Effect of Dietary Fibers on Retinol Binding Protein (RBP, cRBP I, cRBP II) Gene Expression in Rats Fed High Fat Diet (고지방 식이내 식이섬유질이 흰쥐의 레티놀 결합 단백질 (RBP, cRBP I, cRBP II) 유전자 발현에 미치는 영향)

  • Recently it has been reported that vitamin A and retinol binding proteins (RBPs) in blood and urine were changed in the condition of diabetes mellitus or hyperlipidemia. Fruits and vegetables are recommended to consume for the people suffered from these chronic degenerative diseases. The main components of fruits and vegetables are dietary fibers, for example cellulose and pectin, of which function to affect the absorption and excretion of dietary fat and fat-soluble substances. This study was conducted to investigate the effect of dietary fibers on RBPs mRNA expression in liver, small intestine and serum of rat fed high fat diet during 4 weeks. Sprague-Dawley rats, weighing 121g on average, were divided into four groups; (Control; $17\%$ fat & cellulose supplement diet, HF0: $25\%$ fat & fiber free diet, B:.Uc: $25\%$ fat & cellulose supplement diet and HF0: $25\%$ fat & pectin supplement diet) . The rats fed high fat diet groups (HF0, HFC, HFP) tended to consume the food less than the control group, but FER of HF0 groups was significantly higher than the control (p < 0.05) . The weight of adrenal gland in high fat diet groups (HF0, HFC, HFP) was significantly less than the control. Total lipid in feces daily excreted and in liver did not show any significant differences among the groups. Total cholesterol in HFP group was significantly different from that of HFC group. Serum total cholesterol and triglyceride in other group tended to lower than other groups and HDL cholesterol higher. Consequently, AI (atherogenic index) was the lowest in HFP group. Vit A contents in feces daily excreted tended to lower in high fat diet groups (HF0, HFP) compared to the control group. That content in adrenal gland was the lowest in HF0 group, but not in liver. In HFP group were down-regulated cRBPI mRNA in liver and cRBPII mRNA in small intestine and up-regulated RBP and transthyretin expression in serum compared to the other groups. In conclusion, dietary fibers, especially pectin, in high fat diet might down-regulate the expression of CRBP I, CRBP II mRNA in liver and small intestine, but increase the secretion of RBP into serum and therefore inhance the bioavailability of Vit A through the body.   • The purpose of this study was to compare to the effects of regular exercise on body fat contents and plasma lipid composition of college women. This study was conducted from May to August in 2000. The subjects selected for the experiment were composed sixty three female college students. The basal anthropometric measurements were performed. The levels of total cholesterol, HDL-cholesterol, triglyceride and fatty acid composition of plasma were determined. And plasma glucose concentrations was also assayed. The results obtained are summerized as follows : Average height and weight of E(exercise) group were found to be slightly higher than NE(non exercise) group. The percent of body fat and body fat mass (kg) in E group was slightly lowered than that of NE group. There was decreased on the levels of plasma total cholesterol, triglyceride by regular exercise. HDL-cholesterol level was not significantly different between two groups, but LDL-cholesterol level in E group was significantly lowered than that of NE group. Therefore, plasma total cholesterol and LDL cholesterol levels was significantly decreased by regular exercise.

Effect of Onion Powder Supplementation on Lipid Metabolism in High Fat-cholesterol Fed SD Rats

  • This study was performed to examine the effects of onion powder supplementation on lipid metabolism in male SD rats fed a high fat and high cholesterol diet. Experimental groups were control (C), high fat fed group (HF), high fat+onion powder intake (OP), high fat+quercetin intake (Q). The HF diet contained 1% cholesterol, 4% lard, 0.3% Na-taurocholate, and quercetin supplementation level was 0.1 g/kg diet. The OP group showed lower body weight gains compared to the control, while there was no significant difference in food efficiency ratio efficiency. When the proportion of fecal bile acids per total lipids was calculated, there was a significant decrease in the HF group compared to the control group, while the levels of the control group was same as that of the OP group. There was no significant difference between the HF and Q groups in bile acid/total lipid in feces. The amount of total cholesterol in liver increased significantly in HF group compared to the control group, while total cholesterol decreased significantly in the OP group compared to the HF group. There was a significant decrease in GOT (glutamic oxaloacetic transaminase) activity in OP and Q groups compared to the HF group. In conclusion, feeding onion powder to hyperlipidemic rates appeared to control weight gain, significantly lower the level of total cholesterol in the liver, and recover GOT activity. We also demonstrated that onion powder intake was more effective than quercetin intake.

Effect of Acanthopanacis Cortex Boiling Extract Solutions on Fat Accumulation in the Obese Rats Induced by High Fat Dietary (오갈피의 열수추출액이 고지방식이에 의한 비만유도 흰 쥐의 지방 축적에 미치는 영향)

  • This study was carried out to evaluate the effect of Acanthopanacis Cortex boiling extract solutions on the fat accumulation in the obese rats induced by the oral high fat administration for six weeks. Total cholesterol, neutral fat and adipose acid of ACR groups were lower than the control group. During the feeding experiment, LDL and VLDL were increased while HDL was decreased in all groups. Insulin and cortisol were higher than the control group, due to the fat accumulation. Based on the above results, it was shown that it is possible to improve fat accumulation induced by high fat dietary through using the oral administration of Acanthopanacis Cortex boiling extract solutions.

Growth Associated Hormones Response and Fat Metabolism Change in Finishing Pigs Fed with n-Methyl-d, L-Aspartate

  • A trial was conducted to investigate the effect of dietary NMA on several growth associated hormones and fat metabolism in finishing pigs. A total of 84 crossbred finishing pigs (average initial BW of $56{\pm}$0.37kg) were divided into 6 pens, 14 pigs per pen (7 gilts and 7 barrows per pen). 3 pens of pigs were fed with control diet (corn-soybean meal) and the others were fed control diet addition with 50 mg/kg NMA. During the trial, all pigs were given free access to feed and water. After 44 days trial, 8 pigs from each treatment (4 gilts and 4 barrows, weight similar to average group weight, $86.94{\pm}0.71kg$ for control group, and $90.55{\pm}1.51kg$ for NMA treated group) were sacrificed to collect the sample of the liver, longissimus muscle, subcutaneous fat (10th rib). The addition of NMA in diet increased the IGF-I, Insulin, T3, T4 levels in serum by 50.68% (p<0.05), 38.36% (p<0.05), 123.33% (p<0.01), 60.58% (p<0.03), respectively. Meanwhile, IGF-I level in the liver and the muscle were increased with 17.83% (p<0.03) and 26.00% (p<0.03) with addition of NMA. The data from subcutaneous fat (10th rib) analysis showed that supplement of 50 mg/kg NMA decreased the total activities of malic dehydrogenase (MDH) by 20.54% (p<0.05), glucose-6- phosphate dehydrogenase (G-6-DPH) by 16.97% (p<0.05), and decreased the specific activities of MDH and G-6-DPH by 37.46% (p<0.01) and 35.06% (p<0.01), respectively. The hormone sensitive lipase (HSL) total activity was increased by 25.00% (p<0.05) in NMA treated pigs. These results indicated that addition of 50 mg/kg NMA to diet can induce the endocrine great change in finishing pigs, furthermore, inhibit the fat synthesis through suppressing lipogenic enzymes and promote the fat degradation by elevating HSL activity in finishing pigs.

Effects of Wax Guard on Weight, Triglyceride, Leptin and Fat Cell Size in Rats Fed on a High Fat Diet (고지방식이를 섭취시킨 흰쥐에서 동과가 체중, 중성지방, Leptin과 지방세포의 크기에 미치는 영향)

  • The purpose of this study was to investigate the effects of wax gourd on weight, triglyceride, leptin and fat cell size in rats fed a high-fat diet. Male Sprague Dawley rats were fed an experimental diet containing total dietary fat at 40% of calories with wax gourd 0%, 5%, 10% and 15% (w/w) for 4 weeks. Weight gain and triglyceride level fell significantly in the 15% wax gourd group compared to the control group. Epididymal fat pad, abdominal fat and perirenal fat tended to decrease in the 15% wax gourd group. Leptin and free fatty acid level were not significantly different among the groups. Fat cell size significantly decreased in the 10% and 15% wax gourd groups compared to the control group. Weight gain correlated positively with visceral fat masses and the levels of leptin and triglyceride. Fat cell size significantly correlated with visceral fat and leptin level. Therefore, the 15% wax gourd diet substantially reduced weight, triglyceride and fat cell size. (Korean J Nutrition 36(5): 446∼451, 2003)

Effect of Gastrodiae rhizoma Powder on Serum and Liver Lipid Levels of Rats with High Fat Diet (고지방 식이와 병행 섭취한 천마 분말이 흰쥐의 혈청 및 간 조직 지질 함량에 미치는 영향)

  • This study was performed to study the effects of Gastrodiae rhizoma on lipid concentration of serum and liver in rats fed high fat diet. Male Sprague-Dawley rat were randomly assigned to 5 groups such as basal diet only(BDG), high fat diet(HFG) and high fat diet with 5% processed Gastrodiae rhizoma(GRL), high fat diet with 10% processed Gastrodiae rhizoma(GRM), high fat diet with 15% processed Gastrodiae rhizoma(GRH) supply each of the experimental diets for 6 weeks. Food intake was significantly decreased by both high fat intake and high fat with Gastrodiae rhizoma intake compared with basal diet intake but weight gain was not significantly different from each groups. Liver weights were significantly lowered by Gastrodiae rhizoma intake compared with high fat intake. In the serum, total cholesterol was not significantly different with each groups, but triglyceride level was significantly lowered by Gastrodiae rhizoma intake as compared with control. In the liver, lipid levels inceased with the Gastrodiae rhizoma intake, but total cholesterol and triglyceride levels were not significantly different with control. These results showed that dietary Gastrodiae rhizoma as supplementary food may decrease triglyceride in the serum.

Inhibitory Effects of Anthocyanin-rich Fraction from Purple Sweet Potato on High Fat Diet-induced Insulin Resistance and Hepatic Steatosis (자색고구마로부터 분리한 안토시아닌 분획물의 고지방식이로 유도된 인슐린 저항성과 간 지질 축적 개선 효과)

  • Anthocyanins, a class of flavonoids, are natural water-soluble pigments, mainly found in vegetables and fruits. Anthocyanins have attractive pharmacological activities, such as anti-oxidant, anti-inflammatory, anti-cancer, and anti-diabetic effects. The purpose of this study was to investigate the inhibitory effects of the anthocyanin-rich fraction (ANF) from purple sweet potato on high fat diet-induced insulin resistance and hepatic steatosis. C57BL/6J mice were assigned to the following groups (n=8 per group): normal fat diet (NF); high fat diet (HF); high fat diet with ANF 50mg/kg (ANF50). Normal fat or high fat diets were fed for a total of 17 weeks, and ANF was orally administrated for 8 weeks (from 10 to 17 weeks, five times/week). In our results, there were no significant differences in body weight, food intake, and tissue weight upon ANF supplementation. The levels of serum triacylglycerol, total-cholesterol, and glucose were also not affected by ANF supplementation. However, ANF supplementation significantly decreased serum insulin and HOMA-IR levels as well as prevented hepatic fat accumulation in high fat-fed mice. These results show that ANF may be beneficial for improving high fat-induced insulin resistance and protecting against development of hepatic steatosis.

Effects of Chegameyiin-tang extract on the change of the weight, tissue in epididymal fat, blood, leptin and uncoupled protein in visceral fat of obesity rats induced by high fat diet (체감의이인탕(體減薏苡仁湯)이 비만유도 흰쥐의 체중, 지방조직, 혈액변화, leptin과 Uncoupled protein에 미치는 영향)

  • In Oriental medicine, there has been a theory that the deficiency of the Qi(氣) and the Phlegm(濕)-Damp(痰) bring Obesity. And so a clinically representative herb-medicine of the obesity treatments is Chegameyiin-tang . We observed the effects of Chegameyiin-tang on the fat tissues and what the function of Chegameyiin-tang is. These experimental studies were designed to investigate the effects of Chegameyiin-tang on the weight and lipid metabolism of obesity rats induced by high fat diet. And what is changed in the blood and how the leptin and uncoupled protein is affected. The measurement has been performed on (1) the weight of obese rats fed high fat diet, (2) the average size and number of epididymal fat cells, (3) the total cholesterol, triglyceride, glucose. and free fatty acids in the blood. and (4) the leptin and uncoupled protein in the blood are observed. The results are as follows; 1. In the sample group, the weight decrease occured significantly throughout the whole research period than that of control group. 2. In the sample group, epididymal fat weight showed significantly decrease in the 8th and 14th weeks than that of control group.3. In the sample group, epididymal fat cell size was decreased significantly in the 8th and 14th weeks than that of control group. 4. In the sample group, total cholesterol. triglyceride and glucose increased rather than control group in 8 weeks, those decreased significantly in 14 weeks. 5. In the sample group, free fatty acids and insulin increased rather than control group in 8 weeks. those showed some decrease in 14 weeks . 6. In the sample group, leptin decreased significantly than control group in 8, 14 weeks. Uncoupled protein showed some decrease in 8 weeks. that decreased significantly in 14 weeks.

Effect of Chromium Methionine Supplementation on Obesity Index and Serum Lipids in Rats (Chromium Methionine첨가가 비만지수 및 지질 대사에 미치는 영향)

  • Chromium (Cr) plays an important role in body composition. Several human and animal studies of Cr supplementation have reported increases in lean body mass, often with concurrent decreases in body fat. The purpose of the current study was to determine the effects of supplemental Cr as chromium methionine (CrMet) on obesity index, body fat and serum lipids in rats. Thirty-six male Sprague-Dawley rats were divided into four groups of nine rats. Rats received a AIN-76 semipetrified basal diet or high fat diet supplemented with 300 ppb CrCl$_3$and CrMet as well as no added dietary Cr (referred to as Control, HF, HFCr, and HFCrMet). After 6 weeks on the respective diets, animals were killed and analyzed for differences in serum lipid concentrations. Rats supplemented with CrMet had a slightly higher naso-anal length and final body weight was decreased. The lowest obesity index and body fat content were found in CrMet supplemented animals (p〈0,05). Supplemental CrMet decreased the total lipid, triglyceride and total cholesterol. The significant differences observed were total cholesterol concentration for CrMet supplementation. These indicate that supplemental Cr, as chromium methionine, resulted in a significant decrease of obesity index and body fat and influence serum lipid concentrations, possibly by decreasing the body fat.
