Influence of different levels of corn steep liquor (CSL) on chemical composition of urea treated wheat straw (UTWS), ruminal characteristics, digestion kinetics, nitrogen (N) utilization, and nutrient digestibility by ruminally-cannulated buffalo bulls was studied in a $4{\times}4$ Latin Square Design. The CSL was used to ensile 5% UTWS at the rate of 0, 3, 6, and 9% on a dry matter (DM) basis. Total N and neutral detergent fibre (NDF) were increased with increasing level of CSL. Increased NDF content was attributable to increased neutral detergent insoluble nitrogen. Four diets were formulated to contain 20% concentrate and 80% UTWS ensiled with 0, (control), 3 (CSL3), 6 (CSL6), and 9% CSL (CSL9). All diets were mixed daily and fed at 1.5% of body weight twice daily. Ruminal NH3 concentration decreased with level of CSL used to ensile UTWS at 3 and 6 h post prandial, however, at 9 h post parandial it was similar across all diets and at 12 h post prandial was higher with diets containing UTWS ensiled with CSL. Concentrations of total ruminal volatile fatty acid and acetate were increased with the CSL level used to ensile UTWS. Increased rate of disappearance and reduction in lag time of DM and NDF was recorded with diets containing UTWS ensiled with CSL. Dry matter and NDF digestibilities were higher with CSL diets than on the control diet. Buffalo bulls retained more N with diets containing UTWS ensiled with CSL. The present results indicated that UTWS could be ensiled with CSL to improve its nutritive value and N utilization by ruminants.
Knowledge on the functional characteristics and temporal variation of anaerobic bacterial populations is important for better understanding of the microbial process of two-stage anaerobic reactors. However, owing to the high diversity of anaerobic bacteria, close attention should be prioritized to the frequently abundant bacteria that were defined as core bacteria and putatively functionally important. In this study, using MiSeq sequencing technology, the core bacterial community of 98 operational taxonomic units (OTUs) was determined in a two-stage upflow blanket filter reactor treating pharmaceutical wastewater. The core bacterial community accounted for 61.66% of the total sequences and accurately predicted the sample location in the principal coordinates analysis scatter plot as the total bacterial OTUs did. The core bacterial community in the first-stage (FS) and second-stage (SS) reactors were generally distinct, in that the FS core bacterial community was indicated to be more related to a higher-level fermentation process, and the SS core bacterial community contained more microbes in syntrophic cooperation with methanogens. Moreover, the different responses of the FS and SS core bacterial communities to the temperature shock and influent disturbance caused by solid contamination were fully investigated. Co-occurring analysis at the Order level implied that Bacteroidales, Selenomonadales, Anaerolineales, Syneristales, and Thermotogales might play key roles in anaerobic digestion due to their high abundance and tight correlation with other microbes. These findings advance our knowledge about the core bacterial community and its temporal variability for future comparative research and improvement of the two-stage anaerobic system operation.
This experiment was conducted to find out the difference between roughage productivity and feed value by using two domestic forage rice cultivars in a mid-west reclaimed rice field in Korea. During the growing period, Na+ content in soil showed no significant difference between the two cultivars; however, the K+ content in the soil was significantly lower than that in which the Mogwo was grown. The tiller number was 45.5 to 58.8, which was higher than that of Mogyeong, and the leaf number was higher than that of Mogyeong. The dry matter yield per 10 a was 1,338 kg for Moggyang and 1,532 kg for Mogwoo. At the harvest time, the crude protein yield per 10 a produced 108 kg of Mogyang and 170 kg of Mogwoo, and the total digestion nutrient (TDN) yield per 10 a also produced about 617 kg of Mogyang and 880 kg of Mogwoo. To sum up the above results, Mogwo, which has many tiller numbers and leaf numbers, has a high dry matter production, a relatively low neutral detergent fiber (NDF) and acidic detergent fiber (ADF), and a high crude protein yield and TDN yield, and it was better when cultivating the forage rice cultivars in reclaimed land.
Two in vivo experiments were conducted to evaluate the effect of novel urease inhibitor hydroquinone (HQ) on ammonia release rate from urea hydrolysis, nitrogen balance, nutrient digestibility and efficiency of microbial protein synthesis. In Exp. 1, twelve crossbred cannulated lambs were randomly assigned within initial body weight block to one of four HQ treatments, which included 0 (control), 30, 60 or 80 mg HQ/kg DM intake. Ammonia concentration and pH of ruminal fluid were immediately measured at 0, 2, 4, 6 and 8 h after feeding. Increasing the dose of HQ tended (p<0.15) to linearly decrease NH3 formation. The ammonia peak concentration (2 h post-feeding) in animals receiving HQ was approximately one-half of that in animals not receiving HQ (p<0.01), and a relatively sustained ammonia release could be obtained at the dose of 30 or 60 mg HQ/kg DM. In Exp. 2, sixteen intact crossbred lambs (weight $40{\pm}0.8kg$) were used in a $2{\times}2$ factorial design experiment. The four rations consisting of soybean meal-based (SBM) or urea-based (Urea) nitrogen source with or without HQ (S1, S0, U1 and U0) were fed in digestion and N balance trials. Apparent digestibility of major nutrients except that of ADF was not affected by either nitrogen source or addition of HQ. Regardless of nitrogen source, supplementation of HQ significantly improved ADF digestibility (p<0.05). The various ration had no effects on N metabolism in the presence of HQ. There was significant difference between total purine derivatives (PD), estimated efficiency of microbial N synthesis (p<0.05) and urea-N excretion (p<0.01) in the urine for the SBM ration and for the Urea ration. However, HQ had little influence on efficiency of microbial N synthesis as proportion of daily intake of total tract digestible OM (p>0.05). No interactions between main nitrogen source and HQ were measured throughout the trial. Results of this study suggest that addition of HQ to ration may improve ADF digestion with having no negative effect on N metabolism and microbial protein production.
This paper describes a micro total analysis system ($\mu$ TAS) for detecting and digesting the target protein which includes a bead based temperature controllable microchip and computer based controllers for temperature and valve actuation. We firstly combined the temperature control function with a bead based microchip and realized the on-chip sequential reactions using two kinds of beads. The PEG-grafted bead, on which RNA aptamer was immobilized, was used for capturing and releasing the target protein. The target protein can be chosen by the type of RNA aptamer. In this paper, we used the RNA aptamer of HCV replicase. The trypsin coated bead was used for digesting the released protein prior to the matrix assisted laser desorption ionization time of flight mass spectrometer (MALDI TOF MS). Heat is applied for release of the captured protein binding on the bead, thermal denaturation and trypsin digestion. PDMS microchannel and PDMS micro pneumatic valves were also combined for the small volume liquid handling. The entire procedures for the detection and the digestion of the target protein were successfully carried out on a microchip without any other chemical treatment or off-chip handling using $20\;{\mu}l$ protein mixture within 20 min. We could acquire six matched peaks (7% sequence coverage) of HCV replicase.
Four Korean native steers ($511{\pm}17.2kg$; $2{\times}2$ replicated crossover design) fitted with duodenal cannulas were used to investigate the influence of oral administration of soluble whey protein (WP; 82.29% crude protein) on ruminal fermentation, gastrointestinal (GI) hormone secretion in the blood, pancreatic ${\alpha}$-amylase activity in the duodenum, and disappearance rate in each segment of the GI tract. Steers were orally fed the basal diet (control; TMR [total mixed ration] 9 kg/d) or the basal diet with enriched WP (400 g/d) for 14 days. The apparent crude protein disappearance rate in the rumen of the WP was higher than in control (p < 0.05). However, no difference between groups was observed in the apparent crude protein disappearance rate in the intestine and the apparent starch disappearance rates in the rumen, GI tract. The level of cholecystokinin, secretin, and ghrelin in serum and pancreatic ${\alpha}$-amylase activity in the duodenum of the WP also did not change. The changes in the level of blood urea nitrogen related to protein metabolism were higher in the WP than in the control (p < 0.05). However, the levels of total protein, lipid, carbohydrate and mineral metabolites did not change. Consequently, we suggest that the oral administration of WP in steers assisted in ruminal fermentation due to the population increase of microbes in the rumen but did not improve the starch digestion rate in the small intestine because GI hormone secretion in the blood and pancreatic ${\alpha}$-amylase activity did not change.
Experiments for highly concentrated contaminants in pig waste slurry were carried out for the feasibility test of a pilot-scale innovative process scheme of engaging autothermal thermophilic aerobic digestion (ATAD) and expended granular sludge bed (EGSB) followed by sequencing batch reactor (SBR) system. Contaminants in pig waste slurry such as organic substance, total nitrogen (TN), ammonia nitrogen and total phosphorus (TP) contents were successfully reduced in the system. Total volatile solids (TVS) and chemical oxygen demands (COD) for organic matter in the feed were 32.92 g/L and 42.55 g/L respectively, and they were reduced by about 98.7% and 99.2%, respectively in the system. The overall removal efficiencies for TN and ammonium nitrogen were found to be 98.1 and 98.5%, respectively. The overall removal efficiency for total phosphorus was also found to be 92.5%. Faecal coliform density was reduced to <$1.2{\times}10^4CFU/g$ total solids. Biogas and $CH_4$ were produced in the range of 0.39-0.85 and $0.25-0.62m^3/kg$ [VS removed], respectively. The biogas produced in the system comprised of $295{\pm}26ppm$ (v/v) [$H_2S$].
본 연구에서는 국내 엽산 분석 데이터의 질적 양적 향상을 위해 첫째, AOAC법의 trienzyme 추출법과 식품공전의 암모니아 가수분해법을 비교 분석하고 둘째, 엽채류에 있어 trienzyme 추출법의 검증을 위해 국제표준인증물질을 분석하고 정밀성과 정확성을 측정하였다. 마지막으로 검증된 추출법을 적용하여 시판 봄나물(생것, 데친것)의 총 엽산 합량을 L. casei를 이용한 미생물학적 정량법을 적용하여 비교 분석하였다. 연구 결과 CRM 485(mixed vegetable), CRM 487(pig liver), CRM 121(wholemeal flour), 시금치, 돌미나리의 경우 암모니아 추출법보다 trienzyme 추출법을 적용할 경우 엽산 함량이 시료에 따라 1.2에서 5.8배까지 증가하였다. 따라서 나물 등 엽채류 식품에 함유된 천연 엽산 분석에는 trienzyme 추출법 적용이 필요한 것으로 생각된다. Trienzyme 추출법을 적용하여 분석한 봄나물(생것, 데친 것)의 엽산 함량은 습물 기준(wet weight basis) 나물 생시료의 경우 오가피($146.9{\mu}g/100g$), 부지갱이($142.8{\mu}g/100g$), 갯기름($140.4{\mu}g/100g$)의 엽산 함량이 15개의 분석시료 중 높은 값을 나타내었다. 반면 건물 기준(dry weight basis)을 적용한 절대량 비교에서는 민들레($1,643.0{\mu}g/g$), 시금치($1,284.8{\mu}g/g$), 비름나물($1,222.5{\mu}g/g$) 순으로 높은 엽산 함량을 나타내었다. 가죽나물, 참두릅, 세발나물, 돌미나리, 참취나물 경우 $100^{\circ}C$에서 30초간 데쳤을 때 엽산 함량은 차이가 없었으며(P>0.05) 갯기름나물을 제외한 나머지 9개 시료에서는 엽산 함량이 11~63% 감소하였다(건물 기준 비교). 본 연구에 적용된 엽산 trienzyme 추출법은 상대적으로 부족한 국내 다소비 식품의 엽산 분석 연구에 활용될 것으로 생각되며 나아가 국가표준식품성분표 작성을 위한 엽산 데이터베이스 구축의 기초 자료가 될 것이다.
본 연구는 음식물쓰레기의 2상 혐기성 소화에서 메탄가스 발생량을 높이기 위해 고온 가용화 전처리시 pH를 $7.00{\pm}0.50$으로 조정하여 대조군과 비교하였다. pH에 따른 가용화 효율 회분식(Batch) 실험에서 pH가 $4.20{\pm}0.40$ (pH를 조절하지 않은 것)에서부터 $7.00{\pm}0.50$, $12.00{\pm}0.50$으로 증가시킴에 따라 가용화 효율도 각각 26.2, 47.1, 55.6%로 높게 나타났다. 그러나 pH를 $7.00{\pm}0.50$에서 $12.00{\pm}0.50$으로 증가시켰을 때 가용화 효율은 8.5%증가로 큰 차이가 없었다. 실험실규모 2상 혐기성 소화시스템은 가용화 조건에서 pH를 조절하지 않은 Run1 (pH $4.20{\pm}0.40$)과 pH를 $7.00{\pm}0.50$으로 조절한 Run2로 나누어 운전되었다. pH $7.00{\pm}0.50$의 가용화에 의해 Run2시스템에서 전반적으로 높은 SCOD 및 TVFA농도가 측정되었다. 산생성조에서 TVFA농도는 18.4 g/L로 Run1보다 1.8배 높은 결과를 나타내었다. 그 결과 메탄생성조에서 메탄가스 발생량은 0.333 L/gVS로 Run1의 0.282 L/gVS과 비교하여 18% 향상되었다.
The effects of dry roasting whole faba beans (WFB) and whole lupin seeds (WLS) at 110, 130 or $150{^{\circ}C}$ for 15, 30 or 45 min on rumen (RDCP%), estimated intestinal (IDCP%) and total tract disappearance of CP (TDCP%) and intestinal availability (IARUCP%) of rumen undegraded CP (RUCP%) were determined. The RDCP values were estimated by in sacco technique by incubating nylon bags for 8, 12 and 24 h in the rumen of dairy cows. The IDCP and IARUCP values were estimated using a sequence of ruminal incubation, in vitro incubation in acid-pepsin for 1 h and then in pancreatin for 24 h of three-step in vitro procedure technique. Dry roasting at 130 and $150^{\circ}C$ decreased RDCP with correspondingly increasing IDCP. The IDCP value generally increased from 12.3(raw) to 8.6, 14.8 and 39.6% (WFB) and from 28.3 (raw) to 33.7, 36.2 and 56.2% (WLS) at 8 h rumen incubation; from 2.9 (raw) to 2.9, 4.6 and 23.3% (WFB) and from 19.6 (raw) to 19.0, 24.0 and 46.6% (WLS) at 12 h rumen incubation; from 1.3 (raw) to 1.9, 1.7 and 11.0% (WFB) and from 4.4 (raw) to 4.2, 10.7 and 36.7% (WLS) at 12 h rumen incubation as the temperatures rose to 110, 130 and $150{^{\circ}C}$ respectively. The TDCP values were always high and increased by time in the rumen, the average values of which were 97.9, 96.6; 99.2, 96.9 and 99.6, 98.7% for WFB and WLS, respectively, at 8, 12 and 24 h rumen incubation. But within the same retention time, TDCP was generally unchanged. The average IARUCP increased from 87.3 (raw) to 87.4, 88.7 and 92.0% (WFB); from 87.6 (raw) to 88.9, 91.5 and 93.0% (WLS) at roasting temperatures of 110, 130 and $150{^{\circ}C}$, respectively. It was concluded that dry roasting can shift the digestion of CP from rumen to the lower gastrointestinal tract without depressing the digestion of RUCP. The best processing condition in this study was dry roasting at $150{^{\circ}C}$ for 45 min in terms of effects on the disappearances and availability of CP. Research data on intestinal availability of individual amino acids need to be further investigated.
