가염 및 분해기간에 따른 멸치의 가수분해 (Hydrolysis of Anchovy (Engraulis Japonicus) Homogenate with Salting and Digestion Time)

  • 멸치를 마쇄하여 자가소화시킬 경우 무염상태에서 20시간까지는 $50^{\circ}C$, pH 4라 최적조건이었고, 식염을 20% 첨가하였을 때는 $50^{\circ}C$, pH 6에서 가수분해가 가장 잘행해졌다. 한편 식염을 첨가하여 8 일동안 가수분해시킬 경우 최적조건이 $40^{\circ}C$, pH 6으로 변화하는 것으로 밝혀졌다. 상기한 최적조건에서 20시간동안 자가소화 시켰을 때 무염상태에서는 총질소중 60.5%가 TCA-가용성 질소를 가수분해되었으나 식염을 20% 첨가하였을 때는 49.8%로 낮아졌다. 그리고 가염상태에서 8 일간 가수분해 시킨 결과 총질소중 83.1%가 TCA-가용성질소로 분해되었고 계속하여 실온에서 52일간 숙성시킨 결과 총질소중 89.6%가 분해되었다.

파일로트 규모 음식쓰레기 2상 혐기소화 처리공정에 관한 연구 (Pilot Scale Anaerobic Digestion of Korean Food Waste)

  • A 5 ton/day pilot scale two-phase anaerobic digester was constructed and tasted to treat Korean food wastes in Anyang city. The process was developed based on 3 years of lab-scale experimental results on am optimim treatment method for the recovery of biogas and humus. Problems related to food waste are ever Increasing quantity among municipal solid wastes(MSW) and high moisture and salt contents. Thus our food waste produces large amounts of leachate and bed odor in landfill sites which are being exhausted. The easily degradable presorted food waste was efficiently treated in the two-phase anaerobic digestion process. The waste contained in plastic bags was shredded and then screened for the removal of inert material such as fabrics and plastics, and subsequently put into the two-stage reactors. Heavy and light inerts such as bones, shells, spoons and plastic pieces were again removed by gravity differences. The residual organic component was effectively hydrolyzed and acidified in the first reactor with 5 days space time at pH of about 6.5. The second, methanization reactor part of which is filled with anaerobic fillters, converted the acids into methane with pH between 7.4 to 7.8. The space time for the second reactor was 15 days. The effluent from the second reactor was recycled to the first reactor to provide alkalinities. The process showed stable steady state operation with the maximum organic rate of 7.9 $kgVS/m^3day$ and the volatile solid reduction efficiency of about 70%. The total of 3.6 tons presorted MSW containing 2.9 tons of food organic was treated to produce about $230m^3$ of biogas with 70% of methane and 80kg humus. This process is extended to full scale treating 15 tons of food waste a day in Euiwang city and the produced biogas is utilized for the heating/cooling of adjacent buildings.

보리의 효소적(酵素的) 제분(製粉) 및 이용(利用)에 관한 연구 (Enzymatic Milling Process for Barley Flour Preparation)

  • 부분도정한 보리를 미생물효소(Asp. niger 및 oryzae)에 의한 분해반응 및 미세 고형분의 용출, 여과, 침강 또는 원심분리, 건조 등의 과정을 거쳐 제분할 때, 보리가루 획분$(73{\sim}76%)$, 맥강, 배아, 종구등의 잔유물 획분(3.6% 내외) 그리고 가용성물질 획분 등으로 분리하여 얻을 수 있었다. 이 때 가장 좋은 효소반응 조건은 온도 $45^{\circ}C$, 보리와 물의 비율 1 : 1.5(w/v)였고, ${\alpha}-a-mylase$역가 보다는 glucanase 와 Protease의 역가가 높은 효소를 사용할 때 더 좋은 효과를 보였다. 그리고 얻어진 세가지 종류 획분의 성분 조성은 단백질, 지방, 섬유소 등에서 각각 현저한 차이를 보여, 각 획분의 특성별 구분 이용이 가능하였다.

Nutrient digestibility in black soldier fly larva was greater than in adults for pigs and could be estimated using fiber

  • The objectives of the present study were to determine the nutrient digestibility of fish meal, defatted black soldier fly larvae (BSFL), and adult flies and to develop equations for estimating in vitro nutrient digestibility of BSFL for pigs. In vitro digestion procedures were employed to mimic the digestion and absorption of nutrients in the pig intestine. Correlation coefficients between chemical composition and in vitro nutrient digestibility of BSFL were calculated. In Exp. 1, in vitro ileal digestibility (IVID) of dry matter (DM) and crude protein (CP) and in vitro total tract digestibility (IVTTD) of DM and organic matter in defatted BSFL meal were less (p < 0.05) than those in fish meal but were greater (p < 0.05) than those in adult flies. In Exp. 2, CP concentrations in BSFL were negatively correlated with ether extract (r = -0.91) concentration but positively correlated with acid detergent fiber (ADF; r = 0.98) and chitin (r = 0.95) concentrations. ADF and chitin concentrations in BSFL were negatively correlated with IVID of DM (r = -0.98 and -0.88) and IVTTD of DM (r = -1.00 and -0.94) and organic matter (r = -0.99 and -0.98). Prediction equations for in vitro nutrient digestibility of BSFL were developed: IVID of CP (%) = -0.95 × ADF (% DM) + 95 (r2 = 0.75 and p = 0.058) and IVTTD of DM (%) = -2.09 × ADF + 113 (r2 = 0.99 and p < 0.001). The present in vitro experiments suggest that defatted BSFL meal was less digestible than fish meal but was more digestible than adult flies, and nutrient digestibility of BSFL can be predicted using ADF as an independent variable.

스트레스와 소화기능을 활용한 체질별 맞춤 수면관리 (Individualized Sleep Management for Each Sasang Type Using Stress and Digestive Function)

  • Objectives This study aimed to analyze the influence of various sleep-related factors that affect sleep quality by each Sasang type. Methods A total of 400 subjects were included for this study, 108 males and 292 females. Sasang type was diagnosed using the SCAT. Then, the characteristics of each Sasang type were analyzed using HRV, DITI, and PSQI, PSS, and SDFI questionnaires. Logistic regression analysis was used to predict sleep-related factors that affect sleep disorders by Sasang types. Results This study shows that the pathophysiological characteristics for stress and digestive function of each Sasang type can differentiate sleep management through a logistic regression model including subscales of PSS and SDFI. Stress had no effect on the occurrence of sleep disturbance within only So-Eum, since the stress level is originally high in the So-Eum regardless of sleep quality. Rather, decreased appetite and poor eating habits had a significant impact on the decline in sleep quality. In addition, poor digestion and eating habits in So-Yang had a greater impact and poor digestion in Tae-Eum had a greater impact on the decline in sleep quality. Conclusion The stress and subscales of digestive function provide differentiated sleep management in So-Yang, Tae-Eum, and So-Eum types. The individualized sleep management for each Sasang type with statistically validated PSS and SDFI would be useful for sleep-related experts planning safe and effective person-centered health care as well as for Western clinicians who want to incorporate Sasang typology into their treatments as integrative medical technique in the future.

Effects of Exogenous Enzymes on Ruminal Fermentation and Degradability of Alfalfa Hay and Rice Straw

  • This study was conducted to evaluate the use of exogenous enzymes as a potential means of improving the ruminal digestion (i.e., degradability) of alfalfa hay and rice straw. Twenty six enzyme-additives were examined in terms of protein concentration and enzymic activities on model substrates. The exogenous enzymes contained ranges of endoglucanase, xylanase, ${\beta}$-glucanase, ${\alpha}$-amylase, and protease activities. Six of the enzyme additives were chosen for further investigation. The enzyme additives and a control without enzyme were applied to mature quality alfalfa hay substrate and subsequently incubated in rumen batch cultures. Five of the enzyme additives (CE2, CE13, CE14, CE19, and CE24) increased total gas production (GP) at 48 h of incubation compared to the control (p<0.05). The two additives (CE14 and CE24) having the greatest positive effects on alfalfa hay dry matter, neutral detergent fibre (NDF) and acid detergent fibre (ADF) degradability were further characterized for their ability to enhance degradation of low quality forages. The treatments CE14, CE24, a 50:50 combination of CE14 and CE24 (CE14+24), and control (no enzyme) were applied to mature alfalfa hay and rice straw. For alfalfa hay, application of the two enzyme additives, alone and in combination, increased GP compared to the control at 48 h fermentation (p<0.05), whereas only CE14 and CE14+24 treatments improved GP from rice straw (p<0.05). Rumen fluid volatile fatty acid concentrations throughout the incubation of rice straw were analyzed. Acetate concentration was slightly lower (p<0.05) for CE14${\times}$CE24 compared to the control, although individually, CE14 and CE24 acetate concentrations were not different from the control. Increases (p<0.05) in alfalfa hay NDF degradability measured at 12 and 48 h of incubation occurred only for CE14 (at 12 h) and for CE14+24 (at 12 and 48 h). Similarly, ADF degradability increased (p<0.05) with CE14 and CE14+24. As for rice straw, increased DM degradability was observed at 12 and 48 h of incubation for all enzyme treatments with an exception for CE14 at 12 h. The degradability of NDF was improved by all the enzyme treatments at either incubation time, while ADF degradability was only enhanced at 48 h. Overall, the enzymes led to enhanced digestion of mature alfalfa and there was evidence of improved digestibility of rice straw, an even lower quality forage.

쌀의 KOH 농도별 붕괴양상에 따른 품종변이 해석 (Analysis of Varietal Variation in Alkali Digestion of Milled Rice at Several Levels of Alkali Concentration)

  • 우리나라 주요 벼 재배품종 및 신육성계통(자포니카: 25품종, 통일형:8품종)에 대한 쌀 알칼리 붕괴성의 품종적 특이성을 분시검토하여 보고자 KOH 농도를 0.8-1.8까지 0.2% 간격으로 처리하여 농도별 쌀의 알칼리 붕괴반응을 조사하고 주성분분석법을 적용하여 품종적 유형을 분류하였다. 주성분분석에서 상위 2개 주성분치의 변이가 전변이량의 92%을 차지하였고 제1 및 제2 주요분치 좌표상의 품종분포에 따라 크게 4개 유형으로 군집화 시킬 수 있었다. 제 I 군에서는 전 KOH 농도에 걸쳐 거의 비슷하게 알칼리 붕괴성(ADV)가 낮은 도봉벼만 유일하게 분포하였고, 제 II군에는 1.4% KOH 농도에서 중간정도의 ADV 이면서 고-저 알칼리 농도 ADV간차가 중도인 백운찰벼, 신선찰벼 및 수원 34002 등 찰벼가 포함되어 있었다. 제 III군에는 1.4% KOH 농도에서 중-중고의 ADV 이면서 저-고 KOH 농도간 ADV 변화가 심했던 대부분의 통일형 품종과 자포니카 조생계가 분포되어 있었고 육도농림찰 001와 한강 찰벼등은 이 유형에 포함되어 있었다. 제 IV군에는 1.4% KOH 농도에서 ADV가 중고이상으로 높으면서 고-저 알칼리 농도 ADV간차가 중-중고인 중생 및 중만생 자포니카 품종들이 주로 분포 하였으며 조생인 금조벼가 이 유형에 속하였다. 여기서 제 1 주성분은 전 알칼리 농도에서 평균적으로 표현되는 붕괴성 정도였고 제 2 주성분은 저-고 KOH 농도의 ADV간 차 또는 KOH 농도에 따른 ADV 변화의 회귀계수와 밀접하게 관련된 요소였다. 1.2%-1.4% KOH 농도에서의 ADV는 저-고 KOH 농도간 ADV차와 2차 곡선 회귀계수를 나타내었다.

돈분 슬러리 성상에 따른 최적 바이오가스 회수 (Optimum Recovery of Biogas from Pig Slurry with Different Compositions)

  • 돈분 슬러리를 이용한 혐기성 소화과정에서 유기물 농도 및 식종슬러지의 식종비율, 소화조의 혼합강도 등의 최적 실험조건을 도출하고, 돼지의 성장에 따른 사료 급여 특성과 분뇨의 성상이 바이오가스 발생에 미치는 영향을 관찰하였다. 식종슬러지 비율 50%와 TS 농도 1% 수준에서 메탄함량은 45%이었고, 유기물 함량이 3~7 %로 증가할수록 메탄가스 함량도 증가되었다. 반응조의 혼합강도에 따른 총 누적가스 발생량은 식종슬러지의 식종비율에 따른 영향은 관찰되지 않았고, TS 농도 3%와 5%에서 혼합강도를 증가시켰을 때(80 $\rightarrow$ 160 rpm) 바이오가스 발생량도 증가되었다. 돈분폐수의 혐기소화 실험시 바이오가스를 회수하고자 하는 최적의 운전 조건은 투입되는 TS 농도 3~5% 정도의 유기물 농도와 50% 수준의 식종슬러지의 식종비율 그리고 반응조의 적절한 교반강도(120 rpm) 따라 결정될 수 있다. 돈분뇨 종류에 따른 가스발생량은 분만돈 분뇨의 바이오 가스 발생량이 높았고, gas 발생의 peak(20일)도 짧게 나타나 분해율이 가장 좋은 것으로 나타났다. 돼지의 사육 및 소비 형태에 따라 돼지의 사육 환경이 변화되며, 투입되는 사료의 급여 특성도 달라져 바이오가스 발생 특성도 차이가 있는 것으로 나타났다.

혐기성 소화 효율 향상을 위한 초음파 주파수의 순차적 적용에 대한 연구 (A Study on Sequential-Frequency Sonication for Improving Anaerobic Digestion)

  • The effect of ultrasound pretreatment on sludge degragability was investigated at different condotions: ultrasonic frequency(28, 40, 50kHz), intensity(10, 25, 50W), sonication time(10, 20, 30, 60min). Total suspended solid(TSS) and soluble chemical oxygen demand(SCOD) concentration were measured for the evaluation of pretreatment efficiency. The pretreated sludge was used as the feedstock for anaerobic digestion process. Biogas production and volatile suspended solid(VSS) removal were determined for evaluating the process performance. 1. TSS concentration of the sludge decreased at a constant rate as sonication operation was applied. The degradation rate of TSS increased when ultrasound frequency was decreased from 50kHz to 28kHz and intensity was increased from 10W to 50W. Efficiency of TSS degradation per input energy increased as ultrasonic frequency and intensity were decreased. At the frequency of 28 and 40kHz, SCOD concentration rapidly increased during the initial 30min of sonication time, and then it gradually increased. At 50kHz, SCOD concentration constantly increased for 60min of the sonication time. The SCOD production rate increased with increasing intensity under all ultrasound frequencies. 2. The optimum condition of ultrasound treatment was 28kHz, 50W and 60min for maximizing the biogas production, methane fraction, VSS removal. The highest values in biogas production, methane fraction in biogas, VSS removal were 370ml, 70%, 2.45g, respectively. Methane production rate per input energy increased at ultrasonic frequency and intensity decreased. 3. When raw sludge was pretreated at the condition of ultrasonic frequency of 28 and 40kHz in series, sequential-frequency sonication, intensity of 50W and 60min, biogas production, methane fraction, VSS removal were about the same that of 28kHz single-frequency sonication. When sequential-frequency sonication of 28 and 50kHz was applied in series, biogas production, methane fraction, VSS removal were 356 ~ 423ml, 69 ~ 71%, 2.41 ~ 2.78g, respectively. The pretreatment efficiency of 28-50kHz sequential-frequency sonication which sonication time of 28kHz and 50kHz was 40min and 20min was higher than that of 28kHz single-frequency sonication.

Replacing Concentrate with Wheat Straw Treated with Urea Molasses and Ensiled with Manure: Effects on Ruminal Characteristics, In situ Digestion Kinetics and Nitrogen Metabolism of Nili-Ravi Buffalo Bulls

  • To evaluate the effects of replacing concentrate with urea molasses treated fermented wheat straw (FWS) ensiled with cattle manure (CM) on ruminal characteristics, in situ digestion kinetics and nitrogen (N) metabolism was studied in Nili Ravi cannulated buffalo bulls in a $4{\times}4$ Latin Square Design. Wheat straw treated with urea (4%) and molasses (6%) was ensiled with cattle manure (CM) (70:30) and fermented for 40 days. Four iso-nitrogenous and iso-caloric diets were formulated. In the FWS0, FWS10, FWS20 and FWS30 diets 0, 10, 20 and 30% of the concentrate was replaced with FWS, respectively. Daily intake by bulls was restricted to 1.5% dry matter (DM) of body weight. Ruminal ammonia nitrogen concentration was greater (p<0.05) in bulls fed FWS diet than for those fed FWS0 diet at 3, 6, 9 and 12 h post-parandial. Bulls fed FWS 20 and FWS 30 diets had higher ruminal pH at 3 and 6 h post-parandial than bulls fed FWS10 and FWS0. Ruminal total volatile fatty acid (VFA) concentrations 3 h post-parandial were greater (p<0.05) in bulls fed FWS0 than those fed FWS diets. However ruminal VFA tended to increase at 6, 9 and 12 h post-parandial as the level of FWS increased. In situ ruminal DM and neutral detergent fiber (NDF) degradation, rates of disappearance and extent of digestion were higher (p<0.05) for bulls fed FWS30 diet than those fed FWS0. Ruminal DM and NDF lag time tended to decrease (p<0.05) as FWS concentration in the diet increased. Feed intake, nitrogen intake, N-balance and blood urea-N did not differ (p>0.05) in buffalo bulls fed different diets. Wheat straw treated with urea and molasses and ensiled with CM enhanced the nutritive value of wheat straw and improved nutrient utilization in buffalo bulls when up to 30% of the concentrate was replaced with FWS; no adverse effects on ruminal characteristics and nutrients digestibilities were detected.
