The Role of N-Acetyl Transferases on Isoniazid Resistance from Mycobacterium tuberculosis and Human: An In Silico Approach

  • Background:N-acetyl transferase (NAT) inactivates the pro-drug isoniazid (INH) to N-acetyl INH through a process of acetylation, and confers low-level resistance to INH in Mycobacterium tuberculosis (MTB). Similar to NAT of MTB, NAT2 in humans performs the same function of acetylation. Rapid acetylators, may not respond to INH treatment efficiently, and could be a potential risk factor, for the development of INH resistance in humans. Methods: To understand the contribution of NAT of MTB and NAT2 of humans in developing INH resistance using in silico approaches, in this study, the wild type (WT) and mutant (MT)-NATs of MTB, and humans, were modeled and docked, with substrates and product (acetyl CoA, INH, and acetyl INH). The MT models were built, using templates 4BGF of MTB, and 2PFR of humans. Results: On the basis of docking results of MTB-NAT, it can be suggested that in comparison to the WT, binding affinity of MT-G207R, was found to be lower with acetyl CoA, and higher with acetyl-INH and INH. In case of MT-NAT2 from humans, the pattern of score with respect to acetyl CoA and acetyl-INH, was similar to MT-NAT of MTB, but revealed a decrease in INH score. Conclusion: In MTB, MT-NAT revealed high affinity towards acetyl-INH, which can be interpreted as increased formation of acetyl-INH, and therefore, may lead to INH resistance through inactivation of INH. Similarly, in MT-NAT2 (rapid acetylators), acetylation occurs rapidly, serving as a possible risk factor for developing INH resistance in humans.

Glulathione-S-transferases Gene Polymorphism in Prediction of Gastric Cancer Risk by Smoking and Helicobacter Pylori Infection Status

  • Aim: To evaluate the association of glutathione S-transferases gene polymorphisms with the risk of gastric cancer, with reference to smoking and Helicobacter pylori infection. Methods: We conducted a 1:1 matched case-control study with 410 gastric cancer cases and 410 cancer-free controls. Polymorphisms of GSTM1, GSTT1 and GSTP1 were determined using PCR-CTPP. Results: The GSTM1 and GSTT1 null genotypes were significantly associated with the risk of gastric cancer after adjusting for potential confounding factors (OR=1.68, 95% CI=1.32-2.23 for null GSTM1, OR=1.73; 95% CI=1.24-2.13 for null GSTT1). The combination of null GSTM1 and null GSTT1 conferred an elevated risk (OR=2.54, 95% CI=1.55-3.39). However, no association was found for GSTP1 polymorphism The smoking modified the association of GSTM1 and GSTT1 null genotypes with the risk of gastric cancer. Conclusion: GSTM1 and GSTT1 null genotypes are associated with increased risk of gastric cancer, and smoking modifies the association.

Role of Household Exposure, Dietary Habits and Glutathione S-Transferases M1, T1 Polymorphisms in Susceptibility to Lung Cancer among Women in Mizoram India

  • Background: A case-control study was conducted to evaluate the effect of household exposure, dietary habits, smoking and Glutathione S-Transferases M1, T1 polymorphisms on lung cancer among women in Mizoram, India. Materials and Methods: We selected 230 newly diagnosed primary lung cases and 460 controls from women in Mizoram. Multivariate logistic regression analysis was performed to estimate adjusted odds ratio (OR). Results: Exposure of cooking oil fumes (p<0.003), wood as heating source for cooking (p=0.004), kitchen inside living room (p=0.001), improper ventilated house (p=0.003), roasting of soda in kitchen (p=0.001), current smokers of tobacco (p=0.043), intake of smoked fish (p=0.006), smoked meat (p=0.001), Soda (p<0.001) and GSTM1 null genotype (p=0.003) were significantly associated with increased risk of lung cancer among women in Mizoram. Significantly protective effect was observed for intake of bamboo shoots (p=<0.001) and egg (p<0.001). A clear increase in dose response gradient was observed for total cooking dish years. Risk for lung cancer tends to increase with collegial effect of indoor environmental sources (p=0.022). Significant correlation was also observed for interaction of GST polymorphisms with some of dietary habits. Conclusions: We confirmed the important role of exposure of cooking oil emission and wood smoke, intake of smoked meat, smoked fish and soda (an alkali preparation used as food additives in Mizoram) and tobacco consumption for increase risk of lung cancer among Women in Mizoram.

효소를 이용한 아실화 반응의 최근 동향과 전망 (Recent Developments and Prospects in the Enzymatic Acylations)

  • 가수분해 효소(혹은 아실전이효소)를 이용한 알콜과 아민의 아실반응은 에스터의 가수분해 반응(hydrolysis, deacylation)과 더불어 효소를 이용한 유기합성 반응에서 이미 잘 확립된 기술로서, 산업체에서 제약의 합성이나 고분자의 합성에서 널리 응용되고 있다. 이러한 효소를 이용한 아실화 반응은 주로 열역학적인 제한으로 인해 그동안 대부분이 주로 유기용매에서 이루어지고 있다. 최근 들어서, 수용액에서 아실화반응을 전이효소를 이용하여 효율적으로 할 수 있다는 보고와 함께 그 반응 기제에 대한 연구들이, X-ray 구조와 이러한 반응을 가능하게 하는 효소의 단백질 서열 비교 연구, 그리고 계산 화학에 의한 효소의 설계 연구등을 통해 새롭게 밝혀지고 있다. 본 총설에서는 효소를 이용한 아실화반응을 유기용매와 수용액에서의 수행함에 있어서 장단점을 비교해 보면서, 앞으로의 전망도 함께 제시하고자 한다. 특별히 다양한 천연물들의 구조 변화에 아실화 반응 생체촉매를 사용할 수 있는 가능성에 대해 살펴볼 것이다.

Comparative Investigation of Glutathione S-Transferases, Glyoxalase-I and Alliinase Activities in Different Vegetable Crops

  • Glutathione S-transferases(GSTs, EC 2.5.1.18), glyoxalase-I(EC 4.4.1.5) and alliin lyase(alliinase, EC 4.4.1.4) are important enzyme systems in plant bodies. The first two are mainly detoxifying enzymes that utilize glutathione(GSH) in the defense mechanism, and the last one is mainly involved in secondary metabolism and relevant to sulfur compounds derived from GSH. The activities of the three enzymes have been investigated in soluble extracts of vegetable crops, including pumpkin, cabbage, broccoli, radish, carrot, potato, sweet potato, mungbean, and onion. GST activities were detected in all of the vegetables, and the extract of onion bulb exhibited the highest specific activity(648 nmol/min/mgP). The putative GSTs of most of the vegetables were found to be induced by ethanol. The activities of GSTs in onion bulb were found to be markedly inhibited by S-hexyl glutathione and were also inhibited by S-butyl glutathione and S-propyl glutathione. The anti-CmGSTF1 antiserum recognized a thick band for putative onion GST. The estimated glyoxalase-I activity level was also high in onion bulb(4540 nmol/min/mgP), indicating that the thick band detected by Western blot analysis might result from partial recognition of glyoxalase-I by the antiserum. The specific activities for glyoxalase-I were moderate in radish and carrot, and the extracts of other vegetables had rather low levels of activities. The extract of onion also showed the highest specific activity level for alliinase(2069nmol pyruvate/mgP). The extracts of other vegetables also had alliinase activities, although the estimated values were much lower than that of onion.

Quizalofop-Ethyl이 콩과 옥수수의 Glutathione-S-Transferases와 Carboxylesterase의 활성에 미치는 영향 (Effect of Quizalofop-Ethyl on Glutathione-S-Transferases and Carboxylesterase Activity of Soybean and Corn Plants)

  • 콩과 옥수수에서 glutatione-S-transferases와 carboxylesterase의 생화학적 특성과 quizalofopethyl 처리에 따른 식물체내 효소의 활성 변화를 조사한 결과는 다음과 같다. GSTS의 최적 pH는 8.2이고, Km값은 콩 $6.7{\times}10^{-3}M$, 옥수수 $2.5{\times}10^{-3}M$이었으며, Vmax는 콩 50nmole/mg/min, 옥수수 20nmole/mg/min이었다. Carboxylesterase의 최적 pH는 6.2${\sim}$7.0 범위이고, Km값은 콩 $4.2{\times}10^{-4}M$, 옥수수 $2.5{\times}10^{-4}M$이었으며, Vmax는 콩 33 nmole/mg/min, 옥수수 10nmole/mg/min이었다. 콩과 옥수수에 살포된 quizalofop-ethyl은 콩과 옥수수의 GSTs와 carboxy-lesterase의 활성을 감소시켰고, quizalofop-ethyl 80ppm을 처리했을때 콩은 10일이 경과후 회복되었지만, 옥수수는 3일이 경과후 고사하였다.

Proteomic Identification of Differentially Expressed Proteins in Arabidopsis Mutant ntm1-D with Disturbed Cell Division

  • Proteome analysis was performed to identify proteins differentially expressed in an Arabidopsis mutant, ntm1-D. In this mutant the NAC transcription factor NTM1 is constitutively expressed and the resultant phenotypic changes include dwarfism, serrated leaves, and altered floral structures, probably due to reduced cell division. Marked elevation of proteins mediating environmental stress responses, including annexin, vegetative storage proteins, beta-glucosidase homolog 1, and glutathione transferases was observed. Overexpression of annexin was confirmed by RT-PCR and Western blotting. These observations suggest that the reduced growth observed in the ntm1-D mutant is caused by enhancement of its stress responses, possibly resulting in a cost in fitness.

  • Heat shock proteins (HSPs), one of the most highly conserved groups of proteins characterized to date, play crucial roles in protecting cells against environmental stresses, such as heat shock, salinity and oxidative stress. The glutathione S-transferases (GST) have important role in detoxification of oxidative stress, environmental chemicals and environmental stress. GST mRNA expression have been used as biomarkers on environmental stress. The purpose of this study was to investigate the death rate and the gene expression of Hsp70 and GST during air exposure and starvation. Results showed that, the expression of Hsp70 mRNA was significantly changed in the experiment groups, such as air exposure and starvation. GST mRNA expression was significantly increased in the experimental group of starvation. These results suggest that Hsp70 and GST were played roles in biomarker gene on the air exposure and starvation.

  • A gene coding for the GST of cotton (Gh-5) was cloned into Escherichia coli and experssed. The enzyme remained within the cytoplasm of E. coli. An 696 bp open reading frame was in the 988 base pair fragment of the recombinant plasmid pET-30b(+). The deduced protein sequence consists of 232 amino acids and has a molecular mass of 30235.58 Da. The cloned enzyme conjugated reduced glutathione and 1-chloro-2,4-dinitrobenzene (CDNB). Plant GST cDNA was expressed in microbe and produced polypeptide had function as an enzyme.

  • Induction of glutathione S-transferases is associated with cancer chemoprevention. We reported that PD98059, an MKK1 inhibitor, induces glutathione Stransferase A2 (rGSTA2). This report comparatively examines the role of CCAAT/enhancer binding protein (C/EBP) and Nrf-2 in the induction of rGSTA2 by PD98059. PD98059 at the concentrations effective for the inhibition of MKKI increased the rGSTA2 protein and mRNA levels in H4IIE cells. (omitted)
