Effects of Water Temperature, Salinity, Rearing Density and Food Supply on the Growth and Survival of the Surf Clam, Tresus keenae Larvae (수온, 염분, 사육밀도 및 먹이공급이 왕우럭(Tresus keenae) 유생의 성장과 생존에 미치는 영향)

  • This study was conducted to investigate the influence of environmental factors such as water temperature, salinity, rearing density and feeding amount on the survival and development of larvae in surf clam, Tresus keenae. As a result of this study, the optimum conditions of larval rearing were as follows. The optimum range of water temperature is 20℃ to 24℃, the optimum salinity is 30 psu, the optimum rearing density is 5 ind./ml and the supply of mixed feed organism for larva rearing such as Isochrysis galbana, Chaetoceros simplex and Tetraselmis suecica is 10×104 cells/ml. The results of this study suggest that the optimum conditions of environmental factors in larval rearing of Tresus keenae contribute to productivity improvement through rapid growth and high survival.

  • The gametogenesis of the surf clam, Tresus keenae, were investigated by SEM and TEM. Both the testis and the ovary had follicle tubes surrounded by inter-tubal tissue composed of adipogranular cells that provided storage function. In the vitellogenic oocyte, lipid droplets and lipid yolk granules were found in the vacuoles formed by the Golgi apparatus. Proteid yolk granules were formed by the endoplasmic reticulum and cortical granules in the cytoplasm during vitellogenesis. The mature sperm was primitive and resembled a jar with a cover. The sperm heads were approximately $2.00-2.30 {\cal}um$. The acrosomal rod projected in front of the acrosome. In addition, four large mitochondria were in the midpiece.

Effects of Temperature and Salinity on the Survival and Metabolism of Tresus keenae (Mollusca: Bivalvia)

  • We examined the variation in survival and the respiration and filtration rates of Tresus keenae in response to changes in water temperature and salinity. The survivorship of animals exposed to temperatures below $25^{\circ}C$ for 7 days was $80\%$; however, all test animals died on the fourth day at $28^{\circ}C$. The upper lethal temperature over 7 days was $25.9^{\circ}C$. After exposure to lower temperatures, $93\%$ ofthe animals survived at temperatures over $5^{\circ}C$ for 10 days. Survivorship rapidly decreased below $4^{\circ}C$ with all test animals dying at $2^{\circ}C$ on the eighth day. The $LT_{50}$ over 10 days was $4.8^{\circ}C$. The respiration and filtration rates of T. keenae increased as temperature increased. It is believed that energy consumption increases as a result of the increased respiration rate at temperatures above the upper lethal temperature. At temperatures below the lower lethal temperature, the metabolic rate of T. keenae was substantially lowered. In response to changes in salinity, the survivorship of T. keenae was $90\%$ at 30.2 psu after exposure for 5 days; at below 26.8 psu, all test animals died by the fifth day. The $LS_{50}$ was 29.1 psu. As salinity decreased, both the respiration rate and the filtration rate decreased. At 23.5 psu, the respiration and filtration rates decreased by 48 and $34\%$, respectively. These data have implications for increasing efficiency in the production and management of shellfish aquaculture farms.

Degradation capability of macromolecular organic matters and antimicrobial activities of Bacillus species isolated from surf clam (Tresus keenae) (왕우럭(Tresus keenae)에서 분리된 Bacillus species의 고분자 유기물질 분해능력과 항균활성)

  • The production of enzymes that help digestion, assimilation of essential nutrients, and prevent pathogenic bacteria are important for probiotics used in aquaculture. The objective of this study was to investigate enzyme activities for macromolecular organic matters and antimicrobial properties of the selected potential probiotics isolated from gut of surf clam (Tresus keenae) against well-known shellfish-pathogenic bacteria. Among 65 isolates from guts of 60 surf clams, seven Bacillus strains with outstanding degradation capability of macromolecule organic matter were selected as potential probiotics as follows: TKI01 (B. vietnamensis), TKI02, TKI26 (B. thuringiensis), TKI14, TKI32, TKI42 (B. amyloliquefaciens), and TKI18 (B. stratosphericus). After in vitro antimicrobial activity test was performed against five shellfish-pathogenic bacteria including Listonella anguillarum, Vibrio parahaemolyticus, V. splendidus, V. harveyi, V. tubiashii, PCR assay was performed to detect bacteriocin-producing strain. PCR results revealed that the five Bacillus strains possessed diverse bacteriocin genes including ericinA, coagulin, surfactin, iturin, bacyllomicin, fengycin, bacylisin, subtilin, and lantibiotics. In the present study, the selected seven Bacillus strains showed different enzyme activities according to types of macromolecule organic matters. And their antimicrobial activities varied based on the species of pathogenic bacteria. In addition, at least five Bacillus strains had genetic potential to produce several natural lipopeptide antibiotics that may help biological control of surf clam aquaculture. Therefore, mixed use of probiotics might show co-operative effect and increase the efficiency of probiotics rather than separate use. To the best of our knowledge, it is the first report on antimicrobial properties of Bacillus species isolated from surf clam.

  • Seasonal variation of the infection by three species of symbiotic copepods in clam, Tresus keenae was investigated in south coast of Korea. The prevalence and abundance of three symbiotic copepods showed seasonal variation with a peak in winter. Reproduction of the copepods occurs from early autumn to spring as indicated by higher ratio of gravid copepods. The frequency distribution of Lichomolgus inflatus, Pseudomyicola spinosus and Anthessius progectus in their host population could be fitted with negative binominal distribution. Results from the present study suggest that seasonal fluctuation in prevalence, relative density and mean intensity is related to temperature and life history of the symbiotic copepods.

  • Ultrastructure and sperm morphology were investigated Mactra venerifomis, Mactra chinensis, Spisula sacharinensis, Tresus keenae in Mactridae. All the sperm studied were primitive type. They consisted of three parts; the head, the middle piece and the tail part. The acrosome forms were similar among the 4 species. The acrosome was shaped like a modified hat. It was consisted of two different parts in electronic density; the anterior part, electric lucent part (elp), and the posterior part, electric dense part (edp). But nuclei forms were slightly different among the species. All the species studied had 4 mitocondria at middle piece. We guessed that the Mactridae sperm were family-specific with characterful acrosome shape.

  • For the aquaculture industrialization of surf clam (Tresus keenae), it is important to basic data on the marine environment of the habitat of surf clam (T. keenae). In this study, we investigated the marine environment of habitat of surf clam (T. keenae) and sought to basic data for the preparation of surf clam (T. keenae) for artificial seed production. The water temperature of the habitat of surf clam (T. keenae) was the lowest in winter and appeared high in summer. The salt concentration showed it range from 31.2 to 33.9 psu. The pH showed it range from 7.69 to 8.70, with high pH in winter and low pH in summer. The dissolved oxygen(DO) was showed it range from 6.20 to 10.24 mg / L and the autumn was relatively higher than the spring and winter. The species composition of phytoplankton was about 30 to 40 species, and most of them were diatoms. The abundance of seasonal phytoplankton showed it range from 23.5 to 61.3 cells / ml, showing seasonal differences. The expression of dominant species also showed a difference depending on the season. As for the particle size composition of the sediment, sandy silt was the most distributed. Flow velocities appeared at 50-80 cm / s in the southeast direction at ebb tide and at 60-100 cm / s in the northwest direction at flood tide. The results of this study can be used as basic data for providing knowledge about the habitat and marine environment of surf clam (T. keenae) and for studying shellfish that inhabit the sedimentary layer.

  • This study interested on management of broodstock and the effect of raise the temperature for promotion of maturity in Tresus keenae. For management of broodstock, we performed indoor and outdoor culture. In addition, broodstock was put in exposure and immersion condition. This experimental design is aimed at knowing the optimal management of broodstock. In addition, we investigated the promotion of maturity, growth and survival rate when raise the temperature of the breeding seawater of broodstock in winter season. Result that, the management of broodstock was shown to be more effective obesity and survival rate in outdoor culture than indoor culture. In addition, there is no anthropogenic food supply in management and it is easy to management because it is kept under natural environmental conditions. Therefore, It is considered appropriate to management to outdoor culture. In raise the temperature, survival rate was lower in the experimental group than in the control group. However, obesity was significantly increased compared with the control group. Also, gonad showed that the sex maturity was promoted. If sex maturation is not developed due to seasonal water temperature, the promotion of sexual maturity through heating is considered to be an effective method in terms of artificial seed production.
