Regulation of Transforming Growth Factor ${\beta}1$, Platelet-Derived Growth Factor, and Basic Fibroblast Growth Factor by Silicone Gel Sheeting in Early-Stage Scarring

  • Background Hypertrophic scars and keloids are associated with abnormal levels of growth factors. Silicone gel sheets are effective in treating and preventing hypertrophic scars and keloids. There has been no report on the change in growth factors in the scar tissue following the use of silicone gel sheeting for scar prevention. A prospective controlled trial was performed to evaluate whether growth factors are altered by the application of a silicone gel sheet on a fresh surgical scar. Methods Four of seven enrolled patients completed the study. Transforming growth factor (TGF)-${\beta}1$, platelet-derived growth factor (PDGF), and basic fibroblast growth factor (bFGF) were investigated immunohistochemically in biopsies taken from five scars at 4 months following surgery. Results In both the epidermis and the dermis, the expression of TGF-${\beta}1$ (P=0.042 and P=0.042) and PDGF (P=0.043 and P=0.042) was significantly lower in the case of silicone gel sheet-treated scars than in the case of untreated scars. The expression of bFGF in the dermis was significantly higher in the case of silicone gel sheet-treated scars than in the case of untreated scars (P=0.042), but in the epidermis, the expression of bFGF showed no significant difference between the groups (P=0.655). Conclusions The levels of TGF-${\beta}1$, PDGF, and bFGF are altered by the silicone gel sheet treatment, which might be one of the mechanisms of action in scar prevention.

CONTROL OF SCARRING IN ADULT WOUNDS USING ANTISENSE TRANSFORMING GROWTH FACTOR-$\beta$ OLIGODEOXYNUCLEOTIDES

  • Although synthetic antisense oligodeoxynucleotides (ODNs) have been used to dissect gene function in vitro, technical difficulties of targeted delivery prevented the use of this approach for investigating the effect of gene products in vivo. Here we report the use of local delivery of antisense transforming growth factor-${\beta}$l (TGF-${\beta}$1) oligonucleotides to decrease the fibrosis in the skin wound. Adult wounds heal with scar-tissue formation, whereas fetal wounds heal without scarring and with a lesser inflammatory and cytokine response. We reasoned that strategy emptying antisense TGF-${\beta}$1 ODNs complementary to TGF-${\beta}$1 mRNA might decrease the scarring in dermal wound of mouse. To evaluate this concept, we tested the effects of antisense ODNs targeted to TGF-${\beta}$1 mRNA by topical application of the chemical on the skin wound. Phosphorothioate antisense ODNs was employed to retard their degradation. When antisense TGF-${\beta}$1 ODNs were applied into the wound site, there was a maked reduction of scar compared with control wound site, These effects of antisense TGF-${\beta}$1 ODNs on the scar formation were associated with decreased expression of TGF-${\beta}$1 gene. However sense TGF-${\beta}$l ODNs had no effect on expression of TGF-${\beta}$1 gene. Also, control wounds healed with excessive fibrosis, whereas the antisense treated wounds healed with less fibrosis. In conclusion, our results indicate that antisense TGF-${\beta}$1 ODNs could be used for amelioating scar formation during wound healing.

Asian Dust Particles Induce TGF-${\beta}_1$ via Reactive Oxygen Species in Bronchial Epithelial Cells

  • Background: Asian dust storms can be transported across eastern Asia. In vitro, Asian dust particle-induced inflammation and enhancement of the allergic reaction have been observed. However, the fibrotic effects of Asian dust particles are not clear. Production of transforming growth factor ${\beta}_1$ (TGF-${\beta}_1$) and fibronectin were investigated in the bronchial epithelial cells after exposure to Asian dust particulate matter (AD-PM10). Methods: During Asian dust storm periods, air samples were collected. The bronchial epithelial cells were exposed to AD-PM10 with and without the antioxidant, N-acetyl-L-cysteine (NAC). Then TGF-${\beta}_1$ and fibronectin were detected by Western blotting. The reactive oxygen species (ROS) was detected by the measurement of dicholorodihydrofluorescin (DCF), using a FACScan, and visualized by a confocal microscopy. Results: The expression of TGF-${\beta}_1$, fibronectin and ROS was high after being exposed to AD-PM10, compared to the control. NAC attenuated both TGF-${\beta}_1$ and fibronectin expression in the AD-PM10-exposed the bronchial epithelial cells. Conclusion: AD-PM10 may have fibrotic potential in the bronchial epithelial cells and the possible mechanism is AD-PM10-induced intracellular ROS.

  • 본 논문에서는 간섭원리에 기반한 컴퓨터형성홀로그램과 광굴절매질인 BaTiO$_3$를 이용하여 광 정보 보호 시스템을 구현하였다. 먼저 원영상을 재생할 수 있는 이진위상홀로그램을 생성한 후, 이를 암호화할 영상으로 간주한다. 그리고 이 홀로그램에 간섭 원리를 적용하여 암호화된 영상과 기준영상(키 정보)을 얻는다. 복호화과정은 Mach-Zehnder 간섭계에서 암호화된 영상과 기준영상과의 간섭을 통해 간섭세기 정보를 얻고, 이를 LCD를 이용하여 위상 정보로 변환한 후 푸리에 변환함으로써 원영상을 얻는다. 이 과정에서 두 영상의 간섭을 통해 얻어지는 세기 정보는 주위 진동에 상당히 민감하다. 그래서 광굴절매질인 BaTiO$_3$의 SPPCM 특성을 이용하여 안정한 간섭 무늬를 얻는다. 제안된 방법은 원래의 영상 대신 그 영상의 홀로그램을 암호화하므로 이중 영상 암호 효과의 장점이 있다.

R-C 분포회로에 관한 연구 (Research on R-C Distributed Circuits)

  • 어떤 R-C 분포회로의 징분방정식의 해가 기지일 때 이것을 이용하여 다수의 다른 분포회로의 징분방정식의 해를 구하는 방법이 얻어졌다. 임의의 R-C 분포회로를 제작의 편의상 R(x)C(x)=일정인 회로로 등가변환하는 도해적방법이 얻어졌으며 해석적증명도 가능하다. 또 이론적 취급의 통일과 간단을 위하여 임의의 R-C분포회로를 R(x), C(x) 어느 한쪽이 일정한 분포회로로 등가변환하는 방법이 얻어졌다. 이 후자의 회로와 근사적인 집중정수회로를 생각하므로서 분포회로의 근사적인 해석, 합성이 비교적 간단하게 이루어 질 수 있다. 6x A method by which solutions of the differential equations of any other distributed circuits can be obtained is described when the solution of the differential equation of an R-C distributed amplifier is known. A graphical method of transforming any R-C ditributed circuit into an equivalent circuit which has a constant R(x)-C(x) was also obtained. The theoretical verification of this method is possible. For simplicity, any R-C distributed circuit can be transformed into an equivalent circuit which is a distributed circuit of either constant R(x) or C(x). Using this equivalent circuit and considering a lumped circuit, an approximate analysis and synthesis can be made simply.

  • N데이터센터는 정부부처를 대상으로 클라우드 컴퓨팅 서비스를 제공하는 '클라우드 컴퓨팅센터'로 변모를 꾀하고 있으며, 각 부처에 필요한 만큼 정보자원을 서비스 형태로 제공하는 'IT서비스 센터'로 탈바꿈 예정이다. N센터 8중의 보안체계 하에서 클라우드서비스가 이미 정부부처에 제공되고 있으며, 향후 보안을 전제조건으로 민간분야까지 확대할 계획이다. 따라서 보안은 민간분야와의 클라우드 확산에 있어 선결요건이며, 이의 체계적이고 효율적 추진을 위해서 클라우드 데이터 센터로서의 보안수준을 파악하고 적절한 방안을 제시할 필요가 있다. 본 연구에서는 이를 위해 선진 각국의 클라우드 데이터 센터의 보안 요건을 분석하고 클라우드 서비스 형태에 클라우드 컴퓨팅의 취약점을 파악하고, 선진 민간 클라우드 데이터 센터의 보안수준을 파악하고, 현행 N데이터센터 보안 수준과 선진 민간 클라우드 데이터센터와의 갭을 분석하여 보안관점에서 전환을 위한 요건을 제시하였다.

In vitro and in vivo evaluation of tissue-cultured mountain ginseng on penile erection

  • Background: Progressed tissue culture techniques have allowed us to easily obtain mass products of tissue-cultured mountain ginseng over 100 yr old (TCMG-100). We investigated the effects of TCMG-100 extract on erectile function using in vitro and in vivo studies. Methods: To examine the relaxation effects and mechanisms of action of TCMG-100 on rabbit cavernosal strips evaluated in an organ bath. To investigate the long-term treatment effect of TCMG-100, 8-wk administration was performed. After administration of TCMG-100, intracavernosal pressure, cyclic guanosine monophosphate and nitric oxide (NO) levels of cavernosal tissue, serum testosterone level, histological observation of collagen fiber, endothelium, smooth muscle cell, and transforming growth factor-${\beta}1$ were investigated. Results: TCMG-100 extract displayed dose-dependent relaxation effects on precontracted rabbit corporal smooth muscle. The TCMG-100-induced relaxation was significantly reduced by removing the endothelium, and treatment with an NO synthase inhibitor or NO scavenger. Eight weeks of TCMG-100 administration increased intracavernosal pressure in a rat model. The levels of cyclic guanosine monophosphate and NO in the corpus callosum and serum testosterone level were also increased by TCMG-100 treatment. Furthermore, histological evaluation of collagen, smooth muscle, and endothelium showed increases in endothelium and smooth muscle, and a decrease in transforming growth factor-${\beta}1$ expression. Conclusion: These relaxation effects on corporal smooth muscle and increased erectile function suggest that TCMG-100 might be used as an alternative herbal medicine to improve erectile function.

Expression and Activation of Transforming Growth Factor-Beta 2 in Cultured Bone Cells

  • Transforming growth factor-$\beta$ (IGF-$\beta$)s are multifunctional small polypeptides synthesized in most cell types. TGF-$\beta$ exerts pivotal effects on both bone formation and resorption. In addition, increasing lines of evidence implicate TGF-$\beta$ as a potential coupling factor between these two processes during bone remodeling. In the present study, the expression form and the activation mechanism of latent-TGF-$\beta$ were investigated using specific antibodies for each isoform. TGF-$\beta$s were observed to be synthesized and accumulated in a large amount in cultured osteoblastic cells. The estimated molecular weights of intracellular TGF-$\beta$2 and -$\beta$3 were 49 and 55 kDa, respectively. Based on proteolytic digestion study and immunofluorescence observation, these precursor forms seemed to be accumulated in distinct intracellular compartments. To examine whether the internal pool of TGF-$\beta$ was possiblely regulated by external signals, their biological activites were examined in a conditioned media of this cell. Although the intact conditioned media did not contain detectable TGF-$\beta$ activity, heat-treatment or acid-activation of the conditioned media revealed significant TGF-$\beta$ activity. Furthermore, in the presence of estrogen, this activity was dramatically diminished. It is known that activation of latent TGF-$\beta$ can be achieved by different chemical and enzymatic treatments, or by incubation with certain cell types. This extracellular activation was suggested as a key step in the regulation of TGF-$\beta$ activity. In addition to these extracellular activation, this study suggests that the synthesis and intracellular processing are important regulation steps for TGF-$\beta$ action. In addition, this regulation Is specific for TGF-$\beta$ type 2, because the change was not observed in TGF-$\beta$3 in osteoblastic cell line.

시각장애인 인지특성을 기반으로 한 영상자료의 촉각지도 자동 변환 방법 (Automatic Method for Transforming Image into Tactile Map Based on Cognitive Characteristics of Visually Impaired Persons)

  • 본 논문에서는 시각장애인에게 다양한 영상정보를 보다 손쉽게 제공하기 위하여 시각장애인의 인지특성을 기반으로 기존 영상자료를 촉각지도 정보로 자동 변환하는 방법을 제안한다. 제안하는 방법에서는 일반 교과서에 수록된 컬러영상의 밝기값 히스토그램을 분석하여 저복잡도 및 고복잡도 영상으로 분류하고, 이를 각 영상 종류에 적합한 서로 다른 변환과정을 적용하여 시각장애인이 인지할 수 있는 촉각지도 정보로 자동 변환한다. 실험 평가에서는 제안한 자동 변환 방법으로 생성된 촉각지도와 수동으로 생성된 기존 점자교과서의 촉각지도들을 가지고 전맹 시각장애인 5인을 대상으로 인지율과 만족도를 평가하는 실험을 수행하였다. 실험결과 제안한 방법으로 생성된 촉각지도에 대한 인지율과 만족도가 기존 점자교과서의 촉각지도에 대한 것들과 매우 유사하였다. 따라서 시각장애인을 위한 교육자료 제작의 시간과 비용을 획기적으로 절감할 수 있으며, 또한 교육자료 내에 영상자료를 보다 손쉽게 많이 추가하여 교육효과를 크게 향상시킬 수 있다.

후코이단에 의한 인간 폐 섬유모세포의 활성 억제 효과 (Inhibitory Effect of Fucoidan on TGF-β1-Induced Activation of Human Pulmonary Fibroblasts)

  • Fucoidan, one of the dominant sulfated polysaccharides extracted from brown seaweed, possesses a wide range of biological activities. Transforming growth $factor-{\beta}$ ($TGF-{\beta}$) plays a pivotal role in the pathogenesis of pulmonary fibrosis, by stimulating the synthesis of profibrotic factors. In this study, we investigated the in vitro effects of fucoidan on collagen synthesis, ${\alpha}-smooth$ muscle actin (${\alpha}-SMA$) expression, and interleukin (IL)-6 production in $TGF-{\beta}$-stimulated human pulmonary fibroblasts. The expression of type I collagen and ${\alpha}-SMA$ was detected by Western blot, and the production of IL-6 by enzyme-linked immunosorbent assay. $TGF-{\beta}1$ treatment of pulmonary fibroblasts enhanced the expression of ${\alpha}-SMA$, type I collagen, and IL-6 whereas these effects were inhibited in cells pretreated with fucoidan. The activation of Smad2/3, p38 mitogen-activated protein kinases (MAPKs), and Akt was also inhibited in fucoidan-pretreated, $TGF-{\beta}1-stimulated$ human pulmonary fibroblasts. These data demonstrate the anti-fibrotic potential of fucoidan in $TGF-{\beta}-induced$ human pulmonary fibroblasts, via the inhibition of Smad2/3, p38 MAPKs, and Akt phosphorylation. Our results suggest the therapeutic potential of fucoidan in the prevention or treatment of pulmonary fibrosis.
