Quality Characteristics of Cookies prepared with Chinese Sweet Tea Leaf (Rubus suavissimus S. Lee) Powder (첨차(Rubus suavissimus S. Lee) 분말을 첨가한 쿠키의 품질 특성)

  • The aim of this study was to propose a mixing proportion by adding Rubus suavissimus powder, and to determine the characteristic of the product. The spread factor and the hardness, moisture contents, and soluble solids content of the cookies significantly increased with an increase in Rubus suavissimus powder, although the pH of the dough, and L values of the cookies decreased when increasing the Rubus suavissimus powder content (p<0.001). Total phenolic contents and DPPH free radical scavenging activity of cookies significantly increased when more Rubus suavissimus powder was added. The antioxidant activity was highly correlated with the total phenolic composition of sweet tea leaf cookies(r=0.976). Additionally, the consumer acceptability scores for the 6% sweet tea leaf cookie groups ranked significantly(p<0.001) higher than those of the other groups in overall preference. In conclusion, the results of this study suggest that Rubus suavissimus powder is a good ingredient for increasing the functionality of cookies.

Antioxidant Activities of Five Melania Snails of the Genus Semisulcospira in Korea (한국산 다슬기류(Semisulcospira spp.) 5종의 항산화 활성)

  • The in vitro antioxidant activities of five melania snails in the genus Semisulcospira (S. coreana, S. forticosta, S. libertina, S. tegulata and S. gottschei) were tested in detail. The total phenolic contents of the snails ranged from $32.3{\pm}1.0$ to $87.9{\pm}6.9$ mg gallic acid equivalent (GAE)/g dry sample. The $EC_{50}$ values for the 2,2-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities were $2.245{\pm}0.179$ mg/mL for S. coreana and $9.319{\pm}1.276$ mg/mL for S. gottschei and differed significantly (P<0.05) among the tested species. The superoxide dismutase (SOD)-like activity was highest for S. gottschei at 67.2% and lowest for S. forticosta at 4.7%. However, no significant differences among the species were recognized for the peroxynitrite anion scavenging activity. Comparing the correlation coefficients between the total phenolic contents and the DPPH radical and peroxynitrite anion scavenging activities, there was a low level relationship between each activity.

Anti-Glyaction and Radical Scavenging Activities of Ligustri Fructus by Extraction Method (추출 방법에 따른 여정실의 최종당화산물 생성 저해 및 라디칼 소거 활성)

  • Objectives : Ligustri Fructus has been used since ancient times as a medicinal usages in folk medicines against antitumor purpose. Many biological active constituents have been identified from this biomass such as several terpenoids and lignans. In current study, the properties of antioxidant and anti-diabetic complications using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and 2,2'-azino-bis(3-ethylbenzothiazoline-6-sulphonic acid)($ABTS^+$) radicals scavenging, and advanced glycation end products (AGEs) inhibition assays were evaluated by different extraction methods of Ligustri Fructus. Methods : In present continuing research for development of bioactive natural ingredients, antioxidant and AGEs formation inhibitory capacities of Ligustri Fructus extracts using different organic solvents were prepared and the biological potentials were investigated using in vitro bioassays. Antioxidant properties were evaluated employing radical scavenging assays using DPPH and $ABTS^+$ radicals. In addition, the anti-diabetic complications effects of Ligustri Fructus extracts were tested via AGEs formation inhibitory assay. The total phenolic contents were determined using a spectrophotometric method. Results : All the tested extracts exhibited dose-dependent radical scavenging and AGEs formation inhibitory activities. Among the tested samples, hot water extract of Ligustri Fructus was showed the most potent activity with $IC_{50}$ value of $494.8{\pm}6.7{\mu}g/m{\ell}$ against DPPH radical scavenging assay. Also, $ABTS^+$ radical scavenging activity of hot water extract was higher than those of other extracts. In addition, AGEs formation inhibitory effects of each extacts and total phenolic contents were evaluated. Conclusions : These results suggested that Ligustri Fructus can be considered as a new effective source of natural antioxidant and anti-diabetic complications resources.

Antioxidant Properties of Lentinula edodes after Sawdust Bag Cultivation with Different Oak Substrates (참나무 수종별 톱밥재배에 따른 표고의 항산화 특성)

  • We evaluated the antioxidant properties of Lentinula edodes upon sawdust bag cultivation with 5 oak substrates: Quercus acutissima, Q. mongolica, Q. serrata, Q. aliena, and Q. variabilis. We found that the optimal extraction conditions were 70% (v/v) methanol shaken at 150 rpm at $25^{\circ}C$ and 150 rpm for 24 h. The methanolic extracts from L. edodes contained high phenolic and flavonoid contents, they also exhibited stronger antioxidant activities. The total phenolic contents and total flavonoid contents of the mushroom extracts ranged from 2.37 to 3.12 milligrams of gallic acid equivalents per gram of dried mushroom (mg GAE/g) and 0.48 to 0.48 milligrams of quercetin equivalents per gram of dried mushroom (mg QE/g), respectively. In addition, the mushroom extracts exhibited 2,2-diphenyl-1-picrylhydrazyl radical scavenging activity (15.8% to 89.4%) at 2 to 10 mg/mL, ferric reducing antioxidant power (0.153 to 0.425) at 5 to 20 mg/mL, and reducing power (0.078 to 0.359) at 5 to 20 mg/mL, respectively. Q. aliena more effectively increased total phenolic contents, total flavonoid contents, and antioxidant activities than the other oak substrates.

Antibacterial Activities of Phenolic Components from Camellia sinensis L. on Pathogenic Microorganisms

  • Antibacterial activities of the major phenolic components from Camellia sinensis L. were investigated against several pathogenic microorganisms including Gram-positive strains like Staphylococcus aureus ATCC 29213 and Streptococcus pyogens 308A; and Gram-negative strains like Escherichia coli ATCC 25922, Escherichia coli 078, Pseudomonas aeruginosa 9027, and Enterobacter cloacae 1321E. The MIC values demonstrate that both (-)-epicatechin and (-)-epigallocatechin were more considerably toxic against Staphylococcus aureus ATCC 29213 than the other two catechins like (-)-epicatechingallate and (-)-epigallocatechin-3-gallate. (-)-Epicatechingallate and (-)-epigallocatechin-3-gallate were most inhibitory against Escherichia coli ATCC 25922. As a result, (-)-epicatechin showed predominant antibacterial activities among tea varieties. The contents of major polyphenolic components such as four catechins, theaflavin, and quercetin were different according to fermentation processes. The total contents of four catechins were ranged from 13.81 to 1.33%, with (-)-epigallocatechin-3-gallate being dominant among tea varieties; theaflavin was found the characteristic pigment in fully-fermented black tea.

Antioxidant Activity and Total Phenolic Contents of Grape Juice Products in the Korean Market (시판 포도 주스의 항산화 활성 및 총 페놀 함량)

  • The total phenolic content and antioxidant activities of grape and fruit juice products in the Korean market were examined The total phenolic content was measured by the Folin-Ciocalteau method, and antioxidant activities were evaluated by FRAP and DPPH assays. The total phenolic content of the grape juice products were within the range $57.95{\sim}205.64\;mg/L$. Orange juice had the strongest antioxidant activity, apple juice the weakest, and grape juice was intermediate. Grape juice products exhibited a wide range of antioxidant activities. Especially, GU4, GU5, and GU9 exhibited about 80% of the DPPH radical scavenging activities, similar to the antioxidant activities by the FRAP assay. The antioxidant activities by FRAP and DPPH assays were well correlated with the total phenolic content of grape juice products (> 0.97).

Antioxidant and Anti-Inflammatory Effects of Various Cultivars of Kiwi Berry (Actinidia arguta) on Lipopolysaccharide-Stimulated RAW 264.7 Cells

  • The present study evaluated the total phenolic and flavonoid contents as well as total antioxidant capacity (TAC) of three cultivars of Actinidia arguta Planch. kiwi berries; cv. Mansoo (Mansoo), cv. Chiak (Chiak), and cv. Haeyeon (Haeyeon). In addition, the anti-inflammatory effects of the three cultivars of kiwi berries were investigated using a lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cell line. Mansoo had the highest total phenolic content and TAC among the three cultivars, whereas Chiak had the highest total flavonoid content. The total antioxidant capacities of the kiwi berry extracts were more strongly correlated with total phenolic content than with total flavonoid content. The kiwi berry extracts suppressed the secretion of pro-inflammatory cytokines, including interleukin-6 and tumor necrosis factor-α, from LPS-stimulated RAW 264.7 cells. The release of nitrite, an indirect indicator of nitric oxide, was also ameliorated by pre-treatment with the kiwi berry extracts in a dose-dependent manner. Cellular-based measurements of antioxidant capacity exhibited that the kiwi berry extracts had cellular antioxidant capacities. Such cellular antioxidant effects are possibly attributed to their direct antioxidant capacity or to the inhibition of reactive oxygen species generation via anti-inflammatory effects. Our findings suggest that kiwi berries are potential antioxidant and anti-inflammatory agents.

Antibacterial Effect of Allium ampeloprasum and Allium porrum Extracts on Staphylococcus aureus and Pseudomonas aeruginosa

  • Objectives: The most prominent microorganisms that cause hospital infections and acquire antibiotic resistance are Staphylococcus aureus and Pseudomonas aeruginosa. The present study aimed to compare the phenolic and flavonoid compounds of various Allium ampeloprasum and Allium porrum extracts and evaluate the antibacterial effects of these extracts against these two microorganisms. Methods: The total phenolic and flavonoid contents of the acetone, methanol, aqueous, and hexane leeks extracts from A. ampeloprasum and A. porrum were measured. The antibacterial activity of these extracts against S. aureus and P. aeruginosa was tested using the disk diffusion method for 24, 48, and 72 hours. Further, the minimum inhibitory concentrations and the minimum bactericidal concentrations of these extracts for these two bacteria were evaluated and compared with those of common antibiotics. Results: The aqueous extracts showed the highest phenolic and flavonoid contents and at concentrations of 35 and 40 mg per disk, showed the most antibacterial activity against S. aureus and P. aeruginosa; P. aeruginosa showed more sensitivity to the aqueous extracts than S. aureus. Conclusion: Aqueous A. ampeloprasum and A. porrum extracts may prevent the growth of hospital pathogens, especially P. aeruginosa; our findings will aid the discovery of new antimicrobial substances against antibiotic-resistant bacteria.

Anticancer and Antimutagenic Activities after Simulated Digestion of Ethanol Extracts from White, Red and Yellow Onions

  • The beneficial effects of digested onion extracts have been assessed by antimutagenic and anticancer activities by Ames test and SRB test. The total phenolic acids and flavonoids in onion extracts were determined. Red and yellow onions contain more phenolic acids and flavonoids than those in the white onion. Digested, extracts showed antimutagenic activity and anticancer activity, and it appears that the antimutagenic activity of digested extracts of onion against mutagens and anticancer activities were related to their phenols and flavonoids contents. Moreover, the extracts inhibited the proliferation of four human tumorigenic cell lines such as HT-29 (colon), MCF-7 (breast), DU-145 (prostate) and HepG2 (liver), in a dose-dependent manner. Phenolic acids and flavonoids caused oxidative damage to the cancer cell lines and induced apoptosis. Generally, red onion extracts showed effective antimutagenic and anticancer activity, and the digested red onion extracts elicited stronger antimutagenic activity than those of the onion extracts without digestion.

Antioxidative Effect of Ethanolic Extracts of Some Tea Materials on Red Pepper Seed Oil (고추씨 기름에 대한 다류 에탄올 추출물의 항산화 효과)

  • Antioxidative effect of ethanolic extracts of various tea materials(Camellia sinensis, Cassia tora, Lyc ium chinense, Polygonatum odoratum, Schizandrae chinensis) on red pepper seed oil was investigated. Ethanolic extracts were added to red pepper seed oil at a concentration of 0.05%(w/v). Two experimental conditions were employed : 50$\pm$0.1oC for 45 days and 150$\pm$3oC for 24 hours. Oxidation of red pepper seed oil was determined by measuring peroxide value and acid value. Electron donating ability(EDA) and total phenolic contents of each extract were also determined. The result showed that the extracts possess an antioxidative activities. The effectiveness of them was in the following order: C. sinensis>C. tora>P. odoratum>S. chinensis >L. chinense. Ethanolic extracts of C. sinensis showed substantially higher EDA value and total phenol contents than other tea materials. These results indicate that the antioxidative effect was strongly related with EDA and total phenol contents.
