Anti-adipocyte differentiation activity and flavonoid content determination by HPLC/UV analysis of tree sprouts

  • The in vitro anti-obesity activity of 12 species of tree sprouts in differentiated 3T3-L1 cells and the mechanisms underlying their activity were evaluated. (+)-Catechin and quercetin concentrations in the sprouts were analyzed by HPLC/UV at 270 and 254 nm, respectively. Euonymus alatus (EAT) and Fraxinus mandschuria (FMS) extracts at doses of 50 and 100 ㎍/mL inhibited the accumulation of lipid droplets in differentiated 3T3-L1 cells. Moreover, EAT and FMS downregulated the expression of the CCAAT/enhancer-binding protein-α, adipogenesis-related proteins peroxisome proliferator-activated receptor-γ, and adipocyte P-2α in differentiated 3T3-L1 cells. Tree sprouts with an abundant flavonoid content exerted the highest anti-obesity activity. Concentrations of total flavonoids were the highest in FMS (24.281 mg/g DW) sprouts. These findings could be used to develop health-promoting functional foods or supplements derived from tree sprouts.

Antibacterial Effect of Allium ampeloprasum and Allium porrum Extracts on Staphylococcus aureus and Pseudomonas aeruginosa

  • Objectives: The most prominent microorganisms that cause hospital infections and acquire antibiotic resistance are Staphylococcus aureus and Pseudomonas aeruginosa. The present study aimed to compare the phenolic and flavonoid compounds of various Allium ampeloprasum and Allium porrum extracts and evaluate the antibacterial effects of these extracts against these two microorganisms. Methods: The total phenolic and flavonoid contents of the acetone, methanol, aqueous, and hexane leeks extracts from A. ampeloprasum and A. porrum were measured. The antibacterial activity of these extracts against S. aureus and P. aeruginosa was tested using the disk diffusion method for 24, 48, and 72 hours. Further, the minimum inhibitory concentrations and the minimum bactericidal concentrations of these extracts for these two bacteria were evaluated and compared with those of common antibiotics. Results: The aqueous extracts showed the highest phenolic and flavonoid contents and at concentrations of 35 and 40 mg per disk, showed the most antibacterial activity against S. aureus and P. aeruginosa; P. aeruginosa showed more sensitivity to the aqueous extracts than S. aureus. Conclusion: Aqueous A. ampeloprasum and A. porrum extracts may prevent the growth of hospital pathogens, especially P. aeruginosa; our findings will aid the discovery of new antimicrobial substances against antibiotic-resistant bacteria.

Antioxidant activities of Erythrina stricta Roxb.using various in vitro and ex vivo models

  • Erythrina stricta, a deciduous tree widely used traditionally in indigenous system of medicine for various ailments such as rheumatism, fever, leprosy, epilepsy etc. The leaves of Erythrina stricta was extracted with ethanol (70%) and used for the evaluation of various in vitro antioxidant assays which includes H - donor activity, nitric oxide scavenging, superoxide anion scavenging, reducing ability, hydroxyl radical, hydrogen peroxide scavenging, total phenolic content, total flavonoid content, total antioxidant activity by thiocyanate and phosphomolybdenum method, metal chelating, $\beta$-carotene bleaching, total peroxy radical assays. The pro-oxidant activity was measured using bleomycin-dependent DNA damage. Ex vivo models like lipid peroxidation and erythrocyte haemolysis were also used to study the antioxidant property of the extract. The various antioxidant activities were compared with suitable standard antioxidants such as ascorbic acid, butylated hydroxyl toluene, $\alpha$-tocopherol, curcumin, quercetin and Trolox. The generation of free radicals viz. $O_2^{{\cdot}-}$, $OH^{\cdot}$, $H_2O_2$, $NO^{\cdot}$ and peroxyl radicals were effectively scavenged by the ethanolic extract of Erythrina stricta. In all the methods, the extract offered strong antioxidant activity in a concentration dependent manner. The total phenolic content, flavonoid content and total antioxidant activity in Erythrina stricta were determined as microgram (g) pyrocatechol, quercetin and $\alpha$-tocopherol equivalent/mg respectively. The extract did not exhibit any prooxidant activity when compared with ascorbic acid. The results obtained in the present study clearly indicates that Erythrina stricta scavenges free radicals and reduces lipid peroxidation, ameliorating the damage imposed by oxidative stress in different disease conditions and serve as a potential source of natural antioxidant.

Comparison of Bioactive Constituents and Biological Activities of Aronia, Blackcurrant, and Maquiberry (아로니아, 블랙커런트, 마키베리의 기능성 성분 및 생리활성 비교)

  • In this study, bioactive constituents and biological activities of aronia, blackcurrant, and maquiberry were investigated. The 60% ethanol extracts were prepared and evaluated for total polyphenol, total flavonoid, and total anthocyanin contents. Biological activities, including 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activities, ferric reducing antioxidant power (FRAP), reducing power, tyrosinase inhibitory activity, and nitrite scavenging activity were also examined. Maquiberry showed the highest bioactive constituents as shown based on total polyphenol (73.66 mg GAE/g), total flavonoid (28.08 mg/g), and total anthocyanin (12.82 mg/g) contents compared to aronia and blackcurrant. Maquiberry also exhibited the highest DPPH and ABTS radical scavenging activity, FRAP, and reducing power at a concentration of 5 mg/mL, which were superior to those of ascorbic acid (a positive control). Therefore, maquiberry could be used as a potential source of antioxidants and functional food substances.

Biological Activity of Korean Dandelion (Taraxacum coreanum) Extracts and Preparation of Korean Dandelion Tea by Roasting Time (흰민들레 추출물의 생리활성 및 볶음시간에 따른 흰민들레 침출차 제조에 관한 연구)

  • This study was conducted to investigate the antioxidative and antimicrobial activities of Korean dandelion (Taraxacum coreanum). Water extracts, ethanol extracts and methanol extracts were used to examine the free radical scavenging activity, total flavonoid content, total polyphenol content and antimicrobial activity. The free radical scavenging activity, total flavonoid, total polyphenol and total antioxidant activity of the water extracts were higher than those of the other extraction solvents. The antimicrobial activties of Korean dandelion extracts were examined on several food borne illness microorganisms using the paper disc diffusion method. Inhibition zones were observed on Staphylococcus aureus subsp. aureus, Listeria monocytogenes, Bacillus subtilis, Bacillus cereus and Escherichia coli in ethanol extracts. Inhibition zones were also observed on Listeria monocytogenes in water extracts. The physico-chemical properties of Korean dandelion tea according to the roasting time and soaking amount of tea were studied. The pH of the dandelion tea significantly decreased while the soluble solid contents significantly increased with increased roasting time (p<0.01). The lightness of the dandelion tea decreased and the turbidity increased with increased roasting time. In sensory evaluation, the sensory scores for the color, flavor and total acceptability were highest in the 40 min roasted tea. These results suggest that the water extract of Korean dandelion could be used as an antioxidative and antimicrobial functional food source. The optimum roasting time for Korean dandelion tea was 40 min at $200^{\circ}C$.

Comparison of Biochemical Composition and Antimicrobial Activity of Southern-Type Garlic Grown in the Eastern and Western Regions of Jeju (제주 동부와 서부지역 남도마늘의 생화학적 성분 및 항균활성 비교)

  • Components and physiological activity of crops are strongly affected by the natural environments of the growth regions. In this study, we carried out soil analysis and determined the allicin, alliin, total pyruvate, total phenol and flavonoid, and reducing sugar contents of garlic from garlic-cultivating regions in the eastern and western portions of Jeju Province. Significantly higher contents of macronutrients were found in the soil and garlic from the eastern region of Jeju compared to western region, but micronutrients were higher in the western region than eastern region. In addition, the alliin and total phenolic contents were higher in the western region than in the eastern region. Also, allicin, total pyruvate, total flavonoid content, and ${\alpha}$-glucosidase inhibitory activity of the eastern region samples were higher than those of the western region in Jeju. This study promotes our understanding of the different components of garlic according to the cultivation regions of Jeju.

Quality Characteristics and Functionality of Mozzarella Cheese with Wine Concentrate (와인 농축액을 첨가한 모짜렐라 치즈의 품질 특성 및 기능성)

  • The objective of this study was to evaluate the quality characteristics of mozzarella cheese added wine concentrate. Mozzarella cheese was produced with different additives of wine concentrate which were 0~5%. The pH and the total acidity of the Cheese were analyzed. We also examined the radical scavenging activities for the antioxidant effect of samples and evaluated for their total polyphenol, and total flavonoid contents. The pH of Meoru wine cheese (6.28~6.37) was significantly higher and total acidity tended to decrease compared to that of the control. L* (lightness) of the wine cheese decreased with increasing amounts of wine concentrates whereas a* (redness) tended to increase. In the texture profile analysis, Meoru wine cheese showed higher values of hardness, gumminess, and chewiness. The ABTS radical scavenging activity of Meoru wine cheese showed the highest value when wine concentrate amounts were 2%. The DPPH radical scavenging activity was significantly increased in cheese added with wine concentrate. Total polyphenol contents and total flavonoid contents of Meoru wine cheese tended to increase with increasing amounts of wine concentrate. This research result highlights the positive influence of wine concentrate addition in cheese. Also, these results are expected to impact the experience programs in farm wineries.

Anti-oxidant effect of ethanol extract from mixture including Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix (정향 및 목향과 백지를 혼합한 한약재 에탄올 추출물의 항산화 효능)

  • Objectives : Herbal medicinal mixture (JMB) are consisted of Caryophylli Flos, Aucklandiae Radix and Angelicae Dahuricae Radix. Each of herbal medicines has studied on anti-oxidant effect. So this study was conducted to investigate efficacy and potency of JMB on anti-oxidation. Methods : The JMB was extracted at room temperature by 80% ethanol. And total polyphenol contents, total flavonoid contents in JMB ethanol extract were determined by colorimetric method. Also, DPPH, ABTS free radical scavenging capacity and reducing power of JMB ethanol extract were measured at 100, 500, 1000, $5000{\mu}g/m{\ell}$ concentrations by spectrometric assay. Positive control was used BHA (butylated hydroxyanisole). Results : The total polyphenol contents and total flavonoid contents of the extract were 55.38 mg/TAEg, 513.72 mg/RUEg, respectively. Also, DPPH free radical scavenging capacity and reducing power of JMB ethanol extract in treated concentrations (100, 500, 1000, $5000{\mu}g/m{\ell}$) increased dose dependently. In particular, DPPH free radical scavenging capacity of JMB ethanol extract at 500, 1000, $5000{\mu}g/m{\ell}$ was similar to positive control (BHA) at high concentration (50, $100{\mu}g/m{\ell}$). ABTS free radical scavenging capacity of JMB ethanol extract at 500, 1000, 5000 ug/ml was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Also, reducing power was showed that JMB ethanol extract at $5000{\mu}g/m{\ell}$ was similar to BHA at high concentration (50, $100{\mu}g/m{\ell}$). Conclusions : These results suggest that JMB ethanol extract has effects to scavenge free radicals. Therefore, JMB has potential and applicable benefits for development of materials and products to have anti-oxidation functions.

Antioxidant and anti-diabetic effects of Ixeris strigosa extract (선씀바귀 추출물의 항산화 및 항당뇨 효과)

  • Purpose: Ixeris strigosa (IS) is a perennial plant that commonly grows in meadows. The leaves and roots of IS have been used in medicine as a sedative. This study evaluated the antioxidant and carbohydrate-digestive-enzyme inhibitory effects of IS to determine its potential as an essential antioxidant and glycemic inhibitor for type 2 diabetics. Methods: The antioxidative and α-amylase and α-glucosidase inhibitory activities were examined using the water extracts (ISW), ethanol extracts (ISE), and solvent fractions from IS. The antioxidative activities were measured using in vitro methods by measuring the 1,1-diphenyl-2-picrylhydrazyl and 2,2'-azino-bis-3-ethylbenzothiazoline-6-sulfonic acid radical scavenging activity. Results: Investigations of the total polyphenol, flavonoid content, in vitro antioxidant activity, and α-amylase and α-glucosidase inhibitory activities of the IS extract showed that the ISE had higher total phenolic and flavonoid contents than the ISW, as well as high antioxidant activity. The ethanolic extracts of IS (70%) had an α-amylase inhibitory activity of 78.55%. The ethyl acetate fraction (90.56%) showed higher α-glucosidase inhibitory activity than the positive control, acarbose (83.01%). Conclusion: Among the ISE fractions, the ethyl acetate and butanol fractions showed the best digestive enzyme inhibitory activity. Moreover, the antioxidant activity of the extract and the carbohydrate, α-amylase, and α-glucosidase inhibitory effects showed a stronger correlation with the total phenol and flavonoid contents compared to the ISW. As a result, the antioxidant and digestive enzyme inhibitory activities of high ISE are due to the phenolic compounds, particularly the flavonoid compounds. Therefore, ethyl acetate and butanol fractions of the 70% ethanol extract are excellent anti-diabetic functional materials.

Total Polyphenol and Flavonoid of Fruit Extract of Opuntia humifusa and Its Inhibitory Effect on the Growth of MCF-7 Human Breast Cancer Cells (천년초선인장 열매추출물의 폴리페놀, 플라보노이드 함량과 유방암 세포(MCF-7)에 대한 성장 억제효과)

  • Opuntia humifusa, widely distributed in the southern regions of the Korean peninsula, is known to have bioactive functions and medicinal benefits for treating various diseases such as arteriosclerosis, diabetes mellitus, gastritis, and hyperglycemia. In this study total polyphenol and flavonoid contents of fruit and its anticarcinogenic effects on human breast cancer were investigated. As expected, O. humifusa showed high concentrations of total polyphenol as well as flavonoid as compared to other kinds of cactus. Effects of the water extracts of O. humifusa on the proliferation, G1 arrest and apoptosis of the MCF-7 human breast cancer cells were also examined using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) assays, and G1 cycle arrest and apoptotic effect of O. humifusa were analyzed by flow cytometry. When MCF-7 cells were treated with different concentrations of hexane, ethyl acetate and water extracts of O. humifusa, water extracts of the fruit significantly decreased viable cell numbers in a concentration dependent manner. A G1 arrest in MCF-7 cells was induced as well. The overall results indicate that water extracts of fruit of O. humifusa would inhibit MCF-7 human breast cancer cell proliferation and induce G1 arrest.
