Factors Affecting Primary Culture of Nuclear Transfer Blastocysts for Isolation of Embryonic Stem Cells in Miniature Pigs

  • Pluripotent embryonic stem (ES) cells isolated from inner cell mass (ICM) of blastocyst-stage embryos are capable of differentiating into various cell lineages and demonstrate germ-line transmission in experimentally produced chimeras. These cells have a great potential as tools for transgenic animal production, screening of newly-developed drugs, and cell therapy. Miniature pigs, selectively bred pigs for small size, offer several advantages over large breed pigs in biomedical research including human disease model and xenotransplantation. In the present study, factors affecting primary culture of somatic cell nuclear transfer blastocysts from miniature pigs for isolation of ES cells were investigated. Formation of primary colonies occurred only on STO cells in human ES medium. In contrast, no ICM outgrowth was observed on mouse embryonic fibroblasts (MEF) in porcine ES medium. Plating intact blastocysts and isolated ICM resulted in comparable attachment on feeder layer and primary colony formation. After subculture of ES-like colonies, two putative ES cell lines were isolated. Colonies of putative ES cells morphologically resembled murine ES cells. These cells were maintained in culture up to three passages, but lost by spontaneous differentiation. The present study demonstrates factors involved in the early stage of nuclear transfer ES cell isolation in miniature pigs. However, long-term maintenance and characterization of nuclear transfer ES cells in miniature pigs are remained to be done in further studies.

  • The maximun short circuit current of modern power system is becoming so large that circuit breaker is not expected to be able to shut down the current in the future In order cut over-currents, a system composed of a superconducting fault current limiter(SFCL) and traditional breaker seems to provide a promising solution for furture power operation. In present paper, three line-to-ground fault is assumed to happen at the center of 345kV transmission lines in a large capacity electric power system. The superconducting fault current limiter was represented using a commutation type, which consists of a non-inductive superconducting coil and current limiting element (resistor or reactor). from the viewpoint of current limiting performance, the prevention of the voltage drop at the load bus and comparision characteristics for two type SFCL. Desired design specification and operation parameters of SECL were also given qualitatively by the performance.

Implementation of condition monitoring system in underground utility tunnels using inductive coupler (유도성 커플러를 이용한 지하공동구의 상태감시시스템)

  • The incidence of fire in underground utility tunnel is lower than other fires, but the damage caused by fire can cause social loss due to social management paralysis as well as economic loss. Hereupon, this paper presents the results of an empirical test on the construction of the underground utility tunnel condition monitoring system using the leakage coaxial cable installed in the underground utility tunnel. For this reason, a verification test was conducted by connecting a inductive coupler 200 Mbps power line communication modem with insertion loss characteristics of $-6{\pm}2dB$ to the installed the leakage coaxial cable installed in the underground utility tunnel. As a result, We confirmed sending/receiving of IP cameras up to 500 m. Therefore, it is judged that it is possible to construct a condition monitoring system for underground utility tunnel by using the leakage coaxial cables installed in the underground utility tunnels without installing additional communication lines for data transmission.

Health Status of Electric Utility Workers Exposed to Extremely Low Frequency Electromagnetic Field (ELF-EMF) (근로자들의 극저주파 전자파 노출 수준에 따른 인체 영향 평가)

  • Recently, the use of an electrical apparatus has brought up concerns of health risks from exposure to electromagnetic fields. EMF is composed of electric fields and magnetic fields. Heavy exposure to EMF can occur only in the vicinity of high-voltage overhead transmission lines, close to transformers and underground cables, and also close to large electrical machinery. In this thesis I have investigated the hypothesis of the correlation between occupational exposure to ELF-EMF and the risks of leukemia, anemia, cancer. Therefore, the aim of this study is to investigate whether or not ELF-EMF emitted from electric power stations and transformer substations affect some hematological parameters and tumor markers of electric utility workers. The hematological test results and tumor markers under investigation were similar in the two groups but some of parameters such as RBC, AFP, LDH showed significant difference between the two groups from two sample t-test (p<0.05). The exposure group showed increased LDH level compared to the control group by two sample t-tests. In addition, the abnormal LDH level in the exposure group was observed to be clinically significant by ${\chi}^2$-test. However, the levels of RBC, AFP observed were not clinically significant by ${\chi}^2$-test (p>0.05). These results suggested that ELF-EMF does not affect most blood test parameters except LDH of electric utility workers.

A Dual-Band Asymmetrical Metamaterial Antenna for Orthogonal Radiation Patterns (수직한 방사패턴을 형성하는 이중대역 비대칭 배열 메타물질 안테나)

  • A new metamaterial antenna with dual resonant modes is presented using an asymmetrical periodic arrangement for orthogonal radiation patterns. The proposed antenna produces two orthogonal modes by the asymmetrical periodic unit-cell arrangement. The orthogonal resonant mode provides perpendicular radiation patterns without changing the antenna polarization at each resonant mode. The fabricated antenna shows good agreements with the theoretical analysis of the electric-field. The experimental results shows the orthogonal radiation patterns along x- and y-axises, and gains are 3.34 and 3.86 dBi at each radiating resonant mode, respectively. Additionally, slotted ground structures are embedded on the back side of the antenna in order to reduce the size and enhance the radiation efficiency of 12 % and 27 %, respectively.

  • The EMI shielding effectiveness of the shielding layer thickness was analyzed when the metal shielding layer was placed in the near field of the magnetic probe and the noise source. Microstrip lines were used as noise source, and graphite and ferrite were selected as metal shielding materials. The magnetic probe uses the electromagnetic radiation measurement method using the magnetic probe by applying the IEC 61967-6 method. The transmission coefficient between the microstrip line and the magnetic probe was analyzed. The distance between the two was 1 mm for a single shielding layer and 5 mm for a multiple shielding layer. The thickness of the shielding layer was changed to 5 um, 10 um, 30 um, and 50 um. When the frequency was changed from 150 kHz to 1 GHz, a maximum shielding effectiveness (SE) of 44.9 dB was obtained.

Production of ginsenoside aglycone (protopanaxatriol) and male sterility of transgenic tobacco co-overexpressing three Panax ginseng genes: PgDDS, CYP716A47, and CYP716A53v2

  • Background: Protopanaxatriol (PPT) is an aglycone of ginsenosides, which has high medicinal values. Production of PPT from natural ginseng plants requires artificial deglycosylation procedures of ginsenosides via enzymatic or physicochemical treatments. Metabolic engineering could be an efficient technology for production of ginsenoside sapogenin. For PPT biosynthesis in Panax ginseng, damarenediol-II synthase (PgDDS) and two cytochrome P450 enzymes (CYP716A47 and CYP716A53v2) are essentially required. Methods: Transgenic tobacco co-overexpressing P. ginseng PgDDS, CYP716A47, and CYP716A53v2 was constructed via Agrobacterium-mediated transformation. Results: Expression of the three introduced genes in transgenic tobacco lines was confirmed by Reverse transcription-polymerase chain reaction (RT-PCR). Analysis of liquid chromatography showed three new peaks, dammarenediol-II (DD), protopanaxadiol (PPD), and PPT, in leaves of transgenic tobacco. Transgenic tobacco (line 6) contained $2.8{\mu}g/g$ dry weight (DW), $7.3{\mu}g/g$ DW, and $11.6{\mu}g/g$ DW of PPT, PPD, and DD in leaves, respectively. Production of PPT was achieved via cell suspension culture and was highly affected by auxin treatment. The content of PPT in cell suspension was increased 37.25-fold compared with that of leaves of the transgenic tobacco. Transgenic tobacco was not able to set seeds because of microspore degeneration in anthers. Transmission electron microscopy analysis revealed that cells of phloem tissue situated in the center of the anther showed an abnormally condensed nuclei and degenerated mitochondria. Conclusion: We successfully achieved the production of PPT in transgenic tobacco. The possible factors deriving male sterility in transgenic tobacco are discussed.

Apoptosis and inhibition of human epithelial cancer cells by ZnO nanoparticles synthesized using plant extract

  • The present research work reports in-vitro anti-cancer activity of biologically synthesized ZnO nanoparticles (ZnO NPs) against human carcinoma cells viz SCC-40, SK-MEL-2 and SCC-29B using Sulforhodamine-B (SRB) Assay. ZnO NPs were synthesized by a unique and novel biological route using Temperature-gradient phenomenon where the extract of combination of Catharanthus roseus (L.) G. Don (C. roseus), Azadirachta indica (A. indica), Ficus religiosa (F. religiosa) and NaOH solution were used as synthesis medium. The morphology of the ZnO NPs was characterized by Transmission Electron Microscopy (TEM). TEM images reveal that particle size of the samples reduces from 76 nm to 53 nm with the increase in reaction temperature and 68 nm to 38 nm with the increase in molar concentration of NaOH respectively. XRD study confirms the presence of elements and reduction in crystallite size with increase in reaction temperature and NaOH concentration. The diffraction peaks show broadening and a slight shift towards lower Bragg angle ($2{\theta}$) which represents the reduction in crystallite size as well as presence of uniform strain. The FTIR spectra of the extract show transmittance peak fingerprint of Zn-O bond and presence of bioactive molecules These NPs exhibit inhibition greater than 50% for SCC-40, SK-MEL-2 and SCC-29B cell lines and more than 50% cell kill for SCC-29B cells at concentrations < $80{\mu}g/ml$. Nanoparticles with smallest size have shown better anti-cancer activity and peculiar cell-selectivity. The combination of extracts of these plants with ZnO NPs can be used in targeted drug delivery as an effective anti-cancer agent, a potential application in cancer treatment.

  • I began to produce a series of works based upon the plastic element as a visual transmission -such as points, lines, sides, volumes and cubics, movements with in time, and space- which appeared in the optical art, that emerged in the 1950s and the early 1960. My works, which portray the natural scenes such as mountains, trees, and flowers that are commonly seen around us, are completed by drawing, erasing, filling with many points and overlapping. My works also ignore the scenography. All artistic activities must be based upon humanistic values and ultimately need to work for human being. The work tried to express more diverse impression rather than the emotion from the familiar perspective. We hope, therefore, my works give pleasure and satisfaction to these who come to see them.

Measurement of the ICRH antenna phasing using antenna strap probe based diagnostic system in EAST tokamak

  • To operate the ion cyclotron resonance heating (ICRH) antennas in a better heating state and produce relatively low impurities, it is necessary to control the antenna spectrum by changing the antenna phasing. As the electrical length of the antenna feeding transmission lines is changing as a matter of the standing wave pattern at the ceramic supports, 90° elbows, T-connectors and antenna loops, we chose to measure the current at the grounding points of the antenna loops by antenna strap probe. The voltage drops along a small, several millimeter-long paths at the end of the antenna loops give a signal that is proportional to the current in the antenna loop. Through the simulation of the antenna strap probe and the actual measurement of the antenna phasing under vacuum conditions, the reliability of the antenna strap probe based diagnostic system have been successfully proved. Moreover, this system was successfully applied to the ICRH daily experiments in the spring of 2021. In the near future, the active real-time feedback control of the antenna phasing system will be developed based on this diagnostic system in the EAST tokamak.
