Prevalence, Associated Risk Factors, and Phylogenetic Analysis of Toxocara vitulorum Infection in Yaks on the Qinghai Tibetan Plateau, China

  • Toxocara vitulorum has been rarely reported in yaks at high altitudes and remote areas of Sichuan Province of Tibetan Plateau of China. The current study was designed to investigate the prevalence, associated risk factors, and phylogenetic characteristics of T. vitulorum in yak calves on the Qinghai Tibetan plateau. Fecal samples were collected from 891 yak calves and were examined for the presence of T. vitulorum eggs by the McMaster technique. A multivariable logistic regression model was employed to explore variables potentially associated with exposure to T. vitulorum infection. T. vitulorum specimens were collected from the feces of yaks in Hongyuan of Sichuan Province, China. DNA was extracted from ascaris. After PCR amplification, the sequencing of ND1 gene was carried out and phylogenetic analyses was performed by MEGA 6.0 software. The results showed that 64 (20.1%; 95% CI 15.8-24.9%), 75 (17.2; 13.8-21.1), 29 (40.9; 29.3-53.2), and 5 (7.6; 2.5-16.8) yak calves were detected out to excrete T. vitulorum eggs in yak calve feces in Qinghai, Tibet, Sichuan, and Gansu, respectively. The present study revealed that high infection and mortality by T. vitulorum is wildly spread on the Qinghai Tibetan plateau, China by fecal examination. Geographical origin, ages, and fecal consistencies are the risk factors associated with T. vitulorum prevalence by logistic regression analysis. Molecular detection and phylogenetic analysis of ND1 gene of T. vitulorum indicated that T. vitulorum in the yak calves on the Qinghai Tibetan plateau are homologous to preveiously studies reported.

  • Background: Toxocariasis is a common cause of eosinophilia and eosinophilic lung disease in Korea. We analyzed the characteristics of eosinophilic lung disease in toxocariasis. Method: One hundred and forty one patients with eosinophilia caused by a toxocara larval infection were evaluated from September 1, 2001 through March 30, 2006. The plain chest x-ray, chest CT(computed tomography), and bronchoalveolar larvage(BAL) were examined. A diagnosis of toxocariasis was made by ELISA using that secretory-excretory antigen from the T. canis larvae. Results: Toxocarial eosinophilic lung diseases was diagnosed in 32 out of 141 patients. Ground glass attenuation was the main feature on the CT scans in 23 out of 141 patients (71.9%). Thirteen patients (40.6%) had a random in zonal distribution on CT. Pleural effusion was observed in 9 patients (28.1%). Twenty eight patients (87.5%) complained of respiratory symptoms. Eleven patients (34.4%) had gastrointestinal symptoms and 12 patients (37.5%) had liver infiltration. Conclusions: The most common findings of the chest CT in patients with toxocariasis was a randomly distributed ground grass attenuation. A toxocara infection should be considered in a differential diagnosis of patients who exhibit pulmonary infiltration with eosinophilia in Korea.

Histopathological changes of small intestine of cats infected with helminth (연충감염 고양이 소장의 병리조직학적 변화)

  • Helminth and their ova or protozoan oocysts from fecal samples of cats in Chonju and Daejon were examined from December 1995 to January 1996. Out of the 43 samples, 41(95.35%) were positive and 66(153.49%) were cumulative positive for parasite infection. The identified parasites were summerized as follows; Worms of Toxocara cati(62.79%), Taenia taeniformis(48.88%), Spirometra sp(23.25%) and Diphyllobothrium latum (6.97%), eggs of Clonorchis sinensis(2.32%), Ancylostoma tubaeformae(2.32%) and Capillaria sp(4.65%), oocyst of Isospora (2.32%) sp. In haevy infected intestine, mast cells(38 cells/10 villi) were detected in villi and goblet cells(131 cells/$0.1mm^2$) were severely increased in mucosa. Epithelium of intestine was partially deteched B. The end of villi were necrosed and hyperaemia was widely recognized in lamina propria. T taeniaeformis have not the neck, but have 2 layed hooks(anterior : 19, posterior : 19, total : 38).

A Survey on the Prevalence of Internal Parasitism in Dogs of Chonnam Area (전남지방(全南地方) 축견(畜犬)의 내부기생충(內部寄生蟲) 조사(調査))

  • Observations were made on the infection rate of common internal parasites in dogs in Kwangju area from January to March, 1982. Dog's stools were collected from veterinary hospitals, houses, intensive dog-rearing places and sidewalks. On the basis of egg counting, the following results were obtained; 1. Of 464 fecal samples, 73.5% were appeared as positive. 2. Dogs from veterinary hospitals and intensive dog-rearing places have higher infection rates than others. 3. Of the parasites detected, Toxocara canis was the most commonly encountered (23.7%). 4. Ascarids, flukes and coccidia were more prevalent in pups than in mature dogs, whereas bookworms were detected less often in young dogs than in older ones. 5. There were significant (p<0.01) differences among age categories for all parasites except tapeworms.

  • The frequent finding of soil contamination by dogs' feces are remarkable, and such soil contamination may be public health problem. Of 608 samples, 30(4.9%) were positive for nematode ova. Ten paint 3% of sail samples from parks and 7.9% from footpaths were found to contain nematode eggs. Of the helminth eggs detected, eggs of Toxocara spp. were the most commonly encountered, and mostly from the soil of footpaths. No eggs were recovered from the soil of school yards; the level of soil contamination was low in public play areas. Egg recovery rate of the soil to dogs' feces collected from footpaths was approximately 1:5. And the larvae demonstrated from the soil in the present study were nematode larvae, mainly recovered from parks.

A Case Report of Intravitreal Dexamethasone Implant with Exudative Retinal Detachment for Ocular Toxocariasis Treatment

  • Toxocariasis is one of the most common geohelminth infections in several parts of the world. We describe a rare case of ocular toxocariasis with secondary exudative retinal detachment treated with albendazole and an intravitreal dexamethasone implant. A 13-year-old boy with counting finger vision was diagnosed with retinal vasculitis and exudative retinal detachment in his right eye. Fundoscopic examination revealed retinal hemorrhage, retinal vasculitis, and exudative retinal detachment. Serological test using serum and intraocular aqueous humor were positive for anti-Toxocara specific IgG antibodies. He received repeated doses of intravitreal dexamethasone implants combined with oral albendazole. A sequential follow-up optical coherence tomography revealed that the retina was successfully reattached. His visual acuity subsequently improved to 20/400.

Etiologic Survey on the Acute Diarrhea of Puppies in Seoul (서울지역에서 문제되고 있는 강아지의 급성설사증에 관한 병인학적 연구)

  • In Seoul area, there are so many kennel clubs, veterinary hospital, pet establishment and breeding confers that various problems have occurred. They are crowed pet houses, poor sanitation, stress to puppies, sudden environmental changes to puppies and unvaccination against parvovirus, canine distemper virus, parainfluenza virus, infectious hepatitis caused by Adenovirus type I and Leptospira. Several studies were made to survey the infectious agents involved in acute diarrhea of poppies in Seoul are, such as history taking, physical examination, complete blood count and serum chemistry, histopathological finding, bacterial isolation and identification, and hemagglutination test in feces and hemagglutination inhibition test in serum against parvovirus, respectively. The results obtained are summarized as followed. 1. The percent of PCV (30.5$\pm$5.6) and concentration of Total protein(5.0$\pm$0.8) resulted from statistical analysis are significantly lower than normal values (p<0.05), respectively. In addition, fibrinogen (505$\pm$326) was significantly higher an normal value (p<0.001), Band neutrophil (22.9$\pm$12.7) showed signifiant difference (P<0.01). decreased monocyte (3.2$\pm$2.1) and eosinophil (0.7$\pm$0.8) values appeared statistically significant (p<0.001), lymphocyte (16.7$\pm$9), as well. 2. The concentrations of calcium .(8.0$\pm$2.8), glucose (40.1$\pm$31.4), and albumin (2.0$\pm$0.39) were lower than normal values (p<0.01, p<0.01, p<0.001), respectively. Also Inorganic phosphate (7.1$\pm$2.4), pH (p<7.9f:0.2), and Blood Urea Nitrogen (40.41=37.1) were significantly higher than normal values (p<0.001, p(0.001, p<0.05), respectively. 3. Simple and mixed infections occupied 18% and 82% in the distribution of causes in puppies with acute diarrhea, respectively. 4. As puppies got older, incidence of acute diarrhea caused by Staphylococcus aureus was decreased to 13% and infection of canine distemper virus was increased to 53%, but E coli and canine parvovirus always showed high frequency of outbreak in the body weight ranged from 35g to 7.8Kg. 5. As showed in table 5, infections of E coli and Canine Parvovirus showed high outbreak regardless of the age which is classified into three stages, 35~50 day, 60 day and 75~day to 1 year, canine distemper virus appeared increased, but in case of Staphylococcus aureus, visa versa. 6. In comparison wi methods for the laboratory diagnosis diagnosis parvovirus, Hemagglutination test showed positive reaction in 25% and mean serum antibody titer measured by Hemagglutination inhibition test showed 2779 (n=20). In addition, positive reaction was 90% (18/20). 7. In histopathological studies, enteritic and pneumonic lesions were indicated in 53.7% and 39.5% of samples, respectively. 8. Etiologic diagnosis based on the history taking, clinical signs and histopathological findings in puppies with acute diarrhea and vomiting indicated that 50% of puppies were infected by canine parvovirus and distemper virus. 9. In the parasitological examination made by simple flotation with saturated zinc sulfate tour parasites found were Isospora canis, Toxocara canis, Ancylostoma spp and Toxocara spp. Isospora canis and Toxocara canis were more frequently found among those parasites of diarrhoeic causes in puppies ranged from 35 days to 1 year. Their infestation rates were 15% and 13% respectively.

  • It has become possible for small scale laboratories to interpret large scale genomic DNA, thanks to the reduction of the sequencing cost by the development of next generation sequencing (NGS). De novo assembly is a method which creates a putative original sequence by reconstructing reads without using a reference sequence. There have been various study results on de novo assembly, however, it is still difficult to get the desired results even by using the same assembly procedures and the analysis tools which were suggested in the studies reported. This is mainly because there are no specific guidelines for the assembly procedures or know-hows for the use of such analysis tools. In this study, to resolve these problems, we introduce steps to finding whole genome of an unknown DNA via NGS technology and de novo assembly, while providing the pros and cons of the various analysis tools used in each step. We used 350Mbp of Toxocara canis DNA as an application case for the detailed explanations of each stated step. We also extend our works for prediction of protein-coding genes and their functions from the draft genome sequence by comparing its homology with reference sequences of other nematodes.

Prevalence of Toxocariasis and Its Risk Factors in Patients with Eosinophilia in Korea

  • Eosinophilia occurs commonly in many diseases including allergic diseases and helminthic infections. Toxocariasis has been suggested as one cause of eosinophilia. The present study was undertaken to examine the prevalence of toxocariasis in patients with eosinophilia and to identify the risk factors for toxocariasis. This prospective cohort study recruited a total of 81 patients with eosinophilia (34 males and 47 females) who visited the outpatient clinic at Seoul National University Hospital from January 2017 to February 2018 and agreed to participate in this study. The prevalence of toxocariasis was examined by T. canis-specific ELISA, and the various risk factors for toxocariasis were evaluated by a questionnaire survey. Among 81 patients with eosinophilia, 18 were positive for anti-T. canis antibodies (22.2%); 88.9% were male (16/18) and 11.1% were female (2/18). Multivariate statistical analysis revealed that males (OR 21.876, 95% CI: 1.667-287.144) with a history of consuming the raw meat or livers of animals (OR 5.899, 95% CI: 1.004-34.669) and a heavy alcohol-drinking habit (OR 8.767, 95% CI: 1.018-75.497) were at higher risk of toxocariasis in patients with eosinophilia. Toxocariasis should be considered a potential cause of eosinophilia when the patient has a history of eating the raw meat or livers of animals in Korea. A single course of albendazole is recommended to reduce the migration of Toxocara larvae in serologically positive cases with eosinophilia.

  • A total of 480 live buffaloes and 180 visceral samples from Dhaka, Mymensingh, Bogura and Rajshahi were examined for the presence of parasites of water buffaloes in Bangladesh during September, 1988 to August, 1989. The recorded parasites were eight trematodes, two cestodes, fourteen nematodes, two protozoa and two arthropods. The trematodes were Fasciola gigantica (18.9%-46.4%). Paramphistomes (Gigantocotyl explanatum, Ceylonocotyl scoliocoelium, Cotylophoron cotylophorum and Gastrothylax crumenifer (29.5%-48.3%). Schistosoma indicum (1.6%-31.6%), S. spindale (13.9%-27.7%) and S. nasalis (4.6%-8.3%). The cestodes were Hydatid cyst (24.4%), Cysticercus tenuicollis (11.1%). The nematodes were Strongyloides papillosus (14.8%-21.6%), Capillaria spp. (C. bilobata, C. bovis) (8.5%-20.0%), Setaria digitata (7.2%), Onchocerca armillata (27.2%), Thelazia rhodesii (2.3%), Gongylonema pulchrum (3.9%), Oesophagostomum radiatum (6.6%-41.6%), Hookworms (Agriostomum vryburgi, Bunostomum phlebotomum) (8.1%-17.2%), Trichostrongylus axei (11.2%-21.6%), Mecistocirrus digitatus & Haemonchus contortus (15.2%-25.5%) and Toxocara vitulorum (1.1%-9.8%). The protozoa were Eimeria zuerni (2.3%) and Trypanosoma theileri (0.4%). The arthropods were Haemaphysalis bispinosa (8.1%) and Haematopinus tuberculatus (34.6%).
