Identification of Genes Expressed during Conidial Germination of the Pepper Anthracnose Pathogen, Colletotrichum acutatum (고추 탄저병균의 포자 발아 단계 발현 유전자 동정)

  • Genes expressed during conidial germination of the pepper anthracnose fungus Colletotrichum acutatum were identified by sequencing the 5' end of unidirectional cDNA clones prepared from the conidial germination stage. A total of 983 expressed sequence tags (ESTs) corresponding to 464 genes, 197 contigs and 267 singletons, were generated. The deduced protein sequences from half of the 464 genes showed significant matches (e value less than 10-5) to proteins in public databases. The genes with known homologs were assigned to known functional categories. The most abundantly expressed genes belonged to those encoding the elongation factor, histone protein, ATP synthease, 14-3-3 protein, and clock controlled protein. A number of genes encoding proteins such as the GTP-binding protein, MAP kinase, transaldolase, and ABC transporter were detected. These genes are thought to be involved in the development of fungal cells. A putative pathogenicity function could be assigned for the genes of ATP citrate lyase, CAP20 and manganese-superoxide dismutase.

  • Melhylovorus sp. strain SS1 grown on methanol was found to show activities of key enzymes of the linear route, $NAD^+$-linked formaldehyde and formate dehydrogenases, and the cyclic route, hexulose-6-phosphate synthase, glucose-6-phosphate isomerase, glucose-6-phosphate dehydrogenase, and 6-phosphogluconate dehydrogenase, for formaldehyde oxidation. The activities of the cyclic route enzymes were higher than those of the linear route enzymes. The bacterium also exhibited activities of the key enzymes of the ribulose monophosphate and Entner-Doudoroff pathways and transaldolase involved in the formaldehyde assimilation and the enzymes involved in the biosynthesis of extracellular polysaccharide. Cells grown in the presence of 2.3 mM ammonium sulfate were higher in the productivity of extracellular polysaccharide, but lower in the growth yield, than those grown in the presence 7.6 mM ammonium sulfate. The activities of 6-phosphogluconate dehydrogenase, phosphoglucomutase, and UDP-pyrophosphorylase in cells grown under nitrogen-limited condition were higher, but that of 6-phosphogluconate dehydratase/2-keto-3-deoxy-6-phosphogluconate aldolase was lower, than those in cells grown in the presence of sufficient amount of nitrogen source.

Analysis of Differential-expressed Proteins of Acidithiobacillus ferrooxidans Grown under Phosphate Starvation

  • Acidithiobacillus ferrooxidans is one of the most important bacterium used in bioleaching, and can utilize $Fe^{2+}$ or sulphide as energy source. Growth curves for Acidithiobacillus ferrooxidans under phosphate starvation and normal condition have been tested, showing lag, logarithmic, stationary and aging phases as seen in other bacteria. The logarithmic phases were from 10 to 32 hours for Acidithiobacillus ferrooxidans cultivated with normal cultivating condition and from 20 to 60 hrs for Acidithiobacillus ferrooxidans cultivated phosphate starvation. Differences of protein patterns of Acidithiobacillus ferrooxidans growing in case of normal or phosphate starvation were separately investigated after cultivation at $30^{\circ}C$ by the analysis of two-dimensional gel electrophoresis (2-DE), matrix-assisted laser desorption/ionization (MALDI)-Mass spectrometry. There were total 6 protein spots identified, which were Recombination protein recA, RNA helicase, AP2 domain-containing transcription factor, NADH dehydrogenase I chain D, Hyothetical protein PF1669, and Transaldolase STY3758. From the 6 identified protein spots, 3 proteins were found to be decreased in expression at the cultivating condition of phosphate starvation, while another three upregulated.
