Trichophyton tonsurans의 진균학적 성상 (Mycologic Characteristics of Trichophyton tonsurans)

  • Trichophyton(T.) tonsurans has been identified for the first time in Korea in 1995 and has been increasing in number of reported in wrestlers, in judo players, and in korean - style wrestlers. Author tried mycological studies with T. tonsurans is isolated in Daegu Korea. The results are as follows: All 16 strains of T. tonsurans tested for nutritional requirement, showed strongly stimulated growth by thiamine, mild to moderate growth on casamino acid agar, poor growth on ammonium nitrates base agar. All 16 strains of T. tonsurans tested showed positive results for hair perforation and urease test. Gross findings of T. tonsurans showed that fine granular surface with light yellow hue or white color and mahogany brown color on the reverse side. Microscopic findings showed that septated hyphae, chlamydoconidia and microcondia. Microconidia were characteristic in their arrangerment and shape : round or oval shaped microconidia laterally to hyphae, some were match - head like terminal swelling.

  • 저자는 우리나라에서 아직 보고되어 있지 않지만 앞으로 유입될 가능성이 있는 T. tonsurans에 대하여 경험을 넓히고자 미국에서 환자에서 분리한 T. tonsurans에 대상으로 여러가지 진균학적 검사를 실시하여 다음과 같은 결과를 얻었다. 1. 육안적으로 표면은 편평하고 무두질한 양가죽 양상으로 미세한 과립이 얇게 덮혀있고 연한 회색 또는 연한 황색을 나타내었으며 중앙에는 굴곡이 있고 뒷면은 마호가니 갈색을 나타내었다. urease test에서 균주에 따라 다양한 색깔을 나타내었다. 2. 현미경 소견에서 격벽이 있는 균사를 비롯하여 대분생자, 소분생자, spiral hyphae, 후막포자가 발견되었으며 소분생자의 배열과 모양이 특징적으로 균사를 중심으로 양옆으로 배열되어 길고 그 끝이 성냥 끝처럼 부풀어 있었다. 3. 모발천공검사에서 19주중 16주에서 양성소견을 보였다.

백려노근경(白藜蘆根莖) 추출물(抽出物)의 항진균작용(抗眞菌作用) (Antifungal Activity of the Extracts from Veratrum album L. var. grandiflorum Max.)

  • Although numerous drugs are available for the treatment of superficial fungi infections of skin, the clinical effects of the majority of such drugs are not satisfactory. In the hope of searching the effective drugs for superficial fungi infections, authors studied whether Veratrum rhizoma extracts had any effect on fungi, with water extract (VRWE), ethanol extract (VREE) and methanol extract (VRME) from Veratrum album L. var. grandiflorum Max. In in vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained three extracts of Veratrun rhizoma in each concentration of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and the growth of the fungi were observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum canis, Microsporum nanum, Microsporum gypseum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of the studies were as follows: 1. The growth of M. canis, M. nanum, T mentagrophytes and T. tonsurans were slightly inhibited by VRWE $1,000\;{\mu}g/ml$, and with VRWE $5,000\;{\mu}g/ml$, the growth of E. floccosum, M. gypseum and T. rubrum were slightly inhibited, moderate inhibition on the growth of M. canis, M. nanum, M. cookei, T. mentagrophytes and T. tonsurans were showed by VRWE $5,000\;{\mu}g/ml$. 2. With $500\;{\mu}g/ml$ of VREE, the inhibition on growth of E. floccosum, M. nanum and M. gypseum were slight, however significant inhibition on the growth of M. canis, M. cookei, T. mentagrophytes, T. rubrum and T. tonsurans were observed. The growth of M. nanum and M. gypseum were moderately inhibited, and significant inhibition on the growth of E. floccosum, M. canis, M. cookei, T. mentagrophytes, T. rubrum and T. tonsurans were observed by VREE $1,000\;{\mu}g/ml$. By VREE $5,000\;{\mu}g/ml$, the growth of all tested fungi were significantly inhibited except T. verrucosuia being showed slight inhibition. 3. Significant inhibition on the growth of M. canis, T, mentagrophytes, T. rubrum and T. tonsurans were noted, and moderate inhibition of M. nanum, slight inhibition of E. floccosum and M. gypseum in growth were observed by VRME $500\;{\mu}g/ml$. The growth of E. floccosum, M. canis, M. nanum, M. cookei, T. mentsgrophytes, T. rubrum and T. tonsurans were significantly inhibited by VRME $1,000\;{\mu}g/ml$, and that of M. gypseum was moderate. With $5,000\;{\mu}g/ml$ of VRME, significant inhibition on the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T mentagrophytes, T. rubrum and T. tonsurans were observed, and T. verrucosum was showed only slight inhibition. From the above results, it was found that the extracts of organic solvents from Veratrum rhizoma (VREE & VRME) exerted significant antifungal activity, and their effects were probably derived from the pharmacological action of steroidal alkaloids.

토복령알코홀추출물의 항진균작용 (Effects of Chinae Rhizoma Extracts on the Growth of the Dermatophytes)

  • The majority of drugs used in the treatment of superficial fungal infections has limited values due to its low efficacy or development of resistance. For the purpose of searching efficacious agent on the superficial fungal infections induced by dermatophytes which is regarded as the most malicious one, authors examined whether Chinae Rhizoma Extracts have significant on it. Extracts from Smilax china Linne used for the study are water extract (CRWE), ethanol extract (CREE) and methanol extract (CRME). In in vitro studies, the spores of the dermatophytes were inoculated on Sabouraud's glucose agar media which contained three extracts of Chinae Rhizoma in each concentration of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and also $1,000\;{\mu}g/ml$ of salicylic acid and undecylenic acid $1,000\;{\mu}g/ml$ as comparable drugs. The growth of the dermatophytes were observed for 3 weeks. The species of the dermatophytes used in this experiment were Epidermophyton floccosum, Microsporum canis, Microsporum cookei, Microsporum gypseum, Microsporum nanum, Trichophyton mentagrophytes, Trichophyton rubrum, Trichophyton tonsurans and Trichophyton verrucosum distributed from The Institute of Tropical Medicine in Belgium. The results of the studies were as follows: 1. The growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans were slightly inhibited in CRWE $1,000\;{\mu}g/ml$ and CRWE $5,000\;{\mu}g/ml$, and only slight inhibition on the growth of E. floccosum, M. canis and M. gypseum were observed in CRWE $5,000\;{\mu}g/ml$. 2. Complete inhibition of T. rubrum, moderate inhibition of M. nanum & T. tonsurans, and slight inhibition of E. floccosunl, M. canis, M. cookei & T. mentagrophytes in growth were observed in concentration of CREE $500\;{\mu}g/ml$. The growth of M. gypseum was slightly inhibited, moderate inhibition on the growth of M. canis, M. cookei & T. mentagrophytes, and complete inhibition of E. floccosum, M. nanum, T. rubrum & T. tonsurans in growth were observeed by CREE $1,000\;{\mu}g/ml$. With $5,000\;{\mu}g/ml$ of CREE, the growth of E. floccosum, M. canis, M. cookei, M. gypseum, T. mentagrophytes, T. rubrum & T. tonsurans were completely inhibited except T. verrucosum being showed slight inhibition. 3. In CRME $500\;{\mu}g/ml$, slight inhibition of T. verrucosum, moderate inhibition of M. gypseum and complete inhibition of E. floccosum, M. canis, M. cookei, T. mentagrophytes, T. rubrum & T. tonsurans in growth were observed. The growth of E. floccosum, M. canis, M. cookei, M. gypseum, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans were completely inhibited except T. verrucosum being showed moderate inhibition in both CRME $1,000\;{\mu}g/ml$ and CRME $5,000\;{\mu}g/ml$. 4. In $1,000\;{\mu}g/ml$ of undecylenic acid, slight inhibition of T. verrucosum and complete inhibition of E, floccosum, M. canis, M. cookei, M. gypseum, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans in growth were observed. From the above results, it was found that Chinas Rhizoma Alcoholic Extracts(CREE & CRME) exerted significant antifungal activity, and their effects were probably derived from the pharmacological actions of triterpenoidal saponin and steroidal saponin.

  • ITSI-5.8S-ITSII rDNA region was amplified from the reference strains and clinical isolates with ITS1 and ITS4 primers. These primers amplified DNA fragments of 550 bp in Microsporum audouinii and Trichophyton violaceum, 700 bp in Microsporum gypseum, Trichophyton mentagrophytes, Trichophyton rubrum, and Trichophyton tonsurans, and 750 bp in Microsporum ferreugineum and Microsporum canis. The restriction enzyme patterns of PCR products digested with 13 restriction enzyme including PstI were distint among the genera, whereas identical in the same species. Examination of the ITS (Internal Transcribed Spacers)1 nucleotide sequence revealed that there was the genetic difference in each genera and species. Phylogenetic relationship among each species showed that the Trichophyton mentagrophytes was more closely related Trichophyton tonsurans than Trichophyton rubrum, and Microsporum gypseum was less related than Microsporum spp..

In the hope of searching for effective drugs for superficial fungi infections, the authors observed fungistatic effects of Pyrethri Flos, a common herb in Korea, with water extract (PFWE), ethanol extract (PFEE), and methanol extract (PFME) from Pyrethrum cinerariaefolium V. In in vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained Pyrethri Flos extracts in each concentrations of $500\;{\mu}g/ml$, $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$ respectively, and the growth of the fungi was observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum canis, Microsporum nanum, Microsporum gypseum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of these studies were as follows; 1. The growth of M. nanum & T. rubrum was slightly inhibited by PFWE $500\;{\mu}g/ml$, and the growth of M. nanum, M. cookei & T. rubrum was slightly inhibited by PFWE $1,000\;{\mu}g/ml$. With $500\;{\mu}g/ml$ of PFEE, the growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited, and moderate inhibition of M. cookei growth and slight inhibition E. floccosum & M. gypseum were observed. The growth of M. canis, M. nanum, T. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited, and the growth of E. floccosum, M. gypseum & M. cookei was moderately inhibited by PFEE $1,000\;{\mu}g/ml$. Significant inhibitions of the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T. mentagrophytes, T. rubrum & T. tonsurans were observed by PFEE $5,000\;{\mu}g/ml$. 3. The growth of E. floccosum & M. cookei was moderately inhibited, and the growth of M. canis, M. nanum, M. gypseum, M. mentagrophytes, T. rubrum & T. tonsurans was significantly inhibited by PFME $500\;{\mu}g/ml$. But the growth of E. floccosum, M. canis, M. nanum, M. gypseum, M. cookei, T. mentagrophytes, T, rubrum & T. tonsurans was significantly inhibited, and the growth of T. verrucosum was slightly inhibited in both PFME $1,000\;{\mu}g/ml$ and $5,000\;{\mu}g/ml$.

매자나무뿌리 알콜 추출물의 항진균 작용 (Antifungal Activity of Alcohol Extract and Crystal(A) of Berberis Koreana Palibin)

  • The fungistatic effects of extract and crystal (A) of Berberis Koreans Palibin, a common shrub in Korea, were obeserved and compared with undecylenic acid and vegetable oil. In vitro studies, the spores of fungi were inoculated on Sabouraud's glucose agar media which contained compounds of various concentration, and the growth of the fungi was observed for 3 weeks. The species of the fungi used in these experiments were Epidermophyton floccosum, Microsporum gypseum, Microsporum audouini, Microsporum canis, Microsporum nanum, Microsporum cookei, Trichophyton rubrum, Trichophyton mentagrophytes, Trichophyton tonsurans and Trichophyton verrucosum. The results of the studies were as follows: 1. The growth of E. floccosum, M. audouni, M. canis, M. nanum, M. cookei, was inhibited in Sabouraud's glucose agar media containing extract and crystal (A) of Berberis Koreana Palibin 1 mg/ml.

삼나무 정유의 표재성 진균에 대한 항진균 활성 (Antifungal activity of essential oil from Cryptomeria japonica against dermatophytic fungi)

  • 본 연구에서는 표재성 진균에 의한 피부질환 개선용 소재로의 개발을 위해 삼나무 정유의 표재성 알레르기성 진균, Trichophyton schoenleinii, Trichophyton tonsurans, Trichophyton mentagrophytes (interdigitale), Trichophyton rubrum, Epidermophyton floccosum, Aspergillus fumigatus에 대한 항진균 활성을 평가하여 삼나무 정유의 표재성 진균에 의한 피부질환 개선 소재로의 개발 가능성을 평가하고자 하였다. 항진균 활성 평가는 삼나무 정유의 MIC (Minimum Inhibitory Concentration) 측정과 TLC bioassay, 한천희석법을 통해 측정하였다. 또한 활성 성분의 주성분을 확인하기 위해 GC/MS 분석을 실시하였다. 삼나무 정유 crude oil의 MIC는 모든 균에서 500 ppm 이하의 농도 값을 나타냈으며, TLC bioassay를 통해 spot 1에서 항진균 효과를 확인하였다. 한천희석법을 통한 항진균 활성은 A. fumigatus를 제외한 모든 진균에 대해 삼나무 정유 crude oil의 경우, 500 ppm 이상에서 100%의 활성을 나타냈고, 분획 D의 경우, 100 ppm에서도 100%의 활성을 나타냈다. TLC bioassay를 통해 항진균 활성을 나타낸 spot 1과 한천희석법을 통해 활성을 나타낸 분획 D를 GC/MS로 분석한 결과 주성분은 elemol, ${\gamma}$-eudesmol, ${\beta}$-eudesmol이었다. 따라서 elemol, eudesmol 등은 표재성 진균에 의한 피부질환의 개선 소재로의 이용 가능성이 충분하다고 사료된다.

  • The antifungal studies on the triterpenoids from the pericarp of Forsythia viridissima LINDL. were conducted. The pericarp was obtained from the plant which was cultivated at Eusung, Kyungsang-bukkdo, Korea. Substances A and B were isolated by column fractionation and purified by recrystallization. Antifungal actions of substance A, acetyloleanolic acid, show significant inhibition effect against Trichophyton rubrum, T. mentagrophytes, T. tonsurans, Microsporum nanum, M. canis, and M. cookei. Substance B, viridissimic acid, shows negative effect against Candida albicans and shows lower effects against other test fungi.

Study on antifungal activity of herb oils against Trichophyton spp

  • The antifungal activities of the essential oils from Citrus borgamia. Ciderus atlantica. Cymbopogon ditratus, Eucalyptus globulus. Juniperus communis. Lavandula angustifolia. Melaeuca aterinfolia. Pelargonium graveolens. Pogestemon patchouli. Rosmarinus officinalis. Styrax tonkinensis. and Thymus vulgaris, which are recommended for the treatment of microbial infections in aromatherapy and complementary medicines. were tested against Trichophyton spp. The activities were measured by broth dilution method and disk diffusion assay. As the results, most of the test oils inhibited growth of T. tonsurans. T.mentagrophytes. T. ferugineum. and T. rubrum. Eapecially, the essential oils from C. atlantica. C. ditratus. e. globulus, and P. graveolens showed the strongest activity among the tested herb oils showing MICs between < 0.09 and 0.39 mg/ml.
