Hair Growth Effect of TS-SCLF from Schisandra chinensis Extract Fermented with Lactobacillus plantarum

  • This study investigated the hair growth effect of Schisandra chinensis extract (TS-SC) and TS-SC fermented by Lactobacillus plantarum (TS-SCLF) on human dermal papilla cells (hDPCs). The production of vascular endothelial growth factor (VEGF), insulin-like growth factor 1 (IGF-1), keratinocyte growth factor/fibroblast growth factor 7 (KGF/FGF-7) and hepatocyte growth factor (HGF), transforming growth factor beta 1 (TGF-β1) were examined. The secretion rates of VEGF and KGF/FGF-7 were high in TS-SC, and the secretion rates of IGF-1 and HGF were high in TS-SCLF. TGF-β1 was inhibited in a concentration-dependent manner in all samples. Gene expression of VEGF, IGF-1, KGF, HGF and alkaline phosphatase, relevant to hair growth, were examined. The data revealed that TS-SC and TS-SCLF successfully promoted hair growth in hDPCs. The IGF-1 gene was expressed in a dose-dependent manner in TS-SCLF. These results indicate that TS-SC and TS-SCLF fermented extract effectively promoted hair growth and gene expression relevant to hair growth in hDPCs. Used in clinical trials the test substance 'CMK-LPF01' showed a statistically significant increase in the number of hairs at 8 weeks, 16 weeks, and 24 weeks compared to before product use, and a change in hair growth, a secondary efficacy evaluation variable. Through additional research in the future, it is expected that "CMK-LPF01" can be developed as a functional material that can help alleviate symptoms of hair loss.

A sporadic case of Loeys-Dietz syndrome type I with two novel mutations of the TGFBR2 gene

  • A recently recognized connective tissue disorder, Loeys-Dietz syndrome (LDS) is a genetic aortic aneurysm syndrome caused by mutations in the transforming growth factor-receptor type I or II gene (TGFBR1 or TGFBR2). They have distinctive phenotypic abnormalities including widely spaced eyes (hypertelorism), bifid uvula or cleft palate, and arterial tortuosity with aortic aneurysm or dissection throughout the arterial tree. LDS is characterized by aggressive and rapid progression of aortic aneurysm. Therefore, the patients with distinct phenotype, marked aortic dilatation and aneurysm at early age should be suspected to be affected by LDS and rapid TGFBR gene analysis should be done. We report one child diagnosed as LDS due to typical phenotypes and two novel missense mutations of the TGFBR2 gene (c.1526G>T and c.1528A>T).

Upstream Regulators and Downstream Effectors of NADPH Oxidases as Novel Therapeutic Targets for Diabetic Kidney Disease

  • Oxidative stress has been linked to the pathogenesis of diabetic nephropathy, the complication of diabetes in the kidney. NADPH oxidases of the Nox family, and in particular the homologue Nox4, are a major source of reactive oxygen species in the diabetic kidney and are critical mediators of redox signaling in glomerular and tubulointerstitial cells exposed to the diabetic milieu. Here, we present an overview of the current knowledge related to the understanding of the role of Nox enzymes in the processes that control mesangial cell, podocyte and tubulointerstitial cell injury induced by hyperglycemia and other predominant factors enhanced in the diabetic milieu, including the renin-angiotensin system and transforming growth factor-${\beta}$. The nature of the upstream modulators of Nox enzymes as well as the downstream targets of the Nox NADPH oxidases implicated in the propagation of the redox processes that alter renal biology in diabetes will be highlighted.

Novel Association of a Familial TGFBR1 Mutation in Loeys-Dietz Syndrome with Concomitant Hematologic Malignancy

  • Concomitant Loeys-Dietz syndrome (LDS) and hematologic malignancies are exceptionally rare. This is the first report of a patient operated on for aortic root dilation who had been previously diagnosed with LDS and B-cell-lymphoma. After completion of chemotherapy and complete remission, an elective valve-sparing aortic root replacement (using the David-V method) was performed. Due to the positive family history, preoperative genetic counseling was conducted, and revealed LDS with a TGFBR1 (transforming growth factor beta receptor type I) mutation in 6 probands of the family, albeit in 1 of them posthumously. This missense mutation has been previously described in relation to aortic dissection, but a causative relationship to malignancy has so far neither been proposed nor proven.

TGF-β1 Protein Expression in Non-Small Cell Lung Cancers is Correlated with Prognosis

  • To investigate the expression intensity and prognostic significance of TGF-${\beta}1$ protein in non-small cell lung cancer (NSCLC), immunohistochemistry was carried out in 194 cases of NSCLC and 24 cases of normal lung tissues by SP methods. The PU (positive unit) value was used to assess the TGF-${\beta}1$ protein expression in systematically selected fields under the microscope with Leica Q500MC image analysis. We found that the TGF-${\beta}1$ PU value was nearly two-fold higher in NSCLC than in normal lung tissues (p=0.000), being associated with TNM stages (p=0.000) and lymph node metastases (p=0.000), but not to patient age, gender, smoking history, tumor differentiation, histological subtype and tumor location (P>0.05). Univariate analysis indicated that patients with high TGF-${\beta}1$ protein expression and lymph node metastases demonstrated a poor prognosis (both p=0.000,). Multivariate analysis showed that TGF-${\beta}1$ protein expression (RR = 2.565, p=0.002) and lymph node metastases (RR=1.874, p=0.030) were also independent prognostic factors. Thus, TGF-${\beta}1$ protein expression may be correlated to oncogenesis and serve as an independent prognostic biomarker for NSCLC.

Increased α2-6 sialylation of endometrial cells contributes to the development of endometriosis

  • Endometriosis is a disease characterized by implants of endometrial tissue outside the uterine cavity and is strongly associated with infertility. Focal adhesion of endometrial tissue to the peritoneum is an indication of incipient endometriosis. In this study, we examined the effect of various cytokines that are known to be involved in the pathology of endometriosis on endometrial cell adhesion. Among the investigated cytokines, transforming growth factor-${\beta}1$ ($TGF-{\beta}1$) increased adhesion of endometrial cells to the mesothelium through induction of ${\alpha}2-6$ sialylation. The expression levels of ${\beta}$-galactoside ${\alpha}2-6$ sialyltransferase (ST6Gal) 1 and ST6Gal2 were increased through activation of $TGF-{\beta}RI/SMAD2/3$ signaling in endometrial cells. In addition, we discovered that terminal sialic acid glycan epitopes of endometrial cells engage with sialic acid-binding immunoglobulin-like lectin-9 expressed on mesothelial cell surfaces. Interestingly, in an in vivo mouse endometriosis model, inhibition of endogenous sialic acid binding by a $NeuAc{\alpha}2-6Gal{\beta}1$-4GlcNAc injection diminished $TGF-{\beta}1$-induced formation of endometriosis lesions. Based on these results, we suggest that increased sialylation of endometrial cells by $TGF-{\beta}1$ promotes the attachment of endometrium to the peritoneum, encouraging endometriosis outbreaks.

Apolipoprotein A1 Inhibits TGF-β1-Induced Epithelial-to-Mesenchymal Transition of Alveolar Epithelial Cells

  • Background: Idiopathic pulmonary fibrosis (IPF) is a progressive and lethal lung disease characterized by the accumulation of excessive fibroblasts and myofibroblasts in the extracellular matrix. The transforming growth factor ${\beta}1$ (TGF-${\beta}1$)-induced epithelial-to-mesenchymal transition (EMT) is thought to be a possible source of fibroblasts/myofibroblasts in IPF lungs. We have previously reported that apolipoprotein A1 (ApoA1) has anti-fibrotic activity in experimental lung fibrosis. In this study, we determine whether ApoA1 modulates TGF-${\beta}1$-induced EMT in experimental lung fibrosis and clarify its mechanism of action. Methods: The A549 alveolar epithelial cell line was treated with TGF-${\beta}1$ with or without ApoA1. Morphological changes and expression of EMT-related markers, including E-cadherin, N-cadherin, and ${\alpha}$-smooth muscle actin were evaluated. Expressions of Smad and non-Smad mediators and TGF-${\beta}1$ receptor type 1 ($T{\beta}RI$) and type 2 ($T{\beta}RII$) were measured. The silica-induced lung fibrosis model was established using ApoA1 overexpressing transgenic mice. Results: TGF-${\beta}1$-treated A549 cells were changed to the mesenchymal morphology with less E-cadherin and more N-cadherin expression. The addition of ApoA1 inhibited the TGF-${\beta}1$-induced change of the EMT phenotype. ApoA1 inhibited the TGF-${\beta}1$-induced increase in the phosphorylation of Smad2 and 3 as well as that of ERK and p38 mitogen-activated protein kinase mediators. In addition, ApoA1 reduced the TGF-${\beta}1$-induced increase in $T{\beta}RI$ and $T{\beta}RII$ expression. In a mouse model of silica-induced lung fibrosis, ApoA1 overexpression reduced the silica-mediated effects, which were increased N-cadherin and decreased E-cadherin expression in the alveolar epithelium. Conclusion: Our data demonstrate that ApoA1 inhibits TGF-${\beta}1$-induced EMT in experimental lung fibrosis.

인간 자궁내막의 탈락막화에서 HOXA10 유전자의 역할 (Role of HOXA Gene in Human Endometrial Decidualization)

  • 목 적: Small interfering RNA (siRNA)를 이용하여 homeobox (HOXA) 10 유전자의 발현이 억제된 일차배양 자궁내막 세포를 이용하여 자궁내막 탈락막화 (decidualization)에 HOXA유전자를 포함한 세포 내 신호전달기전을 분석하고자 하였다. 연구방법: 본원 산부인과에서 자궁내막 질환 이외의 이유로 전자궁 적출술을 받은 환자의 자궁내막 조직을 채취한다. $37^{\circ}C$에서 20분간 Trypsin-EDTA를 처리하여 단일세포로 분리한 후 10% fetal bovine serum이 첨가된 DMEM/F12 배지를 이용하여 24시간 동안 $37^{\circ}C$ 5% $CO_2$ 배양기 안에서 배양한다. 배양된 자궁내막 세포를 HOXA10 siRNA로 첨가한 후 TGF-${\beta}1$을 10 ng/mL 농도로 48시간 첨가하여 탈락막화를 유도한다. 배양된 자궁내막 세포에서 reverse transcription polymerase chain reaction을 이용하여 HOXA10, prolactin, cyclooxygenase (COX)-2, peroxisome proliferator-activated receptor (PPAR)-$\gamma$ 및 wingless-type MMTV integration site family (Wnt)의 발현을 관찰하였다. 결 과: HOXA10의 경우 transforming growth factor (TGF)-${\bata}1$과 HOXA10 siRNA를 처리하지 않은 대조군에 비하여 TGF-${\beta}1$을 처리한 군에서 약 1.8배 가량 발현양의 증가를 보였다. 자궁내막 탈락막 표지인자로 알려져 있는 prolactin의 경우 TGF-${\beta}1$을 처리한 경우 대조군에 비하여 유의한 발현의 증가를 보였으며 HOXA10 siRNA를 처리한 군에 있어서는 TGF-${\beta}1$을 첨가하더라도 prolactin mRNA의 발현양의 증가를 관찰할 수 없었다. 또한 자궁내막 세포의 분화인자로 알려져 있는 COX-2의 발현 역시 HOXA10 siRNA를 처리한 군에 있어서 mRNA 발현양이 유의하게 감소하였으며 TGF-${\beta}1$을 처리하여도 발현의 증가를 관찰할 수 없었다. Wnt4의 경우 HOXA10 siRNA를 이용하여 HOXA10의 발현을 억제한 경우 대조군에 비하여 유의하게 mRNA의 발현양이 감소하였으며 이러한 발현양의 감소는 TGF-${\beta}1$을 처리하여도 증가됨을 관찰할 수 없었다. PPAR$\gamma$의 발현은 HOXA10 siRNA의 처리와 관계없이 TGF-${\beta}1$에 의하여 감소하는 것을 관찰할 수 있었다. 결 론: Progesterone에 의하여 자궁내막 상피세포에서 분비되는 것으로 알려져 있는 TGF-${\beta}1$에 의한 자궁내막 기질세포의 분화 (탈락막화)는 HOXA10 및 Wnt에 의하여 조절되는 것으로 생각된다.

미세변화 신증후군 환아에서 사이클로스포린 치료 후 Osteopontin과 Transforming Growth Factor-${\beta}$의 발현 (Expression of Osteopontin and Transforming Growth Factor- ${\beta}$ in Childhood Minimal Change Nephrotic Syndrome After Cyclosporine Treatment)

  • 목적 : 사이클로스포린(cyclosporine)은 효과적인 면역 억제제로서 현재에는 장기 이식 후 뿐 아니라 다양한 자가면역성 질병에서 그 사용 범위를 넓히고 있다. 신독성은 사이클로스포린의 대표적인 부작용으로서 그 특징적인 조직학적 소견은 간질의 섬유화와 세 동맥의 초자양 변화이다. 간질 섬유화에는 전통적으로 $TGF-{\beta}$가 관여하는 것으로 알려져 있으며 최근에 osteopontin이라는 단백도 관여한다는 사실이 동물 실험을 통해 알려지고 있다. 사이클로스포린에 의한 간질 섬유화에 osteopontin이 관여한다는 사실이 여러 동물실험을 통하여 알려져 있으나 사람에서는 아직까지 분명한 결론을 얻지 못하고 있다. 동일 환자에서 사이클로스포린 치료 전과 후에 두 번 생검을 시행한 신장 조직에서 osteopontin과 $TGF-{\beta}$의 발현을 조사하여 사이클로스포린과 이들의 관계를 밝히는 것이 본 연구의 목적이다. 대상 및 방법 : 연세의대 세브란스 병원 소아과에서 신장 생검으로 미세변화 신증후군으로 진단 받은 19예를 대상으로 하여 사이클로스포린 치료 전 조직 검사 검체를 대조군으로 하고, 치료 후 조직 검사 검체를 비교군으로 하였다. 사구체 메산지움과 세뇨관에서의 osteopontin 발현과 간질에서의 $TGF-{\beta}$ 발현을 면역조직화학염색으로 측정하였다. 결과 : 사이클로스포린 치료 후 사구체 메산지움과 세뇨관에서 osteopontin의 발현이 의의 있게 증가하였지만 간질에서 $TGF-{\beta}$의 증가는 뚜렷하지 않았다. 또한 세뇨관의 osteopontin과 간질의 $TGF-{\beta}$의 변화는 광학현미경으로 관찰되는 간질 섬유화와는 상관관계가 없었다. 따라서 사이클로스포린 사용시 osteopontin의 발현의 증가와 간질 섬유화 간에는 직접적인 연관이 없는 것으로 사료된다. 결론 : 본 연구의 결과 실험 동물에서와는 달리 사람의 신장에서 osteopontin은 간질 섬유화에 직접 관여하기 보다는 신장 손상에 대한 표지자로서의 의미가 있다고 추정된다.

Functional analysis of Bombyx mori Decapentaplegic gene for bone differentiation in a mammalian cell

  • Bone morphogenetic proteins (BMPs) belong to the transforming growth factor (TGF-${\beta}$) superfamily and are involved in osteoblastic differentiation. The largest TGF-${\beta}$ superfamily subgroup shares genetic homology with human BMPs (hBMPs) and silkworm decapentaplegic (dpp). In addition, hBMPs are functionally interchangeable with Drosophila dpp. Bombyx mori dpp may induce bone formation in mammalian cells. To test this hypothesis, we synthesized the 1,285-base pairs cDNA of full-length B. mori dpp using total RNAs obtained from the fat body of 3-day-old of the $5^{th}$ instar larvae and cloned the cDNA into the pCEP4 mammalian expression vector. Next, B. mori dpp was expressed in C3H10T1/2 cells. The target cells transfected with the pCEP4-Bm dpp plasmid showed biological functions similar to those of osteogenic differentiation induction growth factors such as hBMPs. We determined the relative mRNA expression rates of Runt-related transcription factor 2 (RUNX2), osterix, osteocalcin, and alkaline phosphatase (ALP) to validate the osteoblast-specific differentiation effects of B. mori dpp by performing quantitative real-time RT-PCR. Interestingly, mRNA expression levels of the 3 marker genes except RUNX2, in cells expressing B. mori dpp were much higher than those in control cells and C3H10T1/2 cells transfected with pCEP4. These results suggested that B. mori dpp signaling regulates osterix expression during osteogenic differentiation via RUNX2-independent mechanisms.
