Theoretical Effects of Altered Biological and Chemical Properties on Salinity Tolerance of Acacia seeds

  • Multiple regression equations (Rehman et al. 2000) have been developed to predict the salinity tolerance of Acacia seeds, expressed as the I$\sub$50/ (the concentration of NaCl required to reduce final germination to 50% of the control value in DW). Accurate predictions can be made using one or more chemical and biological seed parameters. In this study the theoretical effect of varying final germination percentage in distilled water, germination rate in distilled water. (Rate), Ca$\^$2+/ or K$\^$+/ contents and their ratios, as independent factors or related factors, on the predicted salinity tolerance (I$\sub$50/) of Acacia species was investigated. Simulation of the effects of changing final germination, rate, calcium and potassium suggest the possibility of practical application of these results to modify the salinity tolerance of seeds. The predicted I$\sub$50/ increased with increasing final germination. Similarly, the higher the rate of germination, the higher the predicted salt tolerance of Acacia species. The Ca$\^$2+/ content of seeds was found to be positively correlated with I$\sub$50/. Species with higher Ca$\^$2+/ contents had a higher I$\sub$50/. This suggests that I$\sub$50/ might be increased by increasing the Ca$\^$2+/ contents of seeds by pretreatment with calcium salts or by supplying these to the mother plants.

  • A great deal of attention has been paid to the quality of life in citizens due to the increased number of long term life expectancy. reported as effective in the health promotion of the senior citizens. In this study, the principal investigator developed an exercise program for the senior citizens, which could be educated and managed by nursing staff. This kind approach may support the need of exercise program on regular base via the social organization, which may mean the intentional change of life style. The exercise program consisted of exercise, health education, and a direct the population of the senior A regularexercise has been nursing care. A quasi -experimental study was conducted to investigate the effects of excercise program on health of the elderly in senior citizen's center. Independent variable was a 9 weeks exercise program. Dependent variables were: 1) physical health [e.g. physical fitness(cardiopulmonary endurance, back muscle strength, grip strength, muscular tolerance, flexibility and body fat proportion), physiologic parameters(blood pressure, pulse, respiration. and blood glucose), and perceived physical health status; 2) mental health measured by depression score; and 3) cognitive perception of exercise measured by usefulness and self-efficacy. A total of 37 subjects was randomly assigned into either the control group (without the exercise program: n=18) or the intervention group(with the exercise program: n=19). The results of the study analyzed using a SAS, were as follows: 1) In physical fitness of physical health, there was a significant improvement in cardiopulmonary tolerance. back muscle strength. muscular tolerance and flexibility in the intervention group, compared to the control group, while no difference in grip strength and body fat proportion. The exercise program resulted in decreases in blood pressure, pulse, respiration and blood glucose within normal range and improvement of perceived physical health status in the intervention group. 2) There was a slight increase of mental health(depression score) in the intervention group compared to the control group. but without statistical significance. 3) There was a significant improvement in cognitive perception of exercise(e.g. usefulness and self -efficacy) in the intervention group, compared to the control group. It was concluded that the exercise program employed in this study was appropriate for women senior citizens and had a positive effect on health in general.

  • 전역 클럭 없이 외부 입력에 따라서 값이 변하는 비동기 카운터는 우주용 메모리 등 현대 디지털 시스템에서 널리 사용된다. 본 논문에서는 우주 방사능 누적에 기인하는 크리티컬 레이스 고장이 존재하는 비동기 카운터를 위한 고장 극복 기법을 제안한다. 크리티컬 레이스는 비동기 디지털 회로 설계 과정에서 발생하는 대표적인 고장으로서 회로의 다음 안정 상태가 고정되지 않고 여러 값 중 하나로 나오는 비결정적인 특성을 보인다. 이번 연구에서는 비동기 순차 머신에 대한 교정 제어 기법을 이용하여 크리티컬 레이스를 극복할 수 있는 상태 피드백 제어기의 설계 과정을 제시한다. 또한 비동기 카운터 교정 제어 시스템을 VHDL 코드로 구현하고 실험을 통하여 제안된 제어 시스템이 크리티컬 레이스 고장을 극복하는 과정을 예시한다.

고지방식이로 유발된 비만 백서에서 가미감비제습탕이 비만 유발에 미치는 영향 및 기전 연구 (Anti-obesity Effects and Mechanism of Original and Modified Gambejaeseup-tang in Female Rats with Diet-induced Obesity)

  • Gambejaeseup-tang (GBJST) have recently been used as an anti-obesity herbal medicine but their effect and mechanism of action have not been studied. We modified ingredients of GBJST based on the previous experiments about exploring herbs to suppress triglyceride accumulation in 3T3-L1 adipocytes. We investigated the effects of modified GBJST on energy, glucose and lipid homeostasis using female rats with diet-induced obesity and their action mechanism was also determined. Rats fed a high-fat diet (HFD) were divided into 3 groups: rats in each group received 0.2 or 2 g water extracts of modified GBJST (L-GBJST or H-GBJST) or 2 g cellulose per kg body weight (a negative control) on a daily basis. A further group was fed a low-fat diet (LFD) as a positive control. We found that modified GBJST dose-dependently decreased body weight and mesenteric and retroperitoneal fat more than the control. This decrease was due to the reduction in energy intake and the increase of energy expenditure. HFD increased fat oxidation more than LFD and modified GBJST further increased fat oxidation as a major energy source more than the control in a dose-dependent manner. In addition, H-GBJST improved glucose tolerance without changing serum insulin levels during an oral glucose tolerance test. H-GBJST also suppressed the increase of serum total and LDL cholesterol and triglyceride levels by HFD. In conclusion, modified GBJST have a good anti-obesity effect by decreasing energy intake and increasing energy expenditure mainly as fat in female rats with diet-induced obesity. It also improves glucose tolerance and lipid metabolism.

내당능 장애 모델 흰쥐에서 Vitamin E의 과잉섭취가 지질 분포와 혈청 Insulin 농도변화에 미치는 영향 (Effect of Vitamin E Supplementation on Lipid Distribution and Serum Insulin in Experimental Model Rats with Impaired Glucose Tolerance)

  • The aim of the study was to determine whether vitamin E supplementation in three experimental model rats with impaired glucse tolerance could change serum insulin concentration and lipid distribution. The three groups were adult(AS) and neonatal (NS) streptozotocin-induced groups, and a high sucrose diet(HS) group. Each group was divided into control and vitamin E supplementatino groups at the age of 9 weeks. The level of vitamin E supplementation was 5g/kg diet. Blood and organ samples were taken at 5 and 10 weeks and were examined for changes in the level of serum insulin, glucose, lipids, liver lipids, and oxidative status. Vitamin E supplementation significantly reduced serum insulin in the HS group and caused the significant beneficial changes in serum lipids and triglycerides in As grouop at 10 weeks . In all groups, serum vitamin E was increased and malondialdehyde(MDA) in serumand liver were decreased significantly by vitamin E supplementation. The results suggest that vitamin E supplementation improves lipid distribution in adult streptozotocin-induced rats and serum insulin concentration in high sucrose diet-induced rats. Vitamin E might prevent on reduce oxidative injury in all experimental model rats with impaired glucose tolerance.

Expression of Cholera Toxin B Subunit and Assembly as Functional Oligomers in Silkworm

  • The nontoxic B subunit of cholera toxin (CTB) can significantly increase the ability of proteins to induce immunological tolerance after oral administration, when it was conjugated to various proteins. Recombinant CTB offers great potential for treatment of autoimmune disease. Here we firstly investigated the feasibility of silkworm baculovirus expression vector system for the cost-effective production of CTB under the control of a strong polyhedrin promoter. Higher expression was achieved via introducing the partial non-coding and coding sequences (ATAAAT and ATGCCGAAT) of polyhedrin to the 5' end of the native CTB gene, with the maximal accumulation being approximately 54.4 mg/L of hemolymph. The silkworm bioreactor produced this protein vaccine as the glycoslated pentameric form, which retained the GM1-ganglioside binding affinity and the native antigenicity of CTB. Further studies revealed that mixing with silkworm-derived CTB increases the tolerogenic potential of insulin. In the nonconjugated form, an insulin : CTB ratio of 100 : 1 was optimal for the prominent reduction in pancreatic islet inflammation. The data presented here demonstrate that the silkworm bioreactor is an ideal production and delivery system for an oral protein vaccine designed to develop immunological tolerance against autoimmune diabetes and CTB functions as an effective mucosal adjuvant for oral tolerance induction.

Expressing the Tyrosine Phosphatase (CaTPP1) Gene from Capsicum annuum in Tobacco Enhances Cold and Drought Tolerances

  • As one way to approach to cold defense mechanism in plants, we previously identified the gene for protein-tyrosine phosphatase (CaTPP1) from hot pepper (Capsicum annuum) using cDNA microarray analysis coupled with Northern blot analysis. We showed that the CaTPP1 gene was strongly induced by cold, drought, salt and ABA stresses. The CaTPP1 gene was engineered under control of CaMV 35S promoter for constitutive expression in transgenic tobacco plants by Agrobacterium-mediated transformation. The resulting CaTPP1 transgenic tobacco plants showed significantly increased cold stress resistance. It also appeared that some of the transgenic tobacco plants showed increased drought tolerance. The CaTPP1 transgenic plants showed no visible phenotypic alteration compared to wild type plants. These results showed the involvement of protein tyrosine phosphatase in tolerance of abiotic stresses including cold and drought stress.

Hsp20, a Small Heat Shock Protein of Deinococcus radiodurans, Confers Tolerance to Hydrogen Peroxide in Escherichia coli

  • The present study shows that DR1114 (Hsp20), a small heat shock protein of the radiation-resistant bacterium Deinococcus radiodurans, enhances tolerance to hydrogen peroxide ($H_2O_2$) stress when expressed in Escherichia coli. A protein profile comparison showed that E. coli cells overexpressing D. radiodurans Hsp20 (EC-pHsp20) activated the redox state proteins, thus maintaining redox homeostasis. The cells also showed increased expression of pseudouridine (psi) synthases, which are important to the stability and proper functioning of structural RNA molecules. We found that the D. radiodurans mutant strain, which lacks a psi synthase (DR0896), was more sensitive to $H_2O_2$ stress than wild type. These suggest that an increased expression of proteins involved in the control of redox state homeostasis along with more stable ribosomal function may explain the improved tolerance of EC-pHsp20 to $H_2O_2$ stress.

Comparative Performance of Three Tropical Turfgrasses Digitaria longiflora, Axonopus compressus and St. Augustinegrass under Simulated Shade Conditions

  • Shade affects turf quality by reducing light for photosynthesis. The shade tolerance of the tropical grasses, Digitaria longiflora and Axonopus compressus were evaluated against Stenotaphrum secundatum (St. Augustinegrass). The grasses were established under shade structures that provide 0%, 50%, 75% or 90% shade level for 30 days. A suite of leaf traits, recorded from similar leaf developmental stage, displayed distinct responses to shade conditions. Leaf length, relative to control, increased in all three species as shade level increased. The mean leaf extension rate was lowest in St. Augustinegrass (80.42%) followed by A. compressus (84.62%) and D. longiflora (90.78%). The higher leaf extension rate in D. longiflora implied its poor shade tolerance. Specific leaf area (SLA) increased in all species with highest mean SLA increase in D. longiflora ($348.55cm^2mg^{-1}$)followed by A. compressus ($286.88cm^2mg^{-1}$) and St. Augustinegrass ($276.28cm^2mg^{-1}$). The highest SLA increase in D. longiflora suggested its lowest performance under shade. The percent green cover, as estimated by digital image analysis, was lowest in D. longiflora (53%) under 90% shade level compared to both species. The relative shade tolerance of the three turfgrasses could be ranked as St. Augustinegrass > A. compressus > D. longiflora.

Expression of Catalase (CAT) and Ascorbate Peroxidase (APX) in MuSI Transgenic Tobacco under Cadmium Stress

  • The MuSI is known as a multiple stress resistant gene with several lines. A previous study using RT-PCR showed that the expression of MuSI gene in tobacco plant induced its tolerance to Cd stress. This study was conducted to examine the enhanced Cd tolerance of the MuSI transgenic tobacco plant through germination test and to understand the role of the involved antioxidant enzymes for the exhibited tolerance. Germination rate of MuSI transgenic tobacco was more than 10% higher than that of wild-type tobacco, and seedlings of MuSI transgenic tobacco grew up to 1.6 times larger and greener than seedlings of wild-type tobacco at 200 and 300 ${\mu}M$ Cd. From the third to the fifth day, CAT activities at 100 and 200 ${\mu}M$ Cd and APX activities at 100, 200 and 300 ${\mu}M$ Cd of MuSI transgenic tobacco were up to two times higher than those of wild-type tobacco. MuSI gene is shown to enhance the activities of antioxidant enzymes resulting in higher tolerance to oxidative stress compared with the control plant.
