The purpose of the present study is to measure the effect of Tuber Pinelliae on EP(epinephrine) induced contraction of isolated rat thoracic aorta. Contractile force was measured with force displacement transducer under 1.5g loading tension. The mean percent increasing of rCBF was 11.4% (p<0.05) after 10㎎/㎏ Tuber Pinelliae. Contractions evoked by EP (ED50) and KCI 65.4mM were decreased significantly by Tuber Pinelliae. L-NNA, ODQ and atropine significantly altered the effect of Tuber Pinelliae, but propranolol and indomethacin did not change the relaxation of Tuber Pinelliae. These results indicate that Tuber Pinelliae can relax EP and KCI induced contraction of isolated rat thoracic aorta and that this decreasing contraction related to epithelium, nitric oxide, and parasympathetics.
Objectives : A morphological classification among Pinelliae Tuber, Typhonii Flagelliformis Rhizoma and Arisaematis Rhizoma was made through microscopic observation. Method : The slice of the tested material made by paraffin section technique was colored with Safranine Malachite Green contrast methods, and then observed and photographed by olymphus-BHT. Result : 1. Pinelliae Tuber is like a ball with a $1{\sim}1.5cm$ diameter, with a concave apex and pockmarked root trace around. 2. Typhonii Flagelliformis Rhizoma is somewhat oval or like a cone, with a $4{\sim}8mm$ diameter, a $7{\sim}15mm$ height, a projected round apex, and a pin-pointed back. 3. Arisaematis Rhizoma is somewhat flat round with a $2{\sim}2.5cm$ diameter, a little than Pinelliae Tuber, and its apex has incomplete lobopodium and root trace. 4. Fundamental tissues of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma have starch grains and mucilage cells, and their mucilage cells include the raphides of calcium oxalate. Conclusion : Internal forms of Pinelliae Tuber, Typhonii Flagelliformis Rhizoma, and Arisaematis Rhizoma are difficult to distinguish from one another, but Pinelliae Tuber and Arisaematis Rhizoma can be distinguished by their external forms and sizes, and Pinelliae Tuber and Typhonii Flagelliformis Rhizoma were distinguished by the forms of apex.
Purpose: Endometriosis has been thought to be related with blood stasis in uterus. Pinelliae Tuber is a herb which has effectiveness of dispelling phlegm which can be transformed into blood stasis, so is used in gynecology blood stasis-desease. Therefore in the present study, the effects of Pinelliae Tuber on endometriosis were investigated. Also retention enema therapy acts on the limited part directly, so is used frequently in gynecology clinic. This study was to examine the comparative effects on the surgical induction of endometriosis in the rat between orally and rectally administered with Pinelliae Tuber extract. Methods: The endometrial tissue was autografted to Rat's small intestine. Rats with surgically induced endometriosis were orally and rectally administerd with Pinelliae Tuber extract for 40 days. Size of ectopic uterine implants at the serosal wall and concentration of progesterone, estradiol, $TNF-{\alpha}$ and IL-2, IL-4, IL-6, IL-I0 in serum were examined and compared with the control group. Results: There was no difference in the size of ectopic uterine implants of experimental group compared with control group. The concentration of IL-10 level was significantly decreased in orally administered experimental group and the concentration of progesterone, IL-6 level was significantly decreased in rectally administered experimental group. There was no significant difference between orally and rectally administerd with Pinelliae Tuber. Conclusion: Judging from the above results, it can be suggested that Pinelliae Tuber should be not a useful agent for inhibiting the proliferation of uterine endometrial tissue. There is no difference between orally and rectally administerd with Pinelliae Tuber extract.
Objects : The daily dose of Pinelliae Tuber in ≪Treatise on Cold Damage Diseases≫ is half seung in volume, two and half ryang in weight, and fifteen in total number. But the daily dose should be a whole number. So I found out the background of this setting and correct solution. Methods : I searched Classics of Traditional Medicine, found out the background of the daily dose setting, solution. Results : The daily dose of Pinelliae tuber in ≪Hangdi's Internal Classic Miraculous Pivot≫, ≪Bohenggyuljangbuyongyakbeobyo≫ is half seung. ≪Treatise on Cold Dameage Diseases≫ followed the same daily dose of that because it referred to these books. In ≪Synopsis of Prescription of the Golden Chamber≫, the daily dose of that is half seung, one or two seung. The half seung of the Pinelliae Tuber is thirty three mL, but the diameter is 1~1.5 cm that accurate measurement by volume is difficult. The daily dose by weight is correct considering the unity of marking of the daily dose, accuracy of measurement, the fact that Pinelliae Tuber is currently distributed by cutting. So, two ryang is correct which is thirteen gram. Conclusions : Considering the traditionality, the convenience of measurement, the daily dose of Pinelliae Tuber in the ≪Treatise on Cold Damage Diseases≫ is half seung, but considering the unity, accuracy, current state of distribution, it is correct that the daily dose of it is two ryang. It corresponds to thirteen gram.

Objectives : The purpose of this study is to report that applying the genetic discrimination method to Pinelliae Tuber is suitable as a countermeasure for the limitations of morphological identification announced publicly in the Ministry of Food and Drug Safety(MFDS). Methods : Randomly selected fifty samples in Pinelliae Tuber imported from China were used for morphological and genetic identification. The morphological identification was applied method announced publicly by the MFDS. The traits of morphological identification were classified as Pinellia ternata, P. tripartita, Pinellia pedatisecta, and Typhonium flagelliforme, according to the formation of tuberous root and tuber morphology. The genetic identifications were conducted by Sequence Characterized Amplified Region(SCAR) marker and DNA barcoding analysis for cross-validation, respectively. SCAR marker was verified according to the presence or absence of amplicon through PCR amplification using species-specific primers. DNA barcoding analysis used sequence information of the matK region. Results : As a result of the morphological identification, 27 out of 50 samples were identified as original species 'P. ternata' of genuine 'Pinelliae Tuber', and 23 were identified as adulterant species 'P. pedatisecta'. Unlike this, the genetic identification was identified as the original species 'P. ternata' in all 50 samples in the SCAR marker and matK regional sequence analysis. Conclusions : Pinelliae Tuber of morphological mutant that can not be classified by morphological identification is imported from China. The SCAR marker would be used as accurate and efficient assays for species identification of the morphological mutant.

Objectives : Pinelliae Tuber has been used as a typical unauthentic herbal medicines. Due to the morphological similarity between Pinelliae Tuber and adulterants, the correct authentication is very difficult. Therefore, we introduced DNA barcode to establish a powerful tool for the authentication of Pinelliae Tuner from adulterants. Methods : To obtain DNA barcode regions, genomic DNA was extracted from nineteen specimens of Pinellia ternata, Pinellia pedatisecta, Pinellia tripartita, and Typhonium flagelliforme, and matK and rbcL genes were amplified. For identification of species specific sequences and analysis phylogenetic relationship, a comparative analysis were performed by the ClastalW and UPGMA based on entire sequences of matK and rbcL genes, respectively. Results : In comparison of two DNA barcode sequences, we elucidated the phylogenetic relationship showing distinct four groups depending on species and identified 40 and 20 species specific nucleotides enough to distinguish each species from matK and rbcL gene, respectively. The sequence differences at the corresponding positions were avaliable genetic marker nulceotides to discriminate the correct species among analyzed four species. These results indicated that phylogentic and comparative analysis of matK and rbcL genes are useful genetic markers to authenticate Pinelliae Tubers. Conclusions : The marker nucleotides enough to distinguish P. ternata, P. tripatrita, P. peditisecta, and T. flagelliform, were observed at 40 positions in matK gene and 20 positions in rbcL gene sequence, respectively. These differences can be used to authenticate Pinelliae Tuber from adulterants as well as discriminate each four species.
MeOH extracts of sixteen herbal drugs were tested for the effects on bovine adrenal tyrosine hydroxylase and dopamine ${\beta}-hydroxylase$. The MeOH extracts of Paeoniae Radix and Pinelliae Tuber showed 65% inhibition on the tyrosine hydroxylase activity at the concentration of 100 $\mu$g in the enzyme reaction mixture. Those of Paeoniae Radix, Pinelliae Tuber and Evodiae Fructus showed 87, 84 and 62%, respectively, on the dopamine ${\beta}-hydroxylase$ activity.
This study intends to report the significance of several experimental results obtained from analysing the genes extracted from the plants and herbal medicine such as P. temalta (Thunb.) Breit, A. amurense var serratum Nakai, A. erubescens (Wall.) Schott, Pinelliae Tuber and Arisaematis Tuber, mainly by the method of RAPD(randomly amplified polymorphic DNA) and the method of RFLP(restriction fragment length polymorphism) on ITS(internal transcribed spacer) region. Genomic DNA could be extracted from both original plants and dried materials. DNA fragments of P. temata kind and A. amurense kind showed the same aspect separately within the same species under the method of RAPD using random primer, while various aspects(polymorphism) were discovered among different species. In RAPD analysis by uniprimer, common bands were extracted from all types of P. temata in the case of uniprimer #4, which were distinguished from the kind of A. amurense. Other polymorphic bands appeared in between different A. amurense species as well. In the case of uniprimer #11, particular band came out in the kind of P. temata. On the other hand, in the case of uniprimer #5, #6, and #8, various bands(polymorhism) were revealed in both kinds of P. temata and A. amurense. Although further study is needed to ascertain whether these results are due to the differences of species, kinds, or growing place, the results could be used as a scientific method of identifying the substitutes for A. amurense genus. The author believes that as if P. temata class of plants used in this experiment are different among themselves in terms of the shape, size and property, those are clearly a class of P. temata or belong to the same genus.

Objectives : The processing of Pinelliae Tuber and Arisaematis Rhizoma is a crucial step to reduce the severe acrid irritation mainly due to the needle-like crystals (raphides). Ginger, alum and bile juice have been used as adjuvant materials for the processing. Methods : Bibliographic research on ancient processing and experimental processing was performed to investigate the toxicity reduction mechanisms of the processing with ginger, alum and bile juice. Results : Ginger has been a major adjuvant for the processing of Pinelliae Tuber, followed by alum and bile juice since Song (宋) and Myeong (明) dynasties, and Arisaematis Rhizoma has been mainly used as Damnamseong (膽南星). The raphides consisting of calcium oxalate, lectin, agglutinin and polysaccharides can induce acrid irritation and the inflammatory reactions. The lipophilic components in the ginger denatured the structure of raphides and 6-gingerol-contained ginger extract attenuated the inflammatory reaction. The calcium ion (Ca2+) of calcium oxalate was substituted to the aluminium ion (Al3+) of the alum, which damaged the calcium oxalate structure. Lectin attached to the surface of raphides was dissolved in alum solution and consequently its structure was denatured. The cholate in the bile juice formed the complex with the oxalate anion or the calcium cation. Moreover, the enzymes activated by Lactobacillus or Bifidobacterium during the fermentation promoted the fragmentation of oxalate. Conclusion : The adjuvant materials damaged the raphides by denaturing or degrading the calcium oxalate, resulting in the reduction of acrid irritation. Further experimental studies would support the toxicity reduction mechanism of the processing.

A mouse whole animal bioassay was employed to screen for potential mutagenicity of ethanol/water extracts of 16 Chinese herbal drugs that are commonly prescribed in Korea. Specific cytogenetic toxicity was measrured by recording evidence of clastogenesis toxicity was measured by recording evidence of clastogenesis via the mouse bone marrow micronucleus test. Male ICR mice administered ethanol extract of Pinelliae tuber (Pinellia eternata Breitenbach, ARACEAE, 양복) and ddY female mice administered extract of Angelica Koreanae radix(Angelica Koreana Maximowicz, UMBELLIFERAE, ) (both by oral administration, at a dose of 600 mg/kg), in a short-term dosing schedule, demonstrated significant increase in micronucleated polychromatophilic erythrocytes, indicating the increase of clastogenicity.
