Inhibition of Osteoclast Differentiation by Tanshinones from the Root of Salvia miltiorrhiza Bunge

  • We screened natural products to find compounds with anti-osteoporotic potential using a coculture-based system by which osteoclast differentiation is effectively achieved. We found that methylene chloride soluble fraction of the root of Salvia miltiorrhiza Bunge (Labiatae) suppressed osteoclast differentiation. Five tanshinones, tanshinone IIA (1), tanshinone I (2), cryptotanshinone (3), 15,16-dihydrotanshinone I (4), and ferruginol (5) were subsequently isolated from fraction. Among the five compounds, compounds 1-4 reduced the formation of TRAP­positive multinuclear osteoclasts. These results suggest that the identified tanshinones may be useful candidates for development of therapeutic agents to treat osteoporosis and other bone-resorptive diseases.

  • The isolation and purification of ferritin from the cricket, Gryllus bimaculatus haemolymph were accomplished by anion exchange column chromatography using HiTrap Q column (1.6 $\times$ 4 cm, Amersham Phamacia Co.), 7$0^{\circ}C$ heat and acid treatment, and gel filtration column chromatography using G4000SW column (0.75 $\times$ 60cm, TOSOH Co.) by FPLC system.   • We present a patient with anomalous origin of the left coronary artery from the pulmonary artery (ALCAPA), which was diagnosed and corrected in her 60 s. The patient is the oldest documented survivor of ALCAPA who underwent a surgical repair. ALCAPA should be corrected surgically to restore the dual coronary system at any age and this case shows that the surgical procedure may be performed safely even in an elderly patient.

  • KEPRI has studied planar type SOFC stacks using anode-supported single cells and kW class co-generation systems for residential power generation. A 1kW class SOFC system consisted of a hot box part, a cold BOP part and a water reservoir. A hot box part contains a SOFC stack made up of 48 cells with $10{\times}10cm^2$ area and ferritic stainless steel interconnectors, a fuel reformer, a catalytic combustor and heat exchangers. Thermal management and insulation system were especially designed for self-sustainable operation. A cold BOP part was composed of blowers, pumps, a water trap and system control units. When a 1kW class SOFC system was operated at $750^{\circ}C$ with hydrogen, the stack power was 1.2kW at 30 A and 1.6kW at 50A. Turning off an electric furnace, the SOFC system was operated using hydrogen and city gas without any external heat source. Under self-sustainable operation conditions, the stack power was about 1.3kW with hydrogen and 1.2kW with city gas respectively. The system also recuperated heat of about 1.1kW by making hot water. Recently KEPRI developed stacks using $15{\times}15cm^2$ cells and tested them. KEPRI will develop a 5 kW class CHP system using $15{\times}15cm^2$ stacks by 2010.

  • The information stored in the computer system needs to be protected from unauthorized access, malicious destruction or alteration and accidental inconsistency. In this paper, we propose an intrusion detection system based on sensor concept for defecting and preventing malicious attacks We use software sensor objects which consist of sensor file for each important directory and sensor data for each secret file. Every sensor object is a sort of trap against the attack and it's touch tan be considered as an intrusion. The proposed system is a new challenge of setting up traps against most interception threats that try to copy or read illicitly programs or data. We have implemented the proposed system on the Linux operating system using loadable kernel module technique. The proposed system combines host~based detection approach and network-based one to achieve reasonably complete coverage, which makes it possible to detect unknown interception threats.

  • As diesel engines have high power and good fuel economy on top of less $CO_2$ emissions, their market shares are increasing not only in commercial vehicles but also in passenger cars. LNT, urea-SCR and combination of them have been developed for after-treatment of the exhaust gas to reduce NOx on diesel vehicles. The aim of this study is to investigate the $NH_3$ generation characteristics of LNT catalyst downstream. It was found from the experiments of the LNT catalyst that $H_2$ was useful as a reductant in SCR catalyst because it can enhance the de-NOx performance and improve $NH_3$ selectivity. The $NH_3$ generation of the LNT, when hydrothermally aged at $900^{\circ}C$ for 18 hr, increased to about 90ppm at $300^{\circ}C$ due to Pt sintering and Ba agglomeration. LNT catalyst was most sulfur poisoning at $500^{\circ}C$. The $NH_3$ slip increased due to the reduction of residence time according to SV increase.

  • The increasing numbers of automobiles keep causing air-pollution problems worse than ever. Nowadays, research on catalyst converter and filter trap as a modern technology is very active because PM is designated as a major cancer material and stringent regulations on this are necessary and required. This research emphasized on the development of Continuously Regenerating DPF technology which was the best particulate matters removing technology of current existing technology because of its superior comparability and high applicability. This experimental study has been conducted with equipped and unequipped a Continuously Regenerating DPF ona displacement 7,000cc diesel engine and compared in terms of engine performance and emission. To measure the emission, D-13 mode is applied and measured quantities of the exhaust gases, particularly in CO, HC, PM, and NOx. Therefore, this research is focused on engine performance and characteristics on exhaust emissions with the application of a Continuously Regenerating DPF in a heavy-duty diesel vehicle.

  • Background: The Hypothalamo-Pituitary-Adrenal (HPA) axis is an important regulator for the body's stress response. As a primary stress responsive system, HPA-axis secretes various neurotransmitters, hormones, and cytokines, which regulates the immune system. Natural killer (NK) cell which is plays an important role in the innate immune response, is specially decreased their numbers and loose cytolytic activity in response to stress. However, the effect of HPA-axis secreted proteins on NK cell activity has not been defined. Herein, we studied the effect of adrenal secreted adiponectin on NK cell cytotoxicity. Adiponectin which is well-known metabolic control protein, plays important roles in various diseases, including hypertension, cardiovascular diseases, inflammatory disorders, and cancer. Methods: Signal sequence trap was used to find stress novel secretory protein from HP A-axis. Selected adiponectin was treated mouse mature primary NK cells and then examined the effect of adiponectin to NK cell cytotoxicity and cytokine expression level. Results: We found that adiponectin which is secreted from adrenal gland, suppress IL-2 induced NK cell cytotoxicity. And also investigated cytolytic cytokines are suppressed by adiponectin. Conclusion: These data suggest that adiponectin inhibites NK cell cytotoxicity via suppression of cytotoxicity related target gene.

  • Performance of electrostatic diesel PM filtration systems (E-DPS) with different types has been tested using the carbon particles generated by spark discharge in laboratory. Among the five electrostatic precipitators, the multiple wires cylindrical E-DPS with the highest collection efficiency and relatively lower differential pressure at the flow rate of $1\;m^3$/min, as an applicable device to diesel engine as an after treatment system, has been combined with another collection cylinder to improve the collection efficiency of diesel particulate matters generated from diesel engines. The multiple wires cylindrical E-DPS combined with the cylindrical collector showed the collection efficiency of more than 60% at the engine speed of 2,000 rpm with the engine loads of 25 and 50%.
