Stachys riederi var. japonica Extract Reduces Cytochrome C Release from Mitochondria in UVA-irradiated Human Dermal Fibroblasts

  • This study was performed to investigate the cytoprotective effects of Stachys riederi var. japonica ethanol extract (SREE) to control oxidative stress induced by UVA-irradiation by examining antioxidant capacity and gene expression of cytochrome c using human dermal fibroblasts. The total polyphenolics and flavonoids in the SREE were 41.2 and 25.4 mg/g, respectively. At concentrations of 500 and $1000{\mu}g/mL$, the electron-donating ability of SREE was 48.6% and 82.0%, respectively, and the 2,2'-azino-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity was 62.3% and 78.8%, respectively. These findings showed that SREE has a fairly good antioxidant capacity. As determined by an MTT assay, the maximum permissible level for treating SREE to human dermal fibroblasts was shown to be over $200{\mu}g/mL$. SREE ($200{\mu}g/mL$) significantly decreased cytochrome c mRNA and protein expression by 31.1% (p<0.001) and 38.8% (p<0.01), respectively. These findings suggest that SREE may protect human skin cells against mitochondrial-dependent apoptosis. Therefore, SREE seems to be a natural antioxidant to protect cells against oxidative stress induced by UVA-irradiation.

추출 조건을 달리한 감잎과 녹차의 물 및 에탄올 추출물이 노령쥐의 지방대사와 항산화능에 미치는 영향 (Effect of Water and Ethanol Extracts of Persimmon Leaf and Green Tea Different Conditions on Lipid Metabolism and Antioxidative Capacity in 12-month-old Rats)

  • This study was performed to investigate effects of dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea on lipid metabolism, lipid peroxidation and antioxidative enzyme activity in 12-month-old rats. Fifty-four male Sprague-Dawley rats weighing 542$\pm$4.5g were blocked into groups according to their body weight and were raised for four weeks with the diets containing 5%(w/w) dried leaf powders of persimmon(Diospyros kaki Thunb) and green tea(Camellia Sinensis O. Ktze), water or 75% and 95% ethanol extracts from same amount of each dried tea powder. Food intake was not significantly different among all groups, but weight gain of green tea powder group was significantly lower than that of control group. Plasma and liver lipid levels of all the tea diet groups were lower than those of control group. Especially, 75% ethanol extract of persimmon leaf decreased total lipid and triglyceride concentrations in plasma and 95% ethanol extract of persimmon leaf decreased liver total lipid level. However, there was no difference between 75% ethanol extracts groups and 95% ethanol extracts groups in lipid metabolism. Superoxide dismutase(SOD) and catalase activities in erythrocyte were remarkably increased by all the green tea diets. SOD, catalase and glutathione peroxidase activities in liver were increased by the feeding of ethanol extracts from green tea and persimmon leaf powder. Liver xanthine oxidase activity was not different among all groups. Plasma Thiobarbirutic acid reactive substance(TBARS) concentrations of all the green tea diet groups were significantly low. It was thought that high flavonoids in green tea inhibited plasma lipid peroxidation by promoting SOD, catalase activities in erythrocyte. 95% ethanol extract of persimmon leaf also inhibited plasma lipid peroxidation by high vitamin E and beta-carotene. Persimmon leaf powder decreased liver TBARS concentration by vitamin E, betacarotene and vitamin C and by increasing activities of antioxidative enzymes with flavonoids. In conclusion, dried leaf powders, water, 75% and 95% ethanol extracts of persimmon leaf and green tea were effective in lowering lipid levels and inhibiting lipid peroxidation in 12-month-old rats. Above all, ethanol extracts of persimmon leaf decreased plasma and liver lipid levels and persimmon leaf powder effectively inhibited liver lipid peroxidation. Extracts of green tea leaf inhibited plasma lipid peroxidation. In lowering lipid levels and inhibiting lipid peroxidation, ethanol extracts were more effective than water extracts, but there was no difference between 75% ethanol extracts and 95% ethanol extracts in lipid metabolism. 장미 에탄올 추출물의 항산화성 및 항균성 (The Antioxidative and Antimicrobial Ability of Ethanol Extracts from Rosa hybrida)

  • 본 연구는 현재 버려지고 있는 장미(Rosa hybrida L. cv. Mary Devor)를 이용하여 기능성을 조사한 뒤 그 이용 가능성에 대해 탐색하고자 하였다. 이를 위하여 장미의 일반성분, 식이섬유 및 총 flavonoids 함량을 분석하고 총산화 효과와 추출수율을 측정하였다. 또한 에탄올의 농도(0, 75, 85, 95%)를 달리하여 추출하고 추출물의 첨가농도(0.02, 0.05%)와 저장기간에 따라 과산화물가, 산가, TBA가를 측정하였다. 과산화물가를 측정하여 산화유도기간을 살펴본 결과, 장미 추출물 첨가군이 대조군이나 ${\alpha}$-tocopherol, BHT 첨가군보다 유도기간이 길게 나타나 항산화 효과가 높은 것으로 나타났다. 특히 85% 에탄올 추출물을 0.05%의 농도로 첨가했을 때 가장 긴 유도기간을 나타냈다. 산가 및 TBA가는 장미 추출물 및 BHT 첨가군이 대조군 및 ${\alpha}$-tocopherol 첨가군보다 저장기간이 길어짐에 따라 낮게 나타났다. 즉, 장미 추출물은 BHT와 유사한 높은 항산화 효과를 나타내어 천연항산화제로서의 이용 가능성을 보여주었다. 시험균주(Escherichia coli, Salmonella typhimurium, Staphylococcus aureus)에 대해 장미 추출물은 대체로 높은 항균활성을 나타내었으며, 특히 85% 에탄올 추출물은 E. coli에 대해 유의적으로 높은 항균효과를 나타내었다. 이상의 결과로 볼 때 장미 추출물을 천연항산화제 및 항균제로 사용할 수 있으며 버려지는 장미를 이용한다는 면에서 경제적 효율성도 기대할 수 있다. 장미추출물의 항산화성과 항균성이 확인되어 그 이용성이 기대됨에 따라 추출물의 유효성분을 확인하는 연구가 필요하다.

재배인삼의 연령별 생리활성 차이 연구 (Differential Physiological Activity in Different Ages of Panax ginseng)

  • 인삼을 지하부는 물론 지상부 경엽 부위를 채소용 또는 식품첨가물로 이용하기 위해 낮은 연령 인삼의 지상부 및 지하부의 메탄올 추출물을 이용한 생리활성물질 함량, 항산화성 및 세포독성을 분석하였다. 연령별 인삼의 초장, 근장, 근직경, 지상부 및 지하부 생체중을 기준으로 생육의 차이가 뚜렷하였다. Folin-Denis 방법에 따라 인삼의 메탄올 추출물로부터 총페놀함량을 측정한 결과, 지상부(22.0 - 76.3 mg $kg^{-1}$)가 지하부(19.0 - 28.3 mg $kg^{-1}$)보다 높은 함량을 보였다. 특히, 지상부는 연령이 낮을수록 지하부는 연령이 높을수록 높은 함량을 보였다. 한편, 연령별 인삼의 총플라보노이드 함량은 역시 지상부(23.3 - 138.6 mg $kg^{-1}$)가 지하부(0 - 10.8 mg $kg^{-1}$) 보다 유의적으로 높은 함량을 보였다. 특히 지상부는 연령이 낮을수록 높은 함량을 보였고 지하부는 뚜렷한 경향을 보이지 않았다. 인삼연령별 항산화성을 분석한 결과 인삼의 지상부는 낮은 연령에서, 지하부는 높은 연령에서 높은 DPPH 라디컬 소거능을 보였다. 특히, 지상부의 경우 3년생이 추출물 2,500 mg $kg^{-1}$에서 82.8%로 가장 높은 활성을 보였고 그 다음이 l년생 75.3%, 2년생 68.6% 순으로 높은 활성을 보였으나 나머지 4, 5, 6년생은 40% 이하의 낮은 활성을 보였다. 한편, 지하부(14.8-39.4%)는 지상부 보다 낮은 활성을 보였고, 5년생, 6년생, 4년생, 3년생, 2년생, 1년생 순으로 높은 활성을 보였다. 인삼연령별 세포독성은 연령과 무관한 것으로 나타났으며 폐암세포주(Calu-6)에 대한 5년근의 세포생장 억제율은 52.8%로 가장 높았고, 대장암 세포주(HCT-116)에 대한 억제율은 3년생에서 79.1%로 가장 높게 나타났다. 연령별 생체중은 생리활성물질 함량 및 항산화성에 관련이 있으며, 특히 항산화성을 나타내는 DPPH 라디컬 소거능($r^2=0.7366-0.7870$)과 아질산염 제거능($r^2=0.5604-0.8794$)은 총페놀함량과 각각 높은 상관관계를 보여 직접적으로 관련이 있는 것으로 나타났다.

원적외선 처리가 포도씨의 기능성분에 미치는 영향 (The Effect of Far-Infrared Irradiation on Functional Components of Grape Seed)

  • 원적외선 조사가 포도씨 기능성분의 변화에 미치는 영향을 알아보고자 가열출력(0, 900, 1800 W) 및 가열시간(0, 20, 40 min)을 달리하여 원적외선 조사 후 마이크로파를 이용하여 포도씨 추출물을 제조하고 기능성분으로서 가용성 고형분, 기계적 색도, 카테킨, 총 페놀, 총 플라보노이드 함량 및 전자공여능과 아질산염 소거능을 각각 측정하였다. 가용성 고형분 함량 및 백색도 및 적색도는 유의적인 차이가 없는 것으로 나타났다. Cathechin류는 가열시간에 따라 catechin, procyanidin $B_2$, epicatechin 함량의 변화가 있음을 확인하였으며, 총 catechin 함량은 총 카테킨 함량은 900 W 및 20분 처리 시 가장 높게 나타났다. 총 페놀성화합물 및 플라보노이드 함량 또한 가열출력 900W 및 가열시간 20분 일 때 가장 높게 나타났다. 그에 따라 전자공여능도 가열출력 900 W 및 가열시간 20분 일 때 가장 높은 것으로 나타났다. 이상의 결과는 원적외선 처리에 의해 총 페놀성 화합물 함량, 총 플라보노이드 함량이 증가됨을 볼 수 있었으며, 특히 cathechin류의 변화가 있음을 확인할 수 있었다. 또한 이로 인해 항산화능도 증가됨을 알 수 있었다. 또한 포도씨의 원적외선 가열처리 최적조건을 설정하고자 가열출력 및 가열시간을 달리하여 중심합성실험계획으로 반응표면분석한 결과 가열출력 621.82~818.18 W, 가열시간 16.3~19.83 min의 범위가 최적 가열조건범위로 설정되었다.

고수 잎 분말과 브로콜리 줄기 분말을 혼합한 파운드 케이크의 항산화 활성과 품질 특성 (Antioxidant Activity and Characteristics of Pound Cakes prepared with Coriandrum satovim L. Leaves Powder and Brocolli's Stem Powder)

  • A study was undertaken to examine the effect of the addition of coriander leaves(Coriandrum satovim L.) and brocolli stem powder addition on the quality attributes of pound cakes. Coriander leaves & brocolli stem powder was added to the batter, at a ratio of 10, 12.5 and 15%, respectively. The antioxidant activity was estimated by a DPPH free radical scavening activity, the total phenolic compounds content and flavonoid content in coriander leaves & brocolli stem powder and pounds cakes. Antioxidative activity had correlated highly with the total flavonoids contents of coriander leaves & brocolli stem powder of pound cakes, respectively(r=0.9096). The quality characteristics of coriander leaves & brocolli stem powder pound cakes were estimated in terms of the specific load volume, hardness, hunter value of crumb and crust, and the sensory quality of the cakes. The specific volume decreased significantly, with increasingly substituted level of coriander leaves & brocolli stem powder(p<0.01). The lightness was significantly decreased with increases in the coriander leaves & brocolli stem powder of pound cake crust and crumb(p<0.01). The hardness, chewiness and gumminess tended to reduce, and the cohesiveness decrease in both of powder. The consumer acceptability score for 10% coriander leaves & brocolli stem powder of pound cakes ranked significantly(p<0.01) higher than those of the other groups in taste, flavor and color. These results showed that coriander leaves & brocolli stem powder are good ingredients to increase the consumer acceptability and the health.

알코올을 처리한 HepG2 세포에서 참죽나무 잎 추출물의 세포 보호 및 항산화 효과 (Effects of the Cedrela sinensis A. Juss. Leaves on the Alcohol-Induced Oxidative Stress in the Human Hepatic HepG2 Cells)

  • All the parts of the Cedrela sinensis A. Juss., including the seeds, roots, and leaves, have been known to exert medicinal effects. The C. sinensis and its major compound, quercetin, were previously reported to exhibit the anti-inflammatory and anti-oxidative activities. However, the hepatoprotective effects of the C. sinensis leaves against the alcohol-induced oxidative stress in the HepG2 cells have not been studied. In this study, we investigated the antioxidant activities and analyzed the flavonoid contents of the C. sinensis-leaf extract (CE). The total flavonoid contents of the CE is 1,874.5 mg/100 g dry weight (DW), while the total quercetin 3-O-rhamnoside (quercitrin) contents, which was identified as the major flavonol in the CE, is 1,456.0 mg/100 g DW. In the ethanol-stimulated HepG2 cells, the CE effectively prevented the cytotoxic effect and increased the gene expression of the antioxidant enzymes, such as the heme oxygenase-1 (HO-1) and the glutathion peroxide (GPx). The level of the reactive oxygen species (ROS) production was significantly decreased in the CE-treated HepG2 cells. In conclusion, the C. sinensis extract suppressed the alcohol-induced oxidative stress in the HepG2 cells via the induced GPx and HO-1 gene expressions. It is expected the CE positive effects will likely be attributed to the flavonoids, like the quercetin, within the CE.

Tabebuia avellanedae 추출물의 운동능력 향상 효과 (Stimulatory Effects of Extracts of Inner Bark from Tabebuia avellanedae on Exercise Endurance Capacity)

  • 능소화과인 Tabebuia avellanedae인 타히보는 중앙아메리카와 남아메리카의 아열대 우림지역에서 서식하며 이들은 민간 약제로 사용되어 왔다. 본 연구는 타히보 추출물의 라디칼 소거활성에 기초한 항산화 활성과 운동능력 향상 효과에 대한 활성을 검토하였다. 타히보로부터 얻은 두 추출물 타히보 열수 추출물(TAW)과 타히보 80% 에탄올 추출물(TAE)의 운동능력 향상 효과와 총 폴리페놀과 플라보노이드 함량과 항산화 활성을 평가하였다. 운동능력 향상 효과는 TAW가 대조군 대비 2.0배, TAE는 1.8배 증가하였다. 두 추출물의 항산화 활성은 라디칼 소거활성(DPPH와 ABTS)으로 평가하였다. 동일 농도에서 TAE는 TAW와 비교하여 라디칼 소거활성이 높게 났고 TAE의 총 폴리페놀 함량이 항산화 활성과 연관되어 있는 것으로 나타났으며, 이는 운동으로 인한 산화스트레스를 방어함으로써 마우스의 운동능력 향상 효과를 나타낸 것으로 사료된다.

사매 에탄올 추출물의 항산화 및 항염증 효과 (Antioxidative and Anti-inflammatory Effect of Ethanol Extract from Duchesnea chrysantha)

  • Oxidative stress has been implicated in cutaneous damage in various inflammatory skin diseases, including atopic dermatitis. The present study was undertaken to investigate the antioxidative and anti-inflammatory activities of the extract of Duchesnea chrysantha (DCE). DEC was prepared by extracting with 80% ethanol. Total flavonoids and polyphenols were measured by a colorimetric assay. The free radical scavenging activity of the extract was analyzed by the DPPH (1,1-diphenyl-2-picryl hydrazyl), ABTS (2,2'-azino-bis(3-ethylbenzthiazoline-6-sulfonic acid) and Griess reagent assay. An oxidative product of nitric oxide (NO), was measured in the culture medium by the Griess reaction. The level of prostaglandin $E_2$ ($PGE_2$) was measured by enzyme-linked immunosorbent assay. The expressions of inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2) were measured by Western blot analysis. Total flavonoid and polyphenol contents of DCE were included $24.73{\pm}0.45$ and $178.77{\pm}2.65$, respectively. DCE significantly increased electron donating ability (DPPH), nitrite scavenging (NO) and ABTS reducing activity in dose dependant. We investigated the anti-inflammatory effects of DCE on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. DCE significantly suppressed NO and prdstaglandin $E_2$ ($PGE_2$) in dose dependant. Furthermore, the levels of iNOS and COX-2 protein expressions were markedly suppressed by the treatment with DCE in a dose dependent manner. These results suggest that DEC may has value as natural product with its high quality functional components, antioxidative and anti-inflammatory activities.

증숙 더덕 에탄올 추출물에 대한 항산화·항균 활성 (Antioxidative and Antimicrobial Effects of Ethanol Extract of Codonopsis lanceolata by Steaming Times)

  • This study was investigated the contents of total polyphenol, flavonoids and antioxidant and antimicrobial activities of Codonopsis lanceolata extracts according to different steaming times. The contents of total polyphenol and flavonoid proportionally increased from 6.45 mgGAE/g to 18.26 mgGAE/g and 2.01 mgRE/g to 6.12 mgRE/g according to ethanol extracts at EDS7. DPPH radical scavenging activity was found to have been 15.26~65.2% and showed the highest level of antioxidant activity at EDS7 was 65.2%. The activity of ABTS radical scavenging and SOD-like activity were also the same result. DPPH and ABTS radical scavenging activity was related to the number of steaming, and the scavenging activity was increased up to 7 times of steaming. The antimicrobial activity of EDS7 had strong antioxidant activity. Antimicrobial activities were examined against 5 microorganisms related to pathogens and food poisoning. The antimicrobial Activity was different depending on the bacteria, but it was effective at the concentration of 300 mg/mL rather than 150 mg/mL. These results showed that Codonopsis lanceolata extracts with a different number of steaming would be conducted to confirm the possibility of developing antimicrobial and antioxidant. It will be helpful in the study of component analysis of Codonopsis lanceolata extracts processed products.
