Biochemical Classification of Coliforms Isolated from Drinking Water (식수에서 분리한 대장균군의 생화학적 성상에 의한 균종별 분포)

  • A total of 136 coliform bacteria isolated from spring water (112 strains) and ground water (24 strains), submitted to Seoul Health and Environmental Research Institute from June to July in 1997, were characterized biochemically and microbiologically. Colonical characteristics of each isolate were also noted, including color and texture on EMB agar. Among the 136 isolates, 50.7% were greenish metallic sheen color, 44.2% were pink and 5.1% were violet. The sixty four percent were smooth, 34.6% were mucoid and 0.7%. were rough. Twenty three bacterial species were identified by IMViC and API 20E test. Among the 136 coliform bacteria known to species, 39 isolates (28.6%) were Escherichia spp., 32 isolates (23.5%) were Klebsiella ssp., 30 isolates (22.1%) were Enterobacter spp., 19 isolates (14.0%) were Serratia spp., 6 isolates (4.4%) were Citrobacter spp., 4 isolates (3.0%) were Kluyvera spp. and 7 isolates (5.1%) were other bacterial species. Strains, which were gas-positive in lactose broth but gas-negative in Kligler Iron Agar were Ent. intermedium, Ser. liquefaciens, Ser. marcescenes and Salmonella arizoae. Strains, which were H2S production were also Kleb. pneumoniae, Kleb. oxytoca, Kleb. ornithinolytica, Ent. sahazahii, Ent. cloacae, Ser. liquefaciens, Ser. fica ria, Cit. freundii and Sal. arizoae. In the present study, most of coliform isolated from spring and ground water were E. coli, Klebsiella spp. and Enterobacter spp. Since coliform with pink colony in EMB agar was isolated as frequent as coliform with greenish metallic sheen colony, coliform with pink colony should be considered as important colony. Our results suggested that new coliform strains may be emerging on the basis of biochemical and microbiological testes.

Fermentation quality and in vitro methane production of sorghum silage prepared with cellulase and lactic acid bacteria

  • Objective: The effects of lactic acid bacteria (LAB) and cellulase enzyme on fermentation quality, microorganism population, chemical composition and in vitro gas production of sorghum silages were studied. Methods: Commercial inoculant Lactobacillus plantarum Chikuso 1 (CH), local selected strain Lactobacillus casei (L. casei) TH 14 and Acremonium cellulase (AC) were used as additives in sorghum silage preparation. Results: Prior to ensiling Sorghum contained $10^4LAB$ and $10^6cfu/g$ fresh matter coliform bacteria. The chemical compositions of sorghum was 26.6% dry matter (DM), 5.2% crude protein (CP), and 69.7% DM for neutral detergent fiber. At 30 days of fermentation after ensiling, the LAB counts increased to a dominant population; the coliform bacteria and molds decreased to below detectable level. All sorghum silages were good quality with a low pH (<3.5) and high lactic acid content (>66.9 g/kg DM). When silage was inoculated with TH14, the pH value was significantly (p<0.05) lower and the CP content significantly (p<0.05) higher compared to control, CH and AC-treatments. The ratio of in vitro methane production to total gas production and DM in TH 14 and TH 14+AC treatments were significantly (p<0.05) reduced compared with other treatments while in vitro dry matter digestibility and gas production did not differ among treatments. Conclusion: The results confirmed that L. casei TH14 could improve sorghum silage fermentation, inhibit protein degradation and decrease methane production.

The Environmental Factors Affecting the Distribution and Activity of Bacteria in the Estuary of Naktong River (낙동강 하구의 세균분포와 활성에 미치는 환경요인)

  • From July 1985 to December 1986, 28 variables of phycal-chemical factors, bacteria and heterotrophic activity were investigated 17 times at 3 stations in the estuary of Naktong River and the influences of environmental factors to bacterial population and heterotrophic activity were analyzed through multiple regression. The results of multiple regression were as follows. At station 1, total bacteria and heterotrophic bacteria(Z-25) could explain 57% of the variation of maximum uptake velocity for glucose and 54% of turnover time for glucose was explained by total coliform bacteria and MBOD, Sixty four percent of the variation of Kt+SN was accounted for salinity, MBOD-N and inorganic phosphate. Turnover rate for acetate was also accounted for the change of MBOD-P by 56%. At station 2 maximum uptake velocity for glucose depends on MBOD-N by 81%; turnover time on bacteria by 50%; Kt+Sn on avilable nutrient by 61%. More than 50% of maximum uptake velocity and turnover time for glucose were influenced by bacteria and that of Kt+Sn by the change of nutrient in the surface water of station 3. In the bottom water of station 3, the change of maximumuptake velocity, turnover time and Kt+Sn for glucose was controlled by total bacteria and available nutrient, bacteria, the change of nutrient salts respectively. On the whole, more than 50% of maximum uptake velocity and turnover time for glucose could be due to the change in the number of bacetria and the value of Kt+Sn was affected by the change of nutrient salts. Turnover rate for acetate was controlled by available phosphate at station 1 and by bacteria at station 2 and 3, which showed a distinct difference between the environmental factors which govern the rate of glucose and acetate uptake in the Naktong esturine ecosystem. And bacterial communities were controlled by available nutrients at station 1, by nutrient salts and salinity at station 2 and in the surface water of station 3 and by salinity in the bottom water of station 3.

Research on Bacterial Contamination Levels in Apartment Tower Elevators (주거용 아파트 엘리베이터의 미생물 오염도와 영향요인 조사)

  • Objectives: The aim of this study was to investigate the microbial contamination levels in elevators in apartment buildings and to provide information on such microbial contamination. Methods: A total of 144 samples, including from the exterior buttons, interior buttons, elevator handrails, walls, ventilators and airborne bacteria were collected in the morning and afternoon from July to August 2013 for six different elevators. The samples were used to detect sanitary indicator bacteria (total bacteria, coliform, and Escherichia coli), pathogenic bacteria (E. coli O157, L. monocytogenes, Salmonella spp., B. cereus, S. aureus) and fungi. Results: Contamination levels of total bacteria were 0.3-3.8 and 0.0-2.4 log CFU/100 $cm^2$ in the morning and afternoon, respectively. In the case of coliform bacteria, the levels were 0.0-3.7 log CFU/100 $cm^2$ in the morning and 0.0-0.3 log CFU/100 $cm^2$ in the afternoon. However, E. coli was not detected among all samples. Bacillus cereus, pathogenic bacteria, was only detected in 13 (11%) among 144 samples. E. coli O157, L. monocytogenes, Salmonella spp. and S. aureus were not detected among all samples. Comparing the samples collected in the morning and afternoon, we could confirm that the samples in the afternoon were cleaner. Conclusions: This study indicates that the samples in the afternoon were cleaner because these samples were collected following routine cleaning. Also, the levels of contamination in the elevators were low and the sanitary conditions were comparatively well-managed. Therefore it is deemed necessary for elevators be cleaned regularly to provide good conditions for people using elevators.

STUDIES ON SANITARY QUALITY OF JARHI-DOM CHROMIS NOTATUS (자리돔의 식품위생학적 연구)

  • This study was carried out to evaluate the sanitary quality of Jarhi-dom, Chromis notatus, and its products, during the period from May to October, 1975. And bacterial density of the sea water in the estuary of Sogwipo was compared. The number of general bacteria and coliform group in the sea water appeared lower than that of other areas. Both Jarhimuchim-hoe and Jarhimul-hoe showed higher bacterial densities than the fresh fish when caught and sold in the market although Jarhimuchim-hoe showed slightly higher density than Jarhimul-hoe. Vibrio parahaemolyticus was detected in some samples of sea water and Jarhi-dom, but it was not regarded for food poisoning. Bacterial densities in total plate counts, coliform group and Vibrio parahaemolyticus was highest in August and September.

Effectiveness of 70% Alcohol Solution and Hand Washing Methods on Removing Transient Skin Bacteria in Foodservice Operation (급식시설의 손 세척을 위한 70% 알콜 소독제 사용 및 세척방법의 적용효과 분석)

  • Hand washing is an important component of hygiene program for food handlers. Hands can be a source of direct or indirect contamination of foods with pathogenic microorganisms. In this study, the effectiveness of hand washing methods and the use of 70% alcohol solution against transient skin bacteria was tested in an university foodservice facility. 70% alcohol solution is sprayed for 5 seconds automatically when hands are placed in the dispenser. Samples were taken using swab technique in meat cutting area, vegetable trimming area, and vegetable cutting area: before and after washing hands according to planned methods, and after being sprayed with 70% alcohol solution after washing hands. The bacteriological analysis of total plate counts, coliform, fecal coliform of food handlers' hands was done. Statistical data analysis was completed with Mann-Whitney U test and Kruskal-Wallis model using the SPSS program. The levels of initial contamination of workers' hand were significantly different by the work areas($x^2$=9.156, p<0.01). Workers in the vegetable trimming area had more heavily soiled hands than in the other work areas. Mean of TPC counts and coliform was 8.97×$10^5$ CFU/12.4$cm^2$, 2.93×$10^2$ MPN/12.4$cm^2$ respectively, but fecal coliform was not detected. Transient bacteria were removed from hands after washing and using 70% alcohol solution but were not removed completely. Mean reduction percentage in TPC varied among work areas and ranged from 93.19% to 94.99%, and in coliforms from 97.31% to 100%. A significant difference in TPC was found between before and after hand disinfection (Z=-2.714, p<.01) and between standardized hand washing procedures and un-standardized hand washing procedures(z=-2.301, p<.01). Subjects using the hand sanitizer showed a great elimination of TPC(99.45% reduction), but this effect was valid only after following proper washing procedures. Based on the results, the most effective hand washing method was recommended as the combination of the standardized hand washing procedure with warm-water and use of the 70% alcohol solution. The results can be used to develop hand hygiene programs and training strategies for enhancing hand hygiene practices for food handlers in foodservice operations.

The Conductance Determination of Total, Coliform and Psychrotrophic bacteria Counts in Raw Milk by Using Malthus (Malthus를 이용한 원유(原乳)내의 총균수, 대장균군수, 저온성균수 측정)

  • This study was performed to obtain fast, consistant and reliable estimation system of bacterial counts of raw milk, which effectively related to the quality of sanitaion and the condition of production at the farm. This study compared regression equation and correlation coefficient relationship between standard plate counts and data of Malthus conductance method for the detection time of total, psychrotrophs, coliform bacterial counts in raw milk. Regression equation (RE) between conductance detection time (Y) and total bacterial log counts (X) was Y=18.27651 - 2.07550X, with correlation coefficient -0.95(n=201). In coliform, RE was Y=9.320848 - 1.15598X with correlation coefficient -0.90 (n=207). Psychrotrophs had the RE of Y=29.96008-3.02487 with correlation coeffecient -0.9 (n=201). This conductance method gave results more quickly and was less labor-intensive than traditional standard plate count method.

Distribution of Marine Bacteria and Coliform Groups in Puksin Bay, Korea (북신만의 대장균군 및 해양세균의 분포)

  • Puksin Bay located in the northwestern pan of Tongyeong, Korea has been contaminated by municipal wastewaters. In Puksin Bay, red tides have occured almost every year since the early 1980's. This experiment was carried out two times in a month in the winter in 1990, the summer' 1991, and the winter in 1994 so as to clarify the distribution marine bacteria and coliform groups in Puksin Bay. The water quality of Puksin Bay was not only move polluted than that of any other costal area around Tongyeong, but also its water quality was investigated to keep going bad. Viable cell counts in Puksin Bay were $4.9\times10^3/ml$ in 1990's winter, $3.6\times10^6/ml$ in 1991's summer, and $2.1\times10^4/ml$ in 1994's winter. The variation of seasonal total and fecal coliform MPN/100ml in Puksin Bay were $6.7\times10^2\;and\;2.6\times10^2$ in 1990's winter, $1.5\times10^4$ and $5.4\times10^3$ in 1991's summer, and $1.5\times10^3$ and $5.6\times10^2$ in 1994's winter, respectively. The changes of stational total coliform MPN/100m1 from station 1 to station 8 in Puksin Bay were 95,000, 1600, 1,000, 182, 151, 94, 43 and 13 in winter, and 110,000, 29,000, 2,400, 4,100, 1,700, 1,700, 810 and 150 in 1991's summer, and 3,381, 1928, 1582, 256, 161, 59 and 23 in 1994's winter. During the study period, the number of viable cell was ranged from $10^4\;to\;10^7/ml$ and 307 bacteria strains were isolated from Puksin Bay. The dominant species were Acinetobacter spp. 86 $(28.3\%)$, Pseudomonas spp. 51 $(16.6\%)$, Flavobacterium spp. 41 $(13.4\%)$, Escherichia coli 36 $(11.7\%)$, and Vibrio spp. 27$(8.8\%)$. The results obtained in this study indicate that this bay is getting to contaminate far more with municipal wastewaters and cultivation of the shellfish and finfish in this bay is not proper. When municiple wastewaters keep flowing into this bay, any other coastal area around Tongyeong may be contaminated.

  • Seasonal distribution of various microorganisms was investigated in the effluent water in various sewage treatment plants from January to November in 2007. The target microorganisms were composed of total heterotrophic bacteria, Escherichia coli, coliform and enteric bacteria, Staphylococcus aureus, Yeast and Mold. The number of total bacteria and the indicator microorganisms, such as E. coli, coliform, and Enterobacteriacea, were increased during the summer season. S. aureus, however, was not influenced by the weather, showing higher number even in November and January in most sewage treatment plants investigated. Meanwhile, disinfection methods employed in various plants had no direct relationship with the growth of microorganisms, and small-scaled plants were found to be more effective than large-scaled ones in the sterilization of microorganisms.

Effect of Chlorine Dioxide Treatment on Microbial Growth and Qualities of Chicken Breast

  • Chlorine dioxide $(ClO_2)$ treatment was evaluated for microbial growth inhibition and its effects on the quality of vacuum-packaged chicken breasts. Chicken breast samples were treated with 3, 50, and 100 ppm of $ClO_2$ solution, respectively. After $ClO_2$ treatment, chicken breast samples were individually vacuum-packaged and stored at $4^{\circ}C$, a typical storage temperature for meat and meat product, for 7 days. The vacuum-packaged chicken breasts treated with $ClO_2$ had significantly lower total bacteria, yeast and mold, total coliform, and Salmonella spp. were significantly reduced by $ClO_2$ treatment. $D_{10}-values$ of total bacteria count, yeast and mold, total coliform, and Salmonella spp. in vacuum-packaged chicken breasts was 93, 83, 85, and 50 ppm, respectively. The pH of vacuum-packaged chicken breasts decreased with increasing $ClO_2$ concentration. Thiobarbituric acid reacted substance (TBARS) values of vacuum-packaged chicken breasts increased during storage, regardless of $ClO_2$ concentration. $ClO_2$ treatment caused negligible changes in Hunter L, a, and b values in the vacuum-packaged chicken breasts. Sensory evaluation of the vacuum-packaged chicken breasts showed that there were no significant changes among the samples treated with various $ClO_2$ concentration. These results indicate that $ClO_2$ treatment could be useful in improving the microbial safety and quality of meat products.
