THE LOWERING EFFECT OF PANAXYDOL PURIFIED FROM KOREAN RED GINSENG ON BLOOD HIGH CHOLESTEROL LEVELS IN RATS

  • The lowering effect of cholesterol in Sprague Dawley rats was investigated with panaxydol which was purified from the petroleum ether soluble fraction(PESF) of Korean red ginseng. The level of total cholesterol(TC), Triglyceride(TG) and low density lipoprotein(LOL)-cholesterol in serum was reduced by $48\%,\;47\%\;and\;41\%,$ respectively while high density lipoprotein(HDL)-cholesterol was in creased up to $29\%$ as compared with their control values when the panaxydol(20 umoles. 5 mq/kq/day) was adminstered by intraperitoneal rout for 3 consecutive days along with a $1\%$ cholesterol diet. The hepatic ester cholesterol content which was increased in proportion to the cholesterol content of the diet in the control, clearly decreased with panaxydol administration to about $40\%$ regardless of the two diet cholesterol content. $1\%\;or\;2\%.$ A threshold of supression on the serum lipid levels in both administration routes was observed: the maximium suppression in i.p. and p.o. administration was observed to be at 5mq/kq b.w. and in the range of 50 - 100 mg/kg b.w., respectively. Panaxydol may reduce serum lipid and cholesterol levels by inhibiting cholesterol absorption and/or by modulating the cholesterol metabolism. at least in part.

가와사끼병에서 혈청 지질 대사의 변화 및 관상동맥류에 관한 연구 (A study of serum lipid level and relation between the serum lipid level and coronary aneurysm after kawasaki disease)

  • We measured plasma concentrations of high density lipoprotein, total cholesterol and triglycerides in 31 patients with history of kawasaki disease during acute stage and convalescence stage. High density lipoprotein cholesterol and total cholesterol concentrations were significantly lower(P<0.001) in samples taken within 11 days of the onset of illness($28.7{\pm}12.4mg/dl$ and $145.20{\pm}29.60mg/dl$) than in the second samples taken 1-2months after onset of disease($51.5{\pm}15.2mg/dl$ and $175.4{\pm}29.0mg/dl$). Change of triglycerides was not significant. There was no correlation between the serum cholesterol cencentration and coronary aneurysm and continued long term surveillance of much population is necessary to monitor lipid level and their relation to development of premature coronary atherosclerosis.

Changes of Plasma Cardiovascular Disease Risk Factors according to the Health Practice and Dietary Habits in Healthy Male University Studnets

  • This cross-sectional study was conducted to describe the changes of plasma cardiovascular disease(CVD) risk factors in Korea. Overnight fasting plasma levels of total cholesterol, high density lipoprotein(HDL)-cholesterol, triacylglycerol and glucose were analyzed. Blood pressure and anthropometric data were also measured. Health practice factors such as smoking status, alcohol consumption and frequency fo exercise were evaluated by a self-administered questionnaire. Questions regarding dietary habits and food preferences were also asked. Seventy eight percent of the subjects had more than one CVD risk factor. Plasma total cholesterol, triacylglycerol, and fasting blood glucose were significantly increased according to the subjects body mass index$(kg/m^2$, BMI), whereas HDL-cholesterol, low density lipoprotein(LDL)-cholesterol and blood pressure showed no significant differences with BMI. Current smokers had significantly high plasma total cholesterol, LDL-cholesterol and triacylglycerol levels. Alcohol consumption significantly increased plasma total cholesterol and fasting blood sugar, but regular exercise had no effects on the plasma CVD risk factors. Overeating and frequency of fast food consumption were positively correlated with the CVD risk score, whereas intake of grains, meats and vegetables were negatively correlated with that score. A stepwise multiple regression analysis was performed to examine the effects of specific dietary factors on plasma lipid levels. For plasma total cholesterol level, the frequency of fast food intake explained 8% of the variance, followed by habitual overeating, frequency of grain intake and high cholesterol food intake(Model $R^2$=22.4%). For plasma triacylglycerol level, preference of oily foods accounted for 7.5% of the variance, followed by eating breakfast, preference of fruit and frequency of grain intake(Model $R^2$=22.0%). The findings suggest that intervention programs to reduce the risk of CVD should focus on health practice through reducing BMI, smoking cessation and moderate or no alcohol drinking. Moreover, desirable dietary habits such as eating breakfast, not overeating and reduced intake of fast food may improve CVD risk.

Effects of Lifestyle and Dietary Behavior on Cardiovascular Risks in Middle-aged Korean Men

  • Lifestyle and dietary behavior intervention as the primary prevention of lipid disorder seems safe and compatible with other treatments of cardiovascular diseases. Cross-sectional associations between lifestyle factors and dietary behavioral factors with plasma lipid and lipoprotein levels were analyzed in 189 middle-aged men in Suwon, Korea. Overnight fasting plasma levels of total cholesterol, high-density lipoprotein(HDL)-cholesterol, triacylglycerol and glucose were analyzed. Blood pressure and anthropometric data were also measured. Lifestyle factors such as smoking status, alcohol consumption and frequency of physical exercise were evaluated by a self-administered questionnaire. Questions regarding dietary behavior were also asked. The subjects were 43.8%${\pm}$7.9 years old, and 23.8%${\pm}$2.6kg/m$^2$. From stepwise regression analyses, significant correlates with total cholesterol level were body mass index(BMI), alcohol intake(negative), age and coffee drinking(model R$^2$=14.3%). BMI, breakfast-skipping, age, and sleeping hours were significant for triacylglycerol level(model R$^2$=15.8%). BMI, alcohol drinking(negative), age, and coffee drinking were significant for low-density lipoprotein(LDL)(model R$^2$=11.7%). Age(negative), BMI(negative), alcohol drinking, stress level(negative), physical exercise, and cigarette smoking(negative) were significant for high-density lipoprotein(HDL)(model R$^2$=12.1%). From stepwise regression analyses, excluding BMI and age as factors in the model, alcohol intake(negative) and coffee drinking were significantly correlated with total cholesterol level(model R$^2$=4.4%) : breakfast-skipping with triacylglycerol(model R$^2$=3.2%) : alcohol intake (negative) with LDL level(model R$^2$=3.4%) : alcohol intake, physical exercise and stress level(negative) with HDL level(model R$^2$=6.3%). The findings suggest that a healthy daily lifestyle and dietary behavior may have an anti-atherogenic effect by altering plasma lipid and lipoprotein levels in middle-aged Korean men.   • Effects of Bupleuri Radix, Paeonie Radix or Uncariae Ramulus et rncus on experimental yperlipidemia were studied. Hyperlipidemia was induced on male Wistar rats by feeding them on high holesterol diet for one week, as previously described by the authors. Blood lipid profile was verified on these ats by measuring total cholesterol(TC), triglyceride(TG), high density lipoprotein cholesterol(HDL) and low ensity lipoprotein cholesterol(LDL). Then, the rats were followed by feeding normal diet. At the same time, ethanol extracts of the three individual herb medicines were orally administered to the rats for 4 weeks and the parameters above mentioned were monitored. Methanol extract of 5Bupleuri radix reduced the TC value significantly at 2 week point and Paeoneae Radix reduced the TC value at 4 week point in compare to control group, suggesting the antihyperlipidemic effect of the two herbal medicines in vivo. The extract of Uncariae Ramulus et Uncus however did not show antihyperlipidemic effect in our experiment.

Protective Effect of Curcumin and Aqueous Extract of Onchengyeum on CCI4-induced Hepatotoxicity

  • An aqueous extract of oriental herbal composition named Onchengyeum and curcumin, an antioxidant isolated from turmeric (Curcuma Zonga L.) reduced hepatotoxicity induced by carbon tetrachloride ($CCI_4$). Improved liver function was observed by measuring the activities of aspartate aminotransferase (AST), alanine aminotransferase (ALT), alkaline phosphatase (ALP), blood urea nitrogen (BUN), creatinine (CRE), total cholesterol (T-CHO), triglyceride (TG), low density lipoprotein cholesterol (LDL-CHO), high density lipoprotein cholesterol (HDL-CHO), total protein (TP), albumin (ALB) and total bilirubin (BIL) in serum. Hepatic parameters monitored were levels of cholesterol (CHO), triglyceride (TG), and malondialdehyde (MDA) and activities of cytochrome P450 (CYP), NADPH-CYP reductase, superoxide dismutase (SOD), catalase (CAT), glutathione (GSH), glutathione S-transferase (GST), glutathione reductase (GR), and glutathione peroxidase (GPx). The histopathological examination showed that the treatment of Onchengyeum and curcumin relieved the ballooning degeneration of hepatocytes which had been generated by $CCI_4$. The results suggested that hepatoprotective effects of Onchengyeum and curcumin possibly are due to their promising antioxidative activity.

Protective Effect of Sachungwhan against CCl4-induced Hepatotoxicity

  • Sachungwhan reduced hepatotoxicity induced by carbon tetrachloride($CCl_4$). 소형견종에서 Homocysteine과 비만, 당 관련 인자, 지방 관련인자의 상관관계에 대한 연구 (The Relationship between Homocysteine, Obesity, Glucose and Lipid Profiles in Small-Breed Dogs)

  • 본 연구는 건강한 개에서 혈장 homocysteine농도가 비만이나 비만 관련인자(예, 지방, 인슐린, 혈당, 글루카곤, fructosamine)들과 어떤 상관관계가 있는지를 확인하기 위해 실시되었다. 이를 위해, 절식시 혈당, 지방지수(예, total triglycerides [TG], total cholesterol [TC], high-density lipoprotein cholesterol [HDL-C]과 low-density lipoprotein cholesterol [LDL-C]), fructosamine, insulin 및 glucagon 농도를 각각 측정하였다. 실험에 사용된 개들은 body condition score (BCS)에 따라 분류하였다. 평균 혈장 homocysteine 농도는 비만한 개 집단이 정상이나 마른 개 집단에 비해 상당히 높았다. 또한 혈장 homocysteine농도는 HDL-C농도 뿐아니라 TG, TC, 혈장 glucagon 및 fructosamine 농도와도 밀접한 상관관계를 가지고 있었다. 반대로 LDL-C 농도, 절식시 혈당농도 및 insulin농도는 아무런 상관관계가 확인되지 않았다. 본 연구 결과, homocysteine의 농도 상승이 비만견에서 생물학적으로 중요한 역할을 하는 것으로 추정된다.

항혈전작용 및 콜레스테롤 대사에 관한 흰쥐 식이내 삶은 계란 급여 효과 (Effects of the Dietary Boiled Eggs on the Antithrombotic Activity and Cholesterol Metabolism in Rats)

  • The influence of the ditary containing boiled eggs on the plasma cholesterol level and antithrombotic activity in rats was studied. Rats were fed basal diet(0% boiled eggs) as a control group or diets containing 25% and 50% boiled eggs or a mixed diet with 95% boiled eggs plus 5% $\alpha$-cellulose powder as a experimental groups for 30 days. The bleeding time and whole blood clotting time were significantly(P<0.05) increased by feeding diet containing 25% boiled eggs compared to groups of basal diet, 50% or 95% boiled eggs diets. The plasma clotting time was high in group of 25% boiled eggs diet. However, there were no difference in plasma clotting time among rats fed the dietary boiled eggs. The levels of plasma total cholesterol(TC) and low density plus very low density lipoprotein cholesterol(LDL, VLDL-C) were significantly(P<0.05) highest in group 95% boiled eggs diet compared to others. There were no differences in high density lipoprotein cholesterol(HDL-C) among rats fed the dietary boiled eggs. The levels of plasma TC, HDL-C, LDL$.$VLDL-C and the ratios of HDL-C/TC were not significant among the basal diet, 25% and 50% boiled eggs diets. These results suggest that the intakes of the dietary boiled eggs have the antithrombotic activity and plasma cholesterol lowering effect.

제2형 당뇨병 환자의 대혈관 합병증 유무별 혈중 지질농도와 Lipoprotein(a) 비교 (Plasma Concentrations of Lipid Profiles and Lipoprotein(a) In Type 2 Diabetes Mellitus Patients with Macrovascular Complications)

  • The purpose of this study to examine lipid profiles and lipoprotein (a) concentrations and identify the risk factors of type 2 diabetes mellitus patients with macrovascular complications. The subjects were consisted of 618 out patients with type 2 diabetes mellitus from beginning of March through the end of April in 2001, who visited at the endocrinology department at Kangnam St. Mary's Hospital of Catholic University in Seoul. The patient's clinical laboratory data and the occurrence of chronic complications of diabetes were assessed at medical record review. The data were analyzed using for t-test. chi-square test and logistic regression. The results were as follows : 1. There were no significant differences in age. duration of diabetes. body mass index, sex, fasting blood glucose, 2-hour postprandial blood glucose and $HbA_{1}c$ level between macrovascular complication group and non macrovascular complication group. 2. There were significant differences in the level of total cholesterol. triglyceride. HDL(High density lipoprotein) cholesterol, LDL(Low density lipoprotein) cholesterol and lipoprotein(a) between macrovascular complication group and non macrovascular complication group. 3. Significant factor associated with macrovascular complication in the logistic regression best gut model was HDL cholesterol.
