Antioxidant activity of silkworm powder treated with protease

  • The antioxidant activity of silkworm powder treated by proteolytic enzyme was investigated. Total protein content of silkworm power was assayed using BCA, Bradford assays and SDS-polyacrylamide gel electrophoresis (PAGE) with alkaline protease treatment conditions including temperature and pH. The optimum condition of alkaline protease treatment for silkworm powder was found to be $60^{\circ}C$ and pH 7. The alkaline protease treatment resulted in increased contents of free amino acids, total polyphenol and total flavonoid compared to control group. The silkworm hydrolysates showed excellent antioxidant activities in various in vitro models such as 2,2 diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity, 2,2 - azino-bis(3-ethylbenzthiazoline-6)-sulfonic acid (ABTS) radical scavenging activity. These results provide useful information for using silkworm powder as an ingredient in functional foods and for exploiting alkaline protease treatment to improve the extractability and bioactivity of a raw material.

Antioxidant Activity of Salad Vegetables Grown in Korea

  • The antioxidant activity of forty two kinds of salad vegetables grown in Korea was evaluated. Methanol extract of freeze-dried vegetable was assayed by radical scavenging activity using 1,2-diphenyl-2-picrylhydrazyl (DPPH) and Fe^{2+}$-catalyzed lipid peroxidation inhibition by TBA method. Total phenolics were determined colorimetrically using Folin-Ciocalteu reagent. The highest radical scavenging activity was expressed by perilla leaf, followed by dandelion leaf, red and green leafy lettuce, of which $IC_{50}$ was less than 0.10 mg/mL. Angelica leaf showed the highest inhibitory action for lipid peroxidation with $95\%$, and then dandelion leaf, water spinach, and perilla leaf inhibited over $80\%$. However, lettuce (Iceberg) and young Chinese cabbage exhibited the lowest antioxidant activity based on both assay methods. Highly positive correlations between antioxidative activities and total phenolics were observed (p < 0.001). The results suggested that salad vegetables, especially perilla leaf, leafy lettuce, dandelion or angelica, could be used for easily accessible sources of natural antioxidants.

In vitro anti-oxidant activity of the leaves of Dillenia indica

  • The methanol extract of Dillenia indica was tested for antioxidant activity as determined by free radical scavenging of DPPH radical scavenging assay, reducing power, total antioxidant capacity measured by phosphomolybdenum method, total phenolic content and total flavonoids content determination assays. The extract showed significant activities in all antioxidant assays compared to the standard antioxidant in a dose dependent manner. In DPPH radical scavenging assay the $IC_{50}$ value of the extract was found to be 100.53${\mu}g/ml$ while ascorbic acid has the $IC_{50}$ value 58.92${\mu}g/ml$. Dillenia indica extract showed strong reducing power and total antioxidant capacity. Moreover, methanol extracts also possess high amount of phenolics and flavovonoids and expressed as gallic acid and rutin equivalent respectively. The remarkable activities exhibited in reactive oxygen species scavenging may attributed to the high amount of hydrophilic phenolics present in Dillenia indica.

Antioxidant Activities and Total Phenolics of Ethanol Extracts from Several Edible Mushrooms Produced in Korea

  • Eight edible mushrooms grown in Korea were extracted with ethanol at room temperature for 24 hr. The extracts were investigated for their antioxidant activities as measured by 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical cation scavenging activities. Among the mushroom extracts evaluated in this study, the ethanolic extracts from Ganoderma lucidum and Pleurotus eryngii showed the greatest potential antioxidant activity, by producing 85 and 88% inhibition in DPPH radical scavenging method and 219 and 165 mg ascorbic acid equivalent antioxidant capacity (AEAC), respectively. Total phenolics and total flavonoids in the ethanolic extracts were determined by spectrophotometric method. Positive correlations were found between total phenolic contents in the extracts and their antioxidant activities, suggesting that phenolic contents in the mushrooms extracts are mainly responsible for their antioxidant activities.

Screening of antioxidant and antimicrobial activities of Caesalpinia bonducella Flem., leaves (Caesalpiniaceae)

  • The study was aimed at evaluating the antioxidant and antimicrobial activities of methanol extract of Caesalpinia bonducella leaves (MECB) (Family: Caesalpiniaceae). The effect of MECB on antioxidant activity, reducing power, free radical scavenging (DPPH radical, nitric oxide radical, superoxide anion radical, hydroxyl radical and hydrogen peroxide radical scavenging), total phenolic content and antimicrobial activities were studied. The antioxidant activity of MECB increased in a dose dependent manner. About 50, 100, 250 and 500 g of MECB showed 53.4, 61.2, 69.1 and 76.2 % inhibition respectively on peroxidation of linoleic acid emulsion. Like antioxidant activity, the effect of MECB on reducing power increased in a dose dependent manner. The free radical scavenging activity of MECB was determined by DPPH radical scavenging method. The potency of this activity was increased with increased amount of extract. MECB was found to inhibit the nitric oxide radicals generated from sodium nitroprusside $(IC_{50}\;=\;102.8\;g/ml)$ whereas the $IC_{50}$ value of curcumin was 20.4 g/ml. Moreover, the MECB was found to scavenge the superoxide generated by photoreduction of Riboflavin. MECB was also found to inhibit the hydroxyl radical generated by Fenton reaction, where the $IC_{50}$ value is 104.17 g/ml compared with catechin 5 g/ml, which indicates the antioxidant activity of MECB. The MECB capable of scavenging hydrogen peroxide in a concentration-dependent manner. The amounts of total phenolic compounds were also determined. Antimicrobial activities of MECB were carried out using disc diffusion methods with five Gram positive, four Gram negative and four fungal species. The results obtained in the present study indicate that MECB leaves are potential source of natural antioxidant and antimicrobial agents.

Radical Scavenging Activity and Content of Cynarin (1,3-dicaffeoylquinic acid) in Artichoke (Cynara scolymus L.)

  • The contents of total phenol and total flavonoid of artichoke (Cynara scolymus L.) were measured. The antioxidant activity of the artichoke was evaluated based on its potential as a scavenging the ABTS radical. These results showed the antioxidant activity of artichoke has a close relationship with the total flavonoid content. The compound showing antioxidant activity was isolated from the artichoke by repeated column chromatography and recrystallization. Based on the spectrometric studies, the compound was identified as 1,3-dicaffeoylquinic acid, known as cynarin. The content of cynarin from heads and leafs of the artichoke determined by $C_{18}$ reversed phase HPLC (high-performance liquid chromatography) coupled with photodiode array detector was 10.15 and 0.67 mg/g, respectively. This compound showed potent antioxidant activities against DPPH and ABTS radicals ($EC_{50}$ = 14.09 and 28.85 ${\mu}M$, respectively).

Antioxidant Activities of Methanol Extracts from Root Parts of Korean Salad Plants

  • Phenolics level, total flavonoid content, and antioxidant were determined from the methanol extracts of the eight medicinal plants using roots. Total phenolics were found as the highest levels in the methanol extracts of Arctium lappa, and followed by Youngia sonchifolia and Cirsium japonicum. Total amount of the each phenol compounds were detected in C. japonicum extracts ($319.2\;mg\;kg^{-1}$) as the greatest component, and followed by A. lappa ($96.3\;mg\;kg^{-1}$) and Y. sonchifolia ($22.9\;mg\;kg^{-1}$). Total flavonoid content showed the highest amount in methanol extracts from A. lappa (68.1 mg $100\;g^{-1}$) and followed by Y. sonchifolia (11.2 mg $100\;g^{-1}$) Lycoris radiate extracts (87.2%) had the highest nitrite scavenging activity and followed by A. lappa (81.5%) and Y. sonchifolia (77.5%). Methanol extracts of A. lappa at 25 mg $100\;g^{-1}$ exhibited the highest DPPH radical scavenging activity by 90.8%, even though less activity than synthetic antioxidants Vitamin C or butylated hydroxytoluene (BHT). Level of polyphenols was highly correlated with anti oxidative activity ($r^2$ = 0.85). The results suggest that several medicinal plants selected had the potent biological activities, and that their activities were differential depending on plant species.

  • 헛개나무를 부위별 추출하여 total polyphenol과 flavonoid, Vitamin C equivalent antioxidant capacity를 연구한 결과 헛개나무의 수피와 열매에서 가장 많이 조사되었고 DPPH free radical scavenging activity에서도 수피와 열매에서 활성이 뛰어났다. 식물 추출물인 Polyphenol성 물질이 많이 존재 할 수록 항산화 활성은 뛰어났다.

In Vitro Antioxidant and Antiproliferative Activities of Novel Orange Peel Extract and It's Fractions on Leukemia HL-60 Cells

  • In the present work, novel orange peel was extracted with 100%EtOH (ethanol) and fractionated into four fractions namely F1, F2, F3, F4 which were eluted from paper chromatographs using 100%EtOH, 80%EtOH, 50%EtOH and pure water respectively. The crude extract and its four fractions were evaluated for their total polyphenol content (TPC), total flavonoid content (TFC) and radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl) assay. Their cytotoxic activity using WST assay and DNA damage by agarose gel electrophoresis were also evaluated in a human leukemia HL-60 cell line. The findings revealed that F4 had the highest TPC followed by crude extract, F2, F3 and F1. However, the crude extract had the highest TFC followed by F4, F3, F2, and F1. Depending on the values of $EC_{50}$ and trolox equivalent antioxidant capacity, F4 possessed the strongest antioxidant activity while F1 and F2 displayed weak antioxidant activity. Further, incubation HL-60 cells with extract/fractions for 24h caused an inhibition of cell viability in a concentration-dependent manner. F3 and F4 exhibited a high antiproliferative activity with a narrow range of $IC_{50}$ values ($45.9-48.9{\mu}g/ml$). Crude extract exhibited the weakest antiproliferative activity with an $IC_{50}$ value of $314.89{\mu}g/ml$. Analysis of DNA fragmentation displayed DNA degradation in the form of a smear-type pattern upon agarose gel after incubation of HL-60 cells with F3 and F4 for 6 h. Overall, F3 and F4 appear to be good sources of phytochemicals with antioxidant and potential anticancer activities.

수박(Citrullus vulgaris L.) 외피의 항산화 활성 탐색 (Screening of Antioxidant Activity from Exocarp of Watermelon (Citrullus vulgaris L.))

  • Watermelon (Citrullus vulgaris L.) is a summer fruit typical to help fatigue systemic absorption is getting better. The goal of this study is to screen antioxidant activity to ensure the possibility as a functional material for exocarp of watermelon. Watermelon was extracted with 70% methanol, 70% ethanol, chloroform:methanol (CM, 2:1, v/v). Total phenol contents were 12.01 mg/g, 8.89 mg/g, 3.53 mg/g in the 70% methanol, 70% ethanol, CM, in that order, respectively. Total flavonoid content, DPPH radical scavenging activity, ABTS radical scavenging activity, ferric reducing antioxidant power (FRAP), ${\beta}$-carotene bleaching assay were 70% methanol extract remarkably higher than the other extracts. And these results showed the same trend of total phenol content. From the above results shows that watermelon was effective on the antioxidative activity.
