Korean Tricholoma matsutake Strains that Promote Mycorrhization and Growth of Pinus densiflora Seedlings (균근 형성과 소나무 유묘 생장이 우수한 송이 균주의 선발)

  • Domestic and international production of Tricholoma matsutake has decreased owing to matsutake forests being left alone, host plant disease, forest fires, climate change, and so on. In order to identify strains that are suitable for the production of T. matsutake-inoculated seedlings, Pinus densiflora seedlings were inoculated with T. matsutake after in vitro rooting and mycorrhization was examined in the roots of T. matsutake-inoculated seedlings after 6 months. The mycorrhization rate was greater than 80% for 5 strains (NIFoS 421, 434, 1681, 1984, and 2001) out of 19 total strains. Seven strains (NIFoS 434, 441, 561, 562, 1016, 1807, and 1812) showed shoot/root ratios of less than 3.0 and had a seedling shoot biomass of 2.0 to 4.8 times higher than that of the root. Eight strains (NIFoS 441, 561, 562, 1016, 1807, 1812, 1984, and 2001) stimulated increases in shoot volume and three stains (NIFoS 441, 562, and 1812) promoted the growth of root biomass by mycorrhizal formation. In conclusion, 4 strains (NIFoS 434, 561, 1984, and 2001) out of 19 total strains tested showed higher mycorrhization rates and seedling growth than those of the other strains. We expect that the use of these four strains may contribute to T. matsutake-inoculated seedling production.

Growth-promoting effect on Tricholoma matsutake mycelium by Terrabacteria isolated from pine mushroom habitats in Korea (국내 송이 자생지에서 분리된 Terrabacteria에 의한 송이균사체 생장촉진 효과)

  • To cultivate pine mushroom (Tricholoma matsutake) artificially, co-cultivation with microorganisms has been introduced. Here, experiments were performed to assess the growth-promoting effect of bacteria on T. matsutake mycelia. Bacteria were isolated from soil samples collected in Yangyang County, Korea. Four of the bacterial isolates (Y22_B06, Y22_B11, Y22_B18, and Y22_B22) exhibited a growth-promoting effect on T. matsutake mycelia (154.67%, 125.91%, 134.06%, and 158.28%, respectively). To analyze the characteristics of the bacteria, especially the antifungal activity, 𝛼-amylase and cellulase activity assays were performed. In comparison with the controls, the isolated bacteria exhibited low 𝛼-amylase and cellulase activity. 16S rRNA gene sequencing was performed to identify the four bacterial isolates. The isolates belonged to the Terrabacteria group and were identified as Microbacterium paraoxydans, Paenibacillus castaneae, Peribacillus frigoritolerans, and P. butanolivorans. These bacterial isolates are expected to have contributed to the growth promotion of T. matsutake mycelia and the artificial cultivation of T. matsutake.

Studies on the Extraction Method and Polysaccharide of Tricholoma matsutake using the Supersonic wave and Microwave (초음파와 극초단파를 이용한 송이버섯의 추출법과 다당체에 관한 연구)

  • In order to optimize the extract condition and improve physiological activity of the extract form Tricholoma matsutake, experiments related to extraction methods, totale yield, content of total soluble polysaccharide, SOD-like activity, total polyphenol amount, and volatile flavor compound and the others were carried out, results were obtained as following: Compare with traditional hydrothermal extraction method (Hot water extraction : HWEW), it illustrates that the low temperature extraction method which combines a supersonic waves and microwave (Supersonic microwave extraction : SMEW) causes of increasing the total yield, total soluble polysaccharide. As to the anti-oxident effect, SMEW method leds to increasing of the SOD-like activity, total polyphenol amount as well. Also, cytotoxic effect and growth inhibitory effect against cancer cell line are much higher in SMEW method than HWEW method, especially SMEW5 extracts treated by supersonic 15 min. and microwave 120W, 3 min. and 2 times. The main volatile flavor compound and infinitesimal volatile flavor compound both increase significantly by SMEW method. It is concluded the main components of the volatile flavor compounds extracted from Tricholoma matsutake are 1-octen-3-0l, Methyl cinnamate, 2-octeno1 et al. alcohol typies. Consequently, SMEW5 method is considered as the most effective one for anti-oxidant and is prior to any other methods. And the optimun conditions of this method are : supersonic waves (supersonic, 25KHz, 50W) 15 minutes, microwave spectroscopy (microwave, 2,450MHz, 120W) 3 minutes, and every treatment is performed once followed twice repeats.

  • Tricholoma matsutake (Pinus mushroom, PM) is one of the most valued ectomycorrhizal fungi in Asia because it is an expensive forest product with a unique flavor and taste. Therefore, many studies have tried to successfully cultivate Tricholoma matsutake artificially in Korea and other countries. However, its physiological and ecological characteristics are still unknown. Thus, we need to understand the diversity and clusters of microorganisms related to Tricholoma matsutake and to identify their core microorganisms related to their growth and production. In this study, we obtained an average of 11,661 fragments from three pine mushrooms with metagenome (an assemblage of genes of all microorganisms in the natural world) analysis from a pine forest located in Pohang, Gyeongsang-Bukdo. Of these, the valid reads were on average 5,073 per sample available for analysis, and the average length of a read was 456 bp. There were an average of 33.3 phyla in the metagenome analysis. Firmicutes phylum made up on an average 46% of the phyla and was dominant among the phyla. The next dominant phylum was Proteobacteria at 27% followed by Bacteroidetes at 17%, Actinobacteria at 5% and Verrucomicrobia at 2%. The Proteobacteria phylum consisted of the γ-proteobacteria class at 54% followed by β-proteobacteria at 37%, α-proteobacteria at 6%, δ-proteobacteria at 2% and ζ-proteobacteria at 0%. The metagenome consisted of the Ruminococcaceae family at 17% followed by Pseudomonadaceae at 13%, Burkholderiaceae at 7%, Bacteroidaceae at 7%, Lachnospiraceae at 7% and Clostridiaceae at 6%.

Growth promoting effect on Tricholoma matsutake mycelium by bacteria from fairy Ring in Bonghwa-gun, Korea (국내 봉화 송이 자생지 내 균환 유래 토양세균의 송이균사체 생장촉진 효과)

  • As a member of ectomycorrhizal fungi, Tricholoma matsutake has a symbiotic relationship with its host, Pinus densiflora. To cultivate T. matsutake artificially, the co-cultivation of T. matsutake mycelia and bacteria from shiro was introduced. In this study, bacteria were isolated from soil samples in Bonghwa-gun, and seven bacterial isolates (B22_7_B05, B22_7_B06, B22_7_B07, B22_7_B08, B22_7_B10, B22_7_B13, and B22_7_B14) promoted the growth of T. matsutake mycelia (147.48, 232.11, 266.72, 211.43, 175.17, 154.62, and 177.92%, respectively). Sequencing of the 16S rRNA region of the isolated bacteria was performed. B22_7_B05 and B22_7_B10 were identified as Bacillus toyonensis, B22_7_B06 and B22_7_B08 as Paenibacillus taichungensis, B22_7_B07 and B22_7_B14 as P. gorilla, and B22_7_B13 as P. odorifer. These bacterial isolates were associated with the shiro community and are expected to contribute to the cultivation of T. matsutake.

Mycelial Growth and in vitro Ectomycorrhizal Synthesis on Pinus densiflora Seedlings of Tricholoma bakamatsutake in Korea (한국산 가송이(가칭)의 균사생장 특성과 소나무에 외생균근 형성)

  • Tricholoma bakamatsutake is one of the edible ectomycorrhizal mushrooms as an allied species of Tricholoma matsutake. This is the first report on physical characteristics of T. bakamatsutake strains collected from Quercus mongolica forests in Korea. The pure cultures of these strains were isolated from the tissues of fruit bodies and the culture characteristics were investigated under different conditions (media, temperatures, nitrogen sources). Most strains showed the highest mycelial growth on potato dextrose agar (PDA) at 20 or $25^{\circ}C$. Two strains of T. bakamatsutake preferred the ammonium-form rather than the nitrate-form as an inorganic nitrogen source. T. bakamatsutake showed significantly slower mycelial growth when compared with T. matsutake from a Korean forest, although the optimum culture conditions for the two allied species were similar. We also tested the ability to form mycorrhizae as well as cellulase activity of T. bakamatsutake. All strains showed cellulase activity on a carboxymethylcellulose (CMC) agar plate. The mycorrhizae on axenic Pinus densiflora seedlings were formed by two strains of T. bakamatsutake after 3 or 8 months of inoculation. P. densiflora seedlings inoculated with T. bakamatsutake had a much higher biomass than un-inoculated seedlings.

Fairy Ring Growth of Tricholoma matsutake in 65-year-old Pine (Pinus densiflora) Forest Stand (65년생 소나무림에서 송이 균환 생장)

  • This study was carried out to evaluate annual growth of fairy ring of Tricholoma matsutake. The edge growth of active mycelial zone of fairy ring during $1999{\sim}2000$ and the distance between sporocarp-fruiting positions of four years ($1997{\sim}2000$) and the edge of fairy ring of 2000 were measured. The fairy ring of T. matsutake moved outward about 11.3 cm annually ($1999{\sim}2000$) in the 65 years old P. densiflora stand, and the growth was coincided with the average distance of sporocarp-fruiting positions for four years ($1997{\sim}2000$). In addition, the sporocarp-fruiting positions were about 13.8 cm apart from the edge of active mycelial zone in year of 2000. Therefore, the sporocarp-fruiting position in this year was within the mycelial region one year ago. It is strongly recommended that the sporocarp of T. matsutake should be harvested apart about 50 cm from sporocarp in order to protect the fairy ring.

Dynamics of Root Systems in the Fairy Rings of Tricholoma matsutake (송이(松栮) 균환(菌環)에서의 근계(根系) 동태(動態))

  • The dynamics of root system in the fairy rings of Tricholoma matsutake was investigated in four zones divided into 'zone of decayed mycorrhizae', 'zone of mycorrhizae for fruiting', 'zone of physiologically active mycorrhizae' and 'zone of roots free from mycorrhizal infection'. The roots of Pinus densiflora in fairy rings, which occupied 70% of upper crown, comprised about 60% and 87% of total roots and alive roots, respectively. The ratio of fine-roots of P. densiflora over total roots was relatively higher than other species since that of P. densiflora was about 45% while that of the other species was about 13% on research of fine-roots. Especially, the roots of pine comprised about 70% of total root in the zone of mycorrhizae for fruiting and the zone of physiologically active mycorrhizae, which indicated that the pine roots were closely related to the fairy rings of T. matsutake. The fine roots of P. densiflora in the zone of physiologically active mycorrhizae was about 60.7%(1,087mg/100g soil) which was about twice compared to that of other zones. It allowed us to suppose that the fine roots of P. densiflora can make active growth in the zone of physiologically active mycorrhizae, and the growth was promoted by the fairy ring formation of T. matsutake. In addition, we found 3~5 times higher amount of fine roots than that of medium roots of P. densiflora in this zone, which indicated that the fairy rings of T. matsutake locate in the region of active growth of P. densiflora' roots.

Sustainable Production Strategy of Pine Mushroom (Tricholoma matsutake) using the Maximum Entropy Technique (최대 엔트로피 기법으로 도출한 지속 가능한 송이 생산 전략)

  • Pine mushroom (Tricholoma matsutake) is one of the most profitable forest products in Korea. We postulated a hypothesis that a high rate of returns to labor input could make the harvest of pine mushroom off the optimum level. In the view of developing a sustainable production strategy for pine mushroom producers, production of pine mushroom collectors and pine mushroom growth function were estimated using maximum entropy method. Annual pine mushroom production and labor input were the data used in the estimation of production function of pine mushroom collectors and pine mushroom growth function. The level of sustainable maximum production derived from the estimated function. The production function estimated shows that production of pine mushroom is affected more by the resource of pine mushroom stocked in the forests than by labor that households put in forestry business. The production function of mushroom collectors and the estimated growth function indicate that pine mushroom harvests for the period of 2005-2011 did not reach the potential level of maximum sustainable production. Therefore, we suggest that pine mushroom harvest should be controlled until the resource stock of pine mushroom in the forests increases to the level of maximum sustainable production.

The effect of mushroom components on the proliferation of HeLa cell line in vitro

  • In order to find out nutritional effects of Korean edible mushrooms on the multiplication of tissue cells, the alcohol extracts and acid bydrolysates of eleven species of mushrooms were added to Earle's ESS. A comparison of the respective multiplications of HeLa cells in this solution and in control solution of TC=199. Yielded the following result: The alcohol extracts and acid hydrolysates of a Coprinus comatur, Agaricus campestris, Agaricus bisporus, Lentinus edodes, Tricholoma matsutake, Pleurotus ostretus, Ramaria botrytis and Pholiota nameko influenced favorably the maintenance of the normal form and monolayer of HeLa cells. The growth curves of HeLa cells in the cultures containing, respectively, the alcohol extracts and acid hydrolysates of these eight mushrooms showed that five species, i.e., Coprinus comatus, Agaricus compestris, Agaricus bisporus, Lentinus edodes and Tricholoma matsutake effected an excellent multiplication and that the other three species were less effective than those five species. As to the effects on the cell multiplication, no marked difference was observed between the alcohol extracts and the acid hydrolysates of the mushrooms tested.
