Solvent effect on enzymatic steroid transformation

  • As part of our endeavor to increase the productivity of steroid by enzymatic transformation of corticosteroids, attempts have been made to increase the solubility of steroids by using some organic solvents. When the solubility of steroids is the rate limiting factor in the steroid transformation, it was found that the use of solvents significantly improved the yield. Hydrocorisone as a substrate and 3-ketosteroid .DELTA.$^{1}$ dehydrogense as an immobilized whole cell enzyme were employed as the model system for this study. It was found that the yield of product, prodnisolone, goes through a maximum with an increase in the solvent concentration. At a high solvent concentration, the solvent showed a toxic effect and it causes a decrease in the product yield by the second order inhibition mechanism. Among the solvents evaluated, methanol and ethanol were found to be the best. These alcohols are not only good solvents but also showed minimal toxic effect. Based on the experimental results, it was concluded that the productivity of steroid can be increased by usign well selcted solvents systems for the enzymatic transformation of steroids.

  • It has been used so many kinds of architectural materials and interior products in current building construction, and use of composite architectural materials is increasing with the development of chemical technology. As the green architecture has become the center of public interest, much effort is conducted in advanced countries on the LCA point of view, such as restriction of architectural materials that emitting pollution substances, developing of Non-Toxic architectural materials, and recycling of used materials, etc. with the cooperation of related organizations, material manufacture companies, and construction companies. Because the kinds of materials to be used in building constructions are so various, there might be some possibility of personal and subjective choice at the time of materials selection resulting the missing the requirements of building components and the choice of harmful materials to human. One way to resolve the material problem is to present the green architectural materials which coincide with the quality performance at service and not harmful to man and nature. At this point of view, this study aims to develop the material classification model by investigating the major labelling system about green architectural materials in both domestic and abroad and to implement an efficient material selection system by making a powerful database of environmental standard and quality basis of building requirements.

냉동, 냉장 시스템에서 NH3 누출 사고 시 장외영향평가를 위한 피해범위 및 대피거리 산정에 관한 연구 (Recommended Evacuation Distance for Offsite Risk Assessment of Ammonia Release Scenarios)

  • An accident of an ammonia tank pipeline at a storage plant resulted in one death and three injuries in 2014. Many accidents including toxic gas releases and explosions occur in the freezing and refrigerating systems using ammonia. Especially, the consequence can be substantial due to that the large amount of ammonia is usually being used in the refrigeration systems. In this study, offsite consequence analysis has been investigated when ammonia leaks outdoors from large storages. Both flammable and toxic effects are under consideration to calculate the affected area using simulation programs for consequence analysis. ERPG-2 concentration (150 ppm) has been selected to calculate the evacuation distance out of various release scenarios for their dispersions in day or night. For offsite residential, the impact area by flammability is much smaller than that by toxicity. The methodology consists of two steps as followings; 1. Calculation for discharge rates of accidental release scenarios. 2. Dispersion simulation using the discharge rate for different conditions. This proactive prediction for accidental releases of ammonia would help emergency teams act as quick as they can.

Adsorptive and kinetic studies of toxic metal ions from contaminated water by functionalized silica

  • The objective of the study, to develop adsorbent based purifier for removal of radiological and nuclear contaminants from contaminated water. In this regard, 3-aminopropyl silica functionalized with ethylenediamine tetraacetic acid (APS-EDTA) adsorbent prepared and characterized by Fourier Transform Infrared Spectroscopy (FT-IR), Scanning Electron Microscopy (SEM) and X-ray Diffraction (XRD). Prepared APS-EDTA used for adsorptive studies of Cs(I), Co(II), Sr(II), Ni(II) and Cd(II) from contaminated water. The effect on adsorption of various parameters viz. contact time, initial concentration of metal ions and pH were also analyzed. The batch method has been employed using metal ions in solution from 1000-10000 ㎍/L, contact time 5-60 min., pH 4-10 and material quantities 50-200 mg at room temperature. The obtained adsorption data were used for drawing Freundlich and Langmuir isotherms model and both models were found suitable for explaining the metal ions adsorption on APS-EDTA. The adsorption data were followed pseudo second order reaction kinetics. The maximum adsorption capacity obtained 1.3037-1.4974 mg/g for above said metal ions. The results show that APS-EDTA have great potential to remove Cd(II), Co(II), Cs(I), Ni(II) and Sr(II) from aqueous solutions through chemisorption and physio-sorption.

도시유역의 부유고형물 유출평가를 위한 쓸림모형 개발 (A Development of Washoff Model for Suspended Solids in Urban Areas)

  • Suspended Solid (SS) is one of the main pollutants and discharges with attached other pollutants such as heavy metal and toxic substance. It is very important to estimate and forecast the release characteristics of SS for water quality improvement. The current studies assumed that SS release rate is proportional to the rain intensity and suggested exponential washoff models. These models related to the shear force of flow. In this study, a new washoff model is suggested based on relation with SS release rate and mean flow rate of the basin surface which is closely related to the shear force. The proposed model is applied to the Goonja drainage district in Seoul, Korea. The new washoff model simulates the SS discharge more accurately in the various rainfall types. The model can be widely applied to the real problems such as the management of non-point source pollutant and the design of treatment facilities.

Developmental toxicity and anti-inflammatory effect of the soft coral Dendronephthya gigantea collected from Jeju Island in zebrafish model

  • Recent in vitro studies have demonstrated that extract of soft coral Dendronephthya gigantea (SCDE) had strong anti-inflammatory activities. However, the direct effects of SCDE on anti-inflammatory activities in vivo model remained to be determined. Therefore, the present study was designed to assess in vivo anti-inflammatory effect of SCDE using lipopolysaccharide (LPS)-stimulated zebrafish model. We also investigated whether SCDE has toxic effects in zebrafish model. The survival, heart beat rate, and developmental abnormalities were no significant change in the zebrafish embryos exposed to at a concentration below $100{\mu}g/ml$ of SCDE. However, lethal toxicity was caused after exposure to 200 and $400{\mu}g/ml$ of SCDE. Treating zebrafish model with LPS treatment significantly increased the reactive oxygen species (ROS) and nitric oxide (NO) generation. However, SCDE inhibited this LPS-stimulated ROS and NO generation in a dose-dependent manner. These results show that SCDE alleviated inflammation by inhibiting the ROS and NO generation induced by LPS treatment. In addition, SCDE has a protective effect against the cell damage induced by LPS exposure in zebrafish embryos. This outcome could explain the profound anti-inflammatory effect of SCDE both in vitro as well as in vivo, suggesting that the SCDE might be a strong anti-inflammatory agent.

가압 염소포화액체 저장탱크의 2상 흐름 누출에 대한 유해위험거리의 예측을 위한 결과영향 모델링 방법론 (Consequence Modeling Methodology for Prediction of Hazard Distance for Two-phase Flow Release from the Pressurized Chlorine Saturated Liquid Storage Tank)

  • 본 연구는 화학장치설비중 가압 염소포화액체 저장탱크의 2상흐름 연속누출에 대한 유해위험거리(또는 독성완충거리)를 정량적으로 예측하기 위한 결과영향 모델링 방법론을 개발하기 위한 것이다. 누출원 모델링은 미환경청의 가이드라인에 근거한 정교한 해석방법과 SuperChems 모델의 자체계산에 의하여 각각 수행되었다. 유해위험성 평가에서 법적 독성규제농도로서 사용되는 STEL, IDLH 및 ERPGs (ERPG-2와 ERPG-3) 농도들에 대하여 유해위험거리를 예측하였다. 비상대응계획 수립시 유해위험성 평가의 가이드라인으로 활용하기 위하여 특히 ERPG-2 농도에 대하여 누출원특성 및 기상변화들의 유해위험거리에 미치는 영향을 고찰하였다.

  • 무거운 독성가스의 소규모 누출에 대한 가우시안 연속모델의 적용성을 평가하기 위하여 염소가스의 소규모 누출에서 염소농도의 실험값과 여러 가지 분산계수를 사용한 가우시안 연속모델에 의해 산출한 이론값을 비교하였다. 그 결과, 염소가스 분산은 분산계수와 대기안정도에 따라 상당히 차이가 있었으며, 염소농도는 Briggs의 분산계수와 유효 누출높이를 사용한 가우시안 연속모델에 의해 비교적 정확하게 산출할 수 있었다.

Toxicity Evaluation of Complex Metal Mixtures Using Reduced Metal Concentrations: Application to Iron Oxidation by Acidithiobacillus ferrooxidans

  • In this study, we investigated the inhibition effects of single and mixed heavy metal ions ($Zn^{2+},\;Ni^{2+},\;Cu^{2+},\;and\;Cd^{2+}$) on iron oxidation by Acidithiobacillus ferrooxidans. Effects of metals on the iron oxidation activity of A. ferrooxidans are categorized into four types of patterns according to its oxidation behavior. The results indicated that the inhibition effects of the metals on the iron oxidation activity were noncompetitive inhibitions. We proposed a reduced inhibition model, along with the reduced inhibition constant ($\alpha_i$), which was derived from the inhibition constant ($K_I$) of individual metals and represented the tolerance of a given inhibitor relative to that of a reference inhibitor. This model was used to evaluate the toxicity effect (inhibition effect) of metals on the iron oxidation activity of A. ferrooxidans. The model revealed that the iron oxidation behavior of the metals, regardless of metal systems (single, binary, ternary, or quaternary), is closely matched to that of any reference inhibitor at the same reduced inhibition concentration, $[I]_{reduced}$, which defines the ratio of the inhibitor concentration to the reduced inhibition constant. The model demonstrated that single metal systems and mixed metal systems with the same reduced inhibitor concentrations have similar toxic effects on microbial activity.

α-Mangostin and Apigenin Induced Cell Cycle Arrest and Programmed Cell Death in SKOV-3 Ovarian Cancer Cells

  • Ovarian cancer is the fifth main cause of pre-senescent death in women. Although chemotherapy is generally an efficient treatment, its side effects and the occurrence of chemotherapeutic resistance have prompted the need for alternative treatments. In this study, ${\alpha}$-mangostin and apigenin were evaluated as possible anticancer alternatives to the chemotherapeutic drug doxorubicin, used herein as a positive control. The ovarian adenocarcinoma cell line SKOV-3 (ATCC No. HTB77) was used as model ovarian cancer cells, whereas the skin fibroblast line CCD-986Sk (ATCC No. CRL-1947) and lung fibroblast line WI-38 (ATCC No. CCL-75) were used as model untransformed cells. Apigenin and doxorubicin inhibited the growth of SKOV-3 cells in a dose- and time-dependent manner. After 72 hr exposure, doxorubicin was mostly toxic to SKOV-3 cells, whereas apigenin was toxic to SKOV-3 cells but not CCD-986Sk and WI-38 cells. ${\alpha}$-Mangostin was more toxic to SKOV-3 cells than to CCD-986Sk cells. A lower cell density, cell shrinkage, and more unattached (floating round) cells were observed in all treated SKOV-3 cells, but the greatest effects were observed with ${\alpha}$-mangostin. With regard to programmed cell death, apigenin caused early apoptosis within 24 hr, whereas ${\alpha}$-mangostin and doxorubicin caused late apoptosis and necrosis after 72 hr of exposure. Caspase-3 activity was significantly increased in ${\alpha}$-mangostin-treated SKOV-3 cells after 12 hr of exposure, whereas only caspase-9 activity was significantly increased in apigenin-treated SKOV-3 cells at 24 hr. Both ${\alpha}$-mangostin and apigenin arrested the cell cycle at the $G_2/M$ phase, but after 24 and 48 hr, respectively. Significant upregulation of BCL2 (apoptosis-associated gene) and COX2 (inflammation-associated gene) transcripts was observed in apigenin- and ${\alpha}$-mangostin-treated SKOV-3 cells, respectively. ${\alpha}$-Mangostin and apigenin are therefore alternative options for SKOV-3 cell inhibition, with apigenin causing rapid early apoptosis related to the intrinsic apoptotic pathway, and ${\alpha}$-mangostin likely being involved with inflammation.
