 햄스터에 대한 구강점막자극시험)

  • In order to evaluate the safety of hydroxyapatite sinter produced from tuna bone in Syrian hamsters, oral mucous membrane irritation test was carried out. Oral mucous membrane irritation test was infected in Syrian hamsters as dose of 5 g/kg BW with hydroxyapatite sinder under pentobarbital sodium anesthesia. Each animal`s left cheek pouch was everted, and the samples were loosely placed at the bottom of the pouch with a double-suture technique for 14 days. Hamsters of control group were treated without inserting the hydroxylapatite sinter. Any abnormal clinical signs in both cheek pouches of control and treatment group were not observed for 14 days. There were no significant differences in body weight changes between hamsters of control and treatment group. Therefore, it suggest that hydroxyapatite sinter produced from tuna bone has no particular changes of oral mucous membrane irritation in Syrian hamsters.

Sintering Properties of Hydroxyapatite Derived from Tuna Bone

  • Hydroxyapatite (HA) ceramic was prepared using tuna bone powders and was characterized on properties of sintered samples. The aim was to prepare dense and bulk HA sintered suitable for clinical applications. Thermal analysis, X-ray diffraction analysis, microscopic observation, and mechanical evaluation were applied to characterize sintered HA to find optimum processing condition. The major phase of sintered HA was observed up to 1300$\^{C}$ and $\beta$-TCP was observed as temperature increased. The density of sintered HA was increased up to 1350$\^{C}$ and decreased with further increasing temperature. The flexural strength of sintered HA at 1350$\^{C}$ showed 58 MPa as the highest value in this work. The results suggest that the crystal phase and transformation characteristics of HA prepared from tuna bone are the same as the behavior of HA powders made by chemical synthesis.

  • This paper conducted research on identifying the process of change in fishing gear and organizing the function of periodically-used fishing gear types through net plan and computer simulation by selecting the design of the four types of fishing gear used for the Korea tuna purse seiner in the Western and Central Pacific Ocean, which 1,000 G/T class and whose length over all 60 m class. In the late 1980s, the length of the tuna purse gear was 1,939 m and the design depth was 160 m, but currently, the length and the design depth are 2,515 m and 230 m, respectively. As a result of the simulation, the expansion of the fishing gear increased buoyance, sinking force, sinking depth, surrounded area, and purse wire continuously. Recently, the maximum tension of the currently used purse wire of tuna purse seiner is 23.5 tons and is close to 25.4 tons which is the maximum lift capacity of WS454 winch. The way to improve fishing gear should be proceeded to increase sinking speed rather than expand the size of fishing gear.
