2007년 봄 인천만 갯벌에 있어 폭풍으로 인한 갯벌의 침식작용에 의해 대기 중으로 노출된 바지락 (Ruditapes philippinarum)의 조직병리학적 특성에 관한 연구 (Pathologic condition of the stranded Manila clam Ruditapes philippinarum by storm-originated sediment erosion in spring 2007 in Incheon Bay off the west coast of Korea)

  • In Incheon bay, mass mortalities of Manila clam associated with winter storms have been reported. In the present study we have monitored pathologic condition of the clams stranded on the tidal flats by the winter storms occurred in late March to early April in 2007. The field surveyed indicated that mortality of the Manila clam in the study areas ranged 10-15%. Condition index, a ratio of tissue weight to the shell weight, of the stranded clams was significantly lower than the non-stranded normal clams collected from the same locations (p < 0.05), indicating that the stranded clams were comparatively in poor physiological condition. Perkinsus olseni, the protozoan parasite was observed most of clams used in the analysis and the infection prevalence ranged 77-90%. The infection intensity of P. olseni determined using Ray's fluid thioglycollate medium (RFTM) cultivation and the 2M NaOH digestion assay indicated that the clams collected during late March and early April in 2007 involved 67,182-1,124,727 P. olseni cells/g tissue. The infection intensity of clams from Gung-Pyeung was significantly higher than the intensities observed from Dae-Bu and Young-Heung (p < 0.05). No clear correlation was found between the infection intensities of P. olseni in the non-stranded normal clams and the stranded clams. The stranded Manila clams were also infected with trematode parasite with the prevalence ranged 5 (Young-Heung) to 12.5% (Dae-Bu). The trematode-infected clams exhibited castrated follicles in the gonad, a typical sign of trematode infection. It was believed that mass mortality of Manila clam observed in this study was associated with the poor physiological condition as indicated by CI, although impacts of the parasite infection cannot be ruled out.

Infection Status of Zoonotic Trematode Metacercariae in Fishes from Vientiane Municipality and Champasak Province in Lao PDR

  • The infection status of fishborne zoonotic trematode (FZT) metacercariae was investigated in fishes from 2 localities of Lao PDR. Total 157 freshwater fishes (17 species) were collected in local markets of Vientiane Municipality and Champasak Province in December 2010 and July 2011, and each fish was examined by the artificial digestion method. Total 6 species of FZT metacercariae, i.e., Opisthorchis viverrini, Haplorchis taichui, Haplorchis yokogawai, Haplorchis pumilio, Centrocestus formosanus, and Procerovum varium, were detected in fishes from Vientiane Municipality. The metacercariae of O. viverrini were detected in 50 (49.5%) out of 101 fishes (6 species), and their average number was 154 per fish infected. The remaining 5 species of heterophyid metacercariae were detected in 36.8%, 65.8%, 9.4%, 23.9%, and 5.1% fishes examined, and their average densities were 12, 1,038, 4, 15, and 13 per infected fish, respectively. In fishes from Champasak Province, 3 species of FZT metacercariae, i.e., O. viverrini, H. taichui, and H. yokogawai, were detected. Only 2 O. viverrini metacercariae were found in only 1 Barbonymus schwanefeldi. The metacercariae of H. taichui and H. yokogawai were detected in 60.0% and 50.0% of fishes examined, and their average densities were 47 and 28 per fish infected. By the present study, it has been confirmed that several species of FZT metacercariae are prevalent in fishes from Vientiane Municipality, with P. varium being a new member of FZT in Lao PDR. In comparison, FZT metacercariae are less prevalent in fishes from Champasak Province.

Infections with Zoonotic Trematode Metacercariae in Yellowfin Goby, Acanthogobius flavimanus, from Coastal Areas of Republic of Korea

  • To investigate the infection status of zoonotic trematode metacercariae in yellowfin goby, Acanthogobius flavimanus, from coastal areas of the Republic of Korea (Korea), we examined total 344 gobies from 21 localities with an artificial digestion method from 2013 to 2017. The metacercariae of Stictodora lari were most frequently detected in 45.3% goby from 7 localities, i.e., Taean-gun (Chungcheongnam-do), Gochang-gun (Jeollabuk-do), Muan-gun, Shinan-gun, Haenam-gun (Jeollanam-do), Hadong-gun and Goseong-gun (Gyeongsangnam-do). Their infection rates were 90.0%, 66.7%, 46.7%, 8.0%, 3.3%, 26.7%, and 86.7% and intensities were 1,090, 6.2, 1.6 1.0, 2.0, 2.0, and 7.2 metacercariae per fish infected respectively. Heterophyopsis continua metacercariae were found in 38.2% goby from 6 localities, i.e., Gochang-gun, Muan-gun, Shinan-gun, Gangjin-gun, Boseong-gun (Jeollanam-do) and Goseong-gun, and their intensities were relatively low, 1-21 metacercariae. Stictodora fuscata metacercariae were detected in 61.3% goby from 4 localities, i.e., Taean-gun, Gochang-gun, Hadong-gun and Goseong-gun. Their infection rates were 90.0%, 53.3%, 5.9%, and 73.3% and intensities were 1,081, 3.1, 3.0, and 10.2 metacercariae per fish infected respectively. Heterophyes nocens metacercariae were found in 55.0% goby from Muan-gun and Shinan-gun. Total 3 metacercariae of Isthmiophora hortensis were detected in 2 (8.0%) gobies from Shinan-gun. Total 15 metacercariae of Centrocestus armatus were detected in 5 gobies (33.3%) from Gyeongpo-ho (ho means lake) in Gangneung-si, Gangwon-do. The present study suggests that yellowfin goby, A. flavimanus, acts as the infection sources of zoonotic intestinal flukes in western and southern coastal areas than in eastern coastal areas of Korea.

Morphological and Molecular Characterization of Apharyngostrigea pipientis Faust, 1918 (Trematoda: Diplostomidea) from Ardeacinerea jouyi, Egretta intermedia and Nycticorax nycticorax in Korea

  • An intestinal strigeiid trematode, Apharyngostrigea pipientis, was detected from Ardea cinerea jouyi, Egretta intermedia and Nycticorax nycticorax between July, 2006 and June, 2020 in Daejeon metropolitan city and Kangwon-do, Republic of Korea. It was observed with a light and scanning electron microscope (SEM), and molecularly analyzed with 18S rRNA genes. The infection rate of the strigeiid trematode was 25.9% (7/27). The body is cylindrical in whole mount, greatly elongated in outline and shows a strong dorsal curvature. The overall measurements are: length 4.1-4.8 mm; fore-body 1.00-1.20 mm by 0.8-0.9 mm; hind-body 3.2-3.6 mm by 0.43-0.46 mm. The body is covered by a relatively thick cuticle, devoid of spines. The oral sucker leads into a short esophagus that is devoid of a muscular pharynx. The holdfast organ is enormously developed. The vitelline follicles distributed dorsal wall of fore-body and hind-body. This strigeiid trematode especially differs from other species of genus Apharyngostrigea in morphology. Although these A. pipientis are several morphological distinctions from Apharyngostrigea cornu, the partial sequence of 18S rRNA showed the sequence homology of A. pipientis (99.9%) and A. cornu (99.8%), and this result revealed their conspecific relationship between A. cornu and A. pipientis. This is the first report of A. pipientis in the Republic of Korea.

조개류에 기생하는 흡충류에 관한 연구 - Cercaria tapidis, Cercaria harengulae 의 기생율 변동 (Studies on parasitic trematodes in bivalves -Variation of infection rate of Cercaria tapidis and Cercaria harengulae-)

  • 1980~1982 년에 서해안 금강 하구산 바지락과 맛조개의 생식소내에 기생하는 흡충류의 유충인 Cercaria tapidis 와 Cercaria harengulae 의 기생율을 보고 한 후 동지역에서 13~14 년이 경과한 1994~1995 년 1 월까지 바지락과 맛조개에서 흡충류 유충의 기생율 변동 상황을 조사하였다. 먼저 바지락에 기생한 Cercaria tapidis 는 년평균 기생율이 1981 년의 5.7% 보다 2.4 배가 많은 14.0% 로 증가하였고, 월 기생율이 가장 높았던 달은 10 월의 32.0% 로 1981 년 12 월의 23.3% 보다 훨씬 높은 기생율을 나타냈다. 또 바지락에서의 Cercaria harengulae 의 년평균 기생율은 1980 년의 5.0% 보다 감소한 2.3% 이었으며, 월 기생율이 가장 높았던 1994 년 5 월의 6.7% 는 1980 년 3 월의 19.0% 보다 훨씬 감소된 기생율이었다. 맛조개에서의 Cercaria harengulae 의 년평균 기생율은 1981~1982 년의 10.2% 보다 감소된 6.9% 이었고, 월 최고 기생율도 1981 년에는 8 월에 23.4% 이었으나, 1994 년에는 4 월의 13.3% 로 감소 되었다. 또 Cercaria tapidis 는 1980 년 조사시와 같이 크기가 클수록 기생율이 높았으나, Cercaria harengulae 는 오히려 각장이 작을수록 기생율이 높아지는 경향을 나타냈다.

Endemicity of Zoonotic Trematode Metacercariae in Fish from Deokcheon-gang (River) in Sancheong-gun, Gyeongsangnam-do, Republic of Korea

  • The endemicity of zoonotic trematode metacercariae (ZTM) was investigated with total 871 freshwater fishes (19 species) from Deokcheon-gang (a branch stream of Gyeongho-gang) in Sancheong-gun, Gyeongsangnam-do, Korea for 3 years (2018-2020). All fishes were examined with the artificial digestion method. The metacercariae of Clonorchis sinensis (CsMc) were detected in 233 (36.3%) out of 642 fish in 11 positive fish species (PFS), and their infection intensity was 27 per fish infected (PFI). Especially, in index fish, Puntungia herzi, of CsMc infection, prevalence was 64.2% and infection intensity was 37 PFI. Metagonimus spp. metacercariae (MsMc) were found in 760 (87.5%) out of 869 fish in 18 PFS and their infection intensity was 228 PFI. In sweet smelt, Plecoglossus altivelis, the prevalence of MsMc was 97.6% and their infection intensity was 3,570 PFI. Centrocestus armatus metacercariae were detected in 209 (29.4%) out of 710 fish in 8 PFS and their infection intensity was 1,361 PFI. Echinostoma spp. metacercariae were found in 293 (42.6%) out of 688 fish in 15 PFS and their infection intensity was 5 PFI. Metacercariae of Clinostomum complanatum and Metorchis orientalis were also detected in 2.7% and 21.2% fish in 4 PFS and their infection intensities were 3.1 and 3.4 PFI respectively. By the present study, it was confirmed that some species of ZTM including CsMc and MsMc are more or less prevalent in fishes from Deokcheon-gang in Sancheong-gun, Gyeongsangnam-do, Korea.

Survey of Zoonotic Trematode Metacercariae in Fish from Irrigation Canal of Togyo-jeosuji (Reservoir) in Cheorwon-gun, Gangwon-do, Republic of Korea

  • The infection status of zoonotic trematode metacercariae (ZTM) was investigated in total 568 freshwater fishes (19 species) from the irrigation canal of Togyo-jeosuji (Reservoir) in Cheorwon-gun, Gangwon-do, the Republic of Korea for 3 years (2018-2020). All fishes were examined using the artificial digestion method. The metacercariae of Clonorchis sinensis (CsMc) were detected in 180 (43.8%) out of 411 fish of positive species, and their infection intensity was 38 per fish infected (PFI). Especially, in 2 fish species, i.e., Pseudorasbora parva and Puntungia herzi, the prevalence was 82.1% and 31.3%, and the infection intensity with CsMc was 88 and 290 PFI, respectively. Metagonimus spp. metacercariae (MsMc) were found in 403 (74.1%) out of 544 fish of positive species, and their infection intensity was 62 PFI. In the pale chub, Zacco platypus, the prevalence of MsMc was 98.6%, and their infection intensity was 144 PFI. Centrocestus armatus metacercariae were detected in 171 (38.9%) out of 440 fish of positive species, and their infection intensity was 1,844 PFI. Echinostoma spp. metacercariae were found in 94 (19.6%) out of 479 fish of positive species, and their infection intensity was 3 PFI. Metorchis orientalis metacercariae were detected in 43 (29.3%) out of 147 fish of positive species, and their infection intensity was 4 PFI. By the present study, it has been confirmed that some species of ZTM, including CsMc and MsMc, are prevalent in fishes from the irrigation canal of Togyo-jeosuji in Cheorwon-gun, Gangwon-do, Korea.

Molluscicidal Activity and Clinico-pathological Effect of Agave lophantha

  • Dry powder and different extracts of Agave lophantha were tested against Biomphalaria alexandrina. The results showed that the butanol extract has high molluscicidal activity. The activity of the dry powder has been found to be stable under the effect of some simulated field conditions. Also the toxicological effect of the plant on mice was tested through determination of certain parameters such as total protein, aspartate aminotransferase, alanine aminotransferase, alkaline phosphatase and acid phosphatase enzymes as well as histopathological study on liver and kidney.

Molecular Description of Macroorchis spinulosus (Digenea: Nanophyetidae) Based on ITS1 Sequences

  • We performed a molecular genetic study on the sequences of 18S ribosomal RNA (ITS1 region) gene in 4-day-old adult worms of Macroorchis spinulosus recovered in mice experimentally infected with metacercariae from crayfish in Jeollanam-do Province, Korea. The metacercariae were round, $180{\mu}m$ in average diameter, encysted with 2 layers of thick walls, but the stylet on the oral sucker was not clearly seen. The adult flukes were oval shape, and $760-820{\mu}m$ long and $320-450{\mu}m$ wide, with anterolateral location of 2 large testes. The phylogenetic tree based on ITS1 sequences of 6 M. spinulosus samples showed their distinguished position from other trematode species in GenBank. The most closely resembled group was Paragonimus spp. which also take crayfish or crabs as the second intermediate host. The present study is the first molecular characterization of M. spinulosus and provided a basis for further phylogenetic studies to compare with other trematode fauna in Korea.

Parasitic Infections Based on 320 Clinical Samples Submitted to Hanyang University, Korea (2004-2011)

  • We analyzed 320 clinical samples of parasitic infections submitted to the Department of Environmental Biology and Medical Parasitology, Hanyang University from January 2004 to June 2011. They consisted of 211 nematode infections, 64 trematode or cestode infections, 32 protozoan infections, and 13 infections with arthropods. The nematode infections included 67 cases of trichuriasis, 62 of anisakiasis (Anisakis sp. and Pseudoterranova decipiens), 40 of enterobiasis, and 24 of ascariasis, as well as other infections including strongyloidiasis, thelaziasis, loiasis, and hookworm infecions. Among the cestode or trematode infections, we observed 27 cases of diphyllobothriasis, 14 of sparganosis, 9 of clonorchiasis, and 5 of paragonimiasis together with a few cases of taeniasis saginata, cysticercosis cellulosae, hymenolepiasis, and echinostomiasis. The protozoan infections included 14 cases of malaria, 4 of cryptosporidiosis, and 3 of trichomoniasis, in addition to infections with Entamoeba histolytica, Entamoeba dispar, Entamoeba coli, Endolimax nana, Giardia lamblia, and Toxoplasma gondii. Among the arthropods, we detected 6 cases of Ixodes sp., 5 of Phthirus pubis, 1 of Sarcoptes scabiei, and 1 of fly larva. The results revealed that trichuriasis, anisakiasis, enterobiasis, and diphyllobothriasis were the most frequently found parasitosis among the clinical samples.
