Oudemansiella radicata, one of edible mushrooms belonging to Tricholomataceae of Basidiomycota, has been known to exhibit outstanding therapeutic effects on the hypertension caused by high blood pressure and inhibitory effects on the sarcoma 180 and Erhrlich carcinoma of mice. As one of preliminary experiments for producing fruiting-body of O. radicata, this study was carried out to obtain the basic information for culture conditions of mycelial growth of the fungus. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and pH 6, respectively. The medium for favorable mycelial growth of O. radicata was shown in the Lilly medium, whereas compact mycelial density was found in Hamada medium. The carbon and nitrogen sources promoting for mycelial growth of O. radicata were xylose and alanine, respectively. The optimum C/N ratio was about 20 : 1 in case that 3% glucose was supplimented to the basal medium as a carbon source.
Tricholoma giganteum, belongs to Tricholomataceae of Tricholoma, is also well-known as the medicinal mushroom in Taiwan. "Backryon" was the first variety developed by intra-specific crossing in Korea. It was improved with hybridization between monokaryotic strain derived from MKACC50852 and MKACC 50853. The optimum temperature of mycelial growth and fruiting body development were $25{\sim}30^{\circ}C$ and $20{\sim}25^{\circ}C$, respectively. The color of fruitingbody was pure white and cap type was umbrella. It suggested that 'Backryon' was new commercial variety for small-sized cultivator during the summer season.
Lepista nuda is a world-wide species which has and international reputation as a excellent edible species. In this study, we investigated the genetic variation and taxonomic relationship of L. nuda and other five Tricholomataceae species were analyzed by random amplied polymorphic DNA (RAPD). 15 kinds of random primers were used. The distance matrix was calculated using UPGMA and phyolgenetic relationship were inferred by neighnor-joining (NJ) method. Various bands of 100bp∼1600bp were observed in electrophoretic patterns of RAPD. Nei's genetic distance was calculated using a total of 228 DNA bands identified, and phylogenetic tree was made. The Nei's genetic variations of L. nuda, Lepista surdida, Collybia peronata, Collybia confluens, Lyophyllum cinerascens, Laccara laccata were 0∼21.3%, 21.2∼28.0%, 15.4∼23.0%, 14∼21.8%, 16.5∼34.6%, and 12.4∼27.4%, respectively The consistency index, the retention index and homoplasy index were 0.5217, 0.5769 and 0.5156, respectively. Also, two groups could be made by NJ tree. The genetic distance between L. nuda and C. confluens was closer than that between L. nuda and L. sordida.

To find antitumor components in Korean basidiomycetes, the carpophores of Armillariella mellea which were collected in Gyeong Gi Province were extracted with distilled water at $90{\sim}100^{\circ}C$ for eight hours. The hot water extract was concentrated under reduced pressure, mixed with three-fold volumes of ethanol and allowed to stand at $4^{\circ}C$ overnight. The precipitate was centrifugated and lyophilized to yield a protein-polysaccharide fraction. It was examined for antitumor activity against sarcoma 180 implanted in ICR mice. The fraction showed 75.7%, 83.9%, and 94.1% of tumor inhibition ratios at the doses of 10, 20 and 50 mg/kg/day, respectively. The chemical analysis of the fraction showed that it contained a polysaccharide(41.3%) and a protein (35.0%). The hydrolyzates of the polysaccharide moiety contained fucose (4.5%), xylose (1.1%), galactose (17.4%), glucose (55.4%), mannose(19.4%), and one unknown monosaccharide. The protein moiety contained seventeen amino acids. The protein-polysaccharide from A. mellea was administered, i.p., to mice and caused an influx of polymorphonuclear leukocytes (PMN) at $5{\sim}24$ hours which was followed by an accumulation of macrophages and disappearance of the PMN at $48{\sim}72$ hours.

Mushroom fruiting was investigated in pine and oak dominated forest stands in Songni National Park located in central Korea for six years from 2003 to 2008, in order to understand the relationship between mushroom diversity and the environmental factors, precipitation, temperature, soil moisture and vegetation. The most frequent fruiting families were those of ectomycorrhizal mushrooms, Tricholomataceae, Amanitaceae, Russulaceae, Cortinariaceae, and Boletaceae. The frequency of mushroom fruiting varied from 94 to 167 species per year, with July and August having the highest(13~90 species). Mushroom fruiting was positively correlated to precipitation(r=0.897), using Palmer Drought Severity Index for the long term period and Standard Precipitation Index for short term period. Soil moisture content also affected mushroom fruiting, with Lactarius chrysorrheus and Russula virescens fruiting only at soil moisture content higher than 20%. Positive correlation between mushroom fruiting and temperature was also noted(r=0.77), with optimum rates at $21{\sim}25^{\circ}C$. Tricholoma flayayirens, Amanita gymnopus, Lactarius piperatus, Inocybe asteropora and Xerocomus chrysenteron were able to fruit at temperatures higher than $25^{\circ}C$. However, Laccaria amethystea, Amanita virosa and Russula mariae fruited at relatively wide temperature range. The influence of vegetation on mushroom fruiting was likewise noted, with 38 species, including Suillus bovinus and Boletopsis leucomelas being specific to pine dominated stands, while 42 species, including Polyporus arcularius and Hericium erinaceum were specific to oak dominated stands. On the other hand, around 50 species, including Laccaria laccata and Lycoperdon parlatum, were able to fruit in both types of vegetation. In conclusion, mushroom fruiting greatly varies with changes in precipitation, soil moisture, temperature and vegetation.
Wild Mushrooms collected in Mt. Halla of Jeiu Island around altitude of $200{\sim}1000\;m$ from 1999 to 2000 were studied to investigate utilization as the natural breeding sites by wild drosophilids. Thirty-five mushroom species belonging to the 19 genera of 12 families from 2 orders were used as natural breeding sites by drosophilids. Adult flies emerged from 14 species of mushrooms belonging to the 11 genera of 8 families from 2 orders were identified. From the rest of 21 species of mushrooms, only larvae or/and pupae were reaped. Among 35 species of mushrooms, 34 species belonged to order Agaricales and only one species, Sarcodon scabrosum, belonged to order Aphyllophorales. In terms of composition of mushroom families, total 35 species belonging to 12 families were composed of 10 species of Russulaceae, 6 species of Amanitaceae, 4 species of each Tricholomataceae and Agaricaceae, 3 species of Boletaceae, 2 species of Strophariaceae and each one species of rest 6 families. Concerning the composition of the genus, 35 species belonging to 11 genera were composed of 8 species of Russula, 6 species of Amanita, 4 species of Agaricus, 2 species of Lactarius, and each one species of rest 7 genera. In terms of the mushroomsize preference of drosophilid flies for breeding sites, most flies preferred comparatively large mushrooms above 5 cm of pileus size in diameter, Fourteen species of drosophilids were identified as mushroom-breeding species. Among these, one and five species belonged to genera Mycodrosophila and Hirtodrosophila respectively, The genus Drosophila included the most abundant 8 species. Drosophila bizonata and Hirtodrosophila sexvittata bred on 6 species of mushrooms respectively, D. unispina bred on 3 species of mushrooms, rest species of Drosophila bred on one or two species of mushrooms. From the result of this study and other literatures, it could be presumed that much more mushroom species could serve as the natural breeding sites for greater number of drosophilid species than those examined in this study on Jeju Island.
Tricholoma gigantium, an edible mushroom, belongs to Tricholomataceae of Tricholoma and is distributed at Jeju-Do in Korea. It is also well-known as the medicinal mushroom in Taiwan. The cultivation method using the compost was developed in Korea in 1995. To develop a mass cultivation method and a superior strain, four strains were collected and tested. To establish the optimal cultivation conditions, various examinations were accomplished. Bag cultivation was more effective than box cultivation and the optimal relative humidity was more than 80%. Although the mycelial growth was tested in the substrate supplemented with different additives, such as rice bran and wheat bran, there's no significant difference between them. It suggested that the environmental conditions were more important than the substrate additives for cultivation.
This study was carried out to investigate the genetic relationship of Flammulina velutipes with other species. The ribosomal DNA cluster containing 4 rRNA genes from F. velutipes 4154 were sequenced. The length of the rDNA cluster sequence was estimated at 7,403 bp long and consisted of 1,806 bp of SSU rDNA, 245 bp of ITS 1 region, 159 bp of 5.8S rDNA, 308 bp of ITS 2 region, 3,402 bp of LSU rDNA, 1,400 bp of IGS 1 region, and 83 bp of 5S rDNA. The F. velutipes 4154 genes were contained in the rDNA cluster of F. velutipes in the order of SSU rDNA - ITS 1 - 5.8S rDNA - ITS 2 - LSU rDNA - IGS 1 - 5S rDNA. The phylogenetic relationships of 20 strains of Tricholomataceae and Physalacriaceae were analyzed by conducting distance analysis using the Neighbor-joining (NJ) method. The 20 strains used in this study were divided into three groups and the strains of the genus Flammulina were related very closely to strains of Physalacria bambusae.
Oudemansiella mucida, an edible and medicinal mushrooms belonging to Tricholomataceae of Basidiomycota, has been known to produce antifungal substances to inhibit the mycelial growth and spore germination of the plant pathogenic fungi. To produce good amount of antifungal substances from culture media, the optimal culture conditions of O. mucida were investigated. The most favorable conditions for the mycelial growth were $25^{\circ}C$ and pH 5 in potato dextrose agar. The most favorable carbon and nitrogen sources promoting mycelial growth were maltose and calcium nitrate, respectively. The optimum C/N ratio was about 20 : 1 in case that 3% glucose was supplemented to the basal medium as a carbon source. The optimal mycelial growth of O. mucida was found in the Hennerberg medium. The crude extract from submerged culture of potato dextrose broth exhibited inhibition of mycelial growth of Colletotrichum acutatum, Botrytis cinerea and Pyricularia oryzae but, fungicidal activity is not good enough to compared with commercially available fungicides tested. Therefore, the antifungal substances extracted from submerged culture of O. mucida might have a potential to be used for biocontrol agent of fungal diseases of plants.

Armillaria mellea, one of edible and medicinal mushroom belonging to Tricholomataceae of Basid-iomycota, has been known to have outstanding inhibitive effects on the sarcoma 180 and Erhrlich carcinoma of mice. Neutral salt soluble (0.9% NaCl), hot water soluble and methanol soluble substances (hereinafter referred to Fr. NaCl, Fr, HW and Fr. MeOH, respectively) were extracted from the mushroom. In vitro cytotoxicity tests, crude polysaccharide were not cytotoxic against cancer cell lines such as NIH3T3 and Sarcoma 180 at the concentration of $1000{\mu}g/ml$. Intraperitoneal injection with crude polysaccharides exhibited life prolongation effect of $60{\sim}67.5%$ in mice inoculated with Sarcoma 180, respectively. Fr, NaCl improved the immunopotentiation activity of B lymphocyte by increasing the alkaline phosphatase activity by $1.8{\sim}3.0$ folds, respectively. In case of Fr. NaCl, the numbers of peritoneal exudate cells and circulating leukocytes were increased by 10 and 2 folds, respectively.
