Contents of Total Flavonoid and Biological Activities of Edible Plants (식물성 식품중 총플라보노이드 함량과 생리활성 탐색)

  • The contents of total flavonoids, total antioxidant status and superoxide scavenger activity of edible plants were measured. Among twenty one samples of leafy vegetables and teas, mugwort (Artemesia capillaris) and green teas gave high total flavonoids contents, 48.2 mg/g sample and 44.7 mg/g sample, respectively. The highest concentration of total flavonoids among eighty nine samples analyzed was 52.1 mg/g sample from propolis. When samples were tested for total antioxidant status, a kind of groundsel showed the highest Total Antioxidant Status(TAS) value, 13.4 mM among leafy vegetables and teas and green tea gave high TAS value, 11.84 mM. On the other hand, purslane gave the lowest TAS value, 0.01 mM. The superoxide scavenge activity was measured from the extracts of all edible plants. The superoxide scavenge activity of polygonatum(81.1%) and chrysantemum (78.5%) were the highest among leafy vegetables, teas and medicinalplants. On the other hand, ginseng, poria, cassiae semen and dioscoreae rhizoma showed no superoxide scavenge activities.

Polyphenol Contents and Antioxidant Activities of Fractions from Ssanghwa-tang and Fermented Ssanghwa-tang (쌍화탕과 발효쌍화탕 분획물의 폴리페놀함량 및 항산화 활성)

  • The aim of this study is to compare antioxidant activity and total polyphenol contents between before and after fermentation of Ssanghwa-tang according to solvent partition. Ssanghwa-tang was fermented with Lactobacilus fermentum. Ssanghwa-tang and the fermented Ssanghwa-tang were fractioned by solvent partition with ethyl acetate, butanol and water. The Ssanghwa-tangs and their solvent fractions were evaluated for total polyphenol contents and DPPH radical scavenging activity. The antioxidant activity as well as the total polyphenol contents were highest in each ethyl acetate fraction and significantly (p<0.05) increased after fermentation.

Polyphenolic Contents and Antioxidant Activities of Underutilized Grape (Vitis vinifera L.) Pomace Extracts

  • Grape pomace is an abundant source of underutilized winery by-products. Polyphenols were extracted from grape pomace using cellulase and gluco-amylase enzymes. 2,2-diphenyl-1-picrylhydrazyl (DPPH) and Folin-Ciocalteu's assays were used to measure antioxidant activity and total polyphenolic contents. Both cellulase, and gluco-amylase digested grape pomace showed efficient radical scavenging activity. In addition, the total polyphenolic contents of cellulase digested grape pomace showed lower concentrations were effective compared to higher concentrations, whereas glucoamylase enzyme did not show remarkable variations. The DPPH radical scavenging activity and total polyphenolic contents were significantly higher in the cellulase digested grape pomace compared to the gluco-amylase digested and the not digested grape pomace. It is notable that enzymatic digestions were efficient for extracting polyphenols from grape pomace. The underutilized grape pomace polyphenols can be further used for food safety as a natural antioxidant.

Antioxidant activity and anti-inflammatory activity of ethanol extract and fractions of Doenjang in LPS-stimulated RAW 264.7 macrophages

  • BACKGROUND/OBJECTIVES: Fermentation can increase functional compounds in fermented soybean products, thereby improving antioxidant and/or anti-inflammatory activities. We investigated the changes in the contents of phenolics and isoflavones, antioxidant activity and anti-inflammatory activity of Doenjang during fermentation and aging. MATERIALS/METHODS: Doenjang was made by inoculating Aspergillus oryzae and Bacillus licheniformis in soybeans, fermenting and aging for 1, 3, 6, 8, and 12 months (D1, D3, D6, D8, and D12). Doenjang was extracted using ethanol, and sequentially fractioned by hexane, dichloromethane (DM), ethylacetate (EA), n-butanol, and water. The contents of total phenolics, flavonoids and isoflavones, 2,2-diphenyl-1 picryl hydrazyl (DPPH) radical scavenging activity, and ferric reducing antioxidant power (FRAP) were measured. Anti-inflammatory effects in terms of nitric oxide (NO), prostaglandin (PG) E2 and pro-inflammatory cytokine production and inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expressions were also measured using LPS-treated RAW 264.7 macrophages. RESULTS: Total phenolic and flavonoid contents showed a gradual increase during fermentation and 6 months of aging and were sustained thereafter. DPPH radical scavenging activity and FRAP were increased by fermentation. FRAP was further increased by aging, but DPPH radical scavenging activity was not. Total isoflavone and glycoside contents decreased during fermentation and the aging process, while aglycone content and its proportion increased up to 3 or 6 months of aging and then showed a slow decrease. DM and EA fractions of Doenjang showed much higher total phenolic and flavonoid contents, and DPPH radical scavenging activity than the others. At $100{\mu}g/mL$, DM and EA fractions of D12 showed strongly suppressed NO production to 55.6% and 52.5% of control, respectively, and PGE2 production to 25.0% and 28.3% of control with inhibition of iNOS or COX-2 protein expression in macrophages. CONCLUSIONS: Twelve month-aged Doenjang has potent antioxidant and anti-inflammatory activities with high levels of phenolics and isoflavone aglycones, and can be used as a beneficial food for human health.

Antioxidant Activity of the Various Extracts from Different Parts of Lotus (Nelumbo nucifera Gaertner)

  • This study was conducted to investigate the antioxidant activity of the extracts of lotus (Nelumbo nucifera Gaertner). The total phenolic contents in leaf, stem, and root were 165, 74, and 30 tannic acid equivalent mg/g of dried extract or fraction respectively. The butanol and ethylacetate fractions of lotus parts showed higher 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity than other fractions. 2,2'-Azinobis(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation radical scavenging activity also showed the similar result as the DPPH radical scavenging activity. The antioxidative capacity of the ethylacetate fraction was the highest among fractions and its fraction showed higher contents of total polyphenol.

Analysis of the Phenolic Content and Antioxidant Activities of Soybean Extracts from Different Regions and Cultivars

  • Phenolic compounds such as isoflavones contribute to the antioxidant properties of soybean. This study investigated differences in the phenolic content and antioxidant activity of ten soybean cultivars grown in two different locations in Korea. We observed a wide range of variation in total isoflavone and phenolic content by location and cultivar. The total isoflavone content of cultivars grown at Pyeongchang (sub-highland) was significantly higher than that in cultivars grown at Gangneung (lowland). In contrast, the total phenolic content of cultivars grown at Gangneung was greater than that of cultivars grown at Pyeongchang. The radical scavenging activity of DPPH was similar to that of the total phenolic content rather than that of the total isoflavone content. These results suggest that antioxidant activity of soybean was associated with phenolic compounds rather than isoflavones. To identify the individual antioxidant components, we used an on-line HPLC-$ABTS^+$-based assay system, ESI/MS, and NMR. The results showed that the strongest antioxidant activity was linked to epicatechin.

Alteration in Phenolic Compounds and Antioxidant Activities of Aronia melanocarpa Ethanol Extracts following Fermentation Using Different Strains of Leuconostoc mesenteroides to Develop Natural Antibiotic Alternative (항생제 대체 천연물질을 위한 아로니아 주정 추출물 개발에 있어 다양한 Leuconostoc mesenteroides 균주를 이용한 발효가 페놀계 화합물 및 항산화활성 변화에 미치는 영향)

  • Antioxidant activity is important for reducing oxidative stress that causes various metabolic disorders. Metabolic disorders are highly related to loss of productivity in livestock. Therefore, development of effective antioxidant compounds originating from plants is important for organic agriculture. Phenolic compounds in edible plants are regarded as major components relevant to antioxidant activity. The present study investigated the changes in antioxidant activity and phenolic compound profiles of Aronia (Aronia meloncarpa) by fermentation using different strains of Leuconostoc mesenteroides. A total of 5 strains of L. mesenteroides were used as starter cultures and their ${\beta}$-glucosidase activities were measured. A total of 6 experiment runs were prepared, one for control (uninoculated) and the others (inoculated) for treatments. For biological activity, antioxidant and antibacterial activities were measured. For phenolic compound profiling, TLC and HPLC analysis were performed. The strains of KACC12313 and KACC12315 showed greater enzyme activity than others. Treatment with KCCM35046 showed strong and broad antibacterial activity against to Listeria monocytogenes. Treatments with KCCM35046 and KACC12315 showed the highest total polyphenol content. The highest antioxidant activity was found in KACC12315 treatment. No remarkable alteration was found in thin layer chromatography (TLC) analysis. In phenolic compound profiling analysis, KCCM35046 showed notable alteration in compound area ratio compared to others and also showed the highest caffeic acid content. In chlorogenic acid, treatments with KCCM35046 and KACC12315 showed great content than others. Treatment with KACC12315 showed the greatest content of trans-ferulic acid. As a result of relative performance indexing analysis, L. mesenteroides KCCM35046 and KACC12315 were selected as the best strain for the fermentation of Aronia.

Comparison between the Antioxidant Activity and the Index Content of ACTS002 according to the Extraction Solvent (추출용매에 따른 ACTS002의 항산화 활성 및 지표성분의 함량 비교)

  • Objectives: Samul-tang is commonly used to alleviate the side effects of chemotherapy. This study aimed to establish an efficient method of extracting ACTS002 based on Samul-tang using the yield, high-performance liquid chromatography (HPLC), and antioxidant assay. Methods: ACTS002 was extracted from each extraction solvent, and the contents of 5-hydroxymethyl-2-furaldehyde (5-HMF), paeoniflorin, and ferulic acid were quantitatively analyzed and compared using HPLC. Moreover, the antioxidant activities of ACTS002 were measured using total flavonoids, total phenolic compounds, 1,1-diphenyl-2-picrylhydrazyl (DPPH), 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS), and Ferric reducing/antioxidant power (FRAP). Results: All of the components were set as the index contents because they were easy to process. The antioxidant activity of total flavonoids was the highest in 70% ethyl alcohol extracts, and total phenolic compounds were the highest in 50% ethyl alcohol extracts. In DPPH, 50% ethyl alcohol extracts showed the highest activity, and in ABTS 70% ethyl alcohol extracts were the highest. In FRAP, 70% ethyl alcohol extracts showed the highest activity. Conclusions: ACTS002 can control quality by setting 5-HMF, paeoniflorin, and ferulic acid as the index contents. The antioxidant activity measurement was relatively high in the 50% and 70% ethyl alcohol extracts. Our results can predict the possibility of a pharmacological activity and the standardization of ACTS002.

Antioxidant and Free Radical Scavenging Potential of Justicia gendarussa Burm. Leaves in vitro.

  • Antioxidant activity of 70% aqueous ethanolic extract of leaves of Justicia gendarussa (EJ) was evaluated. EJ was prepared by cold maceration method. The antioxidant potency of EJ was investigated employing various established in vitro systems, such as DPPH radical scavenging, nitric oxide (NO) scavenging, ${\beta}-carotene$ linoleic acid module system (${\beta}$ CLAMS), hydroxyl (OH) radical scavenging, anti lipid peroxidation. $IC_{50}$ values were determined in each experiment. Also, ferric ion reduction capacity of extracts in presence and absence of chelating agent (EDTA) and total antioxidant capacity were determined. Preliminary phytochemical investigation was carried out to know the nature of constituents present in the leaves and correlate it with antioxidant activity. Further total phenolic content was determined in EJ. $IC_{50}$ values of EJ were 123.09 ${\pm}$ 3.01, 643.0 ${\pm}$ 61.10, 132.3 ${\pm}$ 6.03, 68.5 ${\pm}$ 11.5 and 68.13 ${\pm}$ 1.38 ${\mu}g/mL$ in DPPH radical scavenging, NO scavenging, ${\beta}$ CLAMS, OH radical scavenging and anti lipid peroxidation activity respectively. In total antioxidant capacity assay, ascorbic acid equivalent value was found to be 205.56 ${\pm}$ 4.69 ${\mu}g/mg$ of extract. Total phenolic content was found to be 43.76 ${\pm}$ 4.27 ${\mu}g$ equivalent of gallic acid per mg of extract. Phytochemical investigation reveals the presence of flavonoids. The results indicate that EJ possess antioxidant activity and flavonoids are responsible for this activity.

Comparison of Biological Activities of Dendropanax morbiferus by Different Cultivation Areas in Korea

  • Dendropanax morbifera H.Lev is an evergreen tree that lives in subtropical climates. About 75 species of D. morbifera live in East Asia, but only one species live in Korea(Korean dendropanax). D. morbifera belonging to Aralicacae family 98% of D. morbifera are distributed in Jeollanam-do in the Korea and the grow wild in Gyeongsangnam-do and Jeju Island. The harvest time and usage of D. morbifera were recorded in traditional medicinal books. The roots and stems of D. morbifera had been used for traditional medicine to treat migraine, menstrual irregularity and skin disease. And D. morbifera leaves are contain flavonoids and polyacetylene compounds. In this study, we were investigated the physiological activity of D. morbifera by different areas collected at the same time, and compared to characteristics of plants. D. morbifera collected from Jeollanam-do (Goheung-gun), Gyeongsangnam-do (Namhae-gun) and Jeju Island, and dried at $50^{\circ}C$ for three days. We used dried D. morbifera powder for antioxidant tests. Each sample was extracted with hot water under the same conditions. The contents of total polyphenols and total flavonoids from D. morbifera were identified. Also, we performed to DPPH radical scavenging activity, ABTS cation radical scavenging activity and Superoxide anion scavenging activity efficacy for antioxidant activity determination. The contents of total polyphenols and total flavonoids in hot water extract of D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do were higher than Jeju. However, D. morbifera harvested from Gyeongsangnam-do and Jeollanam-do showed no significant difference those content of total polyphenols and total flavonoids. And the antioxidant capacity was showed the similar patterns in antioxidant activity.
