Adult stem cell lineage tracing and deep tissue imaging

  • Lineage tracing is a widely used method for understanding cellular dynamics in multicellular organisms during processes such as development, adult tissue maintenance, injury repair and tumorigenesis. Advances in tracing or tracking methods, from light microscopy-based live cell tracking to fluorescent label-tracing with two-photon microscopy, together with emerging tissue clearing strategies and intravital imaging approaches have enabled scientists to decipher adult stem and progenitor cell properties in various tissues and in a wide variety of biological processes. Although technical advances have enabled time-controlled genetic labeling and simultaneous live imaging, a number of obstacles still need to be overcome. In this review, we aim to provide an in-depth description of the traditional use of lineage tracing as well as current strategies and upcoming new methods of labeling and imaging.

Process Design in Coining by Three-Dimensional Backward Tracing Scheme of Rigid-Plastic Finite Element Method (강-소성 유한요소법의 3차원 역추적 기법을 적용한 코이닝 공정설계)

  • The backward tracing scheme of the finite element analysis, which is counted to be unique and useful for process design in metal forming, has been developed and applied successfully in industry to several metal forming processes. Here the backward tracing scheme is implemented for process design of three-dimensional plastic deformation in metal forming, and it is applied to a precision coining process. The contact problem between the die and workpiece has been treated carefully during backward tracing simulation in three-dimensional deformation. The results confirm that the application of the developed program implemented with backward tracing scheme of the rigid plastic finite element leads to a reasonable initial piercing hole configuration. It is concluded that three-dimensional extension of the scheme appears to be successful for industrial applications.

  • This paper presents the evaluation of an angular spectrum-based method used to calculate scattering pattern of a three-dimensional object modelled as a collection composed of vertical sectional two-dimensional images. This is done via comparing a proposed method with two existing methods, i.e., a Fresnel hologram method and a ray-tracing method, in terms of computatioal complexities and reconstructed results. Maathematical derivations for each methods are reviewed and implementing procedures are described in detail, along with the amount of computaions required from the implementation point of view, rather than from the mathematical point of view. We show simulation results in which the Fresnel holoram method dose not exhibit promising results although it requires the least computation. Moreover, it is also shown that the proposed method, even with much less computational requirement than the ray-tracing method, produces good performances asmuch as the ray-tracing method does.

  • Ray-tracing based method for computing projection image calculates the exact amounts of the intersection between voxels and a ray. Among several different implementations of the ray tracing based methods, Siddon's method is the earliest one. Later faster implementation such as Jacobs's method, Zhao's method, were investigated. To our knowledge, Zhao's method is the fastest one among these. We improve the speed of the Zhao's method by predicting the number of the same intersection length between voxel and a ray. In our experiment, the proposed method showed significantly faster computation speed than Zhao's method.

  • A finger-gesture game controller using the single camera is implemented in this paper, which is based on the recognition of the number of fingers and the index finger moving direction. Proposed method uses the CAMShift algorithm to trace the end-point of index finger effectively. The number of finger is recognized by using a double circle tracing method. Then, HSI color mode transformation is performed for the CAMShift algorithm, and YCbCr color model is used in the double circle tracing method. Also, all processing tasks are implemented by using the Intel OpenCV library and C++ language. In order to evaluate the performance of the proposed method, we developed a shooting game simulator and validated the proposed method. The proposed method showed the average recognition ratio of more than 90% for each of the game command-mode.

  • In this study, algorithm for the selecting the optimum monitoring location and leakage detection algorithm based on back tracing calculation method were developed and verified by the experiments in pilot plant of water distribution system. First of all, optimum monitoring locations were selected and pressure changes were measured due to artificial leakage by pressure gauges in pilot plant. Simulations of leakage detection was performed for the verification of back tracing calculation method as a leakage detection method. From the results, it was found that leakage locations and leakage amount were exactly estimated. Various leakage amount from $0.0005m^3/s$ to $0.0018m^3/s$ were reproduced and leakage location was detected by back tracing calculation method. It was verified that back tracing calculation method as a leakage detection method is effective.

  • This paper is willing to propose a method of wall tracing, a moving algorithm between two points, when a Cleaning robot between two points moves. We use the information about obstacles and wall side in calculating different weight vector the each infrared sensors in cleaning robot. Therefore the cleaning robot navigates the wall. In the algorithm of wall tracing, the value of error in angle and distance between starting point and ending point should be zero to navigate the wall safely. The propriety of algorithm of the wall tracing is simulated as this method by using Visual C++. The result simulated proved to the simulation.

  • We proposed a new algorithm to improve the calculation speed of forward ray tracing method which was quite a problem in conventional Monte Carlo algorithm. To verify the accuracy and the effect of improving calculation speed, we directly compared integrating sphere characteristics with conventional ray tracing algorithm under the same condition which was referred in a reference paper. By applying new algorithm to an absolute reflectometer, we calculated the degree of errors which were caused by baffle and port characteristics to find optimal system design condition.

A Study on a Minimizing Method of Baseline Wandering in ECG (심전도 기저선 변동의 최소화방법에 관한 연구)

  • In this paper, we propose a method to minimize the baseline wandering that make hard to detect R wave in ECG. This method uses a different signal between ECG and ascending slope tracing waves to minimize the baseline wandering. When the slope of ECG signal maintains the value or falls, the ascending slope tracing wave fellows ECG signal directly, and this wave holds that value of ECG signal when the slope begins to rises in a certain time(=hold time). After this hold time, this wave traces ECG signal again. To evaluate this minimizing method for baseline wandering, the experiments are carried out with 5 ECG data in the database of MIT/BIH. R waves in the proposed different signal are detected by using descending slope trace waves and compared with the annotation file. The results show that the proposed method Is sure to minimize the baseline wandering in ECG.

  • An electric heat tracing system manages the temperature of pipes used in a plant. We propose a Zigbee-based wireless system so called the TESCON(: TEmperature Sensing and CONtrol) to monitor and control the electric heat tracing system in an integrated way. Simulations have been done to analyze the performance of the TESCON system. The performance of the TESCON system is validated by obtaining similar results via testbed operation. We also suggest an extension method of the TESCON system based on the hierarchical tree topology by adopting techniques such as network partition, channel reuse and frame aggregation.
