This study was carried out to investigate the antioxidant activities, total phenol, and flavonoid contents of 70% methanol, 70% ethanol, and chloroform-methanol (CM, 2:1, v/v) extracts from garlic (Allium sativum L.) peels. Antioxidant activity was evaluated by DPPH (2,2-diphenyl-1-picrylhydrazyl) radical and ABTS [2,2′-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) diammonium salt] radical scavenging activity, ${\beta}$-carotene bleaching assay, and ferric reducing antioxidant power. Total phenol and flavonoid were in a range of 32.59-63.05 mg CAE/g, 5.26-9.22 mg QE/g, respectively. Total phenol and flavonoid were predominant with the values of $63.05{\pm}0.20mgCAE/g$ and $9.22{\pm}0.17mgQE/g$ in 70% ethanol extract. Additionally, 70% ethanol extract also displayed the strongest antioxidant activity followed by 70% methanol extract and CM extract in all assays. These results showed that garlic peel can be used as natural antioxidant in nutraceutical preparations to prevent human diseases.
In order to enhance the extraction efficiency of flavonoid from Saururus chinensis, carbohydrate-hydrolyzing enzyme Viscozyme L aided extraction techniques have been studied. Then flavonoid composition, as well as quercetin, were also identified using UV/Vis, HPLC/MS, and 1H-NMR. The results showed that favorable extraction conditions were Viscozyme L concentration of 0.25 mg/g, pH 4.2, reaction at 45 ℃ for 12 h. Under the favorable extraction condition, total flavonoid yield (37.9 mg/g) and quercetin yield (0.86 mg/g) increased by about 2.0 and 9.6 times, respectively, compared to control group. Interestingly, as a significant flavonoid of S. chinensis, flavonoid glycones rutin was hydrolyzed to aglycones quercetin by Viscozyme L. These findings provide scientific and theoretical support for the development quercetin-rich products, which was quickly absorbed by the human body than rutin.
건조방법을 달리한 초석잠(Stachys sieboldii Miq.)을 열수 및 70% 에탄올 용매로 추출하여 제조한 후 추출물 농도에 따른 총 flavonoid 및 총 polyphenol 함량, DPPH radical 소거활성, ABTS radical 소거활성, 산화적 스트레스로부터의 신경세포 보호효과에 미치는 효과를 조사하기 위하여 실시하였다. 초석잠 추출물들의 총 flavonoid와 총 polyphenol 함량은 동결건조 열수 추출물에서 각각 가장 높았다(p<0.05). DPPH 및 ABTS radical 소거활성은 농도의존적으로 증가하였으며, $1,000{\mu}g/mL$ 농도에서는 동결건조 열수추출물에서 유의적으로 가장 높게 나타났으나, 대조군인 비타민 C에 비해 유의적으로 낮았다(p<0.05). 신경세포 보호효과는 열품 및 동결건조 열수추출물에서 농도의존적으로 높게 나타났으며, 대조군인 비타민 C에 비해 낮았다(p<0.05). 본 연구를 통하여 열풍건조에 비해 동결건조 초석잠의 열수추출물이 총 flavonoid와 총 polyphenol 함량이 더 높게 함유되어 있어 항산화 활성 및 산화적 스트레스로부터의 신경세포 보호효과가 우수하게 나타내는 것이 관찰되었다.
This study investigated the phenolic, flavonoid, and vitamin constituents in the main root, root hair, and leaf of ginseng. The total individual phenolic and flavonoid contents were the highest in the leaf, followed by the main root and root hair. Ferulic acid and m-coumaric acid were found to be the major phenolics in the main root and root hair, while p-coumaric acid and m-coumaric acid were the major phenolics in the leaf. Catechin was the major flavonoid component in the main root and root hair, while catechin and kaempferol were the major flavonoid components in the leaf. Pantothenic acid was detected in the highest quantity in the non-leaf parts of ginseng, followed by thiamine and cobalamin. Linolenic acid and menadione were the major components in all parts of ginseng.
The current investigation was carried out to explore the possibility of submerged fermentation of Saccharina japonica as sole substrate using Aspergillus oryzae. In this study we used 2% S. japonica powder as fermentation media for A. oryzae. Fermentation period was optimized by monitoring the fermented sample at regular intervals for a period of 7 days. Results found that a fermentation period of 5 days was effective with maximum desirable characteristics such as total sugar, total phenolic and flavonoid contents. Under optimum fermentation period, fermented extracts showed enhanced antioxidant activity as determined by different assays such DPPH radical scavenging, ABTS scavenging and phosphomolydenum assay. This study provides the information for the enhancement of bioactive molecules in an eco-friendly manner and also paves way towards the development of wide range of seaweed-based functional foods.
Antioxidant activities of Ulmus davidiana root and bark extracts were evaluated by various antioxidant tests, including DPPH radical-scavenging, nitric oxide-scavenging, superoxide anion radical-scavenging, and ABTS radical-scavenging assays, and superoxide dismutase (SOD)-like activity and reducing power analysis, along with the determination of total phenolic and flavonoid contents. Both the extracts showed strong antioxidant activities by these testing methods. Ulmus davidiana root extract possessed strong reducing power and nitric oxide-scavenging activity, and high scavenging activities against free radicals including the superoxide anion, and the ABTS and DPPH radicals, but a weaker scavenging activity of SOD. In contrast, the Ulmus davidiana bark extract exhibited a strong SOD-like activity, but all the other activities were weak. It was observed that the total phenolic and flavonoid contents of the Ulmus davidiana root extract were higher than those of the Ulmus davidiana bark extract.
Purpose: Various plants, herbal medicines, and marine foodstuffs have been used in kimchi preparation to improve its overall quality. Teff, which is rich in minerals and starches, facilitates stable blood glucose levels and is well-suited for use in gluten-free products; hence, it can be used to reinforce the mineral composition of kimchi. In this study, we probed the antioxidant activities of hydrolysates prepared by treatment of brown teff with three proteases under different conditions. Methods: The mineral composition of brown teff was determined by inductively coupled plasma spectrophotometry-mass spectrometry, and we established optimal hydrolysis conditions by determining the total phenol and flavonoid contents of teff hydrolysates obtained using three different proteases (protamax, flavourzyme, and alcalase), two different protease concentrations (1 and 3 wt%), and three different incubation times (1, 2, and 4 h). The antioxidant activity of the hydrolysates was further investigated using 2,2-diphenyl-1-picrylhydrazyl (DPPH) scavenging activity, total antioxidant capacity (TAC), and ferrous reducing antioxidant power (FRAP) assays. Results: Brown teff was rich in I, K, Mg, and Ca, and the highest total phenol content ($24.16{\mu}g/mL$), total flavonoid content ($69.08{\mu}g/mL$), and TAC were obtained for 1 wt% protamax treatment. However, the highest DPPH scavenging activity and FRAP values were observed for hydrolysates produced by alcalase and flavourzyme treatments, respectively. Conclusion: Treatment of brown teff with proteases affords hydrolysates with significantly increased antioxidant activities and high total phenol and flavonoid contents, and these antioxidant activities of teff hydrolysates have the potential to enhance the quality and functionality of kimchi in future applications.
타임, 바질, 로즈마리 등 20종의 허브 식물에 대한 80% 에탄올 추출물의 항산화 활성을 분석하였다. 탐색 대상 허브 식물 중 타임 추출물의 총 폴리페놀 함량은 6.90±0.02 mg CAE/100 g로 높은 값을 보였으며, 총 플라보노이드 함량은 1.71±0.19 mg NE/100 g으로 가장 높았다. 또한 DPPH radical 소거능은 93.14±0.23%로 매우 높은 수준의 항산화 활성을 나타냈다. 타임으로부터 플라보노이드를 추출하기 위하여 용매로서 물, 에탄올, 메탄올, 헥산 중 추출수율이 높은 헥산을 추출용매로 선정하였다. 반응표면분석법을 이용하여 타임 추출물의 최적화 추출조건으로 추출온도는 35.9 ℃, 농도는 1.63%, 시간은 192분이었다. 예측된 조건으로 제조한 타임 n-헥산 추출물의 총 플라보노이드 함량은 예측 값의 96.3%로 반응표면분석법에 의한 추출조건 최적화의 신뢰성이 검증되었다. 이상의 최적화된 추출 조건은 플라보노이드의 생산을 위한 타임의 용매추출 조건으로서 활용할 수 있을 것으로 기대된다.
Background: In this study, microwave extraction was used, which is an effective method to extract useful bioactive substances as it requires low quantities of solvent and short time periods. The aim of this study was to determine the optimal extraction conditions for Agrimonia pilosa Ledeb. Methods and Results: The independent variables were ethanol concentration, microwave power, and extraction time, each of which had five levels. The dependent variables were total polyphenol and total flavonoid content, and DPPH radical scavenging activity. To determine the optimal extraction conditions for bioactive compounds, a response surface methodology was employed. Contour maps were generated from polynomial equations. The optimal conditions were then assumed by superimposing these contour maps. Based on the resulting graph, the optimal microwave extraction conditions for Agrimonia pilosa Ledeb were determined as 42 - 48% ethanol concentration, 240 - 280W microwave power, and 13 - 20 min of extraction time. Conclusions: Ethanol concentration had a significant effect on microwave extraction, in terms of total polyphenol and total flavonoid content, as well as DPPH radical scavenging activity. Microwave power and extraction time influenced the total polyphenol content, but not the total flavonoid content or the DPPH radical scavenging activity.
This study extracted Artemisia capillaris Thunberg with distilled water and ethanol to investigate its antioxidant effect. We then investigated the possibility of producing nanofibers by an electrospinning process by adding the extracts to polyvinyl alcohol (PVA). The electrospinning method used a PVA concentration of 12wt.%, an applied voltage of 10kV, and a tip-to-collector distance of 15cm. Total polyphenol and total flavonoid contents were measured to verify the antioxidant activity of Artemisia capillaris Thunberg extracts (ACEs). The total polyphenol content of the distilled water extract and the ethanol extract were measured as 218.47 and 271.26mg/g, respectively, and the total flavonoid content of the distilled water extract and the ethanol extract were measured as 141.68 and 34.98mg/g, respectively. As the content of the ACEs in the PVA nanofibers increased, the Diameters of the nanofibers and the uniformity of the diameters decreased. The electrospinning process was fabricated in a relatively uniform form without beads, and the diameters of the nanofibers that were produced ranged from 340 to 390nm. The results of FT-IR, XRD and DSC analyses confirmed that the ACEs were well mixed with the PVA molecules and were electrospun.
