담배 인산수송자 유전자를 이용한 벼의 형질전환 (Transformation of Rice (Oryza sativa L.) with Phosphate Transporter cDNA from Tobacco (Nicotiana tabacum L.))

  • 저인산 조건하에서 인산 흡수 효율이 높은 벼 품종을 개발하고자 담배의 인산수송자 유전자를 벼에 형질전환하였다. 동진벼로부터 유도된 callus를 담배의 인산수송자 유전자가 도입된 A. tumefaciens LBA 4404와 공동배 양한 후, 선발배지에서 증식된 callus를 250 mg/L carbenicillin, 2 mg/L kinetin, 0.1 mg/L NAA가 첨가된 MS배지에 옮겨 약 2주 후부터 소식물체를 얻었다. Carbenicillin 250 mg/L 첨가된 MS 기본배지에서 소식물체의 발근을 유도하여 재분화 식물체를 얻었다. 선발된 callus는 약 52%의 식물체 재력화율을 보였다. 선발된 재분화 식물체에 대한 PCR 분석을 수행하여 형질전환 벼 식물체에 담배의 인산수송자 유전자가 삽입되었음을 확인하였다. 형질전환체의 genonlic DNA로부터 PCR에 의해 증폭된 DNA에 대한 Southern blot분석 결과, 대조 식물체에서는 band가 검출되지 않았으나 형질전환 식물체에서는 인산수송자 유전자와 동일한 1.7kb의 band가 검출되어 외래유전자인 담배 인산수송자 유전자가 안전적으로 도입되었음이 확인되었다.

Attempting Tobacco Cessation - An Oral Physician's Perspective

  • Introduction: Tobacco use is a global health care problem. Repetitive exposure to nicotine produces neuroadaptation resulting in nicotine dependence. Smoking is associated with a range of diseases, causing high levels of morbidity and mortality and is one of the leading causes of preventable deaths, with more than 4.6 million smokers worldwide dying each year from smoking related illnesses. Stopping smoking has major health benefits. Quitting at any age provides both short and long term benefits. Materials and methods: 45 patients attending the outpatient department at the Oxford Dental College, Bangalore, were randomly allocated to three groups of interventions namely placebo, counseling and nicotine replacement therapy (NRT). Initially each one was assessed for carbon monoxide levels using a breath analyser (pico smokerlyser bedfont UK). They were followed up for six months and the carbon monoxide levels were again assessed using the same instrument. The paired t test was used to compare the results before and after the intervention. Results: The scores before the initiation of intervention and after treatment were compared and all three interventions were found to be statistically significant after six months. It was noticed that patients with very low or low dependence followed by high dependence had good response in the placebo group (68% and 47.6% respectively), in the counseling group maximum response was seen in the medium followed by the very low group (61% and 59% respectively), and maximum response was seen in very high followed by the very low group with NRT (78.7% and 60.5% respectively). Conclusion: The inference that can be drawn from the present study is that non-invasive, non pharmacological methods like placebo and counseling are effective in low to medium groups, and NRT is effective with higher nicotine dependence.

  • 인삼품질에 대한 전통개념을 역사적 경험, 전통적 품질관리방법 및 분석적 방법과 관련하여 조사하였다. 전통개념은 생명과 우주가 하나라는 한국의 기본사상에 근거한 것으로 보인다. 전통품질관리법에서 모양과 크기는 특정한 조직, 세포 및 세포내의 생리적 활성물질을 규정하기 때문에 전통품질관리법은 분석방법이 활성물질을 찾아가는 지도가 될 것이다. 전통법은 생물활성 물질간의 균형이 한 물질의 함량보다 중요하며 인체의 항샹성을 위한 활성물질의 작용양식도 여러 화합물, 여러 목표체계인 것으로 보인다. 인삼이 클 수록 중심부가 더 빨리 크고 중심부에는 질소화합물이 많기 때문에 전통법은 질소 화합물 특히, 수용성 단백질과 열안정단백질들이 활성물질의 균형에 있어 중요함을 강력히 시사한다. 생물활성물질의 균형은 삼속의 종간 사용방법의 차이와 의미있게 관계를 보인다.

Discrimination of biological and artificial nicotine in e-liquid

  • As the use of e-liquid cigarettes is rapidly increasing worldwide, it multiplies the potential risk undisclosed to the health of non- and smokers. To reduce the hazard, each country has its own set of regulations for controlling e-liquids. In Korea, the narrow definition of tobacco makes it difficult and have been steadily occurring tax evasion exploiting the difference in natural and artificial nicotine. Therefore, it is very important to distinguish source of nicotine for their regulation. To find biochemical discriminant markers, this study established analysis methods based on high-performance liquid chromatography coupled with diode array detector (HPLC-DAD) and high-performance liquid chromatography coupled with triple Quadrupole mass spectrometry (HPLC-MS/MS) for nicotine enantiomers and tobacco alkaloids targeted using the difference in pathways of nicotine biosynthesis and chemical synthesis. The method was validated by experimenting linearity (R2 > 0.999), recovery (80.99-108.41 %), accuracy (94.11-109.73 %) and precision (0.04-8.27 %). Then, the results for discrimination of the nicotine obtained from analysis of 65 commercial e-liquid products available in Korean market was evaluated. The method successfully applied to the e-liquids and one sample labelled 'synthetic nicotine' for tax exemption was found to contain a natural nicotine product. This method can be used to determine whether an e-liquid product uses natural or artificial nicotine and monitor non-taxable e-liquid products. The method is more scientific than the existing one, which relies only on field evidence.

Prolonged Opioid Use Among Opioid-Naive Women Undergoing Breast Reconstructive Surgery

  • Background Patients that undergo mastectomy for breast cancer with reconstruction may be prone to prolonged opioid use. As risk factors are not well-established, this article sought to better understand the risk factors that may be associated with this. Methods Patients that underwent breast reconstruction between 2010 and 2018 were identified in PearlDiver, a national insurance claims database. Patient demographics and comorbidities were elucidated, and various complications were then identified. Descriptive statistics as well as a multivariate analysis was used to evaluate the association of risk factors and complications. Results Breast reconstruction patients of 24,765 were identified from this database. Obesity, tobacco use, benzodiazepine use, and anticonvulsant use were all associated with prolonged opioid prescriptions greater than 90 days after both alloplastic and autologous reconstruction. Conclusion Prolonged opioid use continues to remain a topic of concern, and particularly in cancer patients that undergo breast reconstruction. Providers should be aware of potential risk factors for this to reduce this chance following breast reconstruction surgery.

대학생 중복흡연이 우울에 미치는 영향 (The Effects of College Students' Dual Use of Electronic Cigarette and Conventional Cigarette on Depression)

  • Purpose: This study was conducted to find out the association between dual-use of the electronic cigarette (EC) and conventional cigarette (CC) and depression among undergraduate students. Methods: The participants of this study were 225 undergraduate students who were attending two universities located in Ulsan metropolitan city. Descriptive statistics and multiple regression were conducted to analyze the data. Results: Nicotine dependence and depression were significantly higher in the dual-use group than in the single-use group. Dual-use of the EC and CC was significantly associated with depression among undergraduate students. In addition, quality of sleep was significantly associated with depression among undergraduate students. Conclusion: In order to prevent depression among undergraduate students, it is necessary to improve the awareness of the danger of dual use of the EC and CC.

  • Until relatively recently plant scientists have made little use of immunological techniques. Now, however, more and more researchers are discovering the powder of these techniques for the screening of immunomodulators and for the detection, quantitative determination and localization of compounds in plant materials. Especially, the recent developments in the fields of plant biotechnology and plant genetic engineering make it even more important for forkers in the plant sciences to become acquainted with the more sophisticated methods. The possible methods include onestep purification of antigens, visualization in situ by immunocytochemis try and on polyacrylam ids gel s by ni trocellulose Western blotting and quantification by various immunoassay. Among them, in this reviews, the quantitative determination methods are to be reviewed. There are several kinds of methods for the quantitative determination of plant constituents such as colorimetry, TLC, GLC, DCC, UV derivatization, densitometry and HPLC. When the complexity of plant constituents is considered. densitometry and HPLC have many advantages in sensitivity and separation ability. After a 11 some advarltages of two methods meritiorled above, all of these methods have many disadvantages and inconveniences. Previous purification for the application of all these methods make them less sensitive and more tedious. Immunoassay can solve these problems in part. But immunoassay also has some limitations. Specificity of immunoassay, contrary, can be considered to be disadvantages. Including this the advantages and disadvantages of immunoassay are to be discussed.

단백질 분해효소억제제를 이용한 담배의 품질평가 (Evaluation of Cigarette Quality by Use of ??1-Protease Inhibitor)

  • Current studies indicated that emphysema in smokers might be due, in part, to the local suppression of G, -protease inhibitor(u, -Pl) in lung by reactive oxygen species in cigarette smoke or smoke-activated lung neutrophiles. In the present works, we examined the possibility that a measure which inactivated $\alpha$l-Pl by cigarette smoke could be an alternative method to evaluate the cigarette quality, In order to determine the inactivation of $\alpha$1, -Pl, trypsin inhibitory capacity(TIC) was assayed. A rapid loss of $\alpha$1, -Pl activity occurred when $\alpha$1-Pl solutions was exposed the gas phase or total particulate matter(TPM) obtained from various brands. The inactivation of $\alpha$1-Pl by gas phase was dependent upon the number of puffs and the age of the smoke. However, that by TPM was rather decreased since 2 puffs and also showed no more change over 24hrs after exposing. Inactivation of $\alpha$1-Pl determined by our suggested method(5 puffs, 24hours of aging after exposing) using various commercial cigarettes exhibited that high tar brands has inactivated it more strongly than low tar cigarettes. But the ability of some brands to inactivate $\alpha$1-Pl does not correlate with the content of tar or nicotine. These results so여esc that the degree of $\alpha$1-Pl inactivation by cigarette smoke may be a useful index for the evaluation of cigarette quality and that it should be also contribute to the manufacture of less hazardous cigarettes.

  • Up to the 60's cigarette paper commercial grades were simply described as ' non combustible ', ' combustible ' or ' extra-combustible '. It was assumed that combustibility of the paper itself was correlated with cigarette combustibility. Since then, the characteristics of cigarette paper, like natural porosity and burning additives, which influence truly cigarette combustibility, have been described by various authors and it became clear to cigarette designers that paper combustibility was not generally a significant factor governing cigarette combustibility. On the other hand, for Roll Your Own(RYO)wrapping papers, the ECPCI has proposed to the CORESTA RYO Task Force to use paper combustibility, and specially the LCT test, to classify papers between low and high smoke delivery, and a new Task Force on PAPER COMBUSTIBILITY was recently set up to develop a recommended method. The objective of this presentation Is to show results demonstrating that for paper used on cigarettes, there is NO GENERAL CORTRELATION between PAPER and CIGARETTE combustibility. We will also show results confirming that, for the specific group of RYO booklet papers containing no burning additives, there is a correlation between paper combustibility and FCSA's Combustibility/smoke delivery. This apparent discrepancy will be explained by reviewing the role played by the various characteristics of cigarette paper in governing cigarette combustibility.

인삼근 신아의 암하생육에 미치는 영향 (Effect of Temperature on Growth of new Shoot in Panax ginseng under Dark)

  • New shoot growth of Panax ginseng root was investigated comparing with burley and soybean from l0$^{\circ}C$ to 30$^{\circ}C$ under dark. Shoot growth ceased by 12days at 30$^{\circ}C$ and optimum temperature appeared to be 15$^{\circ}C$/20$^{\circ}C$ (15hrs/9 hrs) , and 15$^{\circ}C$/15$^{\circ}C$ for ginseng. Shoot growth seems to be Poor below l0$^{\circ}C$. Temperature for maximum growth 20$^{\circ}C$/20$^{\circ}C$ for barley and 20$^{\circ}C$ /25$^{\circ}C$ for soybean. Barley did not germinate above 25$^{\circ}C$/25$^{\circ}C$, but grow better than soybean below 15$^{\circ}C$/25$^{\circ}C$. Fresh weight of 2 weeks suggesting cessation of water uptake at higher temporal use. Ginseng showed greater root ply s shoot of ginseng was linearly increased at 15$^{\circ}C$ but did not increased at 25$^{\circ}C$ after occurence of die-back of new shoot or root rot above 25$^{\circ}C$.
