Size-dependent toxicity of silver nanoparticles to Glyptotendipes tokunagai

  • Objectives This study aims to evaluate the size-dependent toxicity of spherical silver nanoparticles (Ag NPs) to an endemic benthic organism, Glyptotendipes tokunagai. Methods Ag nanoparticles of three nominal sizes (50, 100, and 150 nm) capped with polyvinyl pyrrolidone (PVP-Ag NPs) were used. Their physicochemical properties, acute toxicity (48 hours), and bioaccumulation were measured using third instar larvae of G. tokunagai. Results The aggregation and dissolution of PVP-Ag NPs increased with exposure time and concentration, respectively, particularly for 50 nm PVP-Ag NPs. However, the dissolved concentration of Ag ions was not significant compared with the median lethal concentration value for $AgNO_3$ (3.51 mg/L). The acute toxicity of PVP-Ag NPs was highest for the smallest particles (50 nm), whereas bioaccumulation was greatest for the largest particles (150 nm). However, larger PVP-Ag NPs were absorbed and excreted rapidly, resulting in shorter stays in G. tokunagai than the smaller ones. Conclusions The size of PVP-Ag NPs significantly affects their acute toxicity to G. tokunagai. In particular, smaller PVP-Ag NPs have a higher solubility and stay longer in the body of G. tokunagai, resulting in higher toxicity than larger PVP-Ag NPs.

감마선 조사에 의한 endosulfan의 면역독성 저감 (Radiation-induced Degradation and Immune Toxicity Reduction of Endosulfan)

  • Endosulfan is an organochlorine pesticide that is widely used throughout the world for higher agricultural production. Its extreme toxicity, however, has caused health and environment concerns that have led to an interest in detoxification. In this study, the radiolytic degradation of endosulfan was investigated. Endosulfan in methanol solution (100 ppm) was irradiated at 0, 10, 30, and 50 kGy, and subsequent changes in immune toxicity and degradation of endosulfan were observed. The concentration of endosulfan that was used in this experiment did not affect the cell proliferation. The irradiation of endosulfan decreased the production of NO, indicating a decrease in the immune toxicity of endosulfan by irradiation. The concentration of endosulfan was significantly reduced by irradiation in a dose-dependent manner. The results suggest that gamma irradiation can degrade endosulfan and can reduce its immune toxicity.

마우스에서 황토국화 추출물의 급성독성 연구 (Acute Toxicity Study on Mud chrysanthemum Indicum in Mice)

  • Objectives : The aim of this study is to collect data for toxicity and safety of Mud chrysanthemum indicum. Methods : In this study, we investigated the acute toxicity for buthanol-extracted Mud schrysanthemum indicum, 25 male and 25 female mice were observed for 14days after one day oral administration of Mud chrysanthemum indicum at the respective doses of 0(control group), 1024, 1280, 1600 and 2000mg/kg. Results : We observed survival rates, general toxicity, change of body weight and autopsy. No abnormality was found for all cases. Conclusions : The data confirmed that Mud chrysanthemum indicum is free from, the toxicity and safety problems. Compared with the control group, we could not find any toxic alteration in all treated groups (1024, 1280, 1600 and 2000mg/kg), In conclusion, LD50 of Mud chrysanthemum indicum was over 2000 mg/kg and it is very safe to mice.

말 태반 가수분해 추출물의 Sprague-Dawley Rat를 이용한 단회 경구투여 독성시험 (Study on a Single Oral Dose Toxicity Test of Horse Placenta Hydrolysate Extract in Sprague-Dawley Rats)

  • Objectives : The objective of this study is to analyze the single-dose toxicity of horse placenta hydrolysate extracts. Methods : Sprague-Dawley rats were chosen for the study. Doses of horse placenta hydrolysate extracts, 2,000 mg/kg, 1,000 mg/kg and 500 mg/kg, were administered to the experimental group, and the same doses of normal saline solution were administered to the control group. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results : In all 4 groups, no deaths occurred, and the horse placenta hydrolysate extracts administered by oral was over 2,000 mg/kg. No significant changes in the weight between the control group and the experimental group were observed. To check for abnormalities in organs and tissues, we used microscopy to examine representative histological sections of each specified organ, the results showed no significant differences in any organs or tissues. Conclusions : The above findings suggest that treatment with horse placenta hydrolysate extracts is relatively safe. Further studies on this subject should be conducted to yield more concrete evidence.

  • Bioluminescence is being used as a prevailing reporter of gene expression in microorganisms and mammalian cells. Bacterial bioluminescence draws special attention from environmental biotechnologists since it has many advantageous characteristics, such as no requirement of extra substractes, highly sensitive, and on-line measurability. Using bacterial bioluminescence as a reporter of toxicity has replaced the classical toxicity monitoring technology of using fish or daphnia with a cutting-edge technology. Fusion of bacterial stress promoters, which control the transcription of stress genes corresponding to heat-shock, DNA-, or oxidative-damaging stress, to the bacterial lux operon has resulted in the development of novel toxicity biosensors with a short measurement time, enhanced sensitivity, and ease and convenient usage. Therefore, these recombinant bioluminescent bacteria are expected to induce bacterial bioluminescence when the cells are exposed to stressful conditions, including toxic chemicals. We have used these recombinant bioluminescent bacteria in order to develop toxicity biosensors in a continuous, portable, or in-situ measurement from for air, water, and soil environments. All the data obtained from these toxicity biosensors for these environments were found to be repeatable and reproducible, and the minimum detection level of toxicity was found to be ppb (part per billion) levels for specific chemicals.

혈관질환 억제 효능이 있는 BDR-29의 백서 신장 독성연구 (Study on Kidney Toxicity of BDR-29 for Treatment Vascular Diseases in Rats)

  • The kidney toxicities of BDR-29 used for improvement of the vascular diseases, was examined using male and female Sprague-Dawley rats. The male and female rats were divided into 4 groups for intragastrical treatment with doses of 0, 5, 50, and 500 mg/kg/day for 13 weeks, respectively. In all male and female rats treated with BDR-29, no mortality and gross pathological findings were shown for 13 weeks. There substantially was no change in body weight in all rats with treatment of BDR-29. The renal functional parameters including urinary volume, urine osmolality, electrolytes excretory rate, creatinine clearance, and solute-free water reabsorption were not exchanged in all rats treated with BDR-29. Taken together, these results suggest that BDR-29 has no toxicity on kidney in all male and female rats.

발효소시호탕의 마우스에 대한 단회투여 경구독성시험 (Single Dose Oral Toxicity Study of Fermented Soshiho-tang Extract in Mice)

  • The aim of this study was to investigate the acute toxicity and safety of fermented Soshiho-tang extract using male and female ICR mice. Mice were treated with fermented Soshiho-tang extract once orally at 1250, 2500 or 5000 mg/kg and observed for two weeks. At the doses used, no mortality or abnormal clinical signs in animals were shown during at the observation period. In addition, no differences were found between control and treated groups in body weight, hematology and biochemical analysis, and other findings. Above data strongly suggest that no observed adverse effect level of fermented Soshiho-tang extract might be over 5000 mg/kg/day in this study.

  • Deficiencies in the early ADMET(absorption, distribution, metabolism, elimination and toxicity) information on drug candidate extract a significant economic penalty on pharmaceutical firms. Microscale cell culture analogue-microscale gastrointestinal(${\mu}CCA-{\mu}GI$) device using Caco 2, L2 and HEp G2/C3A cells, which mimic metabolic process after absorption occurring in humans was used to investigate the toxicity of the model chemical, acetaminophen(AAP). The toxicity of acetaminophen determined after induction of CYP 1A1/2 in Caco 2 cells was not significant. In a coculture system, although no significant reduction in viability of HEp G2/C3A and L2 cells was found, approximately 5 fold increase in the CYP 1A1/2 activity was observed. These results appear to be related to organ-organ interaction. The oral administration of a drug requires addition of the absorption process through small intestine to the current ${\mu}CCA$ device. Therefore, a perfusion coculture system was employed for the evaluation of the absolution across the small intestine and resulting toxicity in the liver and lung. This system give comprehensive and physiologic information on oral uptake and resulting toxicity as in the body. The current ${\mu}CCA$ device can be used to demonstrate the toxic effect due to organ to organ interaction after oral administration,

산화동물모델 흰 쥐 간세포에 대한 자근(紫根)의 황산화 효과 연구 (Study on Antioxidant Effect of Lithospermi Radix on Liver Cells Isolated from Oxidatively Stressed Rat)

  • Objective : This study purposed to investigate the anti-oxidative effect of Lithospermi Radix (root of Lithospermum erythrorhizon S.) on liver cells isolated from oxidatvely stressed rat by AAPH. Method : We investigate effects of Lithospermi Radix(LR) and its fractions on normal liver cells' proliferation. And the amounts of SOD, GSH, catalase, NO, MDA production by liver cells isolated from the oxidatively stressed rat by AAPH also were measured after incubation with various fractions of LR extraction. Results : LR and its fracitons showed no toxicity on the normal liver cells from rat. LR and its fracitons increased the activity of SOD and reduced the amounts of NO and MDA in the liver cells from the oxidatively stressed rat. Conclusion : Lithospermi Radix could be supposed to have antioxidant effect on liver cells with no toxicity.

Single-dose Intramuscular Toxicity of Neutral Natured Blood Stasis Pharmacopuncture in Sprague-Dawley Rats

  • Objectives: This study was performed to analyze the single-dose toxicity of neutral natured blood stasis pharmacopuncture extracts. Methods: All experiments were conducted at Biotoxtech, an institution authorized to perform non-clinical studies, under the regulations of Good Laboratory Practice (GLP). Sprague-Dawley rats were chosen for the pilot study. Doses of neutral natured blood stasis pharmacopuncture extracts, 0.1, 0.5 and 1.0 mL, were administered to the experimental group, and the same doses of normal saline solution were administered to the control group. This study was conducted under the approval of the Institutional Animal Ethics Committee. Results: In all 4 groups, no deaths occurred, and the neutral natured blood stasis pharmacopuncture extracts administered by intramuscular (IM) injection was over 1.0 mL/animal. No significant changes in the body weights between the control group and the experimental group were observed. To check for abnormalities in organs and tissues, we used microscopy to examine representative histological sections of each specified organ; the results showed no significant differences in any organs or tissues. Conclusion: The above findings suggest that treatment with neutral natured blood stasis pharmacopuncture extracts is relatively safe. Further studies on this subject should be conducted to yield more concrete evidence.
