Toxocara canis 충란에 의한 놀이터 토양 오염에 관한 연구 (A Study of Contamination of soil in playgrounds with the eggs of Toxocara canis reduced ocular toxocariasis)

  • 광주광역시 소재 어린이 놀이터 토양에서 채취한 80개의 토양시료와 123개의 견변시료를 통해 Toxocara canis 충란에 의한 오염 실태를 조사하였다. 어린이 놀이터에서 수집한 토양시료의 15%에서 Toxocara canis의 충란이 검출되었으며, 공원(25%)과 주택가 공터(17%) 토양의 충란 검출률이 비교적 높게 나타났다. 놀이터 토양의 충란 오염률은 봄 가을이 여름 겨울보다 상대적으로 높았으며 오염률이 높은 공원과 주택가 공터에서 계절적 변동폭이 컸다. 놀이터에서 수집한 총 123개의 견변에 대한 충란 검출 결과 검출률은 31%로 나타났으며 토양시료의 검출률(21%) 보다 높게 나타났다.

Molecular Diagnosis of an Ocular Toxocariasis Patient in Vietnam

  • An ocular Toxocara canis infection is reported for the first time in Vietnam. A 34-year-old man residing in a village of Son La Province, North Vietnam, visited the National Eye Hospital (NEH) in August 2011. He felt a bulge-sticking pain in his left eye and loss of vision occurred over 3 months before visiting the hospital. The eye examination in the hospital showed damage of the left eye, red eye, retinal fibrosis, retinal detachment, inflammation of the eye tissues, retinal granulomas, and a parasitic cyst inside. A larva of Toxocara was collected with the cyst by a medical doctor by surgery. Comparison of 264 nucleotides of internal transcribed spacer 2 (ITS2) of ribosomal DNA was done between our Vietnamese Toxocara canis and other Toxocara geographical isolates, including Chinese T. canis, Japanese T. canis, Sri Lankan T. canis, and Iranian T. canis. The nucleotide homology was 97-99%, when our T. canis was compared with geographical isolates. Identification of a T. canis infection in the eye by a molecular method was performed for the first time in Vietnam.

서울지역 어린이 놀이터 모래에서 개 회충류의 충란 조사 (Prevalence of Toxocara spp eggs in sands of public playgrounds in Seoul)

  • To demonstrate the prevalence of Toxocara spp eggs in public playgrounds in Seoul city, sand samples collected from March to November in 2001 were examined. Of 2,600 sand samples from 650 playgrounds surveyed, 41 sands from 39 places(6%) had Toxocara rims eggs. Sand samples in apartment complex were more contaminated(9.7%) with the eggs than in residential area(2.4%). Toxocara canis eggs in sands were found in large number of in the spring but other seasons were less found.

개회충에 감염된 생쥐에 있어서 시기별 단백분해효소 활성의 변동 (The dynamics of proteinase activity at different stages of toxocariasis in mice)

  • Toxocariasis is a parasitic zoonosis caused by infestation of humans with larvae of Toxocara canis, the common roundworm in dogs. Two syndromes have been identified : visceral larva migrans and ocular toxocariasis. In this study we were characterized proteinase activity in crude extracts from liver, lung, kidney and heart of mice infected with Toxocara canis and the dynamics of their changes in different stages of disease. The optimal pH was 5.5. In liver of mice infected with Toxocara canis, the maximun activity of proteinase was observed in 5 day post infection. In lung, the activity reached its maximun on 5th day in A group (infected with 100 embryonated eggs), and on 5th week in B group (infected with 50 embryonated eggs). In kidney, the maximum activity was shown at 6th week in A group, and in B group was shown at 10th day. In early infection, the activity reached its maximun in heart of mice infected with Toxocara canis. As we could see, the dynamics of the changes of proteinase activity in mice is similar in the case of the disease with other biochemical and immunological indices observed in toxocariasis.

Prevalence of Toxocara canis, Toxascaris leonina and Dirofilaria immitis in dogs in Chuncheon, Korea (2004)

  • The intestines and hearts of dogs were examined for Toxocara canis, Toxascaris leonina, and Dirofilaria immitis, after necropsy between June 26 and September 29, 2004 in Chuncheon, Korea. Of the 662 dogs examined, 6 were infected with T. canis $(0.9\%),\;86\;with\;T.\;leonina\;(13.0\%)$. Fifty dogs were infected with D. immitis among 500 dogs examined $(10.0\%)$. Five were co-infected with T. canis and T. leonina, and three were co-infected with T. leonina and D. immitis. The cumulative positive infection rate for three species was $134/662(20.2\%)$. Considering previously reported seropositive rates of T. canis excretory-secretory antigen, i.e., $5\%$ in the adult population in Korea, the possibility of toxocariasis caused by T. leonina should be reevaluated.

Characterization of a Toxocara canis species-specific excretory-secretory antigen(TcES-57) and development of a double sandwich ELISA for diagnosis of visceral larva migrans

  • This study describes the isolation of a Toxocara canis species-specific excretory-secretory(ES) antigen and the development of an enzyme-linked immunosorbent assay(ELISA) based on this antigen. Analysis of the ES antigens of T. canis, Toxocara vitulorum, Ascaris lumbricoides and Necator americanus larval antigen was performed by SDS-PAGE followed by western blotting. A 57 kDa T. canis-specific antibody fraction(TcES-57) was identified by western blotting and labelling with anti-Toxocara antibodies(from experimental rabbits and human patients) and tracing with anti-human or anti-rabbit peroxidase conjugate. No protein fraction of 57 kDa was detected in ES or larval antigens collected from T. canis, T. vitulorum, A. lumbricoides and N. americanus. Using TcES-57, a specific anti-serum was produced in rabbits and a double sandwich ELISA was developed. This test was validated using known seropositive sera from toxocariasis patients, sera from A. lumbricoides or N. americanus patients, and 50 serum samples from cats. These tests revealed that TcES-57 antigen is specific to T. canis infection and does not cross react with sera of other related infections. Thus, ELISA based on TcES-57 antigen was proven to be an effective tool in the diagnosis of toxocariasis and studies on the role of T. canis in the epidemiology of human toxocariasis.

대전광역시 관내 단독주택지역내 어린이공원에서 채취한 토양중 개회충란 오염실태 (Contamination of Toxocara canis in soil of playground in a detached dwelling area of Daejeon city)

  • In order to survey the prevalence of Toxocara cams in soil of playground in a detached dwelling area, 150 samples were taken from playground 50 unit. The survey was carried out during 3 months from July to September in 2006. The Egg of Toxocara canis were detected in 6 samples (4.0%) from 150 cases of playground. The prevalence of T canis by month was 6.0% in July, 4.0% in August, 2.0% in September, respectively. This study results suggested that the prevalence of T canis were decreased in 2006 compared to 18.5% (30 positive/162 samples) in 2000.

Prevalence of Toxocara canis in Dogs, North West Bank of Palestine

  • This study was performed to investigate the prevalence of Toxocara canis in local districts in North West Bank of Palestine. Fecal samples from 132 dogs were collected in Nablus, Tulkarm, and Jenin cities from September 2008 to April 2009 and examined for T. canis eggs with the floating technique. The overall infection rate of toxocariasis in dogs was 36.4%. The dogs less than 6 months old showed higher infection rates than those older than 12 months (p=0.04). Vigorous interventions are necessary, such as routine coprological examinations of dogs and prescription of anthelmintics to infected dogs.

Ultrastructural Localization of Toxocara canis Larval Antigen Reacted with a Seropositive Human Serum

  • Excretory-secretory products of Toxocara canis larvae have been considered as a major functional antigen in immune responses against toxocariasis. We studied ultrastructural localization of T. canis second-stage larval antigen using a seropositive human serum under immunogold electron microscopy. High-density gold particles were observed in the secretory cells, excretory duct, intestinal epithelium, and cuticle of the larval worm sections. The distribution of the positive reactions in the larval worms suggests that the nature of the antigen is excretory-secretory antigen including waste metabolites and secretory enzymes.

Vicarious excretion of contrast medium to small intestine in a dog with Toxocara canis infection

  • A male mixed-breed dog of unknown age was presented with a history of bloody diarrhea and cachexia. Toxocara canis in vomitus was identified by a parasitologist. Hematology revealed low hematocrit, eosinophilia, and low albumin. Computed tomography (CT) revealed an enlarged pulmonary artery with an irregular wall, micronodules in the lung, and vicarious excretion of contrast medium to small intestine. CT scan was helpful for identifying lung lesions and the central organs of larval migration and also show vicarious excretion of contrast medium to the small intestine in T. canis infection.
