Interspecific somatic hybrids were obtained by protoplast fusion between Pleurotus ostreatus and Pleurotus sajor-caju. The fusion products between incompatible strains did not form clamp connections. Fruiting body of the clampless fusants was induced by light-dark cycle on saw-dust-rice bran substrate in glass bottles. Out of them, seven somatic hybrids produced fruiting bodies of intermediate morphology of the two species. Light and low temperature were the initiating factors for the development of clamped hyphae from the clampless mycelial colonies. All of these basidiocarps had clamp connections. Eight fusants from the six crosses were analysed with the segregation of genetic characters by random spore isolates. In the three combinations, unexpected alleles were shown. Somatic hybrid between P188 (P. ostreatus 2-1 + P. sajor-caju 2-53) and P. florida 2-3 by triple cross produced fruiting bodies similar to those of fusant between P. ostreatus and P. florida. All the genetic charaters from the three strains were shown to segregate and recombine.
This study has been conducted to estimate the proper amount of phosphatic fertilizer required to increase-yield in growing soybeans on newly reclaimed-sterile land which is short of phosphatic fertilizer. IKSAN, the variety chosen for this study, was planted, using the plant spacing $60cm{\times}30cm$. Phosphatic fertilizer was applied in 4 levels in none, standard, double, and triple amount. and at the same time, nitrogen and postassium were applied only in standard amount. Randomized block design was applied in arranging the experimental plots. A series of results acquired are summarized as follows. 1. The fact was recognized that only the standard amount of phosphatic fertilizer is effective for increasing the pod numbers and the soybean yield per l0a. 2. The difference in stem length and stem diameter was very remarkable even among individual plants in the same plot, Throughout all four of the plots there were difference in stem length and stem diameter. That is to say, any invariable tendency on them due to the amount of phosphatic fertilizer applied could not be observed at all. 3. It was recognized that amount of phosphatic fertilizer applied did not have a considerable influence upon the weight of the seeds and the number of seeds contained in 0.l8ι. 4. It was observed that the weight of 1000 seeds was strikingly increased as far as double the amount phosphatic fertilizer.
This study was carried out to know the effect of seed position on the size, contents of ginsenosides, free sugars, and fatty acids in ginseng seeds. Seed positions were classified by the three portions as center, middle and border in a seed cluster. Seed weight at center was light remarkably in comparison with those of seeds of at border and middle. The weight of embryo plus endosperm was in same tendency as seed weight. Percentage of single-seeded berry was smaller than that of the double-seeded, and the triple-seeded was rare. The percentage of the single-seeded increased from the border to the center. Size of the single -seeded seed was smaller than that of the double- seeded. Rate of dehiscence did not differ among different seed positions. The major ginsenosides in seed were Re, Rb$_1$, and Rb$_2$. The contents of Rb$_2$ and total saponin were highest in border, least in center, but reversed in Re and Rd. Major free sugars in seed were sucrose and glucose. The sucrose content was gradually decreased according to the seed position from border to center. Major fatty acids in the seed were oleic and linoleic acid. Contents of palmitic and linolenic acid were different according to the seed position.
Effective estrus detection and artificial insemination (AI) are necessary for profitable management of dairy herd. In current study, 45 crossbred lactating cows have been selected with the complaint of unobserved oestrus for more than sixty days postpartum. All cows had functional corpus luteum as examined by transrectal ultrasonography. Cows were treated with $PGF_2{\alpha}$ analogue and AI was performed with observed oestrus and then single dose of GnRH was administered. Similar synchronization protocol has been repeated after 14 days in cows that did not repose to first treatment. Remaining cows received additional $PGF_2{\alpha}$ after 14 days of second treatment and timed AI was performed following GnRH administration. Among 45 cows, 28.89% showed estrus after first treatment and 78.79% responded to second hormonal intervention. A higher conception rate (88.89% vs 26.66 and 72.72%) was observed in cows after triple administration of $PGF_2{\alpha}$ and timed AI. We noticed a significant differences in body condition score (BCS, 1~5 scale), postpartum period, and daily milk production between cows that either responded of non-responded following first and second hormonal treatment. In addition, there was a significant positive correlation between daily milk production and BCS, age and postpartum days, milk production and estrus/BCS, and milk production/BCS/estrus and conception rate. Depending upon the findings we conclude that hormonal intervention with $PGF_2{\alpha}$ and GnRH enhances postpartum ovarian cyclicity and help decreasing the days open of dairy herd. Therefore, this finding might provide an excellent guideline for target breeding system for profitable dairy herd management.
Purpose: The purpose of this study was to evaluate the effectiveness of three functional performance tests and one-legged hop test and to establish the reference data of the three functional performance tests of the active young Korean men. Materials and methods: We performed five tests on 40 normal subjects such as one-legged hop test (single hop, triple hop) and the three functional performance tests (carioca test, co-contraction test, shuttle run test). The mean age was 28.4 years. The mean score of Tegner activity scale was 6.2 and Lysholm score 98.2 Mean value and standard deviation were obtained. The correlation of age, height and weight with the results, the relationship between FPT and one-legged hop test and the mean difference between dominant and non-dominant limbs were statistically calculated. Results: All data obtained were normally distributed and there were no significant relationship between age, height and weight. The paired t-test of one legged hop test showed no statistical significance between dominant and non-dominant limbs. The results of one legged hop test had strong relationship with functional performance test. Conclusion; The three functional performance tests can be a useful functional test in ACL-insufficient athletes. Our results can be used as normal reference data of the normal active Korean men.

Herpes simplex virus, Varicella zoster virus and Human herpes virus-6 caused central nervous system infections and latent infections but there is no data of the 3 viruses being tested from the same cerebrospinal fluid samples with aseptic meningitis or encephalitis in adults patients. These viruses produced similar neurologic symptoms but difficulties existed in differentiating of etiologic agents and therefore the viruses needed to be detected in the early state. Herpes simplex virus encephalitis (HSVE) in adults, if not treated promptly was fatal. If treated with antiviral drugs in the early phase of encephalitis, neurologic sequales decreased by 65%. Recently, a PCR method for detection of HSVE with CSF was developed. VZV primary and secondary infections caused neurologic symptoms of encephalitis or meningitis. The second frequency of adult encephalitis that caused VZV were reported. HHV-6 caused CNS latent infection that was studied with normal adults brains. But there is no data of HSV, VZV and HHV-6 for aseptic meningitis and encephalitis of Korean adults through etiologic study. We cultured CSFs on HEp-2 cells and simultaneously tested for HSV PCR, VZV nested PCR and HHV-6 PCR with 8 specific primers. The PCR results of CSF from meningitis Korean adults were 13/19 (68.4%) for HSV, 10/19 (52.6%) for VZV and 12/19 (63.2%) for HHV-67/19 (36.8%) cases were triple infected HSV PCR, VZV PCR and HHV-6 PCR positive; 3/19 (15.8%) cases were dual infected HSV PCR and HHV-6 PCR positive; 1119 (0.5%) cases was VZV PCR positive. Strong viral DNA amplification of CSF means a causative virus may be present in aseptic meningitis or encephalitis patients and may cause clinical neurologic symptoms. HSV and HHV-6 viruses detection rate were higher than VZV by PCR with CSFs.
A potyvirus was isolated from cultivated Iris plants showing leaf streak mosaic symptom. Reverse transcription and polymerase chain reaction (RT-PCR) product of 1 kb long which encoded partial nuclear inclusion B and N-terminal region of viral coat protein (CP) genes for potyviruses was successfully amplified with a set of potyvirus-specific degenerate primers with viral RNA samples from the infected leaves: The RT-PCR product was cloned into the plasmid vector and its nucleotide sequences were determined. The nucleotide sequence of a CDNA clone revealed that the virus was an isolate of Ornithogalum moseic virus (OrMV) based on BLAST search analysis and was denoted as OrMV Korean isolate (OrMV-Ky). To further characterize the CP gene of the virus, a pair of OrMV-specific primers was designed and used for amplification of the entire CP gene of OrMV-Kr, The virus was easily and reliably detected from virus-infected Iris leaves by using the RT-PCR with the set of virus-specific primers. The RT-PCR product of the CP gene of the virus was cloned and its sequences were determined from selected recombinant CDNA clones. Sequence analysis revealed that the CP of OrMV-Kr consisted of 762 nucleotides, which encoded 253 amino acid residues. The CP of OrMV-Ky has 94.1-98.0% amino acid sequence identities (20 amino acid alterations) with that of other three isolates of OrMV, Two NT rich potential N-glycosylation motif sequences, NCTS and NWTM, and a DAC triple box responsible for aphid transmission were conserved in CPs of all the strains of OrMV. The virus has 58.5-86.2% amino acid sequence identities with that of other 16 potyviruses, indicating OrMV to be a distinct species of the genus. OrMV-Ky was the most related with Pterostylia virus Yin the phylogenetic tree analysis of CP at the amino acid level. This is the first report on the occurrence of OrMV in Iris plants in Korea. Data in this study indicate that OrMV is found in cultivated Iris plants, and may have mixed infection of OrMV and Iris severe mosaic virus in Korea.
Purpose: The aim of this study is to investigate the reproducibility of the quantitative assessment of segmental wall motion and systolic thickening provided by an automatic quantitation algorithm. Materials and Methods: Tc-99m-MIBI gated myocardial SPECT with dipyridamole stress was performed in 31 patients with known or suspected coronary artery disease (4 with single, 6 with two, 11 with triple vessel disease; ejection fraction $51{\pm}14%$) twice consecutively in the same position. Myocardium was divided into 20 segments. Segmental wall motion and systolic thickening were calculated and expressed in mm and % increase respectively, using $AutoQUANT^{TM}$ software. The reproducibility of this quantitative measurement of wall motion and thickening was tested. Results: Correlations between repeated measurements on consecutive gated SPECT were excellent for wall motion (r=0.95) and systolic thickening (r=0.88). On Bland-Altman analysis, two standard deviation was 2 mm for repeated measurement of segmental wall motion, and 20% for that of systolic thickening. The weighted kappa values of repeated measurements were 0.807 for wall motion and 0.708 for systolic thickening. Sex, perfusion, or segmental location had no influence on reproducibility. Conclusion: Segmental wall motion and systolic thickening quantified using $AutoeUANT^{TM}$ software on gated myocardial SPECT offers good reproducibility and is significantly different when the change is more than 2 mm for wall motion and more than 20% for systolic thickening.
Gated SPECT can evaluate the regional wall motion of the heart. We evaluated the regional wall motion of the perfusion abnormality in conventional perfusion SPECT with gated SPECT. In case of suspicious perfusion abnormalities, we tried to differentiate the artifact from true abnormality in coronary vascular disease using gated SPECT. We thought that artifacts would have normal wall motion, whereas fixed defects with decreased wall motion would probably represent coronary artery disease. A total of 275 patients who were performed coronary angiography and T1-201 rest/Tc-99m MIBI dipyridamole stress gated SPECT within 2 months were enrolled. In coronary angiography, stenosis more than 50% was considered as coronary artery disease. After injection of 111MBq T1-201 rest image was obtained on triple head SPECT system. 370MBg Tc-99m MIBI was used for the stress image. Eight-frame per-cardiac-cycle gated Tc-99m SPECT studies were done. All the images were analyzed visually. Using perfusion SPECT, the overall sensitivity and specificity were 87% and 55% respectively. Regarding artery territory, sensitivity and specificity were 68% and 73% for left anterior descending artery(LAD), 62% and 78% for right coronary artery(RCA), 42% and 90% for left ciramflex artery(LCX). Using gated SPECT, the overall sensitivity and specificity were 87% and 66% respectively. Sensitivity and specificity were 68% and 78% for LAD, 62% and 79% for RCA, 42% and 90% for LCX. Among 21 false positive cases in perfusion SPECT, 5 cases were interpreted as true negative with gated SPECT. We conclude that gated SPECT provides a valuable adjunct to perfusion SPECT in characterizing perfusion abnormalities and to improve specificity.
We have investigated the effects of the culture broth of lactic acid bacteria (LAB) cultured in herb extract on growth, hematological parameter, nonspecific immune responses and disease resistance of olive flounder (Paralichthys olivacells) and parrot fish (Oplegnathus fasciatus) for 12 weeks. Weight gain of olive flounder fed diet with mixture was not significant among the control group. But In parrot fish, was significantly higher 20g than control group. The feed efficiency of olive flounder were 25% higher in the experimental groups than in the control. There were no significant differences in feed efficiency among each group on parrot fish. Treatment of olive flounder contents of GOT and GPT in serum decreased after 8 weeks. But there were no significant differences in GLU and TP among each group. Also, there was no significant of NBT reduction. The activities of lysozyme were higher in experimental group of olive flounder than in the control after 8 weeks. On the other hand, activities of lysozyme were triple higher in the experimental group of parrot fish than in the control after 12 weeks. In the oliver flounder case, the survival rate (%) after an artificial challenge with $10^7$ CFU/ml of Vibrio anguillarum and Streptococcus iniae per fish, was 18% higher in the experimental groups than the control. The higher survival rate of parrot fish were 17% and 16% in the experimental groups than the control respectively.
