Biological Fixation of Carbon Dioxide by Synechocystis PCC 6803 (Synechocystis PCC 6803에 의한 이산화탄소의 생물학적 고정화)

  • Carbon dioxide is estimated to be responsible for 60% of the global warming effect, and this percentage is tending upward. Studies on removal and fixation of $CO_2$ in the flue gas are recognized as one of the important roles of the future biotechnology. Photobiological systems have considerably higher photosynthetic efficiency than conventional biomass system. The experiment for the photosynthetic fixation of $CO_2$ and the biomass production was performed with various initial cell concentration in a tubular photobioreactor and a bubble column $CO_2$ contactor with a gas sparger of $CO_2$ -enriched air(0.03~20%). Synechocystis PCC 6803 could grow at 10~20% $CO_2$ content under pH control. The highest specific growth rate, 0.0258 $h^{-1}$ , was obtained at 5% $CO_2$-air mixture. The maximum cell production rate, 0.2784 g/L.day, was obtained when the initial cell concentration was 0.45 g/L at 5% $CO_2$ -air mixture. The maximum cell concentration was 2.03 g/L in the tubular photobioreactor when the light intensity was $45.5{\mu}$ $E/m^2$ . s. This system showed 0.482 g $CO_2$ /L . day of the $CO_2$ fixation.

Paricalcitol attenuates indoxyl sulfate-induced apoptosis through the inhibition of MAPK, Akt, and NF-κB activation in HK-2 cells

  • Background/Aims: Indoxyl sulfate (IS) is a uremic toxin and an important causative factor in the progression of chronic kidney disease. Recently, paricalcitol (19-nor-1,25-dihydroxyvitamin D2) was shown to exhibit protective effects in kidney injury. Here, we investigated the effects of paricalcitol treatment on IS-induced renal tubular injury. Methods: The fluorescent dye 2',7'-dichlorofluorescein diacetate was used to measure intracellular reactive oxygen species (ROS) following IS administration in human renal proximal tubular epithelial (HK-2) cells. The effects of IS on cell viability were determined using MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays and levels of apoptosis-related proteins (Bcl-2-associated protein X [Bax] and B-cell lymphoma 2 [Bcl-2]), nuclear $factor-{\kappa}B$ ($NF-{\kappa}B$) p65, and phosphorylation of mitogen-activated protein kinase (MAPK) and protein kinase B (Akt) were determined by semiquantitative immunoblotting. The promoter activity of $NF-{\kappa}B$ was measured by luciferase assays and apoptosis was determined by f low cytometry of cells stained with f luorescein isothiocyanate-conjugated Annexin V protein. Results: IS treatment increased ROS production, decreased cell viability and induced apoptosis in HK-2 cells. IS treatment increased the expression of apoptosis-related protein Bax, decreased Bcl-2 expression, and activated phosphorylation of MAPK, $NF-{\kappa}B$ p65, and Akt. In contrast, paricalcitol treatment decreased Bax expression, increased Bcl-2 expression, and inhibited phosphorylation of MAPK, $NF-{\kappa}B$ p65, and Akt in HK-2 cells. $NF-{\kappa}B$ promoter activity was increased following IS, administration and was counteracted by pretreatment with paricalcitol. Additionally, flow cytometry analysis revealed that IS-induced apoptosis was attenuated by paricalcitol treatment, which resulted in decreased numbers of fluorescein isothiocyanate-conjugated Annexin V positive cells. Conclusions: Treatment with paricalcitol inhibited IS-induced apoptosis by regulating MAPK, $NF-{\kappa}B$, and Akt signaling pathway in HK-2 cells.

Hepatic and renal toxicity study of rainbow trout, Oncorhynchus mykiss, caused by intraperitoneal administration of thioacetamide (TAA) (티오아세트아미드(thioacetamide) 복강투여로 인한 무지개송어, Oncorhynchus mykiss의 간장 및 신장 독성 반응 연구)

  • In veterinary medicine for mammals, studies are being conducted to confirm the effects of antioxidants using pathological toxicity model studies, and are also used to confirm the effect of mitigating liver or kidney toxicity of specific substances. It was considered necessary to study such a toxicity model for domestic farmed fish, so thioacetamide (TAA), a toxic substance that causes tissue damage by mitochondrial dysfunction, was injected into rainbow trout (Oncorhynchus mykiss), a major farmed freshwater fish species in Korea. The experiment was conducted with 40 rainbow trout (Oncorhynchus mykiss) weighting 53 ± 0.6 g divided into two groups. Thioacetamide(TAA) 300mg/kg of body weight was intraperitoneally injected into rainbow trout and samples were taken 1, 3, 5, 7 days after peritoneal injection. As a result, in serum biochemical analysis, AST levels related to liver function decreased 3 and 5 days after intraperitoneal injection and increased after 7 days, and ALT levels also increased after 7 days. In addition, creatinine related to renal malfunction increased 3 and 5 days after TAA injection. In histopathological analysis, pericholangitis and local lymphocyte infiltration were observed in the liver from 1 day after intraperitoneal injection of TAA, and hepatic parenchymal cell necrosis was also observed from 3 days after intraperitoneal injection. Hyaline droplet in renal tubular epithelial cell was observed from 1 day after TAA injection, and acute tubular damage such as tubular epithelial cell necrosis appeared from 3 days after TAA injection. Accordingly, it is thought that it will be able to contribute to studies that require a toxicity model.

  • A tubular tormentor was prepared by packing the wood chips and pumping the yeast solution of Saccharomyces formosensis in a tubular column. Investigations to characterize the ethanol fermentation in the immobilized cell tubular fermentor and to compare such a fermentors with other type fermentors were undertaken. Ethanol productivity of 24.4g EtOH/$\ell$.hr has been obtained from glucose substrate. This productivity is higher or compared favourably with that reported in immobilized bio-reactors.

Expression of Bradykinin Binding Sites within the Mammalian Kidney Tissues (신장 조직의 브라디키닌 결합부위 발현)

  • Based upon the previous experiments showing that kidney and lung tissues of rat had relatively abundant bradykinin binding sites, we tried to characterize and determine the densities of the bradykinin binding sites in the rabbit kidney tissue and proximal tubular cells under different growing conditions. Among the kidney tissue renal medulla segments showed the highest bradykinin binding sites. To determine which growth factors are to add in the serum free culture medium to express selectively the bradykinin binding sites in the rabbit kidney proximal tubular cells, we tried so called hormone-deletion approach and in here insulin, hydrocortisone, transferrin, triiodothyronine and prostaglandin $E_1$ are examined. By performing receptor binding assay and determination of protein concentrations, we may conclude that the most required hormones in the expression for bradykinin binding sites are insulin and transferrin, and fetal bovine serum is shown to be less effective in this regard.

  • The ultrastructure of the gills of Oplegnathus fasciatus was examined by means of light and transmission electron microscopes. The gills have primary and secondary filaments (lamellae). The following cells are identified and described : epithelial cell, pillar cell, chloride cell and mucose cell. The simple epithelial layer consists of squamous epithelium containing a large nucleus and the surface is covered with some of microridges. The lamella pillar structures are characterized by axial microtubules and lateral membrane interdigitations. Chloride cells contain a lot of mitochondria and specifically developed tubular systems. The rough endoplasimic reticulum and golgi complex, and some of mucous granules were observed in immature mucous cells. The mature mucous cells were AB-PAS positive, globular in shape, and had mucous granules of similar size with various electron densities.

  • Performance changes of an anode-supported tubular SOFC including current collectors are analyzed at different current collecting methods using numerical simulation. From the two dimensional numerical model of the solid oxide fuel cell with nickel felts as anodic current collectors and silver wires as cathodic ones, the performance curves and the distributions of temperature, concentration, current density are obtained. Also, the voltage loss of the cell is divided into three parts: activation loss, concentration loss and ohmic loss. The results show that the performance change of the cell is dominantly influenced by the ohmic loss. Although the temperature and concentration distributions are different, the total activation loss and concentration loss are nearly same. And the ohmic loss is divided into each parts of the cell components. The ohmic loss of the anodic current collectorreaches about 60~80% of the cell's total ohmic loss. Therefore, the reduction of the ohmic loss of the anodic current collector is very important for stack power enhancement. It is also recommended that the load should be connected to the both ends of the anodic current collector.

  • The movement of water across cell plasma membranes occurs in all cell types but is particularly rapid in erythrocytes, renal tubular cells. In principle, osmotic/oncotic gradients and hydrostatic pressure difference can drive water across a cell layer by transcellular or paracellular pathways. The aquaporin family of molecular water channels, which now number 10 in mammals and many more in plants and lower organism, are likely to provide a molecular pathway for water transport in some cell membranes.(omitted)
